1Clinical Characteristics of Children with Coronavirus Disease 2019 in Hubei, China显示文摘Since December 2019,COVID-19 has occurred unexpectedly and emerged as a health problem worldw ide.Despite the rapidly increasing number of cases in subsequent weeks,the clinical characteristics of pediatric cases are rarely described.A cross-sectional multicenter study was carried out in 10 hospitals across Hubei province.A total of 25 confirmed pediatric cases of COVID-19 were collected.The demographic data,epidemiological history,underlying discascs,clinical mani festations,laboratory and radiological data,treatments,and outcomes were analyzcd.Of 25 hospitalized patients with COVID-19,the boy to girl ratio was 1.27:1.The median age was 3 years.COVID-19 cases in children aged<3 years,3-6 years,and≥6-years patients were 10(40%),6(24%),and 9(36%),respectively.The most common symptoms at onset of ilness were fever(13[52%]),and dry cough(11[44%]).Chest CT images showed essential normal in 8 cases(33.3%),unilateral involvement of lungs in 5 cases(20.8%),and bilateral involvement in 11 cases(45.8%).Clinical diagnoses included upper respiratory tract infection(n=8),mild pneumonia(n=15),and critical cases(n=2).Two critical cases(8%)were given invasive mechanical ventilation,corticosteroids,and immunoglobulin.The symptoms in 24(96%)of 25 patients were alleviated and one patient had been discharged.It was concluded that children were susceptible to COVID-19 like adults,while the clinical presentations and outcomes were more favorable in children.However,children less than 3 years old accounted for majority cases and critical cases lied in this age group,which demanded extra attentions during home caring and hospitalization treatment.Fang ZHENG Chun LIAO Qi-hong FAN Hong-bo CHEN Xue-gong ZHAO Zhong-guo XIE Xi-lin LI Chun-xi CHEN Xiao-xia LU Zhi-sheng LIU Wei LU Chun-bao CHEN Rong JIAO Ai-ming ZHANG Jin-tang WANG Xi-wei DING Yao-guang ZENG Li-ping CHENG Qing-feng HUANG Jiang WU Xi-chang LUO Zhu-jun WANG Yan-yan ZHONG Yan BAI Xiao-yan WU Run-ming JIN 2020Current Medical Science2020,40,2:10
2Clinicopathological,treatment,and prognosis study of 43 gastric neuroendocrine carcinomas显示文摘AIM To provide more information and therapeutic methods about gastric neuroendocrine carcinomas(G-NECs) which occur rarely but are highly malignant and clinically challenging.METHODS We retrospectively analyzed the clinicopathological characteristics, treatments, and prognosis of 43 G-NEC patients at our hospital between January 2007 and December 2014. The diagnosis was based on the 2010 World Health Organization criteria.RESULTS Forty-three G-NECs containing 39 small cell carcinomas and 4 large cell NECs with Ki67 > 60% were included in this study, accounting for only 0.95% of all gastric carcinomas. The median patient age was 62 years (range, 33-82) and the male-to-female ratio was 4.4:1. All patients underwent surgery, including 38 curative resections and 5 palliative resections. Among these 43 patients, nearly half(48.84%) of these tumors were located in the cardiac region of the stomach, regional lymph node metastasis was found in 31 cases(72.09%), and liver metastasis was found in 6 cases(13.95%). Follow-up information was got for 40 patients. Twentythree die of this disease with a median survival of 31 mo(range 1-90). The 1-year, 2-year, 3-year, and 5-year survival rate was 77.50%, 57.04%, 44.51%, and 35.05%, respectively. Survival was better in patients with tumor located in the cardiac region of the stomach, less than 7 lymph nodes metastasis and no liver metastasis. Five patients did not undergo postoperative chemotherapy, and the median survival time for these patients was 15 mo. For the remaining 34 patients who received postoperative chemotherapy, the median survival time was 44 mo and those received etoposide, cisplatin, and Paclitaxel survived the best. One patient with resected liver metastasis who received postoperative Capecitabine plus Oxaliplatin and Paclitaxel systemic chemotherapy plus octreotide LAR(30 mg intramuscularly, every 4 wk, for 2 years) has survived for 74 mo with no recurrence.CONCLUSION G-NECs are mostly nonfunctioning, which lead to a delay in detection. Local and/or distant metastases were noticed in most patients when diagnosed, and they required postoperative medical treatment. 3Multicenter Clinical Study for Evaluation of Efficacy and Safety of Transdermal Fentanyl Matrix Patch in Treatment of Moderate to Severe Cancer Pain in 474 Chinese Cancer Patients显示文摘Objective: Although a new matrix formulation fentanyl has been used throughout the world for cancer pain management, few data about its efficacy and clinical outcomes associated with its use in Chinese patients have been obtained. This study aimed to assess the efficacy and safety of the new system in Chinese patients with moderate to severe cancer pain. Methods: A total of 474 patients with moderate to severe cancer pain were enrolled in this study and were treated with the new transdermal fentanyl matrix patch (TDF) up to 2 weeks. All the patients were asked to record pain intensity, side effects, quality of life (QOL), adherence and global satisfaction. The initial dose of fentanyl was 25 ?g/h titrated with opioid or according to National Comprehensive Cancer Network (NCCN) guidelines. Transdermal fentanyl was changed every three days. Results: After 2 weeks. The mean pain intensity of the 459 evaluated patients decreased significantly from 5.63?1.26 to 2.03?1.46 (P<0.0001). The total remission rate was 91.29%, of which moderate remission rate 53.16%, obvious remission rate 25.49% and complete remission rate 12.64%. The rate of adverse events was 33.75%, 18.78% of which were moderate and 3.80% were severe. The most frequent adverse events were constipation and nausea. No fatal events were observed. The quality of life was remarkably improved after the treatment (P<0.0001). 4Antigenically dominant proteins within the human liver mitochondrial proteome identified by monoclonal antibodies显示文摘Analysis of the mitochondrial proteome would provide valuable insight into the function of this important organelle, which plays key roles in energy metabolism, apoptosis, free radical production, thermogenesis, and calcium signaling. It could also increase our understanding about the mechanisms that promote mitochondrial disease. To identify proteins that are antigenically dominant in human liver mitochondria, we generated >240 hybridoma cell lines from native mitochondrial proteins after cell fusion, screening, and cloning. Antibodies that recognized mitochondrial proteins were identified by screening human liver cDNA expression libraries. In this study, we identified 6 major antigens that were recognized by at least 2 different monoclonal antibodies (mAbs). The proteins that were antigenically dominant were: acetyl-Coenzyme A acyltransferase 2 (mitochondrial 3-oxoacyl-Coenzyme A thiolase), aldehyde dehydrogenase 1 family member A1, carbamoyl phosphate synthetase 1, dihydrolipoamide S-acetyltransferase (E2 component of pyruvate dehydrogenase complex), enoyl coenzyme A hydratase 1, and hydroxysteroid (11-beta) dehydrogenase 1. We also determined the subcellular localizations of these enzymes within the mitochondria using immunohistocytochemistry. We believe that these well-characterized antibodies will provide a valuable resource for the Human Liver Proteome Project (HLPP), and will make studies aimed at investigating liver mitochondrial function far easier to perform in future. 6Prevalence and Years Lived with Disability for Dental Caries among Children Aged 0-14 Years in China, 1990-2016显示文摘The estimate of dental caries among Chinese children at the microscale level using standard methodology remains unclear.In this study,we assessed and analyzed the disease burden of childhood dental caries in China by extracting data from the Global Burden of Disease,Injuries,and Risk Factors Study 2016(GBD 2016).In 2016,the number of cases,prevalence,years lived with disability(YLD),and age-standardized YLD rate of dental caries was 93.0 million,43.0%,32,200 person years,and 14.8 per 100,000,respectively.Across 33 provincial units,the disease burden was highest in Hubei(YLD rate 28.6 per 100,000),lowest in Macao(9.1 per 100,000),while geographical clustering was not observed.Compared with 1990,the prevalence in 2016 decreased from 46.8% to 43.0%,and the YLD rate decreased from 16.5 per 100,000 to 14.8 per 100,000.Given the slight decrease in dental caries burden,the prevalence and disease burden remained high among Chinese children.Strategies for addressing the spatial inequity of childhood dental caries require geographical targeting.liu wen zhao su ling yu xie xu dong xiang xue rong huang jiao ji ping 2018Biomedical and Environmental Sciences2018,31,5:5
7Cyst(e)ine in nutrition formulation promotes colon cancer growth and chemoresistance by activating mTORCl and scavenging ROS显示文摘Weight loss and cachexia are common problems in colorectal cancer patients;thus,parenteral and enteral nutrition support play important roles in cancer care.However,the impact of nonessential amino acid components of nutritional intake on cancer progression has not been fully studied.In this study,we discovered that gastrointestinal cancer patients who received cysteine as part of the parenteral nutrition had shorter overall survival(P<0.001)than those who did not.Cystine indeed robustly promotes colon cancer cell growth in vitro and in immunodeficient mice,predominately by inhibiting SESN2 transcription via the GCN2-ATF4 axis,resulting in mTORCl activation.mTORCl inhibitors Rapamycin and Everolimus block cystine-induced cancer cell proliferation.In addition,cystine confers resistance to oxaliplatin and irinotecan chemotherapy by quenching chemotherapy-induced reactive oxygen species via synthesizing glutathione.We demonstrated that dietary deprivation of cystine suppressed colon cancer xenograft growth without weight loss in mice and boosted the antitumor effect of oxaliplatin.These findings indicate that cyst(e)ine;as part of supplemental nutrition,plays an important role in colorectal cancer and manipulation of cyst(e)ine content in nutritional formulations may optimize colorectal cancer patient survival.Jiao Wu Sai-Ching Jim Yeung Sicheng Liu Aiham Qdaisat Dewei Jiang Wenli Liu Zhuo Cheng Wenjing Liu Haixia Wang Lu Li Zhongmei Zhou Rong Liu Chuanyu Yang Ceshi Chen Runxiang Yang 2021Signal Transduction and Targeted Therapy2021,6,6:2

9Target alignment in the Shen-Guang Ⅱ Upgrade laser facility显示文摘The Shen-Guang II Upgrade(SG-Ⅱ-U) laser facility consists of eight high-power nanosecond laser beams and one shortpulse picosecond petawatt laser. It is designed for the study of inertial confinement fusion(ICF), especially for conducting fast ignition(FI) research in China and other basic science experiments. To perform FI successfully with hohlraum targets containing a golden cone, the long-pulse beam and cylindrical hohlraum as well as the short-pulse beam and cone target alignment must satisfy tight specifications(30 and 20 μm rms for each case). To explore new ICF ignition targets with six laser entrance holes(LEHs), a rotation sensor was adapted to meet the requirements of a three-dimensional target and correct beam alignment. In this paper, the strategy for aligning the nanosecond beam based on target alignment sensor(TAS) is introduced and improved to meet requirements of the picosecond lasers and the new six LEHs hohlraum targets in the SG-II-U facility. 10QNE1 is a key flowering regulator determining the length of the vegetative period in soybean cultivars显示文摘The soybean E1 gene is a major regulator that plays an important role in flowering time and maturity.However,it remains unclear how cultivars carrying the dominant E1 allele adapt to the higher latitudinal areas of northern China.We mapped the novel quantitative trait locus QNE1(QTL near E1) for flowering time to the region proximal to E1 on chromosome 6 in two mapping populations.Positional cloning revealed Glyma.06G204300,encoding a TCP-type transcription factor,as a strong candidate gene for QNE1.Association analysis further confirmed that functional single nucleotide polymorphisms(SNPs) at nucleotides 686 and 1,063 in the coding region of Glyma.06G204300 were significantly associated with flowering time.The protein encoded by the candidate gene is localized primarily to the nucleus.Furthermore,soybean and Brassica napus plants overexpressing Glyma.06G204300 exhibited early flowering.We conclude that despite their similar effects on flowering time,QNE1 and E4 may control flowering time through different regulatory mechanisms,based on expression studies and weighted gene co-expression network analysis of flowering time-related genes.Deciphering the molecular basis of QNE1 control of flowering time enriches our knowledge of flowering gene networks in soybean and will facilitate breeding soybean cultivars with broader latitudinal adaptation.Zhengjun Xia Hong Zhai Yanfeng Zhang Yaying Wang Lu Wang Kun Xu Hongyan Wu Jinglong Zhu Shuang Jiao Zhao Wan Xiaobin Zhu Yi Gao Yingxiang Liu Rong Fan Shihao Wu Xin Chen Jinyu Liu Jiayin Yang Qijian Song Zhixi Tian 2022Science China(Life Sciences)2022,65,12:1
11Cut-off Values of Diagnostic Indices to Detect Iron Deficiency in Chinese Breast-fed Infants显示文摘Iron deficiency anemia is one of the most prevalent nutritional deficiency worldwide. The commonly used cut-off values for identifying iron deficiency are extrapolated from older children and may not be suitable for infants. Therefore, our study aimed to establish appropriate cut-off values for the evaluation of iron status in Chinese infants. Pregnant women who delivered at ≥37 gestational weeks with normal iron status were recruited. Later, infants with normal birth weight and who were breastfed in the first 4 months were selected. Blood samples were collected to assess hemoglobin, serum ferritin, soluble transferrin receptor, mean corpuscular volume and free erythrocyte protoporphyrin. 14Design and performance of final optics assembly in SG-ⅡUpgrade laser facility显示文摘In high power laser facility for inertial confinement fusion research, final optics assembly(FOA) plays a critical role in the frequency conversion, beam focusing, color separation, beam sampling and debris shielding. The design and performance of FOA in SG-II Upgrade laser facility are mainly introduced here. Due to the limited space and short focal length, a coaxial aspheric wedged focus lens is designed and applied in the FOA configuration. Then the ghost image analysis,the focus characteristic analysis, the B integral control design and the optomechanical design are carried out in the FOA design phase. 16A brain structural connectivity biomarker for autism spectrum disorder diagnosis in early childhood显示文摘Background:Autism spectrum disorder(ASD)is associated with altered brain development,but it is unclear which specific structural changes may serve as potential diagnostic markers,particularly in young children at the age when symptoms become fully estab-lished.Furthermore,such brain markers need to meet the requirements of precision medicine and be accurate in aiding diagnosis at an individual rather than only a group level.Objective:This study aimed to identify and model brain-wide differences in structural connectivity using diffusion tensor imaging(DTI)in young ASD and typically developing(TD)children.Methods:A discovery cohort including 93 ASD and 26 TD children and two independent validation cohorts including 12 ASD and 9 TD children from three different cities in China were included.Brain-wide(294 regions)structural connectivity was measured using DTI(fractional anisotropy,FA)together with symptom severity and cognitive development.A connection matrix was constructed for each child for comparisons between ASD and TD groups.Pattern classification was performed on the discovery dataset and the resulting model was tested on the two independent validation datasets.Results:Thirty-three structural connections showed increased FA in ASD compared to TD children and associated with both autistic symptom severity and impaired general cognitive development.The majority(29/33)involved the frontal lobe and comprised five different networks with functional relevance to default mode,motor control,social recognition,language and reward.Overall,clas-sification achieved very high accuracy of 96.77%in the discovery dataset,and 91.67%and 88.89%in the two independent validation datasets.Conclusions:Identified structural connectivity differences primarily involving the frontal cortex can very accurately distinguish novel individual ASD from TD children and may therefore represent a robust early brain biomarker which can address the requirements of precision medicine.Xi Jiang Xiao-Jing Shou Zhongbo Zhao Yuzhong Chen Fan-Chao Meng Jiao Le Tian-Jia Song Xin-Jie Xu Weitong Guo Xiaoyan Ke Xiao-E Cai Weihua Zhao Juan Kou Ran Huo Ying Liu Hui-Shu Yuan Yan Xing Ji-Sheng Han Song-Ping Han Yun Li Hua Lai Lan Zhang Mei-Xiang Jia Jing Liu Xuan Liu Keith M.Kendrick Rong Zhang 2023Psychoradiology2023,3,1:0

19EPAS1 prevents telomeric damage-induced senescence by enhancing transcription of TRF1,TRF2,and RAD50显示文摘Telomeres are nucleoprotein structures located at the end of each chromosome,which function in terminal protection and genomic stability.Telomeric damage is closely related to replicative senescence in vitro and physical aging in vivo.As relatively long-lived mammals based on body size,bats display unique telomeric patterns,including the upregulation of genes involved in alternative lengthening of telomeres(ALT),DNA repair,and DNA replication.At present,however,the relevant molecular mechanisms remain unclear.In this study,we performed cross-species comparison and identified EPAS1,a well-defined oxygen response gene,as a key telomeric protector in bat fibroblasts.Bat fibroblasts showed high expression of EPAS1,which enhanced the transcription of shelterin components TRF1 and TRF2,as well as DNA repair factor RAD50,conferring bat fibroblasts with resistance to senescence during long-term consecutive expansion.Based on a human single-cell transcriptome atlas,we found that EPAS1 was predominantly expressed in the human pulmonary endothelial cell subpopulation.Using in vitro-cultured human pulmonary endothelial cells,we confirmed the functional and mechanistic conservation of EPAS1 in telomeric protection between bats and humans.In addition,the EPAS1 agonist M1001 was shown to be a protective compound against bleomycin-induced pulmonary telomeric damage and senescence.In conclusion,we identified a potential mechanism for regulating telomere stability in human pulmonary diseases associated with aging,drawing insights from the longevity of bats.Kai-Qin Li Gao-Jing Liu Xiu-Yun Liu Qiong-Fang Chen Xiao-Yan Huang Qiu Tu Jiao Zhang Qing Chang Yun-Hua Xie Rong Hua Dong-Ming Xu Zhen Liu Bo Zhao 2023Zoological Research2023,44,3:0
20Effects of mask wearing duration and relative humidity on thermal perception in the summer outdoor built environment显示文摘During the pandemic,face masks are one of the most significant self-protection necessities,but they also cause heat stress.By using the ERA5(ECMWF Reanalysis 5th Generation)database and the local weather bureau data,the effect of mask wearing on outdoor thermal sensation has been investigated by a survey conducted in the hot summer and cold winter region of eastern China in the summer of 2020.Results show that wearing a face mask for a longer period result in a higher level of discomfort,and the primary source of discomfort is hot and stuffy feelings.The effect of relative humidity is crucial for mask wearers in warm-biased thermal environments,as mean thermal sensation vote(TSV)peaks when environmental relative humidity reaches the range of 70%to 80%and decreases after this range due to the evaporation within the microclimate created by a face mask.Meanwhile,prolonged mask wearing increases participants’hot feelings,especially in warm environments.Specifically,participants wearing face masks for less than 30 min feel hot at a physiological equivalent temperature(PET)value of 34.4℃,but those who wear them for over 60 min express hot feelings even at a PET value of 24.7℃.The participants who wear a face mask while walking slowly outdoors have similar thermal sensations to those who do not wear a mask,but are in a higher activity level.The findings demonstrate that mask wearing has a crucial impact on outdoor thermal comfort assessment in a warm-biased outdoor thermal environment.Rong Hu Jianlin Liu Yongxin Xie Jiao Jiao Zhaosong Fang Borong Lin 2023Building Simulation2023,16,9:0
