1Redox-sensitive prodrug nanoassemblies based on linoleic acid-modified docetaxel to resist breast cancers显示文摘Prodrug nanoassemblies, which can refrain from large excipients, achieve higher drug loading and control drug release, have been placed as the priority in drug delivery system. Reasoning that glutathione(GSH) and reactive oxygen species(ROS) are highly upgraded in tumor tissues which makes them attractive targets for drug delivery system, we designed and synthetized a novel prodrug which utilized mono thioether bond as a linker to bridge linoleic acid(LA) and docetaxel(DTX). This mono thioether-linked conjugates(DTX-S-LA) could self-assemble into nanoparticles without the aid of much excipients. The mono thioether endowed the nanoparticles redox sensitivity resulting in specific release at the tumor tissue. Our studies demonstrated that the nanoassemblies had uniform particle size, high stability and fast release behavior. DTX-S-LA nanoassemblies outperformed DTX solution in pharmacokinetic profiles for it had longer circulation time and higher area under curve(AUC). Compared with DTX solution, the redox dual-responsive nanoassemblies had comparable cytotoxic activity. Besides, the antitumor efficacy was evaluated in mice bearing 4 T1 xenograft. It turned out this nanoassemblies couldenhance anticancer efficacy by increasing the dose because of higher tolerance. 2An Arctic sea ice thickness variability revealed from satellite altimetric measurements显示文摘A modified algorithm taking into account the first year(FY) and multiyear(MY) ice densities is used to derive a sea ice thickness from freeboard measurements acquired by satellite altimetry ICESat(2003–2008). Estimates agree with various independent in situ measurements within 0.21 m. Both the fall and winter campaigns see a dramatic extent retreat of thicker MY ice that survives at least one summer melting season. There were strong seasonal and interannual variabilities with regard to the mean thickness. Seasonal increases of 0.53 m for FY the ice and 0.29 m for the MY ice between the autumn and the winter ICESat campaigns, roughly 4–5 month separation, were found. Interannually, the significant MY ice thickness declines over the consecutive four ICESat winter campaigns(2005–2008) leads to a pronounced thickness drop of 0.8 m in MY sea ice zones. No clear trend was identified from the averaged thickness of thinner, FY ice that emerges in autumn and winter and melts in summer. Uncertainty estimates for our calculated thickness, caused by the standard deviations of multiple input parameters including freeboard, ice density, snow density, snow depth, show large errors more than 0.5 m in thicker MY ice zones and relatively small standard deviations under 0.5 m elsewhere. Moreover, a sensitivity analysis is implemented to determine the separate impact on the thickness estimate in the dependence of an individual input variable as mentioned above. 4Arctic sea ice volume export through the Fram Strait from combined satellite and model data:1979–2012显示文摘By combing satellite-derived ice motion and concentration with ice thickness fields from a popular model PIOMAS we obtain the estimates of ice volume flux passing the Fram Strait over the 1979–2012 period. Since current satellite and field observations for sea ice thickness are limited in time and space, the use of PIOMAS is expected to fill the gap by providing temporally continued ice thickness fields. Calculated monthly volume flux exhibits a prominent annual cycle with the peak record in March(roughly 145 km3/month) and the trough in August(10 km^3/month). Annual ice volume flux(1 132 km^3) is primarily attributable to winter(October through May) outflow(approximately 92%). Uncertainty in annual ice volume export is estimated to be 55 km^3(or 5.7%). Our results also verified the extremely large volume flux appearing between late 1980 s and mid-1990 s. Nevertheless, no clear trend was found in our volume flux results. Ice motion is the primary factor in the determination of behavior of volume flux. Ice thickness presented a general decline trend may partly enhance or weaken the volume flux trend. Ice concentration exerted the least influences on modulating trends and variability in volume flux. Moreover, the linkage between winter ice volume flux and three established Arctic atmospheric schemes were examined. 5Differentiation of neuron-like cells from mouse parthenogenetic embryonic stem cells显示文摘Parthenogenetic embryonic stem cells have pluripotent differentiation potentials,akin to fertilized embryo-derived embryonic stem cells.The aim of this study was to compare the neuronal differentiation potential of parthenogenetic and fertilized embryo-derived embryonic stem cells.Before differentiation,karyotype analysis was performed,with normal karyotypes detected in both parthenogenetic and fertilized embryo-derived embryonic stem cells.Sex chromosomes were identified as XX.Immunocytochemistry and quantitative real-time PCR detected high expression of the pluripotent gene,Oct4,at both the mRNA and protein levels,indicating pluripotent differentiation potential of the two embryonic stem cell subtypes.Embryonic stem cells were induced with retinoic acid to form embryoid bodies,and then dispersed into single cells.Single cells were differentiated in N2 differentiation medium for 9 days.Immunocytochemistry showed parthenogenetic and fertilized embryo-derived embryonic stem cells both express the neuronal cell markers nestin,βIII-tubulin and myelin basic protein.Quantitative real-time PCR found expression of neurogenesis related genes(Sox-1,Nestin,GABA,Pax6,Zic5 and Pitx1) in both types of embryonic stem cells,and Oct4 expression was significantly decreased.Nestin and Pax6 expression in parthenogenetic embryonic stem cells was significantly higher than that in fertilized embryo-derived embryonic stem cells.Thus,our experimental findings indicate that parthenogenetic embryonic stem cells have stronger neuronal differentiation potential than fertilized embryo-derived embryonic stem cells.Xingrong Yan Yanhong Yang Wei Liu Wenxin Geng Huichong Du Jihong Cui Xin Xie Jinlian Hua Shumin Yu Liwen Li Fulin Chen 2013Neural Regeneration Research2013,8,4:4

8Testing the role of genetic variation of the MC4R gene in Chinese population in antipsychotic-induced metabolic disturbance显示文摘Antipsychotic-induced metabolic disturbance(AIMD) is a common adverse effect of antipsychotics with genetics partly underpinning variation in susceptibility among schizophrenia patients. Melanocortin4 receptor(MC4 R) gene, one of the candidate genes for AIMD, has been under-studied in the Chinese patients. We conducted a pharmacogenetic study in a large cohort of Chinese patients with schizophrenia. In this study, we investigated the genetic variation of MC4 R in Chinese population by genotyping two SNPs(rs489693 and rs17782313) in 1,991 Chinese patients and examined association of these variants with the metabolic effects that were often observed to be related to AIMD. Metabolic measures, including body mass index(BMI), waist circumference(WC), glucose, triglyceride, high-density lipoprotein(HDL), and low-density lipoprotein(LDL) levels were assessed at baseline and after 6-week antipsychotic treatment. We found that interaction of SNP×medication status(drug-na?ve/medicated) was significantly associated with BMI, WC, and HDL change %, respectively. Both SNPs were significantly associated with baseline BMI and WC in the medicated group. Moderate association of rs489693 with WC, Triglyceride, and HDL change % were observed in the whole sample. In the drug-na?ve group, we found recessive effects of rs489693 on BMI gain more than 7%, WC and Triglyceride change %, with AA incurring more metabolic adverse effects. In conclusion, the association between rs489693 and the metabolic measures is ubiquitous but moderate. Rs17782313 is less involved in AIMD. 9Exposure measurement, risk assessment and source identification for exposure of traffic assistants to particle-bound PAHs in Tianjin, China显示文摘To investigate the levels of exposure to particulate-bound polycyclic aromatic hydrocarbon(PAH) and to estimate the risk these levels pose to traffic assistants(TAs) in Tianjin(a megacity in North China), a measurement campaign(33 all-day exposure samples, 25 occupational-exposure samples and 10 indoor samples) was conducted to characterize the TAs' exposure to PAHs, assess the cancer risk and identify the potential sources of exposure. The average total exposure concentration of 14 PAHs was approximately 2871 ± 928 ng/m3(on-duty), and 1622 ± 457 ng/m3(all-day). The indoor PAHs level was 1257 ± 107 ng/m3. After 8000 Monte Carlo simulations, the cancer risk resulting from exposure to PAHs was found to be approximately 1.05 × 10-4. A multivariate analysis was applied to identify the potential sources, and the results showed that, in addition to vehicle exhaust, coal combustion and cooking fumes were also another two important contributors to personal PAH exposure. 10Synthesis of Ag@Al_2O_3 Core-shell Structure Nanoparticles and Their Enhancement Effect on Dielectric Properties for Ag@Al_2O_3/Polyimide Nanocomposites显示文摘A novel core-shell structure Ag@Al2O3 nano-particles were synthesized and doped into polyimide as conductive fillers to prepare the composite films with high dielectric properties and low dielectric loss. The morphology and structures of the Ag@Al2O3 nano-particles were characterized by transmission electron microscopy(TEM), X-ray diffraction(XRD), and UV-visible spectroscopy. All the results proved that the Ag@Al2O3 nano-particles had a typical core-shell structure, for the Ag particles were coated by Al2O3 shell and the average sizes of Ag@Al2O3 particles were between 30 to 150 nm. The as-prepared Ag@Al2O3 nanoparticles were doped into the polyimide with different mass fractions to fabricate the Ag@Al2O3/PI composite films via in-situ polymerization process. SEM analysis of composite films showed that the Ag@Al2O3 nanoparticles homogeneously dispersed in polyimide matrix with nanoscale. As dielectric materials for electronic packaging systems, the Ag@Al2O3/PI composites exhibited appropriate mechanical properties and enhanced dielectric properties, including greatly enhanced dielectric constant and just a slight increase in dielectric loss. 12Effects of Suspended Culture of the Seaweed Laminaria japonica Aresch on the Flow Structure and Sedimentation Processes显示文摘The culture of suspended kelp, such as Laminaria japonica Aresch, has arisen in nearshore areas for approximately 30 years since the 1980 s. This long-term activity has significant impact on the regional hydrodynamic and sedimentary environments. In this study the impact was investigated, based on synchronized multi-station data from continuous observations made within and around the culture area. In total, three current velocity profiles were identified inside and on the landward side of the culture area. Based on the current velocity profiles we calculated the boundary layer parameters, the fluxes of erosion/deposition, and the rate of sediment transport in different times at each observation site. Comparison between culture and non-culture periods showed that the presence of suspended kelp caused the reduction in the average flow velocity by approximately 49.5%, the bottom friction velocity by 24.8%, the seabed roughness length by 62.7%, and the shear stress and the flux of resuspended sediment by approximately 50%. From analyses in combination with the corresponding vertical variation of the suspended sediment distribution, it is revealed that the lifted sediments by resuspension is mixed with the upper suspended material, which will modify the regional distribution of suspended sediment. These changes in flow structure and sediment movement will accelerate seabed siltation, which corresponds to the changes in seabed erosion/deposition. 18Operation management of green ports and shipping networks: overview and research opportunities显示文摘Global ports and maritime shipping networks are important carriers for global supply chain networks, but they are also the main sources of energy consumption and pollution. To limit ship emissions in ports and offshore areas, the International Maritime Organization, as well as some countries, has issued a series of policies. This study highlights the importance and necessity of investigating emergent research problems in the operation management of green ports and maritime shipping networks. Considerable literature related to this topic is reviewed and discussed. Moreover, a comprehensive research framework on green port and shipping operation management is proposed for future research opportunities. The framework mainly comprises four research areas related to emission control and grading policies. 20Bioinspired Functional Surfaces for Medical Devices显示文摘Medical devices are a major component of precision medicine and play a key role in medical treatment,particularly with the rapid development of minimally invasive surgery and wearable devices.Their tissue contact properties strongly affect device performance and patient health(e.g.,heat coagulation and slipperiness on surgical graspers).However,the design and optimization of these device surfaces are still indistinct and have no supporting principles.Under such conditions,natural surfaces with various unique functions can provide solutions.This review summarizes the current progress in natural functional surfaces for medical devices,including ultra-slipperiness and strong wet attachment.The underlying mechanisms of these surfaces are attributed to their coupling effects and featured micronano structures.Depending on various medical requirements,adaptable designs and fabrication methods have been developed.Additionally,various medical device surfaces have been validated to achieve enhanced contact properties.Based on these studies,a more promising future for medical devices can be achieved for enhanced precision medicine and human health.Liwen Zhang Guang Liu Yurun Guo Yan Wang Deyuan Zhang Huawei Chen 2022Chinese Journal of Mechanical Engineering2022,35,3:1
