5Oxidized mitochondrial DNA sensing by STING signaling promotes the antitumor effect of an irradiated immunogenic cancer cell vaccine显示文摘Exposure to ionizing radiation,a physical treatment that inactivates live tumor cells,has been extensively applied to enhance the antitumor responses induced by cancer cell vaccines in both animal research and human clinical trials.However,the mechanisms by which irradiated cells function as immunogenic tumor vaccines and induce effective antitumor responses have not been fully explored.Here,we demonstrate that oxidized mitochondrial DNA(mtDNA)and stimulator of interferon genes(STING)signaling play a key roles in the enhanced antitumor effect achieved with an irradiated tumor cell vaccine.Elevations in ROS and oxidized mtDNA 8-OHG content could be induced in irradiated tumor cells.Oxidized mtDNA derived from irradiated tumor cells gained access to the cytosol of dendritic cells(DCs).Oxidized mtDNA,as a DAMP or adjuvant,activated the STING-TBK1-IRF3-IFN-β pathway in DCs,which subsequently cross-presented irradiated tumor cell-derived antigens to CD8^(+)T cells and elicited antitumor immunity.The results of our study provide insight into the mechanism by which an irradiated cell vaccine mediates antitumor immunity,which may have implications for new strategies to improve the efficacy of irradiated vaccines.Chunju Fang Fei Mo Li Liu Jing Du Min Luo Ke Men Feifei Na Wei Wang Hanshuo Yang Xiawei Wei 2021Cellular & Molecular Immunology2021,18,9:2

8Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1

13Mapping of individual sensory nerve axons from digits to spinal cord with the transparent embedding solvent system显示文摘Achieving uniform optical resolution for a large tissue sample is a major challenge for deep imaging.For conventional tissue clearing methods,loss of resolution and quality in deep regions is inevitable due to limited transparency.Here we describe the Transparent Embedding Solvent System(TESOS)method,which combines tissue clearing,transparent embedding,sectioning and block-face imaging.We used TESOS to acquire volumetric images of uniform resolution for an adult mouse whole-body sample.The TESOS method is highly versatile and can be combined with different microscopy systems to achieve uniformly high resolution.With a light sheet microscope,we imaged the whole body of an adult mouse,including skin,at a uniform 0.8×0.8×3.5μm^(3) voxel resolution within 120 h.With a confocal microscope and a 40×/1.3 numerical aperture objective,we achieved a uniform sub-micron resolution in the whole sample to reveal a complete projection of individual nerve axons within the central or peripheral nervous system.Furthermore,TESOS allowed the first mesoscale connectome mapping of individual sensory neuron axons spanning 5 cm from adult mouse digits to the spinal cord at a uniform sub-micron resolution.Yating Yi Youqi Li Shiwen Zhang Yi Men Yuhong Wang Dian Jing Jiayi Ding Qingjie Zhu Zexi Chen Xingjun Chen Jun-Liszt Li Yilong Wang Jun Wang Hanchuan Peng Li Zhang Wenjing Luo Jian Q.Feng Yongwen He Woo-Ping Ge Hu Zhao 2024Cell Research2024,34,2:0
14A Heavy Metal Analysis Device Based on Light-addressable Potentiometric Sensor显示文摘<正>A full automatic device to detect heavy metal Hg,Fe,Cr elements based on thin-film sensitive materials prepared on surface of light-addressable potentiometric sensors (LAPS) by means of pulsed laser deposition PLD) technique is developed. 15Research on error accumulation control of three-dimensional adjustment with offset constraint显示文摘Background Currently,laser tracker is the primary instrument used to carry out three-dimensional position measurement in accelerator alignment.Theoretically,three-dimensional measuring data processed by three-dimensional adjustment are more rigorous,however,error accumulation is found in practice.Purpose In order to control error accumulation and further improve the measurement accuracy of accelerator alignment,this research introduces the laser alignment system into the activity of measurement and data processing.Methods A measurement scheme combining laser tracker and laser alignment system is proposed.To construct the constraint condition,the offset values from the measuring points to the laser straight-line datum were used.To carry out the three-dimensional adjustment with offset constraint,the laser tracker observations were used.Results A three-dimensional adjustment function model of laser tracker observations is given.The construction method of the constraint equation is researched,and the calculation formulas of the three-dimensional adjustment with offset constraint are derived.A 200 m linac tunnel control network is designed,using simulation measurement method,the measuring data of laser tracker and the offset values from the measuring points to the laser straight-line datum were generated.The simulated data are calculated by the method given in this paper and the result is analyzed.Conclusion Simulation result shows introducing the laser alignment system into laser tracker measurement and applying the three-dimensional adjustment with offset constraint can effectively suppress the error accumulation caused by long distance move station measurement.Wang Xiaolong Kang Ling Dong Lan Li Bo Men Lingling Luo Tao Wang Tong Liang Jing He Zhenqiang Ke Zhiyong Ma Na Lu Shang Han Yuanying Yan Luping Zhang Luyan Liu Xiaoyang Yan Haoyue Li Chunhua Wu Lei 2022Radiation Detection Technology and Methods2022,6,4:0
163' Noncoding Region Construction of GHR Gene-luciferase Report Vector and Valuation显示文摘To analyze miR-139 target sites in 3' UTR of GHR gene in dairy cow mammary gland, a GHR 3' UTR- luciferase reporter vector was constructed and the effect of miRNA on its activity was evaluated in dairy cow mammary gland epithelial cells (DCMECs). The miR-139 targeting GHR 3' UTR was predicted by Target Scan 5.1 software, 3' UTR fragment of GHR was amplified by PCR from RNA of DCMECs. PCR products were cloned into SpeⅠ/Hind Ⅲ modified pMIR-Report vector. The luciferase reporter vector and miRNA eukaryotic expression vector were transferred into DCMECs using lipofectamine 2000 transfection reagent. The dual-luciferase reporter assay system was used to quantitiate the reporter activity. The results showed that a 107 bp 3' UTR fragment of GHR gene was successfully cloned into the pMIR-Report vector, which authenticated by SpeⅠ/Hind Ⅲ digestion and DNA sequencing. The luciferase activity of reporter construction treated with miR-139 decreased 20.87% compared with the control group. It was concluded that the GHR3' UTR-luciferase reporter vector had been successfully constructed. 17Facile fabrication of covalent organic framework functionalized superhydrophobic porous sponges for highly efficient water/oil separation显示文摘Considering the serious environmental pollution and economic losses caused by increasing oil spills and chemical leaks,effective and eco-friendly methods of oil/water separation should be urgently developed.Herein,a facile strategy(about 20 min)was developed for integrating covalent organic framework(COF)particles with melamine sponges(MS)by covalent bonding,and then a superhydrophobic COF/MS composite(denoted OA-CP@MS)could be further obtained by following modification with a 1-octadecylamine/water dispersion.Specifically,the synthesis process was developed under a mild condition that avoids the need for sealing and high-temperature treatments.It was proven that the fabricated OA-CP@MS presented a prominent absorption capacity for various organic reagents,in which adsorption capacity of chloroform could be up to 173 g/g.Owing to its excellent chemical robustness in strong alkaline and saline environments and in various organic solvents,the prepared OACP@MS showed a great potential to perform oil/water separation in complex practical conditions.Its rapid preparation process,fluorine-free treatment,and water-based modification would extend the application of COF materials in oil/water separation.WANG Jing WANG FuShan ZHOU YuJian ZHOU YongJun GAO Peng MEN XueHu 2022Science China(Technological Sciences)2022,65,11:0
18Promoting effect of periodic case analysis test on pathophysiology teaching显示文摘AIM:To explore the effect of the periodic case analysis test on pathophysiology teaching.METHODS:We randomly selected two natural classes from Grade 2011 Clinical Medicine Specialty in our university and set them as experimental group and control group.There were 66 and 68 students in the two groups,respectively.For these two groups,the course background,pathophysiology course teaching and final exam conditions were all the same.In the teaching process,we had a written test every 8 teaching hours,5 times in each group.In experimental group,students were arranged to face some typical clinical cases,they must analyze and discuss the specific pathogenesis according to the recent theoretical knowledge.In the same teaching time,the control students need to complete a certain amount of traditional type homework which was mainly basic concepts,basic pathogenesis and no practical case analysis.We compared and analyzed the final test scores and the pass rates in the two groups,and calculated the mean rank sums of the examination results with nonparametric method Kruskal-Wallis test.RESULTS:The average score of the final exam was 82.69 in experimental group,but that in control group was 77.98.The pass rates were 89.5%and 81.2%in the two groups,respectively.The average test rank scores were 171.84 and 136.95,respectively.CONCLUSION:The periodic case analysis test can obviously improve the students'academic achievements,and has promoting effect on pathophysiology teaching.ZHAO Li-jun MEN Xiu-li WU Jing KONG Xiao-yan LI Hong-jie LIU Li-hua 2013中国病理生理杂志2013,29,10:0
