| 1 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. | 2 | Serum cystatin C concentration as an independent marker for hypertensive left ventricular hypertrophy显示文摘Background Serum cystatin C levels can be used to predict morbidity and mortality in patients with cardiovascular disease. However, the clinical relevance of serum cystatin C levels in patients with hypertensive left ventricular hypertrophy (LVH) has rarely been investigated. We designed the present study to investigate whether serum cystatin C levels are associated with cardiac structural and functional alterations in hypertensive patients. Methods We enrolled 823 hypertensive patients and classified them into two groups:those with LVH (n=287) and those without LVH (n=536). All patients underwent echocardiography and serum cystatin C testing. We analyzed the relationship be-tween serum cystatin C levels and LVH. Results Serum cystatin C levels were higher in hypertensive patients with LVH than in those without LVH (P<0.05). Using linear correlation analysis, we found a positive correlation between serum cystatin C levels and interven-tricular septal thickness (r=0.247, P<0.01), posterior wall thickness (r=0.216, P<0.01), and left ventricular weight index (r=0.347, P<0.01). When analyzed by multiple linear regression, the positive correlations remained between serum cystatin C and interventricular septal thickness (β=0.167, P<0.05), posterior wall thickness (β=0.187, P<0.05), and left ventricular weight index (β=0.245, P<0.01). | 3 | Preliminary study on proteomics of gastric carcinoma and its clinical significance显示文摘AIM: To explore the preliminary identification of serum protein pattern models that may be novel potential biomarkers in the detection of gastric cancer.METHODS: A total of 130 serum samples, including 70 from patients with gastric cancer and 60 from healthy adults, were detected by surface-enhanced laser desorption and ionization time-of-flight mass spectrometry (SELDI-TOF-MS). The data of spectra were analyzed by Biomarker Patterns Software (BPS). Thirty serum samples of gastric cancer patients and 30 serum samples of healthy adults were grouped into the training group to build models, and the other 70 samples were used to test and evaluate the models. The samples of the test group were judged only with their peaks'height and were separated into cancer group or healthy control group by BPS automatically and the judgments were checked with the histopathologic diagnosis of the samples.RESULTS: Sixteen mass peaks were found to be potential biomarkers with a significant level of P<0.01.Among them, nine mass peaks showed increased expression in patients with gastric cancer. Analyzed by BPS, two peaks were chosen to build the model for gastric cancer detection. The sensitivity, specificity, and accuracy of the model were 90%, 36/40, 86.7%, 26/30,and 88.6%, 62/70, respectively, which were greatly higher than those of clinically used serum biomarkers CEA (carcinoembryonic antigen), CA19-9 and CA72-4.Stage Ⅰ/Ⅱ gastric cancer samples of the test group were all judged correctly.CONCLUSION: The novel biomarkers in serum and the established model could be potentially used in the detection of gastric cancer. | 5 | Diffusion tensor imaging assesses white matter injury in neonates with hypoxic-ischemic encephalopathy显示文摘With improvements in care of at-risk neonates, more and more children survive. This makes it increasingly important to assess, soon after birth, the prognosis of children with hypoxic-ischemic encephalopathy. Computed tomography, ultrasound, and conventional magnetic resonance imaging are helpful to diagnose brain injury, but cannot quantify white matter damage. In this study, ten full-term infants without brain injury and twenty-two full-term neonates with hypoxic-ischemic encephalopathy(14 moderate cases and 8 severe cases) underwent diffusion tensor imaging to assess its feasibility in evaluating white matter damage in this condition. Results demonstrated that fractional anisotropy, voxel volume, and number of fiber bundles were different in some brain areas between infants with brain injury and those without brain injury. The correlation between fractional anisotropy values and neonatal behavioral neurological assessment scores was closest in the posterior limbs of the internal capsule. | 9 | Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. | 12 | Pb isotope geochemistry of lead, zinc, gold and silver deposit clustered region, Liaodong rift zone,northeastern China显示文摘Pb isotopic analyses were reported for sulfide and hydrothermal carbonate minerals and marble of the Xiquegou lead-zinc, the Zhenzigou zinc-lead and the Gaojiapuzi silver deposits from the Qingchengzi ore field and the Beiwagou zinc-lead deposit in the west, Proterozoic Liaodong rift zone. Pb isotopic ratios of the marble from the Qingchengzi ore field range from 18.24 to 30.63 for 206 Pb/204Pb, 15.59 to 17.05 for207Pb/204Pb and 37.43 to 38.63 for 208Pb/204Pb. The marble gives a Pb-Pb isochron age of 1822±92 Ma, which is interpreted as the age of the metamorphism of the marble. Ore Pb, including Pb of sulfide and hydrothermal car- bonate minerals, from the Qingchengzi ore field shows limited variation with 206 Pb/204Pb=17.66- 17.96, 207 Pb/204Pb=15.60-15.74 and 208Pb/204Pb=37.94-38.60. In contrast, ore Pb from the Beiwagou deposit gives different Pb isotopic ratios with 206Pb/204Pb=15.68-15.81, 207 Pb/204Pb= 15.34-15.45 and 208Pb/204Pb=35.30-35.68. Pb of all deposits from the Liaodong rift zone is derived from the upper crust. Ore Pb of the Qingchengzi deposits is derived from a young upper crust. The model Th/U ratios of 4.40 to 4.74 for ore Pb are significantly different from that of 1.7 to 4.4 given by the marble of the Qingchengzi ore field, suggesting that marble is not the source of the ore Pb. Ore Pb of the Beiwagou deposit is extracted from its source and the deposit is formed at the Paleoproterozoic era. | 14 | SHRIMP zircon age for a K-bentonite in the top of the Laobao Formation at the Pingyin section,Guizhou,South China显示文摘The Lower Cambrian on the Yangtze Platform in South China(internationally equivalent to the Nerreneuvian and 2nd series of the Cambrian)is valuable for understanding the early evolution of life,the global biogeochemical circles,and the major changes of the ocean.In particular,a precise radiometric calibration of the Lower Cambrian of this region is a critical realm in its multidisciplinary studies.A SHRIMP U-Pb geochronology study of the K-bentonite in the topmost Laobao Formation at the Pingyin section,Guizhou,South China yielded an age of 536±5 Ma(MSWD=0.75),suggesting that the K-bentonite here can be correlated with the intensely studied K-bentonite within the middle Zhongyicun Member(Bed 5)of the Zhujiaqing Formation at the Meishucun section in Yunnan.Such a regional correlation result reveals that the Bed 5 K-bentonite also extends into deeper-water environments rather than being deposited merely in the shallow platform interior of the Yangtze Platform.The age of the K-bentonite at the Pingyin section also implies that its overlying polymetallic Ni-Mo layer should be younger than 536±5 Ma.Hence the previous placement of the Precambrian/Cambrian boundary at this layer is inappropriate.Combined with the results of stratigraphic correlations,it is suggested that the K-bentonites in the middle Zhongyicun Member of the Zhujiaqing Formation(or the equivalents)and the base of the Shiyantou Formation(or its equivalents),together with the polymetallic Ni-Mo layer,serve as three important marker beds.Their self-consistent radiometric ages have established an improved geochronologic framework for the Lower Cambrian in South China. | ZHOU MingZhong LUO TaiYi LIU ShiRong QIAN ZhiKuan XING LeCai | 2013 | Science China Earth Sciences2013,56,10: | 10 |
| 15 | A new dynamic pushback control method for reducing fuel-burn costs: Using predicted taxi-out time显示文摘Long departure-taxi-out time leads to significant airport surface congestion, fuel-burn costs, and excessive emissions of greenhouse gases. To reduce these undesirable effects, a Predicted taxi-out time-based Dynamic Pushback Control(PDPC) method is proposed. The implementation of this method requires two steps: first, the taxi-out times for aircraft are predicted by the leastsquares support-vector regression approach of which the parameters are optimized by an introduced improved Firefly algorithm. Then, a dynamic pushback control model equipped with a linear gate-hold penalty function is built, along with a proposed iterative taxiway queue-threshold optimization algorithm for solving the model. A case study with data obtained from Beijing International airport(PEK) is presented. The taxi-out time prediction model achieves predictive accuracy within 3 min and 5 min by 84.71% and 95.66%, respectively. The results of the proposed pushback method show that total operation cost and fuel-burn cost achieve a 14.0% and 21.1%reduction, respectively, as compared to the traditional K-control policy.(3) From the perspective of implementation, using PDPC policy can significantly reduce the queue length in taxiway and taxi-out time. The total operation cost and fuel-burn cost can be curtailed by 37.2% and 52.1%,respectively, as compared to the non-enforcement of any pushback control mechanism. | 16 | Creation of Triple Hierarchical Micro-Meso-Macroporous N-doped Carbon Shells with Hollow Cores Toward the Electrocatalytic Oxygen Reduction Reaction显示文摘A series of triple hierarchical micro-mesomacroporous N-doped carbon shells with hollow cores have been successfully prepared via etching N-doped hollow carbon spheres with CO_2 at high temperatures.The surface areas, total pore volumes and microporepercentages of the CO_2-activated samples evidently increase with increasing activation temperature from 800 to950 °C, while the N contents show a contrary trend from7.6 to 3.8 at%. The pyridinic and graphitic nitrogen groups are dominant among various N-containing groups in the samples. The 950 °C-activated sample(CANHCS-950) has the largest surface area(2072 m^2 g^(-1)), pore volume(1.96 cm^3 g^(-1)), hierarchical micro-mesopore distributions(1.2, 2.6 and 6.2 nm), hollow macropore cores(*91 nm)and highest relative content of pyridinic and graphitic N groups. This triple micro-meso-macropore system could synergistically enhance the activity because macropores could store up the reactant, mesopores could reduce the transport resistance of the reactants to the active sites, and micropores could be in favor of the accumulation of ions.Therefore, the CANHCS-950 with optimized structure shows the optimal and comparable oxygen reduction reaction(ORR) activity but superior methanol tolerance and long-term durability to commercial Pt/C with a 4 e--dominant transfer pathway in alkaline media. | 17 | Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images. | Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv | 2018 | Cancer Communications2018,38,1: | 9 |

| 19 | Characterization and Fine Mapping of a Novel Rice Narrow Leaf Mutant nal9显示文摘A narrow leaf mutant was isolated from transgenic rice(Oryza sativa L.)lines carrying a T-DNA insertion.The mutant is characterized by narrow leaves during its whole growth period,and was named nal9(narrow leaf 9).The mutant also has other phenotypes,such as light green leaves at the seedling stage,reduced plant height,a small panicle and increased tillering.Genetic analysis revealed that the mutation is controlled by a single recessive gene.A hygromycin resistance assay showed that the mutation was not caused by T-DNA insertion,so a map-based cloning strategy was employed to isolate the nal9 gene.The mutant individuals from the F2generations of a cross between the nal9 mutant and Longtepu were used for mapping.With 24 F2mutants,the nal9 gene was preliminarily mapped near the marker RM156 on the chromosome 3.New INDEL markers were then designed based on the sequence differences between japonica and indica at the region near RM156.The nal9 gene was fnally located in a 69.3 kb region between the markers V239B and V239G within BAC OJ1212_C05 by chromosome walking.Sequence and expression analysis showed that an ATP-dependent Clp protease proteolytic subunit gene(ClpP)was most likely to be the nal9 gene.Furthermore,the nal9 mutation was rescued by transformation of the ClpP cDNA driven by the 35S promoter.Accordingly,the ClpP gene was identifed as the NAL9 gene.Our results provide a basis for functional studies of NAL9 in future work. | Wei Li Chao Wu Guocheng Hu Li Xing Wenjing Qian Huamin Si Zongxiu Sun Xingchun Wang Yaping Fu Wenzhen Liu | 2013 | Journal of Integrative Plant Biology2013,55,11: | 8 |
