2Differentiation of embryonic stem cells into corneal epithelium显示文摘Our project was to determine whether embryonic stem (ES) cells could be induced to differentiate into corneal epithelia by superficial corneoscleral limbal stroma. To achieve this goal, ES-GFP cell line D3 was pre-induced by retinoic acid (RA). The pre-induced cells were seeded on deepithelialized superficial corneoscleral slices (SCSS) to form a monolayer, and di-vided into three groups. Group 1 was cultured and passaged in vitro for direct detection. Group 2 was exposed to air-liquid interfaces for 10 days and implanted into the subcutaneous layer of nude mice for 2 weeks for further induction in vivo. Group 3 was cultured in vitro without any in-ducing factors for control. There were no teratomas found in nude mice which were implanted with differentiated ES cells after two weeks. The differentiated cells showed an appearance of epithelia both in vitro and in vivo. Expression of CK3, P63 and PCNA was detected by immuno-histochemical staining in the differentiated cells in group 1 and 2. Microvillis and zonula oc-cludens were observed on the surface of the differentiated cells under an electron microscope. In the control group, ES cells differentiated freely without any inducing factors. Most cells were shed and formed a neuronal dendrite-like structure, and a minority of cells appeared polymorphic. These results demonstrate that ES cells can differentiate into corneal epithelia on the surface of SCSS under the controlled condition. 3Induction of corneal epithelial progenitors from bone-marrow mesenchymal stem cells of rhesus monkeys in vitro显示文摘Bioengineered corneas are substitutes for human donor tissue that are designed to treat severe dis-ease affecting ocular surfaces. However,a shortage of candidate seed cells for bioengineering corneas is still a problem. Bone-marrow mesenchymal stem cells (MSCs) are capable of multilineage differen-tiation. Therefore,we determined whether MSCs differentiate into corneal epithelial cells (ECs). We applied three exoteric-microenvironmental systems to induce MSCs to become ECs. Induced MSC were identified by means of morphologic examination,immunocytochemical analysis,and flow cytometry. MSCs grown in one microenvironment had characteristics similar to those of corneal epithelial pro-genitors. Induced MSCs expressed markers for EC,including integrin β1,Cx43,Pax6,and P63. MSCs were successfully induced to become corneal epithelial progenitors. 4Identification and molecular mechanism of angiotensin-converting enzyme inhibitory peptides from Larimichthys crocea titin显示文摘This study aimed to identify novel ACEI peptides from Larimichthys crocea titin using in silico approaches and to clarify the molecular interaction mechanism.The hydrolyzed peptides of titin were compared with known ACEI peptides in the AHTPDB and BIOPEP-UWM database.Furthermore,peptides were evaluated for their solubility,ADMET properties,ΔG(kcal/mol)values,and in vitro ACEI activity.Molecular mechanism of ACE-peptide was performed by molecular interactions and binding orientation study.The results revealed that IC50 values of Trp-Ala-Arg(WAR)and Trp-Gln-Arg(WQR)were(31.2±0.8)and(231.33±0.02)mol/L,respectively.The docking interactions result suggested that ACE-WAR and ACEWQR complexes have same binding site,including the residues LYS511,TYR520,TYR523,HIS353,and HIS513.Molecular docking of two tripeptides WAR and WQR with ACE studies predicted their binding site and clarified the interaction between ACE and its inhibitors.The molecular docking data are consistent with the ACE inhibitory activity of the studied peptides.The results showed that Larimichthys crocea titin may be a valuable source for developing nutraceutical food.Yue Fan Zhipeng Yu Wenzhu Zhao Long Ding Fuping Zheng Jianrong Li Jingbo Liu 2020Food Science and Human Wellness2020,9,3:5
5Porphyromonas gingivalis exacerbates ulcerative colitis via Porphyromonas gingivalis peptidylarginine deiminase显示文摘Abstract:Ulcerative Colitis(UC)has been reported to be related to Porphyromonas gingivalis(P.gingivalis).Porphyromonas gingivalis peptidylarginine deiminase(PPAD),a virulence factor released by P.gingivalis,is known to induce inflammatory responses.To explore the pathological relationships between PPAD and UC,we used homologous recombination technology to construct a P.gingivalis strain in which the PPAD gene was deleted(Δppad)and aΔppad strain in which the PPAD gene was restored(comΔppad).C57 BL/6 mice were orally gavaged with saline,P.gingivalis,Δppad,or comΔppad twice a week for the entire 40 days(days 0-40),and then,UC was induced by dextran sodium sulfate(DSS)solution for 10 days(days 31-40).P.gingivalis and comΔppad exacerbated DDS-induced colitis,which was determined by assessing the parameters of colon length,disease activity index,and histological activity index,butΔppad failed to exacerbate DDS-induced colitis.Flow cytometry and ELISA revealed that compared withΔppad,P.gingivalis,and comΔppad increased T helper 17(Th17)cell numbers and interleukin(IL)-17 production but decreased regulatory T cells(Tregs)numbers and IL-10 production in the spleens of mice with UC.We also cocultured P.gingivalis,Δppad,or comΔppad with T lymphocytes in vitro and found that P.gingivalis and comΔppad significantly increased Th17 cell numbers and decreased Treg cell numbers.Immunofluorescence staining of colon tissue paraffin sections also confirmed these results.The results suggested that P.gingivalis exacerbated the severity of UC in part via PPAD.Xida Zhao Jingbo Liu Chong Zhang Ning Yu Ze Lu Shuwei Zhang Yuchao Li Qian Li Junchao Liu Dongjuan Liu Yaping Pan 2021International Journal of Oral Science2021,13,3:5
6Chattering analysis for discrete sliding mode control ofdistributed control systems显示文摘The chattering characteristic of sliding mode control is analyzed when it is applied in distributed control systems(DCSs).For a DCS with random time delay and packet dropout, a discrete switching system model with time varying sampling period is constructed based on the time delay system method. The reaching law based sliding mode controller is applied in the proposed system. The exponential stability condition in the form of linear matrix inequality is figured out based on the multi-Lyaponov function method. Then, the chattering characteristic is analyzed for the switching system, and a chattering region related with time varying sampling period and external disturbance is proposed. 7Initial scalar lithospheric magnetic anomaly map of China and surrounding regions derived from CSES satellite data显示文摘The China Seismo-Electromagnetic Satellite(CSES),China’s first satellite to measure geophysical fields with scientific goals in both space and solid earth physics,was launched successfully in February 2018.It carries high-precision magnetometers to measure the geomagnetic field.In this study,the CSES magnetic data were used to extract the signal of the lithospheric magnetic field caused by magnetized rocks in the crust and uppermost mantle.First,an along-track analysis of the CSES magnetic data was undertaken near the Bangui magnetic anomaly in central Africa and the Tarim magnetic anomaly in China,demonstrating that the CSES magnetic data are indeed sensitive to the lithospheric magnetic anomaly field.Then a lithospheric magnetic anomaly map over China and surrounding regions was derived.This map is consistent with the lithospheric part of the CHAOS-7 model.In particular,it clearly reveals four major magnetic anomalies containing long-wavelength signals at the altitude of lowearth-orbiting satellites.Three magnetic highs are located over the Tarim,Sichuan and Songliao basin,the origins of which could be related to large-scale tectonic-magmatic activities during geological history.A prominent magnetic low is otherwise found in the southern Himalayan-Tibetan plateau,possibly caused by the shallow Curie depth in this region.To further improve the precision of the lithospheric magnetic field model,more detailed data processing and multi-source data merging are needed.WANG Jie SHEN XuHui YANG YanYan ZEREN ZhiMa HULOT Gauthier OLSEN Nils ZHOU Bin MAGNES Werner DE SANTIS Angelo HUANG JianPing GUO Feng LIU WenLong YU JingBo 2021Science China(Technological Sciences)2021,64,5:4

9Fabricating antibacterial CoCrCuFeNi high-entropy alloy via selective laser melting and in-situ alloying显示文摘A quasi-equiatomic CoCrFeCuNi high-entropy alloy(HEA) with a broad-spectrum antibacterial ability and good mechanical properties has been fabricated by selective laser melting(SLM) and in-situ alloying of a blend of pre-alloyed CoCrFeNi powder and Cu elemental powder.The as-built HEA alloy has a homogeneous distribution of Cu and presents a single FCC phase.Compared with the same HEA fabricated using the traditional ingot metallurgy(IM) process,the HEA alloy fabricated by SLM releases more Cu ions to prevent growth and biofilm formation by gram-negative Escherichia coli and gram-positive Staphylococcus aureus,which enhances the applicability of the HEA alloy in potential applications that requires antibacterial ability.The results of this study confirm the feasibility of combining the antibacterial CoCrFeCuNi HEA alloy and SLM technology in fabricating complex shaped parts or structures with a strong antibacterial ability to be used in medical application or other environments desired for antibacterial ability.Jingbo Gao Yuting Jin Yongqiang Fan Dake Xu Lei Meng Cong Wang Yuanping Yu Deliang Zhang Fuhui Wang 2022Journal of Materials Science & Technology2022,,7:4

13High-throughput metabolomics reveals the perturbed metabolic pathways and biomarkers of Yang Huang syndrome as potential targets for evaluating the therapeutic effects and mechanism of geniposide显示文摘High-throughput metabolomics can clarify the underlying molecular mechanism of diseases via the qualitative and quantitative analysis of metabolites.This study used the established Yang Huang syndrome(YHS)mouse model to evaluate the efficacy of geniposide(GEN).Urine metabolic data were quantified by ultraperformance liquid chromatography-tandem mass spectrometry.The non-target screening of the massive biological information dataset was performed,and a total of 33 metabolites,including tyramine glucuronide,aurine,and L-cysteine,were identified relating to YHS.These differential metabolites directly participated in the disturbance of phase I reaction and hydrophilic transformation of bilirubin.Interestingly,they were completely reversed by GEN.While,as the auxiliary technical means,we also focused on the molecular prediction and docking results in network pharmacological and integrated analysis part.We used integrated analysis to communicate the multiple results of metabolomics and network pharmacology.This study is the first to report that GEN indirectly regulates the metabolite“tyramine glucuronide”through its direct effect on the target heme oxygenase 1 in vivo.Meanwhile,heme oxygenase-1,a prediction of network pharmacology,was the confirmed metabolic enzyme of phase I reaction in hepatocytes.Our study indicated that the combination of high-throughput metabolomics and network pharmacology is a robust combination for deciphering the pathogenesis of the traditional Chinese medicine(TCM)syndrome.Heng Fang Aihua Zhang Xiaohang Zhou Jingbo Yu Qi Song Xijun Wang 2020Frontiers of Medicine2020,14,5:3
14Biological evaluation and interaction mechanism of beta-site APP cleaving enzyme 1 inhibitory pentapeptide from egg albumin显示文摘Inhibition of beta-site APP cleaving enzyme1(BACE1)is one of the most promising therapeutic approaches for Alzheimer’s disease.To find natural products for the treatment of Alzheimer’s disease,absorption,distribution,metabolism,excretion and toxicity(ADMET)properties and in vitro BACE1 inhibitory activity of the peptides isolated from egg albumin were evaluated.Then,molecular docking and molecular dynamics simulation were used to explain the molecular mechanism of the interactions between BACE1 and peptides.The IC50 value of peptide KLPGF,with satisfactory ADMET properties,against BACE1 was(8.30±0.56)mmol/L.Molecular docking revealed that KLPGF contacted with the residues of BACE1’s active sites through twelve hydrogen bonds interactions,two hydrophobic interactions,one electrostatic interaction,and two Pi-cation interactions.The 5 ns molecular dynamics simulations confirmed that the structure of KLPGF with BACE1 was stable.Peptide KLPGF contacted the residues Lys321,Asp228,and Asn233 with stable hydrogen bonds.KLPGF may be a potential anti-BACE1 candidate.Zhipeng Yu Sijia Wu Wenzhu Zhao Long Ding David Shiuan Fuping Zheng Jianrong Li Jingbo Liu 2020Food Science and Human Wellness2020,9,2:2

16Virtual screening,molecular docking and identification of umami peptides derived from Oncorhynchus mykiss显示文摘Oncorhynchus mykiss is delicious and contains abundant flavor substances.However,few studies focused on umami peptides of O.mykiss.In the current work,umami peptides derived from O.mykiss were identified using virtual screening,molecular docking,and electronic tongue analysis.First,the O.mykiss protein was hydrolyzed using the PeptideCutter online enzymolysis program.Subsequently,water-soluble and toxicity screening were performed by Innovagen and ToxinPred software,respectively.The potential peptides were docked with umami receptor T1R1/T1R3.Furthermore,taste properties of potential peptides were validated by electronic tongue.Docking results suggested that the three tetrapeptide EANK,EEAK,and EMQK could enter the binding pocket in the T1R1 cavity,wherein Arg151,Asp147,Gln52,and Arg277 may play key roles in the production of umami taste.Electronic tongue results showed that the umami value of EANK,EEAK,and EMQK were stronger than monosodium glutamate.This work provides a new insight for the screening of umami peptides in O.mykiss.Wenzhu Zhao Lijun Su Shitong Huo Zhipeng Yu Jianrong Li Jingbo Liu 2023Food Science and Human Wellness2023,12,1:2

18Elutriation characteristics of multi-component mixtures in a fluidized bed pyrolysis process显示文摘Multi-component mixtures were prepared to simulate a bed of walnut shells undergoing pyrolysis in acold fluidized bed.Four mixtures with different ratios of walnut shell,semi-chars,and clean char were used.Batch elutriation experiments using these mixtures were performed in a transparent fluidized bed to allow direct observation of the flow patterns and particle composition.The results indicated that the bubble wake rather than bubble nose was primarily responsible for particle entrainment at higher gas velocities,and that coarser particles would be“transformed”into elutriable particles.Dimensionless gas velocities ranged from 0.2 to 1.0 and an exponential decrease in particle concentration with respect to bed height was observed.The transport disengagement height(TDH)gradually increased with the gas velocity until the entire bed layer enters a pneumatic transport state.Notably,larger TDHs were required when the bed contained a larger fraction of light components.Two characteristic parameters were used to evaluate particle elutriation:the elutriation rate constant(K),and the residual volatile content of the elutriated particles(v′).These parameters were used to optimize operating gas velocity for the fluidized bed.Xing Wu Jingbo Peng Yu Zhang Zhiduo Wang 2021Particuology2021,19,5:2
