1Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5

3Cellulose-hydrogen production from corn stalk biomass by anaerobic fermentation显示文摘Cellulose-hydrogen production from corn stalk by lesser panda manure was carried out in batch tests and a 5 L scale-up continuously stirred anaerobic bioreactor(CSABR),respectively.The bio-pretreatment of corn stalk was found most effective at 25℃ using microbe additive of 7.5 g/kg,in which the yields of soluble saccharides(SS) and lactic acid were 212 mg/g-TS and 21 mg/g-TS,respectively.The maximum cumulative H2 yield(176 ml/g-TS) and H2 production rate(14.5 ml/g-TS h-1) were obtained at pH 5.5,36℃ by treating a substrate of 15 g/L.The hydrogen content in biogas was 57.2% and there was no significant methane gas observed.During the optimal period of H2 production,the ORP values stayed in the lower level ranging from -445 mV to -455 mV.The results show that the bio-pretreatment of the raw materials played a vital role in the effective conversion of corn stalk into cellulose-hydrogen by mixed culture.XING Yan MA HongCui FAN YaoTing HOU HongWei CHEN JingRun 2009Chinese Science Bulletin2009,54,8:3

5Triptolide protects astrocytes from hypoxia/reoxygenation injury显示文摘Astrocytes in an in vitro murine astrocyte model of oxygen and glucose deprivation/hypoxia and reoxygenation were treated with different concentrations of triptolide (250, 500, 1 000 ng/mL) in a broader attempt to elucidate the protection and mechanism underlying triptolide treatment on astrocytes exposed to hypoxia/reoxygenation injury. The results showed that the matrix metalloproteinase-9, interleukin-1β, tumor necrosis factor α and interleukin-6 expressions were significantly decreased after triptolide treatment in the astrocytes exposed to hypoxia/ reoxygenation injury, while interleukin-10 expression was upregulated. In addition, the vitality of the injured astrocytes was enhanced, the triptolide's effect was apparent at 500 ng/mL.
