1Rett syndrome-causing mutations compromise MeCP2- mediated liquid—liquid phase separation of chromatin显示文摘Rett syndrome(RTT),a severe postnatal neurodevelopmental disorder,is caused by mutations in the X-linked gene encoding methyl-CpG-binding protein 2(MeCP2).MeCP2 is a chromatin organizer regulating gene expression.RTT-causing mutations have been shown to affect this function.However,the mechanism by which MeCP2 organizes chromatin is unclear.In this study,we found that MeCP2 can induce compaction and liquid-liquid phase separation of nudeosomal arrays in vitro,and DNA methylation further enhances formation of chromatin condensates by MeCP2.Interestingly,RTT-causing mutations compromise MeCP2-mediated chromatin phase separation,while benign variants have little effect on this process.Moreover,MeCP2 competes with linker histone H1 to form mutually exclusive chromatin condensates in vitro and distinct heterochromatin foci in vivo.RTT-causing mutations reduce or even abolish the ability of MeCP2 to compete with histone H1 and to form chromatin condensates.Together,our results identify a novel mechanism by which phase separation underlies MeCP2-mediated heterochromatin formation and reveal the potential link between this process and the pathology of RTT.Liang Wang Mingli Hu Mei-Qing Zuo Jicheng Zhao Di Wu Li Huang Yongxin Wen Yunfan Li Ping Chen Xinhua Bao Meng-Qiu Dong Guohong Li Pilong Li 2020Cell Research2020,30,5:3
2Characteristics of mass distributions of aerosol particle and its inorganic water-soluble ions in summer over a suburb farmland in Beijing显示文摘Agricultural activity is one of the most important sources of aerosol particles.To understand the mass distribu-tion and sources of aerosol particles and their inorganic water-soluble ions in a suburb farmland of Beijing,particle samples were collected using a microorifice uniform deposit impactor(MOUDI)in the summer of 2004 in a suburb vege-table field.The distribution of the particles and their inor-ganic water-soluble ions in the diameter range of 0.18-18 mm were measured.The dominant fine particle ions were SO_(4)^(2−),NO^(3−),and NH_(4)^(+).The association of day-to-day variation of the concentration of these ions with temperature,humidity,and solar radiation suggested that they are formed by the reac-tion of NH_(3) released from the vegetable field with the acid species produced from photochemical reactions.Fine particle K+is likely from vegetation emission and biomass burning.Coarse particles like Ca^(2+),Mg^(2+),NO_(3)^(−),and SO_(4)^(2−)are sug-gested to come from the mechanical process by which the soil particle entered the atmosphere,and from the reaction of the acid species at the surface of the soil particle.The results show that fertilizer and soil are important factors determining the aerosol particle over agricultural fields,and vegetable fields in suburban Beijing contribute significantly to the aerosol particle.ZHAO Peng ZHU Tong LIANG Baosheng HU Min KANG Ling GONG Jicheng 2007Frontiers of Environmental Science & Engineering2007,1,2:2

14Synthesis and electrochemical properties of a series of novel tetra(4-benzoyl)phenoxyphthalocyanine derivatives显示文摘A novel phthalocyanine,2,9(10),16(17),23(24)-tetra(4-benzoyl)phenoxyphthalocyanine,and its complexes with Zn(II),Cu(II),Co(II),and Ni(II) have been synthesized and characterized by a combination of elemental analysis,IR,1 H NMR,UV-vis spectroscopy and mass spectrometry.All of the materials are very soluble in common organic solvents such as dichloromethane,chloroform,tetrahydrofuran,N,N-dimethylformamide and dimethyl sulfoxide.The Q band wavelengths of the complexes decrease in the order:Zn > Cu > Ni > Co.Redox processes were observed at 1.06,0.74,0.51 and 0.98 V for the free phthalocyanine,at 0.72 and 1.04 V for the Co(II) complex,at 1.24,0.77,0.24,0.61 and 0.91 V for the Cu(II) complex,and at 0.74 and 1.20 V for the Ni(II) complex.The cyclic voltammograms of the phthalocyanine ring of the four species are similar,with reduction and oxidation couples each involving a one-electron transfer process.WANG Kun FU Qiang MA JiCheng LI WeiLi LlWenJu HOU Yang CHEN LiLi ZHAO BaoZhong 2012Science China Chemistry2012,55,9:0

19Selective deletion of zinc transporter 3 in amacrine cells promotes retinal ganglion cell survival and optic nerve regeneration after injury显示文摘Vision depends on accurate signal conduction from the retina to the brain through the optic nerve,an important part of the central nervous system that consists of bundles of axons originating from retinal ganglion cells.The mammalian optic nerve,an important part of the central nervous system,cannot regenerate once it is injured,leading to permanent vision loss.To date,there is no clinical treatment that can regenerate the optic nerve and restore vision.Our previous study found that the mobile zinc(Zn^(2+))level increased rapidly after optic nerve injury in the retina,specifically in the vesicles of the inner plexiform layer.Furthermore,chelating Zn^(2+)significantly promoted axonal regeneration with a long-term effect.In this study,we conditionally knocked out zinc transporter 3(ZnT3)in amacrine cells or retinal ganglion cells to construct two transgenic mouse lines(VGAT^(Cre)ZnT3^(fl/fl)and VGLUT2^(Cre)ZnT3^(fl/fl),respectively).We obtained direct evidence that the rapidly increased mobile Zn^(2+)in response to injury was from amacrine cells.We also found that selective deletion of ZnT3 in amacrine cells promoted retinal ganglion cell survival and axonal regeneration after optic nerve crush injury,improved retinal ganglion cell function,and promoted vision recovery.Sequencing analysis of reginal ganglion cells revealed that inhibiting the release of presynaptic Zn^(2+)affected the transcription of key genes related to the survival of retinal ganglion cells in postsynaptic neurons,regulated the synaptic connection between amacrine cells and retinal ganglion cells,and affected the fate of retinal ganglion cells.These results suggest that amacrine cells release Zn^(2+)to trigger transcriptomic changes related to neuronal growth and survival in reginal ganglion cells,thereby influencing the synaptic plasticity of retinal networks.These results make the theory of zinc-dependent retinal ganglion cell death more accurate and complete and provide new insights into the complex interactions between retinal cell networks.Zhe Liu Jingfei Xue Canying Liu Jiahui Tang Siting Wu Jicheng Lin Jiaxu Han Qi Zhang Caiqing Wu Haishun Huang Ling Zhao Yehong Zhuo Yiqing Li 2023Neural Regeneration Research2023,18,12:0
