| 3 | Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. | 6 | Reconfigurable directional coupler in lithium niobate crystal fabricated by three-dimensional femtosecond laser focal field engineering显示文摘For crystals, depressed cladding waveguides have advantages such as preservation of the spectroscopic as well as non-linear properties and the capability to guide both horizontal and vertical polarization modes, but fabrication is always quite time consuming. In addition, it is usually difficult to couple modes propagating in different depressed cladding waveguides through evanescent field overlap, so it is often required to dynamically reconfigure photonic waveguide devices using external fields for classical or quantum applications. Here, we experimentally demonstrate the single-scan femtosecond laser transverse writing of depressed cladding waveguides to form a 2 × 2 directional coupler inside lithium niobate crystal, which is integrated with two deeply embedded microelectrodes on both sides of the interaction region to reconfigure the coupling. By focal field engineering of the femtosecond laser, we specially generate a three-dimensional longitudinally oriented ring-shaped focal intensity profile composed of 16 discrete spots to simultaneously write the entire cladding region. The fabricated waveguides exhibit good single guided modes in two orthogonal polarizations at 1550 nm. By applying voltage to the deeply embedded microelectrodes fabricated with the femtosecond laser ablation followed by selective electroless plating, we successfully facilitate the light coupling from the input arm to the cross arm and thus actively tune the splitting ratio. | 7 | Warm temperature compromises JA-regulated basal resistance to enhance Magnaporthe oryzae infection in rice显示文摘Changes in global temperatures profoundly affect the occurrence of plant diseases.It is well known that rice blast can easily become epidemic in relatively warm weather.However,the molecular mechanism remains unclear.In this study,we show that enhanced blast development at a warm temperature(22C)compared with the normal growth temperature(28C)is rice plant-determined.Comparative transcriptome analysis revealed that jasmonic acid(JA)biosynthesis and signaling genes in rice could be effectively induced by Magnaporthe oryzae at 28C but not at 22C.Phenotypic analyses of the osaoc1 and osmyc2 mutants,OsCOI1 RNAi lines,and OsMYC2-OE plants further demonstrated that compromised M.oryzaeinduced JA biosynthesis and signaling lead to enhanced blast susceptibility at the warm temperature.Consistent with these results,we found that exogenous application of methyl jasmonate served as an effective strategy for improving blast resistance under the warm environmental conditions.Furthermore,decreased activation of JA signaling resulted in the downregulated expression of some key basal resistance genes at 22C when compared with 28C.Among these affected genes,OsCEBiP(chitin elicitorbinding protein precursor)was found to be directly regulated by OsMYB22 and its interacting protein OsMYC2,a key component of JA signaling,and this contributed to temperature-modulated blast resistance.Taken together,these results suggest that warm temperature compromises basal resistance in rice and enhances M.oryzae infection by reducing JA biosynthesis and signaling,providing potential new strategies for managing rice blast disease under warm climate conditions. | Jiehua Qiu Junhui Xie Ya Chen Zhenan Shen Huanbin Shi Naweed I.Naqvi Qian Qian Yan Liang Yanjun Kou | 2022 | Molecular Plant2022,15,4: | 5 |

| 10 | Separation of chalcopyrite and pyrite from a copper tailing by ammonium humate显示文摘Copper tailings constitute a large proportion of mine wastes. Some of the copper tailings can be recycled to recover valuable minerals. In this paper, a copper tailing was studied through the chemical analysis method, Xray diffraction and scanning electron microscope-energy dispersive spectrum. It turned out that chalcopyrite(Cu) and pyrite(S) were the main recoverable minerals in the tailing. In order to separate chalcopyrite from pyrite in low pulp pH, ammonium humate(AH) was singled out as the effective regulator. The depression mechanism of AH on the flotation of pyrite was proved by FTIR spectrum and XPS spectrum, demonstrating that there was a chemical adsorption between AH and pyrite. By Response Surface Methodology(RSM), the interaction between AH, pulp pH and iso-butyl ethionine(Z200) was discussed. It was illustrated that the optimal dosage of AH was 1678 g·t^(-1) involving both the recovery of Cu and S. | 17 | Preparation and characterization of a novel hybrid chelating material for effective adsorption of Cu(Ⅱ)and Pb(Ⅱ)显示文摘The discharge of heavy metal ions such as Cu^2+and Pb^2+poses a severe threat to public health and the environment owing to their extreme toxicity and bioaccumulation through food chains Herein, we report a novel organic–inorganic hybrid adsorbent, Al(OH)_3-poly(acrylamide dimethyldiallylammonium chloride)-graft-dithiocarbamate(APD), for rapid and effectiv removal of Cu^2+and Pb^2+. In this adsorbent, the 'star-like' structure of Al(OH)3 poly(acrylamide-dimethyldiallylammonium chloride) served as the support of dithiocarbamat(DTC) functional groups for easy access of heavy metal ions and assisted development of larg and compact floccules. The synthesized adsorbent was characterized by scanning electron microscopy(SEM), transmission electron microscopy(TEM), Fourier transform infrared spectroscopy(FTIR), and thermogravimetric analysis(TGA). APD was demonstrated to hav rapid adsorption kinetics with an initial rate of 267.379 and 2569.373 mg/(g·min) as well a superior adsorption capacities of 317.777 and 586.699 mg/g for Cu^2+and Pb^2+respectively. Th adsorption process was spontaneous and endothermic, involving intraparticle diffusion and chemical interaction between heavy metal ions and the functional groups of APD. To assess it versatility and wide applicability, APD was also used in turbid heavy metal wastewater, and performed well in removing suspended particles and heavy metal ions simultaneously through flocculation and chelation.
