4Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response.Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan 2021Cancer Biology & Medicine2021,18,3:11
5Adaptive Neural Network-Based Control for a Class of Nonlinear Pure-Feedback Systems With Time-Varying Full State Constraints显示文摘In this paper, an adaptive neural network(NN)control approach is proposed for nonlinear pure-feedback systems with time-varying full state constraints. The pure-feedback systems of this paper are assumed to possess nonlinear function uncertainties. By using the mean value theorem, pure-feedback systems can be transformed into strict feedback forms. For the newly generated systems, NNs are employed to approximate unknown items. Based on the adaptive control scheme and backstepping algorithm, an intelligent controller is designed. At the same time, time-varying Barrier Lyapunov functions(BLFs)with error variables are adopted to avoid violating full state constraints in every step of the backstepping design. All closedloop signals are uniformly ultimately bounded and the output tracking error converges to the neighborhood of zero, which can be verified by using the Lyapunov stability theorem. 6Chemical characteristics of size-resolved aerosols in winter in Beijing显示文摘Size-resolved aerosols were continuously collected by a Nano Sampler for 13 days at an urban site in Beijing during winter 2012 to measure the chemical composition of ambient aerosol particles. Data collected by the Nano Sampler and an ACSM(Aerodyne Aerosol Chemical Speciation Monitor) were compared. Between the data sets,similar trends and strong correlations were observed,demonstrating the validity of the Nano Sampler. PM10 and PM2.5concentrations during the measurement were 150.5 ± 96.0 μg/m3(mean ± standard variation)and 106.9 ± 71.6 μg/m3,respectively. The PM2.5/PM10 ratio was 0.70 ± 0.10,indicating that PM2.5dominated PM10. The aerosol size distributions showed that three size bins of 0.5–1,1–2.5 and 2.5–10 μm contributed 21.8%,23.3% and 26.0% to the total mass concentration(TMC),respectively. OM(organic matter) and SIA(secondary ionic aerosol,mainly SO42-,NO3-and NH4+) were major components of PM2.5. Secondary compounds(SIA and secondary organic carbon) accounted for half of TMC(about 49.8%) in PM2.5,and suggested that secondary aerosols significantly contributed to the serious particulate matter pollution observed in winter. Coal burning,biomass combustion,vehicle emissions and SIA were found to be the main sources of PM2.5. 8Abnormal energy metabolism and tau phosphorylation in the brains of middle-aged mice in response to atmospheric PM2.5 exposure显示文摘In light of the accelerated aging of the global population and the deterioration of the atmosphere pollution, we sought to clarify the potential mechanisms by which fine particulate matter(PM_(2.5)) can cause cognitive impairment and neurodegeneration through the alteration of mitochondrial structure and function. The results indicate that PM_(2.5) inhalation reduces ATP production by disrupting the aerobic tricarboxylic acid cycle and oxidative phosphorylation, thereby causing the hypophosphorylation of tau in the cortices of middle-aged mice. Furthermore, excessive reactive oxygen species generation was involved in the impairment. Interestingly, these alterations were partially reversed after exposure to PM_(2.5) ended. 10Characterization of Carotenoid Accumulation and Carotenogenic Gene Expression During Fruit Development in Yellow and White Loquat Fruit显示文摘Accumulation of carotenoids in peel and pulp of the yellow-fleshe loquat ‘Zaozhong 6'(ZZ6) and the white-fleshe loquat ‘Baiyu'(BY) were tracked during different fruit development stages, and the expression of 15 carotenogenic genes were analyzed. During loquat fruit ripening the fresh weight content of β-carotene in peel and pulp of ZZ6 increased gradually and peaked at the fully ripe stage, reaching 68.53 μg·g^(-1)FW in the peel and 11.92 μg·g^(-1)FW in the pulp. In BY, the content of β-carotene in the peel increased and peaked at the fully ripe stage, reaching38.89 μg·g^(-1)FW, while it decreased in the pulp from the original 0.47 μg·g~(^(-1))FW and reduced to 0.29 μg·g~(^(-1))FW. The content of β-cryptoxanthin in the peel and pulp of ZZ6 and BY both increased steadily, and peaked at the fully ripe stage; however, the content of lutein decreased in the peel of ZZ6 and increased in the pulp, but in BY, it dropped and then rose in the peel. There was no significan change of β-cryptoxanthin in the pulp of BY. After the breaker stage, the m RNA levels of phytoene synthase(PSY) and chromoplast-specifi lycopene β-cyclase(CYCB) were higher in the peel, while CYCB and β-carotene hydroxylase(BCH) m RNAs were higher in the fles of ZZ6, compared with BY. 14Comparison of Modified Mohr-Coulomb Model and Bai-Wierzbicki Model for Constructing 3D Ductile Fracture Envelope of AA6063显示文摘Ductile fracture of metal often occurs in the plastic forming process of parts.The establishment of ductile fracture criterion can effectively guide the selection of process parameters and avoid ductile fracture of parts during machining.The 3D ductile fracture envelope of AA6063-T6 was developed to predict and prevent its fracture.Smooth round bar tension tests were performed to characterize the flow stress,and a series of experiments were conducted to characterize the ductile fracture firstly,such as notched round bar tension tests,compression tests and torsion tests.These tests cover a wide range of stress triaxiality(ST)and Lode parameter(LP)to calibrate the ductile fracture criterion.Plasticity modeling was performed,and the predicted results were compared with corresponding experimental data to verify the plasticity model after these experiments.Then the relationship between ductile fracture strain and ST with LP was constructed using the modified Mohr-Coulomb(MMC)model and Bai-Wierzbicki(BW)model to develop the 3D ductile fracture envelope.Finally,two ductile damage models were proposed based on the 3D fracture envelope of AA6063.Through the comparison of the two models,it was found that BW model had better fitting effect,and the sum of squares of residual error of BW model was 0.9901.The two models had relatively large errors in predicting the fracture strain of SRB tensile test and torsion test,but both of the predicting error of both two models were within the acceptable range of 15%.In the process of finite element simulation,the evolution process of ductile fracture can be well simulated by the two models.However,BW model can predict the location of fracture more accurately than MMC model.Jianye Gao Tao He Yuanming Huo Miao Song Tingting Yao Wanbo Yang 2021Chinese Journal of Mechanical Engineering2021,34,2:6
15Estimating antiwear properties of esters as potential lubricantbased oils using QSTR models with CoMFA and CoMSIA显示文摘Comparative molecular field analysis and comparative molecular similarity indices analysis were employed to analyze the antiwear properties of a series of 57 esters as potential lubricant-based oils. Predictive 3D-quantitative structure tribo-ability relationship models were established using the SYBYL multifit molecular alignment rule with a training set and a test set. 16The Renminbi as a Trading Currency:Evidence from Selected Countries Participating in the Belt and Road Initiative显示文摘This paper investigates the effects of the renminbi(RMB)exchange rate on trade prices and volumes in selected Belt and Road Initiative(BRI)countries in comparison with the effects of the US dollar.The stylized facts show that the RMB is underused in bilateral trade with selected BRI countries where intermediate goods dominate.By estimating the level of exchange rate pass-through and trade volume elasticity,we find that the RMB is significantly correlated with the volume of imports in the sample countries,predicted by the producer currency pricing(PCP)paradigm.We also regroup intermediate and final goods between China and the BRI countries.The evidence shows that dollar fluctuation affects export volumes,reflecting the role of the US as a final goods destination,whereas the RMB exerts a significant impact on the volume of intermediate goods imported from China to the sample countries due to China s important position in global value chains.Haihong Gao Tingting Li 2020China & World Economy2020,28,5:5
17Germination inhibitors detected in Sapium sebiferum seeds显示文摘This study identifies endogenous inhibitors of germination associated with seed dormancy in Sapium sebiferum,Chinese tallow.Seed extracts were separated into five distinct fractions(petroleum ether,ether,ethyl acetate,methanol,and aqueous)and tested for their effect on germination of cabbage seeds.There were no significant differences in germination percentages and germination index(GI)between the solvents and the control.The five fractions differentially reduced germination percentage and GI values of cabbage seeds.According to the GI results,the greatest inhibition was with the ether fraction followed by methanol,ethyl acetate,petroleum ether,and aqueous fractions.Gas chromatography–mass spectrometry identified the constituents of the four organic fractions.Compared with the mass spectrum of the National Institute of Standards and Technology library,a compound was confirmed,provided it was more than 85%similar.Analysis of ether extracts identified four compounds:butylated hydroxytoluene(BHT;19.7%),1,2-benzenediol(4.1%),phydroxycinnamic acid,ethyl ester(2.8%),and n-hexadecanoic acid(1.4%).Fourteen,20,and 9 compounds were identified in methanol,ethyl acetate,and petroleum ether extracts,respectively.The IC50 of BHT(the concentration at which BHT inhibits radicle growth by 50%)was 54.8 mg/L,indicating that the presence of BHT might be the primary cause of dormancy in S.sebiferum seeds.Tingting Zhao Cunmeng Qian Yunpeng Gao Li Chen Mingwei Zhu Yueting Pan Shuxian Li 2019Journal of Forestry Research2019,30,6:4
18Variation and heritability of morphological and physiological traits among Leymus chinensis genotypes under different environmental conditions显示文摘Intraspecific trait variation and heritability in different environmental conditions not only suggest a potential for an evolutionary response but also have important ecological consequences at the population, community, and ecosystem levels. However, the contribution of quantitative trait variation within a grassland species to evolutionary responses or ecological consequences is seldom documented. Leymus chinensis is an important dominant species in semi-arid grasslands of China, which has seriously suffered from drought and high temperature stresses in recent decades. In the present study, we measured variation and heritability of 10 quantitative traits, namely the number of tillers, maximum shoot height, number of rhizomes, maximum rhizome length, rhizome mass, aboveground mass, root mass, maximum net photosynthetic rate(Pmax), specific leaf area(SLA), and leaf length to leaf width ratio(LL/LW), for 10 genotypes of L. chinensis under one non-stress(Ck) condition and three environmental stress conditions(i.e., drought(Dr), high temperature(Ht), and both drought and high temperature(DrHt)). Result indicated that(1) the interaction of genotype and environmental condition(G×E) was significant for 6 traits but not significant for the other 4 traits as shown by two-way analysis of variance(ANOVA), suggesting that different selection forces were placed for different traits on the factors dominating phenotypic responses to different environmental conditions. Moreover, these significant G×E effects on traits indicated significantly different phenotypic adaptive responses among L. chinensis genotypes to different environmental conditions. Additionally, individuals could be grouped according to environmental condition rather than genotype as shown by canonical discriminant analysis, indicating that environmental condition played a more important role in affecting phenotypic variation than genotype;(2) by one-way ANOVA, significant differences among L. chinensis genotypes were found in all 10 traits under Ck and Dr conditions, in 8 traits under Dr Ht condition and only in 4 traits under Ht condition; and(3) all 10 traits showed relatively low or non-measurable broad-sense heritability(H_2) under stress conditions. However, the lowest H_2 value for most traits did not occur under DrHt condition, which supported the hypothesis of 'unfavorable conditions have unpredictable effects' rather than 'unfavorable conditions decrease heritability'.
