2Genome Engineering in Rice Using Cas9 Variants that Recognize NG PAM Sequences显示文摘CRISPR/Cas9 genome editing relies on sgRNA-target DNA base pairing and a short downstream PAM sequence to recognize target DNA. The strict protospacer adjacent motif (PAM) requirement hinders applications of the CRISPR/Cas9 system since it restricts the targetable sites in the genomes. xCas9 and SpCas9-NG are two recently engineered SpCas9 variants that can recognize more relaxed NG PAMs, implying a great potential in addressing the issue of PAM constraint. Here we use stable transgenic lines to evaluate the efficacies of xCas9 and SpCas9-NG in performing gene editing and base editing in rice. We found that xCas9 can efficiently induce mutations at target sites with NG and GAT PAM sequences in rice. However, base editors containing xCas9 failed to edit most of the tested target sites. SpCas9-NG exhibited a robust editing activity at sites with various NG PAMs without showing any preference for the third nucleotide after NG. Moreover, we showed that xCas9 and SpCas9-NG have higher specificity than SpCas9 at the CGG PAM site. We further demonstrated that different forms of cytosine or adenine base editors containing SpCas9-NG worked efficiently in rice with broadened PAM compatibility. 3Gait Analysis of Quadruped Robot Using the Equivalent Mechanism Concept Based on Metamorphosis显示文摘The previous research regarding the gait planning of quadruped robot focuses on the sequence for lifting o and placing the feet, but neglects the influence of body height. However, body height a ects gait performance significantly, such as in terms of the stride length and stability margin. We herein study the performance of a quadruped robot using the equivalent mechanism concept based on metamorphosis. Assuming the constraints between standing feet and the ground with hinges, the ground, standing legs and robot body are considered as a parallel mechanism, and each swing leg is regarded as a typical serial manipulator. The equivalent mechanism varies while the robot moves on the ground. One gait cycle is divided into several periods, including step forward stages and switching stages. There exists a specific equivalent mechanism corresponding to each gait period. The robot's locomotion can be regarded as the motion of these series of equivalent mechanisms. The kinematics model and simplified model of the equivalent mechanism is established. A new definition of the multilegged robot stability margin, based on friction coe cient, is presented to evaluate the robot stability. The stable workspaces of the equivalent mechanism in the step forward stage of trotting gait under di erent friction coe cients are analyzed. The stride length of the robots is presented by analyzing the relationship between the stable workspaces of the equivalent mechanisms of two adjacent step forward stages in one gait cycle. The simulation results show that the stride length is larger with increasing friction coe cient. 6Needs and concerns of patients in isolation care units - learnings from COVID-19: A reflection显示文摘With strict measures in place to contain the spread of coronavirus disease 2019,many have been isolated as suspected or confirmed cases.Being isolated causes much inconvenience for the patients and family.Patients'and next-of-kins’needs and concerns during isolation will be shared together with suggestions for key process improvements.Our hospital’s Senior Patient Experience Managers contact all patients admitted to the isolation wards on a daily basis to provide some form of support.Common issues raised were gathered and strategies to help with their needs and concerns were discussed.Being in isolation is a challenging period for both patients and family.Nonetheless,we can implement measures to mitigate against the adverse effects of isolation.Patient education,effective and efficient means of communication,close monitoring for signs of distress and anxiety,and early intervention could help patients cope better with the whole isolation experience.Nursing management may want to consider implementing the measures shared in the article to manage patient’s stress while not compromising on staff safety.Peijin Esther Monica Fan Fazila Aloweni Shu Hui Lim Shin Yuh Ang Karen Perera Aik Huan Quek Hwee Koon Susan Quek Tracy Carol Ayre 2020World Journal of Clinical Cases2020,8,10:1

12Elaborative collection of condensed combustion products of solid propellants:Towards a real Solid Rocket Motor(SRM)operational environment显示文摘A novel constant-pressure and constant-quenching distance Condensed Combustion Products(CCPs)collection system was developed,coupled with a timing control system,to collect the CCPs formed in the course of burning of aluminum-based composite propellants.The effects of adiabatic graphite plating,collection zone,quenching distance,time series of collection,and propellant burning rate on the microscopic morphology,particle size distribution and unburned aluminum content of CCPs were investigated.It was verified that the graphite plating can provide a high-fidelity high-temperature environment for propellant combustion.The combustion efficiency is improved by 2.44% compared to the bare propellant case.The time series of collection has a significant effect on the combustion efficiency of aluminum,and the combustion efficiency of aluminum in the thermal state(1.2-2.4 s)is 2.75% higher than that in the cold state(0-1.2 s).Similarly,the characteristics of the CCPs in different collection zones are different.At the quenching distance of 5 mm,the combustion efficiency of aluminum in the core zone(85.39%)is much lower than that in the outer zone(92.07%),while the particle size of the CCPs in the core zone(172μm)is larger than that in the outer zone(41μm).This indicates that the core zone is more likely to produce large-sized and incompletely burned agglomerates during the propellant combustion process.Different burning rates also lead to a significant difference in particle size distribution and combustion efficiency.High burning rates result in higher combustion efficiency.A detailed sequence of the elaborative collection process of CCPs is proposed,mainly including the setting of ignition delay time,burning rate,working pressure,plating length and time series of collection.The findings of this study are expected to provide a reliable tool for the evaluation of the combustion efficiency of solid propellants.Wenchao ZHANG Zhimin FAN Dongliang GOU Yao SHU Peijin LIU Aimin PANG Wen AO 2024Chinese Journal of Aeronautics2024,37,1:0
13Identification of Glyphosate Herbicide with Ion Chromatographic Column Switching Method显示文摘[Objective] The paper was to establish the method for quantitative analysis of glyphosate herbicide and identification of salt type. [Method] Using ion chromatographic column switching method,cations( sodion,ammonium ion,potassium ion,dimethonium ion and isopropylamine ion) and glyphosate anion in glyphosate salt were determined with Thermo IonP ac SCS1 cation exchange column and Thermo IonP ac AS14 anion exchange column,and external standard method was used for quantification. 15Geographical classification of Nanfeng mandarin by near infrared spectroscopy coupled with chemometrics methods显示文摘Near infrared spectroscopy(NIRS),coupled with principal component analysis and wavelength selection techniques,has been sed to develop a robust and reliable reduced-spectrum classifi-cation model for determining the geographical origins of Nanfeng mandarins.The application of the changeable size moving window principal component analysis(CSMWPCA)provided a notably improved lassification model,with correct classification rates of 92.00%,100.00%,90.00%,100.00%,100.00%,100.00%and 100.00%for Fujian,Guangxi,Hunan,Baishe,Baofeng,Qiawan,Sanxi samples,respectively,as well as,a total dassification rate of 97.52%in the wavelength range from 1007 to 1296 nm.To test and apply the proposed method,the procedure was applied to the analysis of 59 samples in an independent test set.Good identification results(correct rate of 96.61%)were also received.The improvement achieved by the application of CSMWPCA method was particularly remarkable when taking the low complexities of the final model(290 variables)into account.The results of the study showed the great potential of NIRS as a fast,nondestructive and environmentally acceptable method for the rapid and reliable determination for geographical classifcation of Nanfeng mandarins.Xuan Zhang Yiping Du Peijin Tong Yuanlong Wei Man Wang 2014Journal of Innovative Optical Health Sciences2014,7,6:0
16Learning from biological attachment devices:applications of bioinspired reversible adhesive methods in robotics显示文摘Many organisms have attachment organs with excellent functions,such as adhesion,clinging,and grasping,as a result of biological evolution to adapt to complex living environments.From nanoscale to macroscale,each type of adhesive organ has its own underlying mechanisms.Many biological adhesive mechanisms have been studied and can be incorporated into robot designs.This paper presents a systematic review of reversible biological adhesive methods and the bioinspired attachment devices that can be used in robotics.The study discussed how biological adhesive methods,such as dry adhesion,wet adhesion,mechanical adhesion,and sub-ambient pressure adhesion,progress in research.The morphology of typical adhesive organs,as well as the corresponding attachment models,is highlighted.The current state of bioinspired attachment device design and fabrication is discussed.Then,the design principles of attachment devices are summarized in this article.The following section provides a systematic overview of climbing robots with bioinspired attachment devices.Finally,the current challenges and opportunities in bioinspired attachment research in robotics are discussed.Kun XU Peijin ZI Xilun DING 2022Frontiers of Mechanical Engineering2022,17,3:0

18Topology Optimization for Harmonic Excitation Structures with Minimum Length Scale Control Using the Discrete Variable Method显示文摘Continuumtopology optimization considering the vibration response is of great value in the engineering structure design.The aimof this studyis toaddress the topological designoptimizationof harmonic excitationstructureswith minimumlength scale control to facilitate structuralmanufacturing.Astructural topology design based on discrete variables is proposed to avoid localized vibration modes,gray regions and fuzzy boundaries in harmonic excitation topology optimization.The topological design model and sensitivity formulation are derived.The requirement of minimum size control is transformed into a geometric constraint using the discrete variables.Consequently,thin bars,small holes,and sharp corners,which are not conducive to the manufacturing process,can be eliminated from the design results.The present optimization design can efficiently achieve a 0–1 topology configuration with a significantly improved resonance frequency in a wide range of excitation frequencies.Additionally,the optimal solution for harmonic excitation topology optimization is not necessarily symmetric when the load and support are symmetric,which is a distinct difference fromthe static optimization design.Hence,one-half of the design domain cannot be selected according to the load and support symmetry.Numerical examples are presented to demonstrate the effectiveness of the discrete variable design for excitation frequency topology optimization,and to improve the design manufacturability.Hongliang Liu Peijin Wang Yuan Liang Kai Long Dixiong Yang 2023Computer Modeling in Engineering & Sciences2023,,6:0
