| 1 | Heat shock protein 90 promotes RNA helicase DDX5 accumulation and exacerbates hepatocellular carcinoma by inhibiting autophagy显示文摘Objective:Hepatocellular carcinoma(HCC),the main type of liver cancer,has a high morbidity and mortality,and a poor prognosis.RNA helicase DDX5,which acts as a transcriptional co-regulator,is overexpressed in most malignant tumors and promotes cancer cell growth.Heat shock protein 90(HSP90)is an important molecular chaperone in the conformational maturation and stabilization of numerous proteins involved in cell growth or survival.Methods:DDX5 m RNA and protein expression in surgically resected HCC tissues from 24 Asian patients were detected by quantitative real-time PCR and Western blot,respectively.The interaction of DDX5-HSP90 was determined by molecular docking,immunoprecipitation,and laser scanning confocal microscopy.The autophagy signal was detected by Western blot.The cell functions and signaling pathways of DDX5 were determined in 2 HCC cell lines.Two different murine HCC xenograft models were used to determine the function of DDX5 and the therapeutic effect of an HSP90 inhibitor.Results:HSP90 interacted directly with DDX5 and inhibited DDX5 protein degradation in the AMPK/ULK1-regulated autophagy pathway.The subsequent accumulation of DDX5 protein induced the malignant phenotype of HCC by activating theβ-catenin signaling pathway.The silencing of DDX5 or treatment with HSP90 inhibitor both blocked in vivo tumor growth in a murine HCC xenograft model.High levels of HSP90 and DDX5 protein were associated with poor prognoses.Conclusions:HSP90 interacted with DDX5 protein and subsequently protected DDX5 protein from AMPK/ULK1-regulated autophagic degradation.DDX5 and HSP90 are therefore potential therapeutic targets for HCC. | Ting Zhang Xinrui Yang Wanping Xu Jing Wang Dawei Wu Zhixian Hong Shengxian Yuan Zhen Zeng Xiaodong Jia Shanshan Lu Rifaat Safadi Sen Han Zhihong Yang Leonard M.Neckers Suthat Liangpunsakul Weiping Zhou Yinying Lu | 2021 | Cancer Biology & Medicine2021,18,3: | 7 |

| 5 | Fecal-associated microbiome differences between traditional Chinese medicine qi-deficiency and balanced constitutions显示文摘Objective:To explore the structural and functional characteristics of the fecal-associated microbiome(FAM)in a traditional Chinese medicine(TCM)qi-deficiency constitution(QDC)by comparing with balanced constitution(BC)and screen the related biomarkers.Methods:In this cross-sectional study,the TCM constitutions of subjects were determined based on published the Classification and Determination of constitution in TCM and further confirmed by a TCM clinician.Clinical characteristics were recorded,and fecal samples were collected for 16S rDNA sequencing using the Illumina Miseq platform.The FAM structure was described using alpha-diversity indexes,beta-diversity indexes,and the relative abundances of the dominant taxa.Differences in the FAM distribution and function were analyzed with a Wilcoxon rank-sum test,MetagenomeSeq,and LEfSe analysis,after which a receiver operating characteristic curve based on the specific operational taxonomic units(OTUs)was constructed to calculate the area under the curve.Results:Our study population was composed of 22 BCs and 9 QDCs.There were no significant differences between the two groups in the distribution of clinical characteristics or alpha-diversity indexes,except for the sweets preference and blood glucose level.In principal coordinate analysis and partial least squares discriminant analysis,the bacterial communities in the BC group samples and QDC group samples clustered separately.Notably,there were 214 OTUs significantly distributed between groups in the MetagenomeSeq analysis,200 OTUs identified by the Wilcoxon rank-sum test,and 6 OTUs found by the LEfSe analysis.Predicted function analysis revealed that six metabolic pathways were distinctly distributed between the two groups.The area under the curve for the receiver operating characteristic curve based on the four specific OTUs was 0.88.Conclusion:Unique FAM structural and related functional characteristics are displayed in individuals with a QDC,and four specific OTUs could be used as QDC biomarkers to assist in clinical diagnosis. | Jianhua Zhen Pengfei Zhao Guangrui Huang Yini Li Lu Zhao Yuhang Zhang Anlong Xu | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,2: | 1 |

| 17 | Structure and function of the fecal-associated microbiome in qi stagnation constitution显示文摘Objective:To clarify the structural and functional characteristics of the gut microbiota in individuals with qi stagnation constitution(QSC)and identify the potential biomarkers related to QSC.Methods:This cross-sectional study involved individuals with QSC and balanced constitution(BC)confirmed by TCM clinicians.The clinical features were recorded,and fecal samples were collected for 16S rDNA sequencing.The structure of the fecal-associated microbiome(FAM)was described by the alpha-diversity indexes,beta-distances,and relative abundances of dominant taxa.The differences in FAM distribution were analyzed by the Wilcoxon rank-sum test,MetagenomeSeq,and LEfSe analysis.The 16S rDNA gene sequences were assigned to the KEGG dataset to predict the functional information of bacterial metabolic pathways by using PICRUSt.Differences in functional pathways between groups were assessed with the Wilcoxon rank-sum test.The ROC curve based on specific operational taxonomic units(OTUs)was constructed,and the AUC was calculated.Results:Twenty-two individuals with BC and 8 with QSC were recruited.Significant differences between the two groups were found in body mass index,health status,and low-density lipoprotein,etc.There was no significant difference in the alpha-diversity index.PCoA showed no evident clustering of bacterial communities according to constitutions.Bacteroidaceae,Lachnospiraceae,Ruminococcaceae,and Prevotellaceae were the four common bacteria with high abundances.Notably,MetagenomeSeq,LEfSe analysis,and the Wilcoxon rank-sum test identified significantly different distributions of 66,42,and 36 OTUs,respectively.Predictive function analysis showed that 13 metabolic pathways were significantly differentially distributed,including those related to fatty acid synthesis.Five specific OTUs were selected as potential biomarkers of a QSC,and the AUC was 0.94.Conclusion:Individuals with QSC have unique FAM structure and related functional characteristics.Five specific OTUs were identified to serve as potentially effective biomarkers related to QSC. | Lu Zhao Pengfei Zhao Jianhua Zhen Guangrui Huang Yini Li Anlong Xu | 2021 | Journal of Traditional Chinese Medical Sciences2021,8,2: | 0 |
| 18 | m^(6)A demethylase ALKBH5 is required for antibacterial innate defense by intrinsic motivation of neutrophil migration显示文摘Neutrophil migration into the site of infection is necessary for antibacterial innate defense,whereas impaired neutrophil migration may result in excessive inflammation and even sepsis.The neutrophil migration directed by extracellular signals such as chemokines has been extensively studied,yet the intrinsic mechanism for determining neutrophil ability to migrate needs further investigation.N6-methyladenosine(m^(6)A)RNA modification is important in immunity and inflammation,and our preliminary data indicate downregulation of RNA m^(6)A demethylase alkB homolog 5(ALKBH5)in neutrophils during bacterial infection.Whether m^(6)A modification and ALKBH5 might intrinsically modulate neutrophil innate response remain unknown.Here we report that ALKBH5 is required for antibacterial innate defense by enhancing intrinsic ability of neutrophil migration.We found that deficiency of ALKBH5 increased mortality of mice with polymicrobial sepsis induced by cecal ligation and puncture(CLP),and Alkbh5-deficient CLP mice exhibited higher bacterial burden and massive proinflammatory cytokine production in the peritoneal cavity and blood because of less neutrophil migration.Alkbh5-deficient neutrophils had lower CXCR2 expression,thus exhibiting impaired migration toward chemokine CXCL2.Mechanistically,ALKBH5-mediated m^(6)A demethylation empowered neutrophils with high migration capability through altering the RNA decay,consequently regulating protein expression of its targets,neutrophil migration-related molecules,including increased expression of neutrophil migration-promoting CXCR2 and NLRP12,but decreased expression of neutrophil migration-suppressive PTGER4,TNC,and WNK1.Our findings reveal a previously unknown role of ALKBH5 in imprinting migrationpromoting transcriptome signatures in neutrophils and intrinsically promoting neutrophil migration for antibacterial defense,highlighting the potential application of targeting neutrophil m^(6)A modification in controlling bacterial infections. | Yang Liu Renjie Song Lu Zhao Zhike Lu Yini Li Xinyi Zhan Fengjiao Lu Jiang Yang Yamei Niu Xuetao Cao | 2022 | Signal Transduction and Targeted Therapy2022,7,7: | 0 |
| 19 | CREDIT-X1local:A reference dataset for machine learning seismology from ChinArray in Southwest China显示文摘High-quality datasets are critical for the development of advanced machine-learning algorithms in seismology.Here,we present an earthquake dataset based on the ChinArray Phase I records(X1).ChinArray Phase I was deployed in the southern north-south seismic zone(20°N-32°N,95°E-110°E)in 2011-2013 using 355 portable broadband seismic stations.CREDIT-X1local,the first release of the ChinArray Reference Earthquake Dataset for Innovative Techniques(CREDIT),includes comprehensive information for the 105,455 local events that occurred in the southern north-south seismic zone during array observation,incorporating them into a single HDF5 file.Original 100-Hz sampled three-component waveforms are organized by event for stations within epicenter distances of 1,000 km,and records of≥200 s are included for each waveform.Two types of phase labels are provided.The first includes manually picked labels for 5,999 events with magnitudes≥2.0,providing 66,507 Pg,42,310 Sg,12,823 Pn,and 546 Sn phases.The second contains automatically labeled phases for 105,442 events with magnitudes of−1.6 to 7.6.These phases were picked using a recurrent neural network phase picker and screened using the corresponding travel time curves,resulting in 1,179,808 Pg,884,281 Sg,176,089 Pn,and 22,986 Sn phases.Additionally,first-motion polarities are included for 31,273 Pg phases.The event and station locations are provided,so that deep learning networks for both conventional phase picking and phase association can be trained and validated.The CREDIT-X1local dataset is the first million-scale dataset constructed from a dense seismic array,which is designed to support various multi-station deep-learning methods,high-precision focal mechanism inversion,and seismic tomography studies.Additionally,owing to the high seismicity in the southern north-south seismic zone in China,this dataset has great potential for future scientific discoveries. | Lu Li Weitao Wang Ziye Yu Yini Chen | 2024 | Earthquake Science2024,37,2: | 0 |
| 20 | From liver fibrosis to hepatocarcinogenesis: Role of excessive liver H_(2)O_(2) and targeting nanotherapeutics显示文摘Liver fibrosis and hepatocellular carcinoma(HCC)have been worldwide threats nowadays.Liver fibrosis is reversible in early stages but will develop precancerosis of HCC in cirrhotic stage.In pathological liver,excessive H_(2)O_(2) is generated and accumulated,which impacts the functionality of hepatocytes,Kupffer cells(KCs)and hepatic stellate cells(HSCs),leading to genesis of fibrosis and HCC.H_(2)O_(2) accumulation is associated with overproduction of superoxide anion(O_(2)^(·-))and abolished antioxidant enzyme systems.Plenty of therapeutics focused on H_(2)O_(2) have shown satisfactory effects against liver fibrosis or HCC in different ways.This review summarized the reasons of liver H_(2)O_(2) accumulation,and the role of H_(2)O_(2) in genesis of liver fibrosis and HCC.Additionally,nanotherapeutics targeting H_(2)O_(2) were summarized for further consideration of antifibrotic or antitumor therapy. | Meiyu Shao Yifan Wang Hongyan Dong Lu Wang Xiaoqing Zhang Xin Han Xianan Sang Yini Bao Mengyun Peng Gang Cao | 2023 | Bioactive Materials2023,,5: | 0 |
