| 4 | OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. | 6 | Compound Danshen injection improves endotoxin-induced microcirculatory disturbance in rat mesentery显示文摘AIM: To investigate the effect of compound Danshen injection on lipopolysaccharide (LPS)-induced rat mesenteric microcirculatory dysfunctions and the underlying possible mechanism by an inverted intravital microscope and high-speed video camera system. METHODS: LPS was continuously infused through the jugular artery of male Wistar rats at the dose of 2 mg/kg per hour. Changes in mesenteric microcirculation,such as diameters of arterioles and venules,velocity of RBCs in venules,leukocyte rolling,adhesion and emigration,free radicals released from post-capillary venules,FITC- albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope assisted with CCD camera and SIT camera. Meanwhile,the expression of adhesion molecules CD11b/CD18 and the production of free radical in neutrophils,and the expression of intercellular adhesion molecule 1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) were quantified by flow cytometry (FACS) in vitro. RESULTS: The continuous infusion with LPS resulted in a number of responses in microcirculation,including a significant increase in the positive region of venulestained with Monastral blue B,rolling and adhesion of leukocytes,production of oxygen radical in venular wall,albumin efflux and enhanced mast cell degranulation in vivo,all of which,except for the leukocyte rolling,were attenuated by the treatment with compound Danshen injection. Experiments performed in vitro further revealed that the expression of CD11b/CD18 and the production of oxygen free radical in neutrophils,and the expression of ICAM-1 in HUVECs were increased by exposure to LPS,and they were attenuated by compound Danshen injection. | 8 | Report on Childhood Obesity in China (9): Sugar-sweetened Beverages Consumption and Obesity显示文摘Objective To explore the associations between sugar-sweetened beverage (SSB) consumption and obesity as well as obesity-related cardiometabolic disorders among children in China. Methods A total of 6974 (boys 3558, girls 3412) children aged 6-13 years participated in the study. Each participant's height, weight, waist circumference, fasting glucose, triglycerides, total cholesterol, high- density lipoprotein cholesterol, and low-density lipoprotein cholesterol were measured. The type of beverage consumption was determined using a self-administered questionnaire. Results SSBs were consumed regularly by 46.1% of the children. The prevalence [adjusted odds ratio (95% confidence internal (CI)] of obesity was 7.6% [as the reference group (ref.)], 10.1% [1.36(1.07, 1.74)], and 11.6% [1.46(1.21, 1.75)], among children who regularly drank milk, other beverages and SSBs, respectively. Regularly drinking SSBs elevated the likelihood of abdominal obesity [adjusted odds ratio (95% CI): 1.36 (1.17, 1.59)]. The prevalence [adjusted odds ratio (95% CI)] of obesity among children who regularly drank sports/caloric beverages, carbonated beverages, sweet tea, and plant protein beverages was 16.8% [2.00(1.31, 3.07)], 12.7% [1.52(1.23, 1.88)], 11.5% [1.52(1.18, 1.95)], and 10.4% [1.41(1.03, 1.94)], respectively, which was higher than that of regular milk drinkers [7.6 % (ref.)]. The prevalence [adjusted odds ratio (95% CI)] of abdominal obesity among children who regularly drank sweet tea, fruit/vegetable juices, and carbonated beverages was 17.7% [1.55(1.26, 1.90)], 16.2% [1.36(1.09, 1.70)], and 15.3% [1.24(1.03, 1.50)], respectively, which was much higher than that of regular milk drinkers [12.8% (ref.)]. Conclusions Regular SSB consumption was positively related to obesity and abdominal obesity. | 9 | Medusoid Fossils from the Sinian Xingmincun Formation of Southern Liaoning Province显示文摘Abundant medusoid soft-bodied metazoan fossils have been found in the Sinian Xingmincun Formation(with an isotopic age of 650 Ma) at Qipanmo, Qidingshan and Yangtun of the Dalian-Jinzhou area, southern Liaoning Province; in addition, a number of macroscopic algal fossils have been discovered for the first time in the Getun Formation overlying the Xingmincun Formation. Medusoid fossils include 3 genera and 6 species(all of which are new). Judging from their biological characters, these medusoid fossils are considered to have an intimate affinity with the Ediacara fauna in the terminal Precambrian of Australia, as their characters are close to each other. The authors suggest that these medusoids and the Edicara fauna might be the products of the same period. | 10 | Wogonin protects glomerular podocytes by targeting Bcl-2-mediated autophagy and apoptosis in diabetic kidney disease显示文摘Diabetic kidney disease(DKD)is one of the microvascular complications of diabetes mellitus and a major cause of end-stage renal disease with limited treatment options.Wogonin is a flavonoid derived from the root of Scutellaria baicalensis Georgi,which has shown a potent renoprotective effect.But the mechanisms of action in DKD are not fully elucidated.In this study,we investigated the effects of wogonin on glomerular podocytes in DKD using mouse podocyte clone 5(MPC5)cells and diabetic mice model.MPC5 cells were treated with high glucose(30 mM).We showed that wogonin(4,8,16μM)dose-dependently alleviated high glucose(HG)-induced MPC5 cell damage,accompanied by increased expression of WT-1,nephrin,and podocin proteins,and decreased expression of TNF-α,MCP-1,IL-1βas well as phosphorylated p65.Furthermore,wogonin treatment significantly inhibited HG-induced apoptosis in MPC5 cells.Wogonin reversed HG-suppressed autophagy in MPC5 cells,evidenced by increased ATG7,LC3-II,and Beclin-1 protein,and decreased p62 protein.We demonstrated that wogonin directly bound to Bcl-2 in MPC5 cells.In HG-treated MPC5 cells,knockdown of Bcl-2 abolished the beneficial effects of wogonin,whereas overexpression of Bcl-2 mimicked the protective effects of wogonin.Interestingly,we found that the expression of Bcl-2 was significantly decreased in biopsy renal tissue of diabetic nephropathy patients.In vivo experiments were conducted in STZ-induced diabetic mice,which were administered wogonin(10,20,40 mg·kg^(−1)·d^(−1),i.g.)every other day for 12 weeks.We showed that wogonin administration significantly alleviated albuminuria,histopathological lesions,and p65 NF-κB-mediated renal inflammatory response.Wogonin administration dose-dependently inhibited podocyte apoptosis and promoted podocyte autophagy in STZ-induced diabetic mice.This study for the first time demonstrates a novel action of wogonin in mitigating glomerulopathy and podocytes injury by regulating Bcl-2-mediated crosstalk between autophagy and apoptosis.Wogonin may be a potential therapeutic drug against DKD. | Xue-qi Liu Ling Jiang Yuan-yuan Li Yue-bo Huang Xue-ru Hu Wei Zhu Xian Wang Yong-gui Wu Xiao-ming Meng Xiang-ming Qi | 2022 | Acta Pharmacologica Sinica2022,43,1: | 20 |
| 11 | Relative effects of phylogeny,biological characters and environments on leaf traits in shrub biomes across central Inner Mongolia,China显示文摘Aims Understanding the drivers for leaf traits is critical to improving our predictions on ecosystems'responses to global changes.Geographic patterns of leaf traits are shaped by phylogenetic,biological and environmental factors simultaneously.However,till now few studies have examined how these factors influenced leaf traits together,and how their effects differed at the within-and among-site levels.Methods We sampled leaf traits from a 1100 km shrub-biome transect across central Inner-Mongolia,including leaf mass per area(LMA),mass-based photosynthetic rate,nitrogen(N)and phosphorus(P)concentrations.We examined the effects of phylogenetic,biological(height and growth rate)and environmental(climate and soil)fac-tors on leaf traits with mixed-model analyses of variance.Variation partitioning method was used to separate the joint and independent effects of these three types of factors.Important Findings(i)Climate and soil fertility(total or available nutrient concentra-tions)together explained 11.4-41.4%of among-site variations,with remarkable difference among traits.(i)Height and height growth rate together explained 0.4--31.9%of trait variations(mostly among-site variations).Our results could only weakly support the ability of leaf traits as predictors for whole-plant growth.(ii)LMA was negatively related to height,which was consistent with the resource-use strategy hypothesis but incon-sistent with the hypotheses proposed for coexisting trees,suggest-ing that the LMA--height relationship is shaped by rather different mechanisms between the within-and among-communities lev-els.(iv)The variation partitioning analysis showed that,the rela-tionships between leaf traits and biological characters largely reflected the differences in both leaf traits and biological char-acters among species that occupying different sites.The relative importance of phylogenetic,biological and environmental fac-tors differed remarkably among leaf traits,between the within-and among-communities levels,and between different biomes.(v)Our results strongly suggest the necessity of examining the three types of factors simultaneously,and at both the within-and among-communities levels,for a better understanding of the drivers for leaf traits patterns. | Chao Liu Xiangping Wang Xian Wu Shuang Dai Jin-Sheng He Weilun Yin | 2013 | Journal of Plant Ecology2013,6,3: | 19 |

| 13 | Automated Counting of Rice Planthoppers in Paddy Fields Based on Image Processing显示文摘A quantitative survey of rice planthoppers in paddy fields is important to assess the population density and make forecasting decisions. Manual rice planthopper survey methods in paddy fields are time-consuming, fatiguing and tedious. This paper describes a handheld device for easily capturing planthopper images on rice stems and an automatic method for counting rice planthoppers based on image processing. The handheld device consists of a digital camera with WiFi, a smartphone and an extrendable pole. The surveyor can use the smartphone to control the camera, which is fixed on the front of the pole by WiFi, and to photograph planthoppers on rice stems. For the counting of planthoppers on rice stems, we adopt three layers of detection that involve the following:(a) the first layer of detection is an AdaBoost classifier based on Haar features;(b) the second layer of detection is a support vector machine(SVM) classifier based on histogram of oriented gradient(HOG) features;(c) the third layer of detection is the threshold judgment of the three features. We use this method to detect and count whiteback planthoppers(Sogatella furcifera) on rice plant images and achieve an 85.2% detection rate and a 9.6% false detection rate. | 17 | Electroacupuncture activates enteric glial cells and protects the gut barrier in hemorrhaged rats显示文摘AIM:To investigate whether electroacupuncture ST36 activates enteric glial cells,and alleviates gut inflammation and barrier dysfunction following hemorrhagic shock.METHODS:Sprague-Dawley rats were subjected to approximately 45% total blood loss and randomly divided into seven groups:(1) sham:cannulation,but no hemorrhage;(2) subjected to hemorrhagic shock(HS);(3) electroacupuncture(EA) ST36 after hemorrhage;(4) vagotomy(VGX)/EA:VGX before hemorrhage,then EA ST36;(5) VGX:VGX before hemorrhage;(6) a-bungarotoxin(BGT)/EA:intraperitoneal injection of a-BGT before hemorrhage,then EA ST36; and(7) a-BGT group:a-BGT injection before hemorrhage.Morphological changes in enteric glial cells(EGCs) were observed by immunofluorescence,and glial fibrillary acidic protein(GFAP; a protein marker of enteric glial activation) was evaluated using reverse transcriptase polymerase chain reaction and western blot analysis.Intestinal cytokine levels,gut permeability to 4-k Da fluorescein isothiocyanate(FITC)-dextran,and the expression and distribution of tight junction protein zona occludens(ZO)-1 were also determined.RESULTS:EGCs were distorted following hemorrhage and showed morphological abnormalities.EA ST36 attenuated the morphological changes in EGCs at 6 h,as compared with the VGX,a-BGT and HS groups.EA ST36 increased GFAP expression to a greater degree than in the other groups.EA ST36 decreased intestinal permeability to FITC-dextran(760.5 ± 96.43 ng/m L vs 2466.7 ± 131.60 ng/m L,P < 0.05) and preserved ZO-1 protein expression and localization at 6 h afterhemorrhage compared with the HS group.However,abdominal VGX and a-BGT treatment weakened or eliminated the effects of EA ST36.EA ST36 reduced tumor necrosis factor-a levels in intestinal homogenates after blood loss,while vagotomy or intraperitoneal injection of a-BGT before EA ST36 abolished its antiinflammatory effects.CONCLUSION:EA ST36 attenuates hemorrhageinduced intestinal inflammatory insult,and protects the intestinal barrier integrity,partly via activation of EGCs. | Sen Hu Zeng-Kai Zhao Rui Liu Hai-Bin Wang Chun-Yu Gu Hong-Min Luo Huan Wang Ming-Hua Du Yi Lv Xian Shi | 2015 | World Journal of Gastroenterology2015,21,5: | 15 |
| 18 | Immunotherapy using IL-2 and GM-CSF is a potential treatment for multidrug-resistant Mycobacterium tuberculosis显示文摘This study investigated the therapeutic effects of interleukin (IL)-2 and granulocyte-macrophage colony-stimulating factor (GM-CSF) coadministrated with antibacterial agents isoniazid (INH) and rifampin (RIF) to treat a mouse model of tuberculosis (TB) infection. A drug-susceptible TB strain, H37Rv was used to infect mice and the effectiveness of IL-2 and GM-CSF was initially evaluated based on survival rate, bacterial counts in lungs and spleens and the pathological condition of the lungs. Next, the therapeutic effect of the immunotherapy regimen was assessed in multidrug-resistant strain OB35-infected mice. In the H37Rv infection model, IL-2 and GM-CSF monotherapies reduced bacterial numbers in the lungs by 0.82 (P<0.01) and 0.58 (P<0.05) lg colony-forming units (CFU), respectively, and in the spleens by 1.42 (P<0.01) and 1.22 (P<0.01) lg CFU, respectively, compared with the untreated group. Mice receiving immunotherapy developed fewer lesions in the lungs compared with mice receiving antibacterial therapy alone. In the OB35 infection model, immunotherapy with either cytokine resulted in a significant reduction of bacterial load in the lungs and spleens and less severe lesions in the lungs compared with the untreated or antibacterial therapy treated mice. Notably, mice receiving immunotherapy with both cytokines had a 30% survival rate which was higher than that in other treated groups, and had significantly less CFUs in the lungs and spleens (1.02 and 1.34 lg CFU) compared with antibacterial therapy alone (P<0.01). | 19 | Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen.
