Human monoclonal antibodies block the binding of SARS-CoV-2 spike protein to angiotensin converting enzyme 2 receptor显示文摘According to the World Health Organization(WHO)newly updated situation report on March 18th,2020,the coronavirus disease 2019(COVID-19)pandemic has confirmed 191,127 cases and claimed 7807 deaths worldwide.1 The etiological agent of COVID-19 has been identified as a novel coronavirus, the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),belonging to Sarbecovirus subgenus(genus Betacoronavirus, family Coronaviridae)and showing 79.6 and 96.2%sequence identity in nucleotide to SARS-CoV and a bat coronavirus(BatCoV RaTG13),respectively.2–4 Like SARS-CoV infection,a substantial fraction of COVID-19 patients exhibits severe respiratory symptoms and has to be hospitalized in intensive care unit.5–8 Although the mortality rate of COVID-19 is significantly lower than that of SARS-CoV infection,SARS-CoV-2 shows much higher human-to-human transmission rate, rapidly leading to a global pandemic declared by WHO on March 11th,2020. | Xiangyu Chen Ren Li Zhiwei Pan Chunfang Qian Yang Yang Renrong You Jing Zhao Pinghuang Liu Leiqiong Gao Zhirong Li Qizhao Huang Lifan Xu Jianfang Tang Qin Tian Wei Yao Li Hu Xiaofeng Yan Xinyuan Zhou Yuzhang Wu Kai Deng Zheng Zhang Zhaohui Qian Yaokai Chen Lilin Ye | 2020 | Cellular & Molecular Immunology2020,17,6: | 17
Impedance Modeling and Stability Analysis of PV Grid-connected Inverter Systems Considering Frequency Coupling显示文摘Impedance analysis is an effective method to analyze the oscillation issue associated with grid-connected photovoltaic systems. However, the existing impedance modeling of a gridconnected photovoltaic inverter usually only considers the effect of a single perturbation frequency, ignoring the coupling frequency response between the internal control loops of a grid-connected inverter, which severely affects the accuracy of the stability analysis.Hence,a method of impedance modeling and stability analysis for grid-connected photovoltaic inverters considering cross-coupling frequency is proposed in this paper.First, the generation mechanism of frequency coupling in gridconnected photovoltaic inverters, and the relationship between the coupling frequency and perturbation frequency are analyzed.Secondly,a sequence impedance model of grid-connected photovoltaic systems considering the coupling frequency is established by using the harmonic linearization method. The impact of DC bus voltage control strategy on frequency coupling characteristics of a grid-connected photovoltaic system is evaluated, and the impact of a coupling frequency term on system stability is quantitatively analyzed. Finally, the advantages of the proposed method are verified by several simulations. The results show that the proposed impedance model can accurately predict the potential resonance points of the system, and the coupling frequency characteristics will become much stronger with smaller DC bus capacitance or larger bandwidth of the DC bus controller. | Shaojian Song Ze Wei Yuzhang Lin Bin Liu Hui Liu | 2020 | CSEE Journal of Power and Energy Systems2020,6,2: | 8
Aggregation-induced Emission-active Hyperbranched Poly (tetrahydropyrimidine)s Synthesized from Multicomponent Tandem Polymerization显示文摘Hyperbranched polymer with highly branched three-dimensional topological structure, a large number of end groups, and multifaceted functionalities have gained much attention, while polymers with aggregation-induced emission(AIE) properties become a group of popular luminescent materials recently. The design and synthesis of AIE-active hyperbranched polymers, which combine the advantages of these two types of materials, are attractive but challenging. In this work, four hyperbranched poly(tetrahydropyrimidine)s were synthesized from the metal-free room temperature multicomponent tandem polymerization of diester group-activated internal alkyne, polyfunctional aromatic amines, and formaldehyde in methanol under the catalysis of acetic acid. Through different monomer combination and controlling the monomer loading order, hyperbranched polymers with various topological structures as well as sequences of different functional groups in the polymer backbone were obtained with high molecular weights(up to 3.0 × 10~4 g/mol) in high yields(up to 98%). The hyperbranched poly(tetrahydropyrimidine) emitted faintly in solution, while its luminescence was notably enhanced in the aggregated state, suggesting its typical aggregation-induced emission property. It is anticipated that the multicomponent polymerization may provide a synthetic platform for the construction of hyperbranched polyheterocycles with diverse structures and functionalities. | Yuzhang Huang Ping Chen Bo Wei Rongrong Hu Ben Zhong Tang | 2019 | Chinese Journal of Polymer Science2019,37,4: | 6

CD49a^(+)CD49b^(+) NK cells induced by viral infection reflect an activated state of conventional NK cells显示文摘Natural killer(NK) cells are important innate effectors that play a pivotal role in the defense against tumors and infections and participate in regulating adaptive immunity. Recent studies have revealed phenotypic and functional heterogeneity of NK cells. Here, using murine models of acute and chronic lymphocytic choriomeningitis virus infection, we observed that a CD49a^(+)CD49b^(+) NK cell subset emerged in the liver and other tissues, and underwent vigorous expansion following viral infection, before progressively decreasing in cell number. These viral infection-induced CD49a^(+)CD49b^(+) NK cells displayed an activated and mature phenotype. Moreover, compared with liver-resident NK cells and conventional NK(cNK) cells, CD49a^(+)CD49b^(+) NK cells showed increased functional competence, as evidenced by higher amounts of IFN-γ production and stronger cytotoxic capabilities during viral infection. Generation of these CD49a^(+)CD49b^(+) NK cells was shown to be independent of the T-bet transcription factor. Adoptive transfer experiments revealed that c NK cells could convert into CD49a^(+)CD49b^(+) NK cells following viral infection. Collectively, these results suggest that viral infection-induced CD49a^(+)CD49b^(+) NK cells represent a transiently activated state of cNK cells. | Wenhan Li Jing Zhou Xianwei Wang Yuzhang Wu Lilin Ye Haiming Wei Rui Sun Zhigang Tian Hui Peng | 2020 | Science China(Life Sciences)2020,63,11: | 2

A Holistic State Estimation Framework for Active Distribution Network with Battery Energy Storage System显示文摘Battery energy storage systems(BESSs)are expected to play a crucial role in the operation and control of active distribution networks(ADNs).In this paper,a holistic state estimation framework is developed for ADNs with BESSs integrated.A dynamic equivalent model of BESS is developed, and the state transition and measurement equations are derived.Based on the equivalence between the correction stage of the iterated extended Kalman filter(IEKF)and the weighted least squares(WLS)regression,a holistic state estimation framework is proposed to capture the static state variables of ADNs and the dynamic state variables of BESSs,especially the state of charge(SOC).A bad data processing method is also presented. The simulation results show that the proposed holistic state estimation framework improves the accuracy of state estimation as well as the capability of bad data detection for both ADNs and BESSs,providing comprehensive situational awareness for the whole system. | Shaojian Song Huangjiao Wei Yuzhang Lin Cheng Wang Antonio Gómez-Expósito | 2022 | Journal of Modern Power Systems and Clean Energy2022,10,3: | 1

The lncRNA Snhgl-Vpsl3D vesicle trafficking system promotes memory CD8 T cell establishment via regulating the dual effects of IL-7 signaling显示文摘The efficient induction and long-term persistence of pathogen-specific memory CD8 T cells are pivotal to rapidly curb the reinfection.Recent studies indicated that long-noncoding RNAs expression is highly cell-and stage-specific during T cell development and differentiation, suggesting their potential roles in T cell programs. However, the key lncRNAs playing crucial roles in memory CD8 T cell establishment remain to be clarified.Through CD8 T cell subsets profiling of lncRNAs,this study found a key lncRNA-Snhgl with the conserved naivehl-effectorlo-memoryh,expression pattern in CD8 T cells of both mice and human, that can promote memory formation while impeding effector CD8 in acute viral infection.Further,Snhgl was found interacting with the conserved vesicle trafficking protein Vps13D to promote IL-7Ra membrane location specifically. With the deep mechanism probing, the results show Snhgl-Vps13D regulated IL-7 signaling with its dual effects in memory CD8 generation, which not just because of the sustaining role of STAT5-BCL-2 axis for memory survival, but more through the STAT3-TCF1-Blimp1 axis for transcriptional launch program of memory differentiation. Moreover,we performed further study with finding a similar high-low-high expression pattern of human SNHG1A/PS13D/IL7R/TCF7 in CD8 T cell subsets from PBMC samples of the convalescent COVID-19 patients. The central role of Snhgl-Vps13D-IL-7R-TCF1 axis in memory CD8 establishment makes it a potential target for improving the vaccination effects to control the ongoing pandemic. | Yanyan Zhang Baohua Li Qiang Bai Pengcheng Wang Gang Wei Zhirong Li Li Hu Qin Tian Jing Zhou Qizhao Huang Zhiming Wang Shuai Yue Jialin Wu Liuqing Yang Xinyuan Zhou Lubin Jiang Ting Ni Lilin Ye Yuzhang Wu | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 0

Screening of HLA-A2 restricted CTL epitopes using molecular simulation显示文摘A new technology has been established to screen CTL epitope by using molecular simulation and mo-lecular dynamics. By using this technology, four peptides of MAGE-2 were screened and further identified by peptide binding assay and cytotoxic assay. Results showed that three candidate peptides had higher affinity to HLA-A2 molecule and were able to pulse CTL activities in vitro in human PBMC. The results indicated that identification of CTL epi-topes with computer assistant could overcome the compli-cated experimental steps, particularly the peptide elution step, and could directly observe peptide-MHC-Ⅰ compound binding pattern and could be used to screen CTL epitopes quickly. | GENG Miao WU Yuzhang LIN Zhihua JIA Zhengcai ZHOU Wei ZOU Liyun | 2003 | Chinese Science Bulletin2003,48,18: | 0
Single cell metabolic phenome and genome via the ramanome technology platform:Precision medicine of infectious diseases at the ultimate precision?显示文摘Due to the limitations of existing approaches,a rapid, sensitive, accurate, comprehensive, and generally applicable strategy to diagnose and treat bacterial and fungal infections remains a major challenge. Here, based on the ramanome technology platform,we propose a culture‐free, one cell resolution, phenome‐genome‐combined strategy called single‐cell identification, viability and vitality tests and source tracking(SCIVVS).For each cell directly extracted from a clinical specimen, the fingerprint region of the D2O‐probed single cell Raman spectrum(SCRS)enables species‐level identification based on a reference SCRS database of pathogen species, whereas the C‐D band accurately quantifies viability, metabolic vitality, phenotypic susceptibility to antimicrobials, and their intercellular heterogeneity. Moreover,to source track a cell,Raman‐activated cell sorting followed by sequencing or cultivation proceeds, producinging an indexed, high coverage genome assembly or a pure culture from precisely one pathogenic cell. Finally,an integrated SCIVVS workflow that features automated profiling and sorting of metabolic and morphological phenomes can complete the entire process in only a few hours.Because it resolves heterogeneity for both the metabolic phenome and genome, targets functions, can be automated, and is orders‐of‐magnitude faster while cost‐effective,SCIVVS is a new technological and data framework to diagnose and treat bacterial and fungal infections in various clinical and disease control settings. | Jian Xu Jianzhong Zhang Yingchun Xu Yi‐Wei Tang Bo Ma Yuzhang Wu | 2023 | iLABMED2023,1,1: | 0
