| 1 | The Qitai radio telescope显示文摘This study presents a general outline of the Qitai radio telescope(QTT)project.Qitai,the site of the telescope,is a county of Xinjiang Uygur Autonomous Region of China,located in the east Tianshan Mountains at an elevation of about 1800 m.The QTT is a fully steerable,Gregorian-type telescope with a standard parabolic main reflector of 110 m diameter.The QTT has adopted an umbrella support,homology-symmetric lightweight design.The main reflector is active so that the deformation caused by gravity can be corrected.The structural design aims to ultimately allow high-sensitivity observations from 150 MHz up to115 GHz.To satisfy the requirements for early scientific goals,the QTTwill be equipped with ultra-wideband receivers and large field-of-view multi-beam receivers.A multi-function signal-processing system based on RFSo C and GPU processor chips will be developed.These will enable the QTT to operate in pulsar,spectral line,continuum and Very Long Baseline Interferometer(VLBI)observing modes.Electromagnetic compatibility(EMC)and radio frequency interference(RFI)control techniques are adopted throughout the system design.The QTT will form a world-class observational platform for the detection of lowfrequency(nano Hertz)gravitational waves through pulsar timing array(PTA)techniques,pulsar surveys,the discovery of binary black-hole systems,and exploring dark matter and the origin of life in the universe.The QTT will also play an important role in improving the Chinese and international VLBI networks,allowing high-sensitivity and high-resolution observations of the nuclei of distant galaxies and gravitational lensing systems.Deep astrometric observations will also contribute to improving the accuracy of the celestial reference frame.Potentially,the QTT will be able to support future space activities such as planetary exploration in the solar system and to contribute to the search for extraterrestrial intelligence. | Na Wang Qian Xu Jun Ma Zhiyong Liu Qi Liu Hailong Zhang Xin Pei Maozheng Chen Richard N.Manchester Kejia Lee Xingwu Zheng Hans J.Kärcher Wulin Zhao Hongwei Li Dongwei Li Martin Süss Matthias Reichert Zhongyi Zhu Congsi Wang Mingshuai Li Rui Li Ning Li Guljaina Kazezkhan Wenming Yan Gang Wu Lang Cui Ming Zhang Haitao Li | 2023 | Science China(Physics,Mechanics & Astronomy)2023,66,8: | 14 |
| 2 | Efficient generation of transgenic chickens using the spermatogonial stem cells in vivo and ex vivo transfection显示文摘The highly efficient novel methods to produce transgenic chickens were established by directly in-jecting the recombinant plasmid containing green fluorescent protein (GFP) gene into the cock's testis termed as testis-medianted gene transfer (TMGT), and transplanting transfected spermatogonial stem cells (TTSSCs). For the TMGT approach,four dosages of pEGFP-N1 DNA/cationic polymer complex were injected intratesticularly. The results showed: (1) 48 h after the injection,the percentages of testis cells expressing GFP were 4.0%, 8.7%, 10.2% and 13.6% in the 50, 100, 150 and 200 μg/mL group, re-spectively. The difference from the four dosage groups was significant (P<0.05). On day 25 after the injection, a dosage-dependent and time-dependent increase in the number of transgenic sperm was observed. The percentages of gene expression reached the summit and became stable from day 70 to 160, being 12.7%, 12.8%, 15.9% and 19.1%, respectively. The difference from the four dosage groups was also significant (P<0.05). (2) 70 d after the injection, strong green fluorescent could be observed in the seminiferous tubules by whole-mount in-situ hybridization. (3) 70 d after the injection, the semen was collected and used to artificially inseminate wild-type females. The blastoderms of F1 and F2 transgenic chicken expressed GFP were 56.2% (254/452) and 53.2% (275/517), respectively. The detec-tion of polymerase chain reaction (PCR) of F1 and F2 transgenic chicken blood genomic DNA showed that 56.5% (3/23) of F1 and 52.9% (9/17) of F2 were positive. Southern blot showed GFP DNA was in-serted in their genomic DNAs. (4) Frozen whole mount tissue sections of F1 and F2 transgenic chicken liver, heart, kidney and muscle showed that the rates of green fluorescent positive were between 50.0% and 66.7%. (5) With the TTSSCs method, SSCs ex vivo transfected with GFP were transplanted into recipient roosters whose endogenic SSCs had been resoluted. The donor SSCs settled and GFP ex-pression became readily detectable in the frozen whole mount tissue sections of recepient testes. Moreover, sperms carrying GFP could be produced normally. The results of artificially inseminating wild-type females with these sperms showed 12.5% (8/64) of offspring embryo expressed GFP and 11.1% (2/18) hatched chicks were tested transgenic. | 3 | Genetic analysis and fine mapping of an enclosed panicle mutant esp2 in rice (Oryza sativa L.)显示文摘The phenomenon of panicle enclosure in rice is mainly caused by the shortening of uppermost internode.Elucidating the molecular mechanism of panicle enclosure will be helpful for solving the problem of panicle enclosure in male sterile lines and creating new germplasms in rice.We acquired a monogenic recessive enclosed panicle mutant,named as esp2 (enclosed shorter panicle 2),from the tissue culture progeny of indica rice cultivar Minghui-86.In the mutant,panicles were entirely enclosed by flag leaf sheaths and the uppermost internode was almost completely degenerated,but the other internodes did not have obvious changes in length.Genetic analysis indicated that the mutant phenotype was controlled by a recessive gene,which could be steadily inherited and was not affected by genetic background.Apparently,ESP2 is a key gene for the development of uppermost internode in rice.Using an F 2 population of a cross between esp2 and a japonica rice cultivar Xiushui-13 as well as SSR and InDel markers,we fine mapped ESP2 to a 14-kb region on the end of the short arm of chromosome 1.According to the rice genome sequence annotation,only one intact gene exists in this region,namely,a putative phosphatidylserine synthase gene.Sequencing analysis on the mutant and the wild type indicated that this gene was inserted by a 5287-bp retrotransposon sequence.Hence,we took this gene as a candidate of ESP2.The results of this study will facilitate the cloning and functional analysis of ESP2 gene. | GUAN HuaZhong DUAN YuanLin LIU HuaQing CHEN ZhiWei ZHUO Ming ZHUANG LiJun QI WenMing PAN RunSen MAO DaMei ZHOU YuanChang WANG Feng WU WeiRen | 2011 | Chinese Science Bulletin2011,56,14: | 6 |
| 4 | Fine mapping of the rice bacterial blight resistance gene Xa-4 and its co-segregation marker显示文摘An F2 population developed from the Xa-4 near isogenic lines, IR24 and IRBB4, was used for fine mapping of the rice bacterial blight resistance gene, Xa-4. Some restriction fragment length polymorphism (RFLP) markers on the high-density map constructed by Harushima et al. and the amplified DMA fragments homologous to the conserved domains of plant disease resistance (R) genes were used to construct the genetic linkage map around the gene Xa-4 by scoring susceptible individuals in the population. Xa-4 was mapped between the RFLP marker G181 and the polymerase chain reaction (PCR) marker M55. The R gene homologous fragment marker RS13 was found co-segregating with Xa-4 by analyzing all the plants in the population. | 5 | Molecular cloning and expression patterns of the cholesterol side chain cleavage enzyme(CYP11A1) gene during the reproductive cycle in goose(Anas cygnoides)显示文摘Background: CYP11A1, a gene belonging to the family 11 of cytochrome P450, encodes a crucial steroidogenic enzyme that catalyzes the initial step in the production of all classes of steroids. Many studies show that CYP11A1 plays a role in ovary function. However, the role of CYP11A1 in goose reproductive cycle remains largely unknown.Results: In this study, full-length CYP11A1 c DNA of Zhedong goose was obtained using reverse transcription polymerase chain reaction(RT-PCR) and rapid amplification of c DNA ends(RACE). The c DNA consisted of a 96-base pair(bp) 5′untranslated region(UTR), a 179-bp 3′UTR and a 1509-bp open reading frame. The open reading frame encodes a putative 503 amino acid protein that shares high homology with CYP11A1 of other birds. The amino acid sequence possesses conserved domains of the P450 superfamily, which include the steroid-binding domain and the heme-binding region. Real-time quantitative polymerase chain reaction(q PCR) analysis revealed CYP11A1 mR NA was expressed ubiquitously in every Zhedong goose tissue analyzed, including the heart, liver, glandular stomach,lung, spleen, kidney, intestinum tenue, intestinum crassum, cerebrum, cerebellum, muscle, oviduct, pituitary,hypothalamus and ovary.. The relatively low levels of CYP11A1 m RNA were detected in pituitary, ovary and oviduct tissues at ovulation when compared with levels at oviposition. Interestingly, higher expression was observed in ovary and oviduct tissues during brooding. | 6 | Population Genetics of SARS-CoV-2: Disentangling Effects of Sampling Bias and Infection Clusters显示文摘A novel RNA virus,the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is responsible for the ongoing outbreak of coronavirus disease 2019(COVID-19).Population genetic analysis could be useful for investigating the origin and evolutionary dynamics of COVID-19.However,due to extensive sampling bias and existence of infection clusters during the epidemic spread,direct applications of existing approaches can lead to biased parameter estimations and data misinterpretation.In this study,we first present robust estimator for the time to the most recent common ancestor(TMRCA)and the mutation rate,and then apply the approach to analyze 12,909 genomic sequences of SARS-CoV-2.The mutation rate is inferred to be 8.69×10^(−4) per site per year with a 95%confidence interval(CI)of[8.61×10^(−4),8.77×10^(−4)],and the TMRCA of the samples inferred to be Nov 28,2019 with a 95%CI of[Oct 20,2019,Dec 9,2019].The results indicate that COVID-19 might originate earlier than and outside of Wuhan Seafood Market.We further demonstrate that genetic polymorphism patterns,including the enrichment of specific haplotypes and the temporal allele frequency trajectories generated from infection clusters,are similar to those caused by evolutionary forces such as natural selection.Our results show that population genetic methods need to be developed to efficiently detangle the effects of sampling bias and infection clusters to gain insights into the evolutionary mechanism of SARS-CoV-2.Software for implementing VirusMuT can be downloaded at https://bigd.big.ac.cn/biocode/tools/BT007081. | Qi Liu Shilei Zhao Cheng-Min Shi Shuhui Song Sihui Zhu Yankai Su Wenming Zhao Mingkun Li Yiming Bao Yongbiao Xue Hua Chen | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 1 |
