1Characterization and Evaluation of OsLCT1 and OsN ramp5 Mutants Generated Through CRISPR/Cas9-Mediated Mutagenesis for Breeding Low Cd Rice显示文摘To explore how rice(Oryza sativa L.) can be safely produced in Cd-polluted soil, OsLCT1 and OsNramp5 mutant lines were generated by CRISPR/Cas9-mediated mutagenesis. One of OsLCT1 mutant(lct1×1) and two of OsNramp5 mutants(nramp5×7 and nramp5×9) were evaluated for grain Cd accumulation and agronomic performances. In paddy field soil containing approximately 0.9 mg/kg Cd, lct1×1 grains contained approximately 40%(0.17 mg/kg) of the Cd concentration of the wild type parental line, less than the China National Food Safety Standard(0.20 mg/kg). Both OsNramp5 mutants showed low grain Cd accumulation(< 0.06 mg/kg) in the paddy(approximately 0.9 mg/kg Cd) or in pots in soil spiked with 2 mg/kg Cd. However, only nramp5×7 showed normal growth and yield, whereas the growth of nramp5×9 was severely impaired. 2Ultra-sensitive Nanoprobe Modified with Tumor Cell Membrane for UCL/MRI/PET Multimodality Precise Imaging of Triple-Negative Breast Cancer显示文摘Triple-negative breast cancer(TNBC)is a subtype of breast cancer in which the estrogen receptor and progesterone receptor are not expressed,and human epidermal growth factor receptor 2 is not amplified or overexpressed either,which make the clinical diagnosis and treatment very challenging.Molecular imaging can provide an effective way to diagnose TNBC.Upconversion nanoparticles(UCNPs),are a promising new generation of molecular imaging probes.However,UCNPs still need to be improved for tumor-targeting ability and biocompatibility.This study describes a novel probe based on cancer cell membrane-coated upconversion nanoparticles(CCm-UCNPs),owing to the low immunogenicity and homologous-targeting ability of cancer cell membranes,and modified multifunctional UCNPs.This probe exhibits excellent performance in breast cancer molecular classification and TNBC diagnosis through UCL/MRI/PET tri-modality imaging in vivo.By using this probe,MDA-MB-231 was successfully differentiated between MCF-7 tumor models in vivo.Based on the tumor imaging and molecular classification results,the probe is also expected to be modified for drug delivery in the future,contributing to the treatment of TNBC.The combination of nanoparticles with biomimetic cell membranes has the potential for multiple clinical applications.Hanyi Fang Mengting Li Qingyao Liu Yongkang Gai Lujie Yuan Sheng Wang Xiao Zhang Min Ye Yongxue Zhang Mingyuan Gao Yi Hou Xiaoli Lan 2020Nano-Micro Letters2020,12,5:5

4Distribution Features of the Nanhua-Sinian Rifts and their Significance to Hydrocarbon Accumulation in the Tarim Basin显示文摘On the basis of reprocessing 34 new two-dimensional spliced long sections(20,191 km) in the Tarim Basin, the deep structure features of the Tarim Basin were analyzed through interpreting 30,451 km of two-dimensional seismic data and compiling basic maps. Seismic interpretation and geological analysis conclude that the Nanhua-Sinian strata are a set of rift-depression depositional systems according to their tectonic and depositional features. The rift valley formed in the Nanhua Period, and the transformation became weaker during the late Sinian Period, which eventually turned into depression. From bottom to top, the deposited strata include mafic igneous, tillite, mudstone, and dolomite. Three major depocenters developed inside this basin during the rift stage and are distributed in the eastern Tarim Basin, the Awati area, and the southwestern Tarim Basin. Among them, the rift in the eastern Tarim Basin strikes in the near east-west direction on the plane and coincides with the aeromagnetic anomaly belt. This represents a strong magnetic zone formed by upwelling basic volcanic rock along high, steep normal faults of the Nanhua Period. Controlled by the tectonic background, two types of sedimentary systems were developed in the rift stage and depression stage, showing two types of sequence features in the Sinian depositional stage. The Nanhua System appears as a wedge-shaped formation, with its bottom in unconformable contact with the base. The rifting event has a strong influence on the current tectonic units in the Tarim Basin, and affects the distribution of source rock in the Yuertus Formation and reservoir beds in the Xiaoerbulake Formation in Lower Cambrian, as well as the gypseous cap rock in Middle Cambrian. The distribution features of the rifts have important and realistic significance for determining the direction of oil and gas exploration in the deep strata of the Tarim Basin. 10A comparison of in vitro anticancerous activity and mechanism of ethanolic extracts from different Ganoderma genus显示文摘Five ethanolic extracts from the mycelia of Ganoderma lucidum,G.tsugae,G.oerstedii,G.subamboinense,and G.resinaceum were respectively studied on their anticancerous activities against leukemic HL-60 cell line in vitro.Results showed that all five extracts potently inhibited HL-60 proliferation.The extract from G.lucidum mycelia exerted the highest activity.Annexin V/PI bivariate flow cytometric analysis further revealed that the five extracts significantly induced early apoptosis in HL-60 cells.The results illustrate that not only G.lucidum but also other Ganoderma species can inhibit cancer cells,and their mechanisms are related to induction of apoptosis.ZHOU Yueqin YANG Xiaotong LI Xuquan FENG Huiqin MI Ke YANG Qingyao 2006Frontiers in Biology2006,1,3:0
11Characterization and prediction of tailpipe ammonia emissions from in-use China 5/6 light-duty gasoline vehicles显示文摘On-road tailpipe ammonia (NH3) emissions contribute to urban secondary organic aerosol formation and have direct or indirect adverse impacts on the environment and human health. To understand the tailpipe NH3 emission characteristics, we performed comprehensive chassis dynamometer measurements of NH3 emission from two China 5 and two China 6 light-duty gasoline vehicles (LDGVs) equipped with three-way catalytic converters (TWCs). The results showed that the distance-based emission factors (EFs) were 12.72 ± 2.68 and 3.18 ± 1.37 mg/km for China 5 and China 6 LDGVs, respectively. Upgrades in emission standards were associated with a reduction in tailpipe NH3 emission. In addition, high NH3 EFs were observed during the engine warm-up period in cold-start cases owing to the intensive emissions of incomplete combustion products and suitable catalytic temperature in the TWCs. Notably, based on the instantaneous NH3 emission rate, distinct NH3–emitting events were detected under high/extra high velocity or rapid acceleration. Furthermore, NH3 emission rates correlated well with engine speed, vehicle specific power, and modified combustion efficiency, which were more easily accessible. These strong correlations were applied to reproduce NH3 emissions from China 5/6 LDGVs. The predicted NH3 EFs under different dynamometer and real-world cycles agreed well with existing measurement and prediction results, revealing that the NH3 EFs of LDGVs in urban routes were within 8.55–11.62 mg/km. 13VALORIZATION OF BIOGAS THROUGH SIMULTANEOUS CO_(2) AND H_(2)S REMOVAL BY RENEWABLE AQUEOUS AMMONIA SOLUTION IN MEMBRANE CONTACTOR显示文摘Upgrading biogas into biomethane not only improves the biogas utilization as vehicle fuel or natural gas substitute,but also reduces the greenhouse gases emissions.Considering the principle of engineering green energy process,the renewable aqueous ammonia(RAA)solution obtained from biogas slurry was used to remove H_(2)S and CO_(2) simultaneously in the hollow fiber membrane contactor.RAA was mimicked in this study using the ammonia aqueous solution mixed with some typical impurities including ethanol,acetic acid,propionic acid,butyric acid and NH4HCO_(3).Compared with the typical physical absorption(i.e.,pure water)removing 48%of H_(2)S from biogas,RAA with 0.1 mol·L^(−1) NH_(3) could remove 97%of H_(2)S.Increasing the NH3 concentration from 0.1 to 0.5 mol·L^(−1) could elevate the CO_(2) absorption flux from 0.97 to 1.72 mol·m^(−2)·h^(−1) by 77.3%.Among the impurities contained in RAA,ethanol has a less impact on CO_(2) absorption,while other impurities like CO_(2) and acetic acid have significant negative impacts on CO_(2) absorption.Fortunately,the impurities have a less influence on H_(2)S removal efficiency,with more than 98%of H_(2)S could be removed by RAA.Also,the influences of operating parameters on acid gases removal were investigated to provide some engineering suggestions.Tao SUN Wenlong LI Jiandong WEI Long JI Qingyao HE Shuiping YAN 2023Frontiers of Agricultural Science and Engineering2023,10,3:0
14Melatonin activates the OsbZIP79–OsABI5 module that orchestrates nitrogen and ROS homeostasis to alleviate nitrogen-limitation stress in rice显示文摘Melatonin(Mel)has previously been reported to effectively alleviate nitrogen-limitation(N-L)stress and thus increase nitrogen-use efficiency(NUE)in several plants,but the underlying mechanism remains obscure.Here,we revealed that OsbZIP79(BASIC LEUCINE ZIPPER 79)is transcriptionally activated under N-L conditions,and its expression is further enhanced by exogenous Mel.By the combined use of omics,genetics,and biological techniques,we revealed that the OsbZIP79–OsABI5(ABSCISIC ACID INSENSITIVE 5)module stimulated regulation of reactive oxygen species(ROS)homeostasis and the uptake and metabolismof nitrogen under conditions of indoor nitrogen limitation(1/16 normal level).OsbZIP79 activated the transcription of OsABI5,and OsABI5 then bound to the promoters of target genes,including genes involved in ROS homeostasis and nitrogen metabolism,activating their transcription.This module was also indispensable for upregulation of several other genes involved in abscisic acid catabolism,nitrogen uptake,and assimilation under N-L and Mel treatment,although these genes were not directly transactivated by OsABI5.Field experiments demonstrated that Mel significantly improved rice growth under low nitrogen(L-N,half the normal level)by the same mechanism revealed in the nitrogen-limitation study.Mel application produced a 28.6%yield increase under L-N and thus similar increases in NUE.Also,two OsbZIP79-overexpression lines grown in L-N field plots had significantly higher NUE(+13.7%and+21.2%)than their wild types.Together,our data show that an OsbZIP79–OsABI5 module regulates the rice response to N insufficiency(N limitation or low N),which is important for increasing NUE in rice production.Meng Jiang Yue Song Ruifang Yang Chenfan Zheng Yunchao Zheng Huali Zhang Shan Li Yuanyuan Tan Jianzhong Huang Qingyao Shu Ruiqing Li 2023Plant Communications2023,4,6:0
