| 3 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 5 | Application of gemstone spectral imaging for efficacy evaluation in hepatocellular carcinoma after transarterial chemoembolization显示文摘AIM: To assess the value of gemstone spectral imaging(GSI) in efficacy evaluation in hepatocellular cancer(HCC) after transcatheter arterial chemoembolization(TACE) treatment.METHODS: Thirty patients with HCC underwent GSI, including nonenhanced, arterial, portalvenous and delayed phase scans, after TACE treatment. Arterial phase images were acquired with GSI for reconstruction of virtual nonenhanced images and color overlay images. Digital subtraction angiography(DSA) was performed in all these patients. Two blinded and independent readers evaluated the data in two reading sessions; standard nonenhanced, arterial, portalvenous, and delayed phase images were read in session A, and the optimal monochromatic images, iodine/water based images and spectrum features were read in session B. Sensitivity and specificity were calculated with the DSA data as the reference standard. The sensitivity and specificity were compared using the χ2 test.RESULTS: DSA revealed 154 lesions in 30 patients, and 100 of them had blood supply. Overall sensitivity and specificity were 72%( 72/100) and 77.8 %(42/54) for session A, and 97%(97/100) and 94.4%(51/54) for session B, respectively. The sensitivity and specificity of the two reading sessions were significantly different(χ2 = 23.04, χ2 = 7.11, P < 0.05).CONCLUSION: Compared with conventional CT, GSI could significantly improve the detection of small and multiple lesions without increasing the radiation dose. | 8 | 中国5地无偿献血者HCV流行特征分析显示文摘目的了解我国献血者丙型肝炎病毒(HCV)的初筛阳性率和确证阳性率。方法采用2种不同ELA试剂对2013年6月-2016年12月我国5家血液中心/中心血站献血者血液标本进行ALT、HBsAg、抗-HCV、抗-HIV、抗-TP进行常规筛查,抗-HCV单试剂或双试剂筛查阳性标本进一步采用MP HCV Blot 3. 0进行确证检测。分别估算首次献血者的HCV流行率及重复献血者的HCV发病率,并采用χ~2检验比较不同人群体、不同献血类型之间HCV确证阳性率。结果在1 276 544人次献血样本中,有3 855 (0. 30%)为抗-HCV ELISA初筛反应性,其中只有30. 18%(904/2 995,)确证为抗-HCV阳性。本次研究中,首次献血者总计648607人次,其中1080份样本确证为抗-HCV阳性,即首次献血者HCV流行率为166. 56 (156. 04—177. 08)/100 000人次,年龄、受教育水平、职业均与HCV确证阳性率相关。重复献血者总计627 937人次,其中126份样本确证为抗-HCV阳性,即重复献血者HCV发病率为13. 04(9. 55—17. 81)/100 000人次,其中重复献血者抗-HCV确证阳性率明显高于单采血小板献血者(0. 02%vs0. 01%)。结论本研究表明我国献血者HCV仍处于较高水平流行状态。尽管现在NAT已成为我国血液检测的常规检测项,但仍需持续有效地对我国输血HCV传播风险进行监测和控制。同时,新的规范中规定可采用1种或2种EIA进行筛查,本意是为了提高检测敏感性,但本研究中EIA筛查阳性样本如此低的确证阳性率提示其特异性差,可能会造成许多合格的献血者丢失。因此,优化献血者HCV筛查策略将有助于防止血液浪费。 | 付萍 Liao Dan 吕运来 张洪斌 何伟兰 黄梅 文秀琼 马红丽 何涛 柯玲 吴秉婷 刘鱼 何苗 Liu Jing Don Brambill 王憬惺 Paul Ness Shan Hua | 2018 | 中国输血杂志2018,31,9: | 10 |
| 9 | Interferon-c and interleukin-4 detected in serum and saliva from patients with oral lichen planus显示文摘Our previous salivary study had demonstrated an apparent T helper 2(Th2)-predominance in saliva of oral lichen planus(OLP) patients and suggested a potential of salivary interleukin-4(IL-4) as a biomarker for monitoring disease severity. To further determine the consistency of Th1/Th2 bias of OLP, this study investigated the expression profile of interferon-c(IFN-c) and IL-4 in serum and the relationship of the serum levels of these cytokines with their saliva partners. Sixty ethnic Chinese patients with OLP(40 of the erythematous/ulcerative form and 20 of the reticular form) were recruited for this study, with 40 age–sex-matched healthy volunteers as control group. IFN-c and IL-4 levels in serum and paired saliva samples were screened by enzyme-linked immunosorbent assay. OLP patient showed a low-level IFN-c but high-level IL-4 expression profile in both serum and saliva, with a lower IFN-c/IL-4 ratio. Serum IL-4 level in the erythematous/ulcerative group was significantly higher than that in the reticular group. Serum levels of IFN-c and IL-4 were significantly and positively correlated with their saliva partners. | 10 | Relationship between pre-TIPS hepatic hemodynamics and postoperative incidence of hepatic encephalopathy显示文摘BACKGROUND: Hepatic encephalopathy (HE) is one of the complications that have limited the effectiveness of transjugular intrahepatic portosystemic shunt (TIPS) most significantly. Up to the present, the predicting factors of HE post-TIPS have been debated controversially. This study was undertaken to verify the relationship between pre-TIPS intrahepatic hemodynamics and the incidence of post-TIPS HE. METHODS: The hepatic blood dynamics was evaluated in 41 patients with liver cirrhosis before TIPS and at one month after TIPS by ultrasonography. The patients were divided into two groups according to Doppler findings before TIPS: group 1, patients with prograde portal flow, and group 2, patients with hepatofugal or back-forth portal flow. The clinical characteristics (age, sex, etiology of liver disease, pre-TIPS Child-Pugh score, incidence of pre-TIPS HE, and portacaval pressure gradient), incidence of post TIPS HE, and pre-/post-TIPS hepatic arterial resistant index (RI) in the two groups were compared. The independent prognostic value of pre-TIPS variables for the onset of HE after TIPS, including age, Child-Pugh score, presence of HE before TIPS, and the pattern of portal flow, was tested with a multiple-factor regression analysis. RESULTS: No significant difference in age, etiology of liver disease, indications of TIPS placement, incidence of HE before TIPS, and portacaval gradient before and after TIPS was observed between the two groups; but liver failure was more severe in group 2 (P<0.05). The incidence of post-TIPS HE in group 2 was significantly lower than that in group 1 (P<0.01). Pre-TIPS, the RI of the hepatic artery in group 1 was significantly higher than that in group 2 (P<0.01). However, TIPS induced a significantly decreased RI in group 1 (P <0.01), but not in group 2. Multiple-factor regression analysis demonstrated that the pattern of portal flow before TIPS was closely associated with the onset of post TIPS HE. CONCLUSIONS: Pre-TIPS intrahepatic hetnodynamics is closely related to the incidence of post-TIPS HE. Hepatic hetnodynamics of patients with hepatofugal portal blood flow only changes a little after TIPS and still provides compensatory blood supply of the hepatic artery, and the hepatic function is less affected. Hence HE is unlikely. Hepatic hemodynamics of patients with prograde portal blood flow changes a lot after TIPS, and dual blood supply of the portal vein and hepatic artery changes into compensatory blood supply of the hepatic artery, and hepatic function suffers greatly in a short time. | 11 | Macrophage migration inhibitory factor as a potential prognostic factor in gastric cancer显示文摘AIM:To investigate macrophage migration inhibitory factor(MIF) expression and its clinical relevance in gastric cancer,and effects of MIF knockdown on proliferation of gastric cancer cells. METHODS:Tissue microarray containing 117 samples of gastric cancer and adjacent non-cancer normal tissues was studied for MIF expression by immunohistochemistry(IHC) semiquantitatively,and the association of MIF expression with clinical parameters was analyzed. MIF expression in gastric cancer cell lines was detected by reverse transcriptionpolymerase chain reaction(RT-PCR) and Western blot. Two pairs of si RNA targeting the MIF gene(MIF si-1 and MIF si-2) and one pair of scrambled si RNA as a negative control(NC) were designed and chemically synthesized. All si RNAs were transiently transfected in AGS cells with OligofectamineTM to knock down the MIF expression,with the NC group and mock group(OligofectamineTM alone) as controls. At 24,48,and 72 h after transfection,MIF m RNA was analyzed by RTPCR,and MIF and proliferating cell nuclear antigen(PCNA) proteins were detected by Western blot.The proliferative rate of AGS cells was assessed by methylthiazolyl tetrazolium(MTT) assay and colony forming assay.RESULTS:The tissue microarray was informative for IHC staining,in which the MIF expression in gastric cancer tissues was higher than that in adjacent noncancer normal tissues(P < 0.001),and high level of MIF was related to poor tumor differentiation,advanced T stage,advanced tumor stage,lymph node metastasis,and poor patient survival(P < 0.05 for all). After si RNA transfection,MIF m RNA was measured by real-time PCR,and MIF protein and PCNA were assessed by Western blot analysis. We found that compared to the NC group and mock group,MIF expression was knocked down successfully in gastric cancer cells,and PCNA expression was downregulated with MIF knockdown as well. | 13 | Honokiol:a promising small molecular weight natural agent for the growth inhibition of oral squamous cell carcinoma cells显示文摘Honokiol (HNK) is a small organic molecule purified from magnolia species and has demonstrated anticancer activities in a variety of cancer cell lines; however, its effect on oral squamous cell carcinoma (OSCC) cells is unknown. We investigated the antitumor activities of HNK on OSCC cells in vitro for the first time. The inhibitory effects of HNK on the growth and proliferation of OSCC cells were demonstrated via in vitro 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and propidium iodide (PI) assays, and the apoptotic cells were investigated by the observation of morphological changes and detection of DNA fragmentation via PI, TdT-mediated dUTP-biotin nick end labeling (TUNEL), and DNA ladder assays, as well as flow cytometry assay. The results showed that HNK inhibited the growth and proliferation of OSCC cells in vitro in a time and dose-dependent manner. The inhibitory effect was associated with the cell apoptosis induced by HNK, evidenced by the morphological features of apoptotic cells, TUNEL-positive cells and a degradation of chromosomal DNA into small internucleosomal fragments. The study also demonstrated here that the inhibition or apoptosis mediated by 15 μg·mL-1 or 20 μg·mL-1 of HNK were more stronger compared with those of 20 μg·mL-1 5-fluorouracil (5-Fu, the control) applied to OSCC cells, when the ratio of OSCC cell numbers were measured between the treatment of different concentrations of HNK to the 5-Fu treatment for 48 h. | 15 | Enhanced Desorption of PAHs from Manufactured Gas Plant Soils Using Different Types of Surfactants显示文摘Surfactant enhanced remediation is thought to be an effective method for the remediation of soils polluted with hydrophobic organic compounds. Desorption of polycyclic aromatic hydrocarbons(PAHs) from an abandoned manufactured gas plant(MGP) soil was evaluated using four eluting agents including Triton X-100(TX100),sodium dodecylbenzene sulfonate(SDBS),rhamnolipid water solution(RWS) and rhamnolipid fermentation broth(RFB). The weight solubilization ratios for acenaphthene and fluorene were in the order of TX100 > SDBS > RWS > RFB. The Sm value,which indicates the maximum amounts of surfactants adsorbed in the soil,was in the order of RWS > RFB > SDBS > TX100. By using 8 g L-1of TX100,SDBS and RWS and 100% of RFB,the T-PAHs removal for the MGP soil contaminated with 207.86 mg T-PAHs kg-1 dry soil was 48.0%,45.7%,1.9%,and 8.6%,respectively,while that decreased to 41.6%,37%,0.38%,and 1.3% for the soil contaminated with 3 494.78 mg T-PAHs kg-1 dry soil. Only 8 g L-1 TX100 could remove all types of the 16 PAHs partly in the MGP soil,and the removal efficiencies of different PAHs ranged from 13% to 77.8%. | 18 | Phaeocystis globosa Bloom Monitoring: Based on P. globosa Induced Seawater Viscosity Modification Adjacent to a Nuclear Power Plant in Qinzhou Bay, China显示文摘The mucus produced by the outbreak of Phaeocystis globosa in the adjacent waters of the Fangchenggang Nuclear Power Plant(FCGNPP) in China has blocked the entrance of the cooling water filtration system of the FCGNPP, and posed a threat to the safe operation of the FCGNPP. At present, there is no related research on whether the changes in seawater viscosity could be used as a new method for monitoring and providing early warning of P. globosa tide. During a complete red tide cycle, the temporal and spatial changes in the hydrological conditions(temperature, salinity, dissolved oxygen), chlorophyll-a(Chl-a), composition and abundance of phytoplankton, number and size of P. globose colonies, concentration of transparent exopolymer particles(TEP) and the seawater viscosity were measured in this study. The results indicate that there was an extremely significant negative correlation between the physical seawater viscosity η_(T,S) and temperature, and the correlation coefficient reached-0.998. The biological seawater viscosity η_(Bio) was positively correlated with the number of P. globosa colonies, and the correlation coefficients is 0.747. Because the increase in phytoplankton abundance, especially during the outbreak of P. globosa and a large amount of mucus produced by the colonies could significantly increase seawater viscosity, we suggest that biological factors were the main reasons for the increase in seawater viscosity.
