| 4 | Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis. | Wang, L. | 5 | Clinicopathological, immunohistochemical, and ultrastructural study of 13 cases of melanotic schwannoma显示文摘Background Melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cellswith ultrastructural features of schwann cells.The description of the course of the tumors differs somewhat, but itis generally considered as a benign lesion.We investigated the clinicopathologic features, immunophenotypes,and ultrastructural features of 13 patients with nonpsammomatous melanotic schwannoma (NPMS).Methods Tumor specimens of each patient were sectioned and stained with hematoxylin-eosin, Fontana-Masson, Prussian blue, and periodic acid-Schiff (PAS).Immunohistochemical markers such as S-100, Leu-7,HMB-45, Melan-A, CK, EMA, vimentin, GFAP, laminin, collagenⅣ and MIB-1 were detected with theEnvision immunohistochemical staining method.Four of the cases were observed by electron microscopy.Results Of the 13 patients, 8 were male and 5 female, aged from 11 to 92 years (mean, 38.6 years).Thetumor sites included the spinal nerve root (5 patients), cranial nerve (1), greater omentum (1), subcutaneoustissue (3), mesentery (1), bone (1) and mediastinum(1).Eleven patients were followed up for over2 years,with a mean of 5.9 years.One patient (9.1%) with a primary tumor in the greater omentum developed anotherprimary tumor of the same type in the subcutaneous tissue of the abdominal wall after the first operation.Localrecurrence of the tumor was seen in2 patients (18.2%).One patient (9.1%) showed the local recurrence andmetastasis.Seven patients (63.6%) showed no evidence of the recurrence or metastasis.Grossly, all tumorswere well-circumscribed and the gross findings were suggestive of melanin-containing tumors.The tumor wascomposed of spindled and epithelioid cells with abundant intracytoplasmic melanin pigments.Nuclei were roundand contained delicate, evenly distributed chromatins as well as small, distinct nucleoli.In some areas, thenucleoli were large and prominent.Rare mitoses were seen in mostlesions exceptthe larger omentumlesion.Thepigment was shown to be positive for the Fontana-Masson and negative for Prussian blue and PAS.Immunohistochemical staining for S-100, Leu-7, HMB-45, Melan-A, and vimentin were strongly positive.Linear immunoreactions of both laminin and collagenⅣ was detected in all patients.Ultrastructurally, numerouselongated tumor-cell processes, duplicated basement membrane and melanosomes were observed in alldevelopmental stages.Conclusions Histologically, melanotic schwannoma is a rare variant of schwannoma composed of melanin-producing cells with ultrastructural features ofschwann cells.Distinguishing between this tumor andmalignant melanoma is of paramount importance inplanning of management. Immunohistochemically,combined use of laminin and collagenⅣ is valuablein distinguishing melanotic schwannoma frommalignant melanoma. Wide local resection andadditional radiotherapy should be advocated.Furtherstudies including cytogenetic or molecular biology arestill required to better delineate melanoticschwannoma from malignant melanoma. | 8 | Human embryonic stem cells-derived endothelial cell therapy facilitates kidney regeneration by stimulating renal resident stem cell proliferation in acute kidney injury显示文摘Endothelial cell therapy has been implicated to enhance tissue regeneration and vascularization in ischemic kidney. However, no published study has yet examined direct effects of endothelial cell treatment in kidney recovery. This study investigated the therapeutic efficacy of endothelial cells in a mouse model with acute kidney injury (AKI). Thus, human embryonic stem cells-derived endothelial cells (hESC-ECs) labeled with a reporter system encoding a double fusion reporter gene for firefly luciferase (Fluc) and green fluorescent protein (GFP) were characterized by Fluc imaging and immunofluoresence staining. Cultured hESC-ECs (1×106) were injected into ischemic kidney shortly after AKI. Survival of the transplanted hESC-ECs was monitored in vivo from day 1 to 14 after endothelial cell transplantation and potential impact of hESC-EC treatment on renal regeneration was assessed by histological analyses. We report that a substantial level of bioluminescence activity was detected 24 h after hESC-EC injection followed by a gradual decline from 1 to 14 d. Human ESC-ECs markedly accelerated kidney cell proliferation in response to ischaemia-induced damage, indicated by an elevated number of BrdU+ cells. Co-expression of Sca-1, a kidney stem cell proliferation marker, and BrdU further suggested that the observed stimulation in renal cell regeneration was, at least in part, due to increased proliferation of renal resident stem cells especially within the medullary cords and arteriole. Differentiation of hESC-ECs to smooth muscle cells was also observed at an early stage of kidney recovery. | 9 | Analysis and Inverse Substructuring Computation on Dynamic Quality of Mechanical Assembly显示文摘Mechanical assembly has its own dynamic quality directly affecting the dynamic quality of whole product and should be considered in quality inspection and estimation of mechanical assembly. Based on functional relations between dynamic characteristics involved in mechanical assembly, the effects of assembling process on dynamic characteristics of substructural components of an assembly system are investigated by substructuring analysis. Assembly-coupling dynamic stiffness is clarified as the dominant factor of the effects and can be used as a quantitative measure of assembly dynamic quality. Two computational schemes using frequency response functions(FRFs) to determine the stiffness are provided and discussed by inverse substructuring analysis, including their applicable conditions and implementation procedure in application. Eigenvalue analysis on matrix-ratios of FRFs before and after assembling is employed and well validates the analytical outcomes and the schemes via both a lumped-parameter model and its analogic experimental counterpart.
