| 1 | Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders. | Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao | 2020 | Cell Research2020,30,9: | 7 |
| 2 | Study on the relative radiometric correction of CBERS satellite CCD image显示文摘The main payload on CBERS-01/02 of China-Brazil Earth Resources Satellite (CBERS) is a push-broom CCD camera with moderate spatial and radiant resolution. Because at lab the data for calibration at satellite assembly stage were unable to be collected, and also because the onboard calibrator after launch was in a different state from imaging, the calibration of CCD image got a series of difficulties involved. In practice, two methods are used in the processing on the ground station: One is extracting calibration data by statistics from the image itself, and the other is the method of histogram match. It was proved that the latter can calibrate the image much better, because it can remove the effect of unstable response of the camera largely and also can overcome the nonlinearity of the camera basically by using Look-Up Table (LUT) calculated from histogram statistics of different temporal images. Considering the problems of CBERS-01, a lot of calibration tests were done before the launch of CBERS-02, in which a set of lab coefficients for relative calibration was formulated after the data collection by using integration-hemisphere in the stage of satellite assembly test. During the on-orbit test, it was found that the calibration result from such coefficients was not satisfying, especially there being response difference between 3 detector arrays, which was attributed to the unstable dark currents of the CCD camera. This paper comes up with a statistic method to remove such response difference. | 3 | Who is the“murderer”of the bloom in coastal waters of Fujian,China,in 2019?显示文摘On May 24-29,2019,a bloom occurring in Pingtan coastal areas of Fujian Province caused mass mortality of cage-cultured fish(Plectorhinchus cinctus and Pagrosomus major).During the bloom,two major causative organisms were present:Prorocentrum donghaiense(at a concentration of 1.46×10^7 cells/L)and an unknown naked dinoflagellate(4.58×10^6 cells/L).The naked dinoflagellate was isolated and cultured in this study,and its morphological features were examined using light microscopy and scanning electron microscopy.The large subunit(LSU)of the rRNA gene and the internal transcribed spacer(ITS)region of the naked dinoflagellate were also sequenced for field bloom samples and lab culture strains(PT-A and PT-B).On the basis of its morphological characteristics and molecular sequences,the unknown naked dinoflagellate was identified as Karlodinium digitatum.According to the phylogenetic analysis,the Karl.digitatum was most closely related to Karlodinium australe and Karlodinium armiger,and the three species clustered into a single clade of Karlodinium with bootstrap/posterior probability values of 95%/0.99 and 86%/0.99 inferred from LSU and ITS sequences,respectively.Karl.digitatum was first reported as Karenia digitata,a new harmful algal species bloomed in Hong Kong,China,in 1998.In present study,we gave a detailed morphological and phylogenetic description of Karl.digitatum and submitted the molecular sequences of this species to GenBank for the first time. | CEN Jingyi WANG Jianyan HUANG Lifen DING Guangmao QI Yuzao CAO Rongbo CUI Lei Lü Songhui | 2020 | Journal of Oceanology and Limnology2020,38,3: | 5 |
| 4 | Evaluation of miR-122-regulated suicide gene therapy for hepatocellular carcinoma in an orthotopic mouse model显示文摘Objective:Intratumoral administration of adenoviral vector encoding herpes simplex virus(HSV)thymidine kinase(TK)gene(Ad-TK)followed by systemic ganciclovir(GCV)is an effective approach in treating experimental hepatocellular carcinoma(HCC).However,hepatotoxicity due to unwanted vector spread and suicide gene expression limited the application of this therapy.miR-122 is an abundant,liver-specific microRNA whose expression is decreased in human primary HCC and HCC-derived cell lines.These different expression profiles provide an opportunity to induce tumor-specific gene expression by miR-122 regulation.Methods:By inserting miR-122 target sequences(miR-122T)in the 3'untranslated region(UTR)of TK gene,we constructed adenovirus(Ad)vectors expressing miR-122-regulated TK(Ad-TK-122T)and report genes.After intratumoral administration of Ad vectors into an orthotopic miR-122-deficient HCC mouse model,we observed the miR-122-regulated transgene expression and assessed the antitumor activity and safety of Ad-TK-122T.Results:Insertion of miR-122T specifically down-regulated transgene expression in vitro and selectively protected the miR-122-positive cells from killing by TK/GCV treatment.Insertion of miR-122T led to significant reduction of tansgene expression in the liver without inhibition of its expression in tumors in vivo,resulting in an 11-fold improvement of tumor-specific transgene expression.Intratumoral injection of Ad vectors mediated TK/GCV system led to a vector dosage-dependent regression of tumor.The insertion of miR-122T does not influence the antitumor effects of suicide gene therapy.Whereas mice administrated with Ad-TK showed severe lethal hepatotoxicity at the effective therapeutic dose,no liver damage was found in Ad-TK-122T group.Conclusions:miR-122-regulated TK expression achieved effective anti-tumor effects and increased the safety of intratumoral delivery of adenovirus-mediated TK/GCV gene therapy for miR-122-deficient HCC. | Gang Wang Xiaoyan Dong Wenhong Tian Yue Lu Jianyan Hu Yunfan Liu Jie Yuchi Xiaobing Wu | 2013 | Chinese Journal of Cancer Research2013,25,6: | 5 |
| 5 | Species-specific detection and quantification of scyphomedusae in Jiaozhou Bay,China,using a quantitative real-time PCR assay显示文摘Over the past decades,jellyfish occurred increasingly and abundantly in coastal areas worldwide.Usually,biomass of jellyfish,especially when they bloom,can be determined by visual counting.However,tiny individuals of jellyfish(e.g.,planulae,polyps,and ephyrae)are difficult to detect in the field.In this study,species-specific quantitative real-time PCR assays(qPCR)(SYBR Green I)targeting the mitochondrial 16S rDNA(mt-16S rDNA)of jellyfish were developed and were used to estimate the distribution and seasonal fluctuations of four jellyfish species(Nemopilema nomurai,Cyanea nozakii,Rhopilema esculentum,and Aurelia coerulea)in Jiaozhou Bay(JZB),China in 2013.The mt-16S rDNA of A.coerulea and N.nomurai was detected in most of the survey months and it peaked in July(1.03×10^(4) copies/L)and September(1.08×10^(6) copies/L),respectively.The mt-16S rDNA of C.nozakii occurred from August to October only with monthly mean values of 7.18-46.17 copies/L and was mainly located from the middle part to the outer part of the bay.The mt-16S rDNA of R.esculentum was the least abundant among the four species and was detected in only one sample(B2 station in March),with a value of 88.49 copies/L.The Spearman correlation test revealed that phytoplankton biomass was significantly and positively correlated with the mt-16S rDNA abundance of A.coerulea(R=0.37,P<0.01)and negatively with the mt-16S rDNA of N.nomurai(R=-0.36,P<0.01).The qPCR assay will enable the identification and quantification of jellyfish species in their whole life history and can be used as an approach in combination of the traditional jellyfish survey. | Jianyan WANG Tiezhu MI Zhigang YU Guoshan WANG Qinsheng WEI Jing YANG Yu ZHEN | 2021 | Journal of Oceanology and Limnology2021,39,4: | 3 |

(Gymnodiniales, Dinophyceae), a novel species isolated from the East China Sea in a dinofl agellate bloom显示文摘Abstract On May 24–29,2019,a harmful algal bloom occurred in Pingtan coastal areas,Fujian,southeast China,and caused mass mortality of cage-cultured fi sh.Two clonal cultures of an unknown naked dinofl agellate were set up from seawater samples taken during the bloom.The cultures were examined for morphological features,ultrastructure characters,photosynthetic pigments,the large subunit(LSU)of the rRNA gene,and the internal transcribed spacer(ITS)sequences,as well as acute toxicity analysis.The cell was unarmored,small-sized,and ovoid,and was characterized by elaborate striations on the epicone and hypocone.The nucleus was large,ellipsoid to oval or kidney-shaped,and centrally located in the cell.A long linear apical groove originated above the sulcus in the ventral epicone and extended to the dorsal side.An elongate,slit-like“ventral pore”was located on the left of the epicone,well away from the apical groove.The chloroplasts were yellowish brown,numerous,band like,and irregularly distributed in the cell periphery.Fucoxanthin was the main accessory pigment composition.Phylogeny topology reconstructed on partial LSU rDNA showed that the unknown dinofl agellate branched as a sister species to Karlodinium sp.(strain IFR981&IFR797,from France)and Karlodinium corrugatum(strain KDGSO08,from Australia),with genetic divergences of 0.6%and 3.3%,respectively.Based on the morphological and molecular phylogenetic analysis,we describe the novel dinofl agellate as Karlodinium elegans sp.nov.A toxicity assay revealed that the clonal culture of K.elegans(strain PTB601)had no adverse eff ect on brine shrimp(Artemia salina)and marine medaka(Oryzias melastigma),indicating it may not be a toxic species. | Jingyi CEN Jianyan WANG Lifen HUANG Yarou LIN Guangmao DING Yuzao QI Songhui LÜ | 2021 | Journal of Oceanology and Limnology2021,39,1: | 1 |
