| 3 | Calcium signaling and T-type calcium channels in cancer cell cycling显示文摘Regulation of intracellular calcium is an important signaling mechanism for cell proliferation in both normal and cancerous cells. In normal epithelial cells, free calcium concentration is essential for cells to enter and accomplish the S phase and the M phase of the cell cycle. In contrast, cancerous cells can pass these phases of the cell cycle with much lower cytoplasmic free calcium concentrations, indicating an alternative mechanism has developed for fulfilling the intracellular calcium requirement for an increased rate of DNA synthesis and mitosis of fast replicating cancerous cells. The detailed mechanism underlying the altered calcium loading pathway remains unclear; however, there is a growing body of evidence that suggests the T-type Ca2+ channel is abnormally expressed in cancerous cells and that blockade of these channels may reduce cell proliferation in addition to inducing apoptosis. Recent studies also show that the expression of T-type Ca2+ channels in breast cancer cells is proliferation state dependent, i.e. the channels are expressed at higher levels during the fast-replication period, and once the cells are in a non-proliferation state, expression of this channel isminimal. Therefore, selectively blocking calcium entry into cancerous cells may be a valuable approach for preventing tumor growth. | 6 | Tumor Antigen Specific Activation of Primary Human T-Cells Expressing a Virally Encoded Chimeric T-Cell Receptor Specific for p185HER2显示文摘We have developed and tested chimeric T-cell receptors (TCR) specific for p185HER2. In these experiments, retroviral vectors expressing the N29γ or N29ζ receptors were constructed in pRET6. Amphotropic viral producer cells were established in the GALV-based PG13 packaging cell line. Ficoll purified human peripheral blood lymphocytes (PBL) were virally transduced using an optimized protocol incorporating activation with immobilized anti-CD3/anti-CD28 monoclonal anti- bodies, followed by viral infection in the presence of fibronectin fragment CH296. Transduced cells were co-cultured with human tumor cell lines that overexpress (SK-OV-3) or underexpress (MCF7) p185HER2 to assay for antigen specific im- mune responses. Both CM+ and CD8+ T-cells transduced with the N29γ or N29ζ chTCR demonstrated HER2-specific anti- gen responses, as determined by release of Th1 like cytokines, and cellular cytotoxicity assays. | 7 | Impact of time to surgery from injury on postoperative infection and deep vein thrombosis in periprosthetic knee fractures显示文摘Purpose:Periprosthetic fracture (PPF) is a serious complication that occurs in 0.3%-2.5% of all total knee arthroplasties used to treat end-stage arthritis. To our knowledge, there are no studies in the literature that evaluate the association between time to surgery after PPF and early postoperative infections or deep vein thrombosis (DVT). This study tests our hypothesis that delayed time to surgery increases rates of postoperative infection and DVT after PPF surgery.Methods:Our study cohort included patients undergoing PPF surgery in the American College of Surgeons National Surgical Quality Improvement Program database (2006-2015). The patients were dichotomized based on time to surgery: group 1 with time ≤2 days and group 2 with time >2 days. A 2-by-2 contingency table and Fisher's exact test were used to evaluate the association between complications and time to surgery groups, and multivariate logistic regression was used to adjust for demographics and known risk factors.Results:A total of 263 patients (80% females) with a mean age of 73.9 ± 12.0 years were identified receiving PPF surgery, among which 216 patients were in group 1 and 47 patients in group 2. Complications in group 1 included 3 (1.4%) superficial infections (SI), 1 (0.5%) organ space infection (OSI), 1 (0.5%) wound dehiscence (WD), and 4 (1.9%) deep vein thrombosis (DVT);while complications in group 2 included 1 (2.1%) SI, 1 (2.1%) OSI, 1 (2.1%) DVT, and no WD. No significant difference was detected in postoperative complications between the two groups. | 16 | Hepcidin contributes to Swedish mutant APP-induced osteoclastogenesis and trabecular bone loss显示文摘Patients with Alzheimer’s disease(AD)often have lower bone mass than healthy individuals.However,the mechanisms underlying this change remain elusive.Previously,we found that Tg2576 mice,an AD animal model that ubiquitously expresses Swedish mutant amyloid precursor protein(APPswe),shows osteoporotic changes,reduced bone formation,and increased bone resorption.To understand how bone deficits develop in Tg2576 mice,we used a multiplex antibody array to screen for serum proteins that are altered in Tg2576 mice and identified hepcidin,a master regulator of iron homeostasis.We further investigated hepcidin’s function in bone homeostasis and found that hepcidin levels were increased not only in the serum but also in the liver,muscle,and osteoblast(OB)lineage cells in Tg2576 mice at both the mRNA and protein levels.We then generated mice selectively expressing hepcidin in hepatocytes or OB lineage cells,which showed trabecular bone loss and increased osteoclast(OC)-mediated bone resorption.Further cell studies suggested that hepcidin increased OC precursor proliferation and differentiation by downregulating ferroportin(FPN)expression and increasing intracellular iron levels.In OB lineage cells,APPswe enhanced hepcidin expression by inducing ER stress and increasing OC formation,in part through hepcidin.Together,these results suggest that increased hepcidin expression in hepatocytes and OB lineage cells in Tg2576 mice contributes to enhanced osteoclastogenesis and trabecular bone loss,identifying the hepcidin-FPN-iron axis as a potential therapeutic target to prevent AD-associated bone loss. | Hao-Han Guo Lei Xiong Jin-Xiu Pan Daehoon Lee Kevin Liu Xiao Ren Bo Wang Xiao Yang Shun Cui Lin Mei Wen-Cheng Xiong | 2021 | Bone Research2021,9,3: | 2 |
