3Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23
4Probabilistic stability analyses of undrained slopes by 3D random fields and finite element methods显示文摘A long slope consisting of spatially random soils is a common geographical feature. This paper examined the necessity of three-dimensional(3 D) analysis when dealing with slope with full randomness in soil properties. Although 3 D random finite element analysis can well reflect the spatial variability of soil properties, it is often time-consuming for probabilistic stability analysis. For this reason, we also examined the least advantageous(or most pessimistic) cross-section of the studied slope. The concept of'most pessimistic' refers to the minimal cross-sectional average of undrained shear strength. The selection of the most pessimistic section is achievable by simulating the undrained shear strength as a 3 D random field. Random finite element analysis results suggest that two-dimensional(2 D) plane strain analysis based the most pessimistic cross-section generally provides a more conservative result than the corresponding full 3 D analysis. The level of conservativeness is around 15% on average. 5Single-atom catalyst: a rising star for green synthesis of fine chemicals显示文摘The green synthesis of fine chemicals calls for a new generation of efficient and robust catalysts. Single-atom catalysts(SACs), in which all metal species are atomically dispersed on a solid support, and which often consist of well-defined mononuclear active sites, are expected to bridge homogeneous and heterogeneous catalysts for liquid-phase organic transformations. This review summarizes major advances in the SAC-catalysed green synthesis of fine chemicals in the past several years, with a focus on the catalytic activity, selectivity and reusability of SACs in various organic reactions. The relationship between catalytic performance and the active site structure is discussed in terms of the valence state, coordination environment and anchoring chemistry of single atoms to the support, in an effort to guide the rational design of SACs in this special area, which has traditionally been dominated by homogeneous catalysis.Finally, the challenges remaining in this research area are discussed and possible future research directions are proposed.Leilei Zhang Yujing Ren Wengang Liu Aiqin Wang Tao Zhang 2018National Science Review2018,5,5:15

7Optimization of clay material mixture ratio and filling process in gypsum mine goaf显示文摘Because there is neither waste rock nor mill tailings in the gypsum mine, and the buildings on the goaf of gypsum mine are needed to be protected, the research proposed the scheme of the clay filling technology. Gypsum, cement, lime and water glass were used as adhesive, and the strength of different material ratios were investigated in this study. The influence factors of clay strength were obtained in the order of cement, gypsum, water glass and lime. The results show that the cement content is the determinant influence factor, and gypsum has positive effects, while the water glass can enhance both clay strength and the fluidity of the filing slurry. Furthermore, combining chaotic optimization method with neural network, the optimal ratio of composite cementing agent was obtained. The results show that the optimal ratio of water glass, cement, lime and clay (in quality) is 1.17:6.74:4.17:87.92 in the process of bottom self-flow filling, while the optimal ratio is 1.78:9.58:4.71:83.93 for roof-contacted filling. A novel filling process to fill in gypsum mine goaf with clay is established. 9Effect of butanol on flotation separation of quartz from hematite with N-dodecyl ethylenediamine显示文摘In order to investigate the effect of butanol on quartz flotation when N-dodecyl ethylenediamine(ND)was used as collector, single mineral flotation and artificial mixed mineral(hematite and quartz were mixed at a mass ratio of 3:2) separation were conducted in the laboratory. Experimental results indicated that addition of butanol could improve the collecting performance of ND on quartz and enhance the floatability of quartz. Best flotation recovery of quartz was obtained when butanol was mixed with ND at a mass ratio of 1:1. Moreover, the molecular structure of alcohols had a significant effect on mineral recovery. Best separation efficiency could be obtained when tert-butanol was added as it had the largest cross-sectional area. Zeta potential measurements indicated that alcohols could strengthen electrostatic adsorption between quartz and collector. 10Preparation, characterization, pharmacokinetics and anticancer effects of PEGylated β-elemene liposomes显示文摘Objective:This study aimed to develop a new polyethylene glycol(PEG)ylatedβ-elemene liposome(PEG-Lipo-β-E)and evaluate its characterization,pharmacokinetics,antitumor effects and safety in vitro and in vivo.Methods:The liposomes were prepared by ethanol injection and high-pressure micro-jet homogenization.Characterization of the liposomes was conducted,and drug content,entrapment efficiency(EE),in vitro release and stability were studied by ultra-fast liquid chromatography(UFLC)and a liquid surface method.Blood was drawn from rats to establish the pharmacokinetic parameters.The anticancer effect was evaluated in a KU-19-19 bladder cancer xenograft model.Histological analyses were performed to evaluate safety.Results:The PEG-Lipo-β-E showed good stability and was characterized as 83.31±0.181 nm in size,0.279±0.004 in polydispersity index(PDI),-21.4±1.06 mV in zeta potential,6.65±0.02 in pH,5.024±0.107 mg/mL inβ-elemene(β-E)content,and 95.53±1.712%in average EE.The Fourier transform infrared spectroscopy(FTIR)and differential scanning calorimetry(DSC)indicated the formation of PEG-Lipo-β-E.Compared to elemene injection,PEG-Lipo-β-E demonstrated a 1.75-fold decrease in clearance,a 1.62-fold increase in half-life,and a 1.76-fold increase in area under the concentration-time curves(AUCs)from 0 hour to 1.5 hours(P<0.05).PEG-Lipo-β-E also showed an enhanced anticancer effect in vivo.Histological analyses showed that there was no evidence of toxicity to the heart,kidney,liver,lung or spleen.Conclusions:The present study demonstrates PEG-Lipo-β-E as a new formulation with ease of preparation,high EE,good stability,improved bioavailability and antitumor effects.Bingtao Zhai Qibiao Wu Wengang Wang Mingming Zhang Xuemeng Han Qiujie Li Peng Chen Xiaying Chen Xingxing Huang Guohua Li Qin Zhang Ruonan Zhang Yu Xiang Shuiping Liu Ting Duan Jianshu Lou Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,1:6
11An ATF24 peptide-functionalized β-elemene-nanostructured lipid carrier combined with cisplatin for bladder cancer treatment显示文摘Objective:In this study,we aimed to develop an amino-terminal fragment(ATF)peptide-targeted liposome carryingβ-elemene(ATF24-PEG-Lipo-β-E)for targeted delivery into urokinase plasminogen activator receptor-overexpressing bladder cancer cells combined with cisplatin(DDP)for bladder cancer treatment.Methods:The liposomes were prepared by ethanol injection and high-pressure microjet homogenization.The liposomes were characterized,and the drug content,entrapment efficiency,andin vitro release were studied.The targeting efficiency was investigated using confocal microscopy,ultra-fast liquid chromatography,and an orthotopic bladder cancer model.The effects of ATF24-PEG-Lipo-β-E combined with DDP on cell viability and proliferation were evaluated by a Cell Counting Kit-8(CCK-8)assay,a colony formation assay,and cell apoptosis and cell cycle analyses.The anticancer effects were evaluated in a KU-19-19 bladder cancer xenograft model.Results:ATF24-PEG-Lipo-β-E had small and uniform sizes(~79 nm),high drug loading capacity(~5.24 mg/mL),high entrapment efficiency(98.37±0.95%),and exhibited sustained drug release behavior.ATF24-PEG-Lipo-β-E had better targeting efficiency and higher cytotoxicity than polyethylene glycol(PEG)ylatedβ-elemene liposomes(PEG-Lipo-β-E).DDP,combined with ATF24-PEG-Lipo-β-E,exerted a synergistic effect on cellular apoptosis and cell arrest at the G2/M phase,and these effects were dependent on the caspase-dependent pathway and Cdc25C/Cdc2/cyclin B1 pathways.Furthermore,thein vivo antitumor activity showed that the targeted liposomes effectively inhibited the growth of tumors,using the combined strategy.Conclusions:The present study provided an effective strategy for the targeted delivery ofβ-elemene(β-E)to bladder cancer,and a combined strategy for bladder cancer treatment.Bingtao Zhai Peng Chen Wengang Wang Shuiping Liu Jiao Feng Ting Duan Yu Xiang Ruonan Zhang Mingming Zhang Xuemeng Han Xiaying Chen Qiujie Li Guohua Li Ying Liu Xingxing Huang Wenzheng Zhang Ting Pan Lili Yan Ting Jin Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,3:5
12Bushen Qiangjin capsule(补肾强筋胶囊) inhibits the Wnt/β-catenin pathway to ameliorate papain-induced knee osteoarthritis in rats显示文摘OBJECTIVE:To evaluate the molecular mechanism underlying the beneficial effect of Bushen Qiangjin capsule(补肾强筋胶囊,BSQJ),a Traditional Chinese Medicine,on knee osteoarthritis(KOA).METHODS:In the present study,32 female Sprague-Dawley rats were randomly divided into four groups:control,KOA,high-dose BSQJ(H-BSQJ),and low-dose BSQJ(L-BSQJ).After successfully establishing the KOA model by intra-articular injection of papain,H-BSQJ and L-BSQJ groups were intragastrically administered 0.243 and 0.122 g/kg BSQJ,respectively,daily for 6 weeks.At the end of the experiment,knee articular cartilage tissues of rats were collected for evaluation by hematoxylin and eosin staining,Safranin O-Fast Green staining,and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay.Serum interleukin-1βand tumor necrosis factor-αlevels of rats were detected with an enzyme-linked immunosorbent assay method.Gene expression of Wnt-4,β-catenin,Frizzled-2,glycogen synthase kinase-3β(GSK-3β),cysteinyl aspartate-specific proteinases 3 and 9(caspases 3 and 9),collagen typeⅡalpha 1(Col2 a1),and matrix metalloproteinases 1 and 13(MMP-1 and MMP-3)of rat knee articular cartilage was quantified by reverse transcription-quantitative polymerase chain reaction analysis.Wnt-4,β-catenin,Frizzled-2,GSK-3β,cleaved caspase-3,and cleaved caspase-9 protein expression in rat knee articular cartilage was determined by western blot analysis.RESULTS:BSQJ obviously reduced pathological damage and matrix degradation of articular cartilage in KOA rats.Compared with the KOA group,H-BSQJ rats exhibited downregulated m RNA and protein expression of Wnt-4,β-catenin,Frizzled-2,and caspase-3,as well as upregulated m RNA and protein expression of GSK-3β.In addition,H-BSQJ significantly increased m RNA expression of Col2 a1 and decreased m RNA expression of MMP-1 and MMP-13.CONCLUSION:BSQJ exerted a beneficial effect on KOA by a mechanism involving downregulation of the Wnt/β-catenin pathway,which inhibited both cartilage extracellular matrix degradation and chondrocyte apoptosis to ameliorate KOA in rats.YAO Nan CHEN Guocai LU Yanyan XU Xuemeng ZHAO Chuanxi HUANG Xuejun LIU Wengang PENG Sha WU Huai 2021Journal of Traditional Chinese Medicine2021,41,6:5

14MEF2C promotes M1 macrophage polarization and Th1 responses显示文摘The polarization of macrophages to the M1 or M2 phenotype has a pivotal role in inflammation and host defense;however,the underlying molecular mechanism remains unclear.Here,we show that myocyte enhancer factor 2 C(MEF2C)is essential for regulating M1 macrophage polarization in response to infection and inflammation.Global gene expression analysis demonstrated that MEF2C deficiency in macrophages downregulated the expression of M1 phenotypic markers and upregulated the expression of M2 phenotypic markers.MEF2C significantly promoted the expression of interleukin-12 p35 subunit(Il12a)and interleukin-12 p40 subunit(Il12b).Myeloid-specific Mef2c-knockout mice showed reduced IL-12 production and impaired Th1 responses,which led to susceptibility to Listeria monocytogenes infection and protected against DSS-induced IBD in vivo.Mechanistically,we showed that MEF2C directly activated the transcription of Il12a and Il12b.These findings reveal a new function of MEF2C in macrophage polarization and Th1 responses and identify MEF2C as a potential target for therapeutic intervention in inflammatory and autoimmune diseases.Xibao Zhao Qianqian Di Han Liu Jiazheng Quan Jing Ling Zizhao Zhao Yue Xiao Han Wu Zherui Wu Wengang Song Huazhang An Weilin Chen 2022Cellular & Molecular Immunology2022,19,4:4
15TBM penetration rate prediction based on the long short-term memory neural network显示文摘Tunnel boring machines(TBMs)are widely used in tunnel engineering because of their safety and efficiency.The TBM penetration rate(PR)is crucial,as its real-time prediction can reflect the adaptation of a TBM under current geological conditions and assist the adjustment of operating parameters.In this study,deep learning technology is applied to TBM performance prediction,and a PR prediction model based on a long short-term memory(LSTM)neuron network is proposed.To verify the performance of the proposed model,the machine parameters,rock mass parameters,and geological survey data from the water conveyance tunnel of the Hangzhou Second Water Source project were collected to form a dataset.Furthermore,2313 excavation cycles were randomly composed of training datasets to train the LSTM-based model,and 257 excavation cycles were used as a testing dataset to test the performance.The root mean square error and the mean absolute error of the proposed model are 4.733 and 3.204,respectively.Compared with Recurrent neuron network(RNN)based model and traditional time-series prediction model autoregressive integrated moving average with explanation variables(ARIMAX),the overall performance on proposed model is better.Moreover,in the rapidly increasing period of the PR,the error of the LSTM-based model prediction curve is significantly smaller than those of the other two models.The prediction results indicate that the LSTM-based model proposed herein is relatively accurate,thereby providing guidance for the excavation process of TBMs and offering practical application value.Boyang Gao RuiRui Wang Chunjin Lin Xu Guo Bin Liu Wengang Zhang 2021Underground Space2021,6,6:4

17Synthesis and electrochemical properties of Li_2MnSiO_4/C nanoparticles via polyol process显示文摘Carbon-coated lithium manganese silicate(Li2MnSiO4/C) nanoparticles were synthesized by polyol process.X-ray diffraction(XRD) patterns of the obtained materials exhibit a good fit with that of the Li2MnSiO4 phase.Field emission scanning electron microscopy(FESEM) images of the obtained samples show that the particle size is only tens of nanometers.The high resolution transmission electron microscopy(HRTEM) analysis shows that the Li2MnSiO4 nanoparticles are surrounded by a very thin film of amorphous carbon.The composite prepared through polyol process shows good performance as cathode materials in lithium cells at room temperature.The charge capacity of the Li2MnSiO4/C samples is 219 mAh/g(about 1.3 Li+ per unit formula extracted),and the discharge capacity is 132 mAh/g(about 0.8 Li+ per unit formula inserted) in the first cycle in the voltage range of 1.5-4.8 V.A good capacity cycling maintenance of 81.8% after 10 cycles was obtained.LIU Wengang XU Yunhua YANG Rong 2010Rare Metals2010,29,5:3
18Seismic behaviors of utility tunnel-soil system: With and without joint connections显示文摘Seismic responses of utility tunnel-soil system were studied via shaking table model tests with considerations of two kinds of double box utility tunnels:with and without joint connections.These two testing utility tunnel models were made of galvanized iron wire and micro-concrete,and the ground was simulated by the dry standard sand through layered tamping treatment.The utility tunnel-soil system was subjected to horizontal vibration in uniaxial direction perpendicular to the longitudinal direction of tunnel model.Via instrumentations of earth pressure gauges,accelerometers and strain gauges,the earth pressure response,acceleration response and bending moment response were measured.The testing results show that the joint connections in the utility tunnel along the longitudinal direction play an important role in determining the characteristic of earth pressure response and bending moment response,whereas the effect of joint connections on acceleration response is less significant.In addition,the partition wall exhibits the consistent acceleration response with the side-wall of double box utility tunnel model under seismic condition.Based on the testing results,it is suggested that the joint connection should be taken reasonably into consideration during design and construction for engineering practice.Liang Han Hanlong Liu Wengang Zhang Xuanming Ding Zhixiong Chen Li Feng Zhenyu Wang 2022Underground Space2022,7,5:3
19Extraction mechanism of rare earths from chloride acidic solution with ammonium-bifunctionalized ionic liquid extractants显示文摘A series of ammonium-bifunctionalized ionic liquid extractants(ammonium-Bif-ILEs) combined with a number of anions, including carboxylic acids and 1,3-diketonates, have been prepared and studied in this report. Their extraction behavior and properties toward rare earth ions(REs(Ⅲ)) are systematically investigated in chloride media as a function of important parameters such as aqueous phase p H, salting-out agent concentrations, and extraction temperature. The separation performance of ammonium-Bif-ILEs toward REs(Ⅲ) are systematically discussed. The results demonstrate that ammonium-Bif-ILEs have a synergistic effect between the cation and anions in separation of REs(Ⅲ). The influences of different anions on separation factor(b) values are further studied.
