ZIKA virus isolated from mosquitoes:a field and laboratory investigation in China,2016显示文摘A field investigation of arboviruses was conducted in Dejiang, Guizhou Province in the summer of 2016. A total of 8,795 mosquitoes, belonging to four species of three genera, and 1,300 midges were collected. The mosquito samples were identified on site according to their morphology, and the pooled samples were ground and centrifuged in the laboratory. The supernatant was incubated with mosquito tissue culture cells(C6/36) and mammalian cells(BHK-21) for virus isolation. The results indicated that 40%(3,540/8,795) were Anopheles sinensis, 30%(2,700/8,795) were Culex pipiens quinquefasciatus, and 29%(2,530/8,795)were Armigeres subbalbeatus. Furthermore, a total of eight virus isolates were obtained, and genome sequencing revealed two Zika viruses(ZIKVs) isolated from Culex pipiens quinquefasciatus and Armigeres subbalbeatus, respectively; three Japanese encephalitis viruses(JEVs) isolated from Culex pipiens quinquefasciatus; two Banna viruses(BAVs) isolated from Culex pipiens quinquefasciatus and Anopheles sinensis, respectively; and one densovirus(DNV) isolated from Culex pipiens quinquefasciatus. The ZIKVs isolated from the Culex pipiens quinquefasciatus and Armigeres subbalbeatus mosquitoes represent the first ZIKV isolates in China's Mainland. This discovery presents new challenges for the prevention and control of ZIKV in China, and prompts international cooperation on this global issue. | Shihong Fu Song Song Hong Liu Yuanyuan Li Xiaolong Li Xiaoyan Gao Ziqian Xu Guoping Liu Dingming Wang Zhenzao Tian Jingzhu Zhou Ying He Wenwen Lei Huanyu Wang Bin Wang Xiaoqing Lu Guodong Liang | 2017 | Science China(Life Sciences)2017,60,12: | 25
Genome sequencing and phylogenetic analysis of Banna virus(genus Seadornavirus, family Reoviridae)isolated from Culicoides显示文摘In an investigation of blood-sucking insects and arboviruses, a virus(YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of Yunnan Province, China. The virus caused cytopathic effect(CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA(dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region(1.12 segments) were17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 protein(RdRp) revealed that YN12243 belonged to genus Seadornavirus of family Reoviridae, and further analysis indicated that YN12243 belongs to the Banna virus(BAV) genotype A2. Additionally, YN12243 was located in the same evolutionary cluster as BAV strains isolated from different mosquito species, suggesting that the BAV isolated from Culicoides does not have species barriers. These results indicate that Culicoides can also be a vector for BAV. In view of the hematophagous habits of Culicoides on cattle, horses, deer, and other large animals, as well as the possibility of spreading and causing a variety of animal arboviral diseases, it is important to improve infection detection and monitor the BAV in large livestock. | Song Song Yuanyuan Li Shihong Fu Wenwen Lei Xiaofang Guo Yun Feng Xiaoyan Gao Xiaolong Li Zhonghua Yang Ziqian Xu Ying He Huanyu Wang Hongning Zhou Bin Wang Xiaoqing Lu Guodong Liang | 2017 | Science China(Life Sciences)2017,60,12: | 8
Effect of Bacillus amyloliquefaciens and Bacillus subtilis on fermentation, dynamics of bacterial community and their functional shifts of whole-plant corn silage显示文摘Background:Bacillus amyloliquefaciens(BA)and Bacillus subtilis(BS)are usually used as feed supplements directly or bacterial inoculants in biological feeds for animals. However, few research have reported the effects of BA and BS on fermentation characteristics and bacterial community successions of whole-plant corn silage during ensiling. If the BA and BS inoculants have positive effects on silages, then they could not only improve fermentation characteristics, but also deliver BA or BS viable cells to ruminants, which would play its probiotic effect. Therefore, the objectives of this study were to investigate the effects of BA and BS on the fermentation, chemical characteristics, bacterial community and their metabolic pathway of whole-plant corn silage. Results:Freshly chopped whole-plant corn was inoculated without or with BA and BS,respectively, and ensiled for1,3,7,14 and 60 d.Results showed that BA and BS inoculations increased lactic acid concentrations of whole-plant corn silages compared with control, and BA inoculation decreased acetic acid concentrations, whereas BS inoculation decreased fiber contents and increased crude protein(CP)content.Higher water-soluble carbohydrate contents and lower starch contents were observed in BA-and BS-inoculated silages compared with that in control. The decreased CP content and increased non-protein nitrogen content were observed in BA-inoculated silage, which was consistent with the higher amino acid metabolism abundances observed in BA-inoculated silage. In addition,it was noteworthy that BA and BS inoculations increased the metabolism of cofactors and vitamins, and decreased the relative abundances of drug resistance:antimicrobial pathways. We also found that the bacterial metabolism pathways were clearly separated into three clusters based on the ensiling times of whole-plant corn silage in the present study. There were no significant differences in bacterial community compositions among the three groups during ensiling. However,BA and BS inoculations decreased the relative abundances of undesirable bacteria such as Acetobacter and Acinetobacter. Conclusion:Our findings suggested that the BS strain was more suitable as silage inoculants than the BA strain in whole-plant corn silage in this study. | Jie Bai Marcia Franco Zitong Ding Lin Hao Wencan Ke Musen Wang Dongmei Xie Ziqian Li Yixin Zhang Lin Ai Xusheng Guo | 2022 | Journal of Animal Science and Biotechnology2022,13,3: | 5
Nuclear Aurora kinase A switches m^(6)A reader YTHDC1 to enhance an oncogenic RNA splicing of tumor suppressor RBM4显示文摘Aberrant RNA splicing produces alternative isoforms of genes to facilitate tumor progression, yet how this process is regulated by oncogenic signal remains largely unknown. Here,we unveil that non-canonical activation of nuclear AURKA promotes an oncogenic RNA splicing of tumor suppressor RBM4 directed by m^(6)A reader YTHDC1 in lung cancer.Nuclear translocation of AURKA is a prerequisite for RNA aberrant splicing, specifically triggering RBM4 splicing from the full isoform(RBM4-FL)to the short isoform(RBM4-S)in a kinase-independent manner. | SiSi Li YangFan Qi JiaChuan Yu YuChao Hao Bin He MengJuan Zhang ZhenWei Dai TongHui Jiang SuYi Li Fang Huang Ning Chen Jing Wang MengYing Yang DaPeng Liang Fan An JinYao Zhao WenJun Fan YuJia Pan ZiQian Deng YuanYuan Luo Tao Guo Fei Peng ZhiJie Hou ChunLi Wang FeiMeng Zheng LingZhi Xu Jie Xu QingPing Wen BiLian Jin Yang Wang Quentin Liu | 2022 | Signal Transduction and Targeted Therapy2022,7,5: | 4

CTGF/CCN2 promotes the proliferation of human osteosarcoma cells via cross-talking with the stromal CXCL12/CXCR4-AKT-αvβ3 signaling axis in tumor microenvironment显示文摘Osteosarcoma (OS) is the most common histological form of primary bone cancer in childhood cancer and young adults. At present, OS is widely investigated because of the interaction between the tumor and bone microenvironment and the effect of such interaction on OS progression and metastasis.1 The connective tissue growth factor (CTGF), also known as cellular communication network factor 2 (CCN2), is a secreted extracellular matrix-associated protein. CTGF is as active as the regulators of signaling activities of several different pathways and an orchestrator of their cross-talk.2 Therefore, we conducted experiments to investigate the effects of CTGF on OS tumor progress and the cross-talk with stromal cells in the tumor microenvironment. | Zhangxu Zhou Shujuan Yan Ruyi Zhang Hao Wang Ziqian Ye Zhilun Zhang Keyu Li Guowei Zuo | 2023 | Genes & Diseases2023,10,2: | 1
Recent Progress of Metal Organic Frameworks-Based Electrocatalysts for Hydrogen Evolution,Oxygen Evolution, and Oxygen Reduction Reaction显示文摘Exploring efficient and cost-saving electrocatalysts is essential to the renewable energy storage and utilization, which is still in its embryonic period.MOFs have drawn tremendous attention due to their adjustability, abundant active sites, and plentiful pores.Notably, satisfactory electrocatalytic performance has been achieved by MOFs-based electrocatalysts comparable to traditional electrocatalysts.State-of-the-art works about the MOFs-based electrocatalysts for hydrogen evolution reaction(HER),oxygen evolution reaction(OER),and ORR were summarized in this review. This review comprises a series of modifying strategies of MOFs and their derivatives, from aspects of structure, composition, and morphology.Furthermore, the active sites and functional mechanisms’recognition are involved in this review expecting to provide reference for rationally designing efficient electrocatalysts. At last, the current status, challenges, and perspectives of MOFs-based electrocatalysts are also discussed. | Yaling Jia Ziqian Xue Yinle Li Guangqin Li | 2022 | Energy & Environmental Materials2022,5,4: | 1
Engineering a folic acid-decorated ultrasmall gemcitabine nanocarrier for breast cancer therapy: Dual targeting of tumor cells and tumor-associated macrophages显示文摘Combination of passive targeting with active targeting is a promising approach to improve the therapeutic efficacy of nanotherapy. However, most reported polymeric systems have sizes above100 nm,which limits effective extravasation into tumors that are poorly vascularized and have dense stroma. This will,in turn, limit the overall effectiveness of the subsequent uptake by tumor cells via active targeting. In this study,we combined the passive targeting via ultra-small-sized gemcitabine(GEM)-based nanoparticles(NPs)with the active targeting provided by folic acid(FA)conjugation for enhanced dual targeted delivery to tumor cells and tumor-associated macrophages(TAMs).We developed an FAmodified prodrug carrier based on GEM(PGEM)to load doxorubicin(DOX),for co-delivery of GEM and DOX to tumors. The co-delivery system showed small particle size of~10 nm in diameter. The ligand-free and FA-targeted micelles showed comparable drug loading efficiency and a sustained DOX release profile. The FA-conjugated micelles effectively increased DOX uptake in cultured KB cancer cells that express a high level of folate receptor(FR),but no obvious increase was observed in 4T1.2 breast cancer cells that have a low-level expression of FR.Interestingly,in vivo, systemic delivery of FAPGEM/DOX led to enhanced accumulation of the NPs in tumor and drastic reduction of tumor growth in a murine 4T1.2 breast cancer model.Mechanistic study showed that 4T1.2 tumor grown in mice expressed a significantly higher level of FOLR2,which was selectively expressed on TAMs.Thus, targeting of TAM may also contribute to the improved in vivo targeted delivery and therapeutic efficacy. | Pearl Moharil Zhuoya Wan Apurva Pardeshi Jiang Li Haozhe Huang Zhangyi Luo Sanjay Rathod Ziqian Zhang Yuang Chen Bei Zhang Christian A.Fernandez Jingjing Sun Song Li | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 1
Performance analysis of full-duplex densely distributed MIMO with wireless backhaul显示文摘In this paper, the spectral efficiency of a full-duplex(FD)densely distributed multi-input multioutput(MIMO)system with wireless backhaul is considered.In-band full-duplex(IBFD)backhauling,in which the backhaul and access transmissions take place on the same spectrum,is exploited for wireless backhauling to enable an efficient spectrum reuse. However, the severe cross-tier interference and crosslink interference reduce the gains of IBFD backhauling. To evaluate the achievable spectral efficiency with imperfect channel state information(CSI),we propose a two-phase channel estimation scheme to estimate the CSI for two wireless links, and the scheme estimates an effective interference CSI between access points(APs)based on beamforming training to perform interference cancelation at APs.Given the estimated CSI,the closed-form expressions of the uplink and downlink achievable rates with maximum ratio transmission beamforming and maximum ratio combining receivers, respectively, are derived with Gamma approximation.Numerical results verify the accuracy of the derived closed-form expressions and the effectiveness of the twophase channel estimation scheme for interference cancelation. Moreover, compared with half-duplex densely distributed MIMO systems,FD systems with interference cancelation have a better performance. | Ziqian WAN Qijun PAN Jiamin LI Pengcheng ZHU Dongming WANG | 2023 | Science China(Information Sciences)2023,66,6: | 1
Cannabis suppresses antitumor immunity by inhibiting JAK/STAT signaling in T cells through CNR2显示文摘The combination of immune checkpoint blockade(ICB)with chemotherapy significantly improves clinical benefit of cancer treatment.Since chemotherapy is often associated with adverse events, concomitant treatment with drugs managing side effects of chemotherapy is frequently used in the combination therapy. However, whether these ancillary drugs could impede immunotherapy remains unknown. Here,we showed that∆9-tetrahydrocannabinol(THC),the key ingredient of drugs approved for the treatment of chemotherapy-caused nausea, reduced the therapeutic effect of PD-1 blockade. The endogenous cannabinoid anandamide(AEA)also impeded antitumor immunity, indicating an immunosuppressive role of the endogenous cannabinoid system(ECS). | Xinxin Xiong Siyu Chen Jianfei Shen Hua You Han Yang Chao Yan Ziqian Fang Jianeng Zhang Xiuyu Cai Xingjun Dong Tiebang Kang Wende Li Penghui Zhou | 2022 | Signal Transduction and Targeted Therapy2022,7,5: | 1
The inhomogeneous diffusion of chemically crosslinked polyacrylamide hydrogel based on poroviscosity theory显示文摘The diffusion of hydrogels is a phenomenon not only profound in novel applications of mechanical engineering but also very common in nature. Comprehensive studies of the swelling properties under stable states have been carried out in the past several years;however, ambiguities in the understanding of the kinetic behaviour of the diffusion phenomenon of hydrogels still remain. The potential applications of hydrogels are confined due to the lack of perceptiveness of the kinetic behaviour of diffusion in hydrogels. Based on our previous work, in this study, we initiate the theoretical kinetic study of the inhomogeneous diffusion of hydrogels. With poroviscosity introduced, we develop a theory for the inhomogeneous diffusion of hydrogels. After implementing this theory into the finite element solution, we could predict the water content in the hydrogel as a function of time and location. The quantitative prediction of the inhomogeneous diffusion and the formulas are given in the numerical study.Furthermore, the corresponding experiments are carried out to substantiate this theory. It can be observed that the theoretical prediction meets fairly well with our experimental data. Finally, we carry out a systematic parameter study to discuss the effect of three important parameters on the diffusion property. The increase of the interaction parameter is seen to constrain the diffusion while increase of the chemical potential is seen to facilitate the process. The change of the diffusion coefficient D, on the other hand, does not affect the process much. By comparing the conclusions above with the experimental data, we can narrow down the range of the values of χ and D. | ZHENG ShouJing LI ZiQian LIU ZiShun | 2019 | Science China(Technological Sciences)2019,62,8: | 1
Recent advances of hydrogel network models for studies on mechanical behaviors显示文摘Current constitutive theories face challenges when predicting the extremely large deformation and fracture of hydrogels, which calls for the demands to reveal the fundamental mechanism of the various mechanical behaviors of hydrogels from bottom up.Proper hydrogel network model provides a better approach to bridge the gap between the micro-structure and the macroscopic mechanical responses. This work summarizes the theoretical and numerical researches on the hydrogel network models, aiming to provide new insights into the effect of microstructure on the swelling-deswelling process, hyperelasticity, viscoelasticity and fracture of hydrogels.Hydrogel network models are divided into full-atom network models, realistic network models and abstract network models.Full-atom network models have detailed atomic structure but small size.Realistic network models with different coarse-graining degree have large model size to explain the swelling-deswelling process, hyperelasticity and viscoelasticity.Abstract network models abstract polymer chains into analytical interactions, leading to the great leap of model size.It shows advantages to reproduce the crack initiation and propagation in hydrogels by simulating chain scission.Further research directions on the network modeling are suggested. We hope this work can help integrate the merits of network modeling methods and continuum mechanics to capture the various mechanical behaviors of hydrogels. | Jincheng Lei Ziqian Li Shuai Xu Zishun Liu | 2021 | Acta Mechanica Sinica2021,37,3: | 1
The genomic characteristics and pathogenicity of a mammalian orthoreovirus within a new lineage from wild pika in plateau显示文摘Emerging and re-emerging viruses from wild animals have seriously threatened the health of humans and domesticated animals in recent years.Herein,we isolated a new mammalian orthoreovirus(MRV),Pika/MRV/GCCDC7/2019(PMRV-GCCDC7),in the Qinghai-Tibet Plateau wild pika(Ochotona curzoniae).Though the PMRVGCCDC7 shows features of a typical reovirus with ten gene segments arranged in 3:3:4 in length, the virus belongs to an independent evolutionary branch compared to other MRVs based on phylogenetic tree analysis. The results of cellular susceptibility, species tropism, and replication kinetics of PMRV-GCCDC7 indicated the virus could infect four human cell lines(A549,Huh7,HCT,and LoVo)and six non-human cell lines, including Vero-E6,LLCMK2,BHK-21,N2a,MDCK,and RfKT cell, derived from diverse mammals,i.e.monkey,mice,canine and bat, which revealed the potential of PMRV-GCCDC7 to infect a variety of hosts.Infection of BALB/c mice with PMRVGCCDC7 via intranasal inoculation led to relative weight loss, lung tissue damage and inflammation with the increase of virus titer, but no serious respiratory symptoms and death occurred. The characterization of the new reovirus from a plateau-based wild animal has expanded our knowledge of the host range of MRV and provided insight into its risk of trans-species transmission and zoonotic diseases. | Kexin Zong Yuanyuan Guo Jingdong Song Maoshun Liu Junfeng Hao Jie Zhang Xin Li Shiyan Zhu Shuting Huo Ziqian Xu Peipei Liu Yingze Zhao Yuhai Bi Jiapeng Qu George F.Gao Jun Liu | 2023 | Virologica Sinica2023,38,6: | 0

Boosted electrochemical performance of Na_(3)V_(2)(PO_(4))_(3) at low temperature through synergistical F substitution and construction of interconnected nitrogen-doped carbonaceous network显示文摘Benefitting from its unique NASICON-type framework, the Na_(3)V_(2)(PO_(4))_(3)(NVP)cathodes have aroused extensive interest and have been deemed as the promising cathode candidate for sodium-ion batteries(SIBs).Unfortunately, the poor electronic conductivity, combined with the undesirable volume variations, seriously hinders the practical application of NVP cathode, especially at low temperatures.Herein,a dual-strategy,F substitution accompanied by V vacancies and the construction of three-dimensional(3D)nitrogen-doped carbonaceous frameworks(NC),were employed for the NVP cathode(F-NVP/C@3DNC).The former can remarkably decrease the particle size and enhance Na^(+)migration capability, increasing the ionic conductivity.Meanwhile, the electronic connection and effective buffering can be obtained from the latter, strengthening the electrode integrity.Consequently,up to 100 cycles at 0.1 A g^(-1),a reversible capacity of 113.8 mAh g^(-1),approaching the theoretical value(117 mAh g^(-1)),is demonstrated,accompa-nied by impressive capacity retentions at 1.0(93.75%after 4800 cycles)and 20.0 A g^(-1)(92.7%after 1000 cycles).More importantly, even at-20℃,a superior specific capacity(102.6 mAh g^(-1) after 100 cycles at 0.1 A g^(-1))and high capacity retention(86.6%at 20.0 A g^(-1) up to 1000 cycles)can still be obtained simul-taneously.Significantly,the design of F-NVP/C@3DNC provides insights for the fabrication of polyanion cathodes for applications at low temperatures with modified structure stability and reaction kinetics. | Jiabao Li Ziqian Li Shaocong Tang Tianyi Wang Likun Pan Chengyin Wang | 2023 | Journal of Materials Science & Technology2023,,19: | 0
