1Immunomodulatory properties of mesenchymal stem cells/dental stem cells and their therapeutic applications显示文摘Mesenchymal stem/stromal cells(MsCs)are widely distributed in the body and play essential roles in tissue regeneration and homeostasis.MsCs can be isolated from discarded tissues,expanded in vitro and used as therapeutics for autoimmune diseases and other chronic disorders.MsCs promote tissue regeneration and homeostasis by primarily acting on immune cells.At least six different types of MsCs have been isolated from postnatal dental tissues and have remarkable immunomodulatory properties.Dental stem cells(DsCs)have been demonstrated to have therapeutic effects on several systemic inflammatory diseases.Conversely,MsCs derived from nondental tissues such as the umbilical cord exhibit great benefits in the management of periodontitis in preclinical studies.Here,we discuss the main therapeutic uses of MSCs/DSCs,their mechanisms,extrinsic inflammatory cues and the intrinsic metabolic circuitries that govern the immunomodulatory functions of MSCs/DSCs.Increased understanding of the mechanisms underpinning the immunomodulatory functions of MSCs/DSCs is expected to aid in the development of more potent and precise MSC/DSC-based therapeutics.Peishan Li Qianmin Ou Songtao Shi Changshun Shao 2023Cellular & Molecular Immunology2023,20,6:4
2CUL4B negatively regulates Toll-like receptor-triggered proinflammatory responses by repressing Pten transcription显示文摘Toll-like receptors (TLRs) play critical roles in innate immunity and inflammation. The molecular mechanisms by which TLR signaling is fine-tuned remain to be completely elucidated. Cullin 4B (CUL4B), which assembles the CUL4B-RING E3 ligase complex (CRL4B), has been shown to regulate diverse developmental and physiological processes by catalyzing monoubiquitination for histone modification or polyubiquitination for proteasomal degradation. Here, we identified the role of CUL4B as an intrinsic negative regulator of the TLR-triggered inflammatory response. Deletion of CUL4B in macrophages increased the production of proinflammatory cytokines and decreased anti-inflammatory cytokine IL-10 production in response to pathogens that activate TLR3, TLR4, or TLR2. Myeloid cell-specific Cul4b knockout mice were more susceptible to septic shock when challenged with lipopolysaccharide, polyinosinic-polycytidylic acid or Salmonella typhimurium infection. We further demonstrated that enhanced TLR-induced inflammatory responses in the absence of CUL4B were mediated by increased GSK3β activity. Suppression of GSK3β activity efficiently blocked the TLR-triggered increase in proinflammatory cytokine production and attenuated TLR-triggered death in Cul4b mutant mice. Mechanistically, CUL4B was found to negatively regulate TLR-triggered signaling by epigenetically repressing the transcription of Pten, thus maintaining the anti-inflammatory PI3K-AKT-GSK3β pathway. The upregulation of PTEN caused by CUL4B deletion led to uncontrolled GSK3β activity and excessive inflammatory immune responses. 4NAD^(+) salvage governs the immunosuppressive capacity of mesenchymal stem cells显示文摘Mesenchymal stem/stromal cells(MSCs)possess robust immunoregulatory functions and are promising therapeutics for inflammatory disorders.This capacity is not innate but is activated or‘licensed’by inflammatory cytokines.The licensing mechanism remains unclear.Here,we examined whether inflammatory cytokines metabolically reprogrammed MSCs to confer this immunoregulatory capacity.In response to stimulation by inflammatory cytokines,MSCs exhibited a dramatic increase in the consumption of glucose,which was accompanied by an enhanced use of nicotinamide adenine dinucleotide(NAD^(+))and increased expression of nicotinamide phosphoribosyltransferase(NAMPT),a central enzyme in the salvage pathway for NAD^(+) production.When NAD^(+) synthesis was blocked by inhibiting or depleting NAMPT,the immunosuppressive function of MSCs induced by inflammatory cytokines was greatly attenuated.Consequently,when NAD^(+) metabolism in MSCs was perturbed,their therapeutic benefit was decreased in mice suffering from inflammatory bowel disease and acute liver injury.Further analysis revealed that NAMPT-driven production of NAD^(+) was critical for the inflammatory cytokine-induced increase in glycolysis in MSCs.Furthermore,the increase in glycolysis led to succinate accumulation in the tricarboxylic acid cycle,which led to hypoxia-inducible factor 1α(HIF-1α)stabilization and subsequently increased the transcription of key glycolytic genes,thereby persistently maintaining glycolytic flux.This study demonstrated that unlike its proinflammatory role in immune cells,NAD^(+) metabolism governs the anti-inflammatory function of MSCs during inflammation.Jiankai Fang Pengbo Hou Shisong Liu Muqiu Zuo Zhanhong Liu Wangwang Chen Yuyi Han Yanan Li Tingting Wang Chao Feng Peishan Li Changshun Shao Yufang Shi 2023Cellular & Molecular Immunology2023,20,10:1

6Theoretical study of the ionospheric dynamo region inside the South Atlantic Anomaly显示文摘The South Atlantic Anomaly(SAA)is a region where the geomagnetic field is significantly lower than that of the surrounding area.On the basis of the models of CHAOS-7.8,Mass Spectrometer Incoherent Scatter Model(NRLMSISE-00),and International Reference Ionosphere 2016(IRI-2016),we theoretically investigated the lower and upper boundaries of the ionospheric dynamo region inside the SAA.In the ionospheric dynamo region,electrons are coupled with magnetic field lines,whereas ions are decoupled from magnetic field lines.Our results showed that the ionospheric dynamo region inside the SAA is higher and larger than that outside the SAA.We also studied the boundary variations of the dynamo region inside the SAA depending on the seasons and solar activities.We found that the dynamo region inside the SAA is the highest and largest in the summer of the southern hemisphere at solar maximum.The larger and higher altitude range of the ionospheric dynamo region in the SAA can contribute to the stronger ionospheric currents in this region.SiQi Yi XiaoJun Xu ZiLu Zhou Qing Chang Xing Wang Lei Luo PeiShan He Hui Li 2023Earth and Planetary Physics2023,7,1:1

11Perioperative dexmedetomidine administration does not reduce the risk of acute kidney injury after non-cardiac surgery:a meta-analysis显示文摘Background:Post-operative acute kidney injury(AKI)is one of the most common and serious complications after major surgery and is significantly associated with increased risks of morbidity and mortality.This meta-analysis was conducted to evaluate the effects of perioperative dexmedetomidine(Dex)administration on the occurrence of AKI and the outcomes of recovery after non-cardiac surgery.Methods:The PubMed,Embase,Web of Science,and Cochrane Library databases were systematically searched for studies comparing the effects of Dexvs.placebo on kidney function after non-cardiac surgery,and a pooled fixed-effect meta-analysis of the included studies was performed.The primary outcome was the occurence of post-operative AKI.The secondary outcomes included the occurence of intra-operative hypotension and bradycardia,intensive care unit(ICU)admission,duration of ICU stay,and hospital length of stay(LOS).Results:Six studies,including four randomized controlled trials(RCTs)and two observational studies,with a total of 2586 patients were selected.Compared with placebo,Dex administration could not reduce the odds of post-operative AKI(odds ratio[OR],0.44;95%confidence interval(CI),0.18-1.06;P=0.07;I^(2)=0.00%,P=0.72)in RCTs,but it showed a significant renoprotective effect(OR,0.67;95%CI,0.48-0.95;P=0.02;I^(2)=0.00%,P=0.36)in observational studies.Besides,Dex administration significantly increased the odds of intra-operative bradycardia and shortened the duration of ICU stay.However,there was no significant difference in the odds of intra-operative hypotension,ICU admission,and hospital LOS.Conclusions:This meta-analysis suggests that perioperative Dex administration does not reduce the risk of AKI after non-cardiac surgery.However,the quality of evidence for this result is low due to imprecision and inconsistent types of non-cardiac operations.Thus,large and high-quality RCTs are needed to verify the real effects of perioperative Dex administration on the occurrence of AKI and the outcomes of recovery after non-cardiac surgery.Bin Hu Tian Tian Xintao Li Weichao Liu Yinggui Chen Tianyu Jiang Peishan Chen Fushan Xue 2022Chinese Medical Journal2022,135,23:0

13Global Deaths Associated with Population Aging—1990–2019显示文摘Introduction:Published global and countryspecific deaths associated with population aging are based on decomposition methods that have significant limitations.Methods:A new decomposition method was developed and its performance was compared with two frequently used methods.The new method was employed to calculate global deaths associated with population aging between 1990 and 2019,using estimates from the Global Burden of Disease Study 2019(GBD 2019).Results:Compared to the two frequently-used existing methods,the new decomposition method generated results that are more consistent with logical expectations.Using the new method,the number of global deaths associated with population aging between 1990 and 2019 was 23.3 million.Upper middleincome countries accounted for 43%of global deaths related to population aging.The most deaths associated with population aging occurred from three types of disease:ischemic heart disease(5.0 million),stroke(3.8 million),and chronic obstructive pulmonary disease(2.2 million).China,India,Japan,the United States of America,and Brazil had the largest number of deaths related to population aging.Loss related to population aging was completely or partially counteracted by the reduction in mortality in 195 of the 200 countries and territories experiencing population aging(97.5%).Conclusions:The new decomposition method achieves more justifiable results associated with population aging than existing methods.Globally,population aging was associated with a substantial increase of deaths between 1990 and 2019,but it was totally or partially offset by the reduction in mortality in 97.5%of countries and territories.Ruotong Li Xunjie Cheng Yang Yang David CSchwebel Peishan Ning Li Li Zhenzhen Rao Peixia Cheng Min Zhao Guoqing Hu 2023China CDC weekly2023,5,51:0

15Urgent Need of Continuing Medical Education:The Key for Patient Awareness of Labor Epidural Analgesia-A Survey of Chinese Perinatal Care Providers显示文摘Objective:This survey was designed to understand the misconceptions about labor epidurals.Methods:This voluntary and anonymous online survey on wenjuan.com was conducted from September 1st,2015 to January 1st,2016 via mainly WeChat groups dedicated to perinatal healthcare providers in China.The questionnaire included items inquiring the knowledge and opinions about labor epidural analgesia related to maternal complications,baby safety,and effect on laboring.Incomplete surveys were excluded from the data analysis.The data was presented as percentages and a Chi-square test or Fisher's exact test,as appreciate,was used to quantitatively compare the results.Results:A total of 1412 respondents completed surveys with 42.9%(606/1412)of them being anesthesiologists,35.1%(495/1412)being obstetricians,11.8%(167/1412)being midwives,3.7%(52/1412)being labor and delivery nurses,and 6.5%(92/1412)being hospital administrators and unspecified.The study revealed a lack of knowledge in labor pain control.Although 82.4%(1164/1412)of respondents were familiar with labor epidural analgesia,8.9%(126/1412)did not know how it works,and 1.1%(15/1412)never heard it in a multiple-choice question.The three main groups(anesthesiologists,obstetricians,and midwives/labor and delivery nurses)were chosen for comparisons.Opinions among these three groups concerning five questions in the three main concerns were evaluated using a statistical significance of P<0.05.Conclusion:The results in our survey indicated an urgent need of continuing medical education to multidisciplinary specialties to improve evidence-based medical practices as these misconceptions have existed for over 10 years in the medical professionals.Lack of public awareness fueled by misconceptions related to labor epidural analgesia may be associated with a lack of professional knowledge.Correct knowledge in professionals needs to be disseminated to the public in order to dispel possible misconceptions and rumors about labor epidural analgesia.This would not only enhance patient understanding of their care but also improve maternal,fetal,and neonatal outcomes.Peishan Zhao Ling-Qun Hu Chunyuan Liu Huiling Li Anna Huang Shuwei Yang Zhenyu Cai 2021Maternal-Fetal Medicine2021,3,3:0
