2Alternative splicing of MaMYB16L regulates starch degradation in banana fruit during ripening显示文摘The alternative splicing of select genes is an important mechanism to regulate responses to endogenous and environmental signals in plants.However,the role of alternative splicing in regulating fruit ripening remains unclear.Here,we discovered that MaMYB16L,an R1-type MYB transcription factor,undergoes alternative splicing and generates two transcripts,the full-length isoform MaMYB16L and a truncated form MaMYB16S,in banana fruit.During banana fruit ripening,the alternative splicing process intensifies with downregulated MaMYB16L and upregulated MaMYB16S.Moreover,MaMYB16L is a transcriptional repressor that directly binds with the promoters of many genes associated with starch degradation and MaDREB2,a positive ripening regulator,and represses their expression.In contrast,MaMBY16S lacks a DNA-binding domain but competitively combines and forms non-functional heterodimers with functional MaMYB16L.MaMYB16L-MaMYB16S heterodimers decrease the binding capacity and transrepression activity of MaMYB16L.The downregulation of MaMYB16L and the upregulation of MaMYB16S,that is,a decreased ratio of active to non-active isoforms,facilitates the activation of ripening-related genes and thereby promotes fruit ripening.Furthermore,the transient overexpression of MaMYB16S promotes banana fruit ripening,whereas the overexpression of MaMYB16L delays this process.Therefore,the alternative splicing of MaMYB16L might generate a self-controlled regulatory loop to regulate banana fruit ripening.Guoxiang Jiang Dandan Zhang Zhiwei Li Hanzhi Liang Rufang Deng Xinguo Su Yueming Jiang Xuewu Duan 2021Journal of Integrative Plant Biology2021,63,7:2

4Direct somatic embryogenesis and related gene expression networks in leaf explants of Hippeastrum ‘Bangkok Rose’显示文摘Hippeastrum, a highly diverse genus in the Amaryllidaceae family, is a valuable ornamental bulbous flowering plant. Somatic embryogenesis(SE) is an efficient method for mass production of Hippeastrum plantlets. Previous studies have been devoted to the in vitro propagation of Hippeastrum, but the SE and its regulatory networks are rarely reported. In this study, we established a direct SE method of Hippeastrum Bangkok Rose' using leaf bases as explants. MS supplemented with 1.00 mg·L^(-1)NAA +1.00 mg·L^(-1)KT + 0.25 mg·L^(-1)TDZ was the optimal medium for SE. Histological observations showed that the bipolar somatic embryo originated from the epidermal cell layer and underwent initiation,globular, scutellar and coleoptile stages. During SE, endogenous hormones of IAA, CTK, ABA, and SA were highly accumulated. Transcriptomic analysis revealed the genes encoding auxin biosynthesis/metabolic enzymes and efflux carriers were induced, while the auxin receptor of TIR1 and ARF transcriptional repressor of Aux/IAA were down-regulated and up-regulated, respectively, leading to suppression of auxin signaling. In contrast, cytokine signaling was promoted at the early stage of SE, as biosynthesis, transport, and signaling components were up-regulated.Various stress-related genes were up-regulated at the early or late stages of SE. Chromatin remodeling could also be dynamically regulated via distinct expression enzymes that control histone methylation and acetylation during SE. Moreover, key SE regulators, including WOXs and SERKs were highly expressed along with SE. 5Rare-earth,nitrogen-rich,oxygen heterocyclic supramolecular compounds(Nd,Sm,and Eu):Synthesis,structure,and catalysis for ammonium perchlorate显示文摘Three novel rare-earth,nitrogen-rich and oxygen heterocyclic supramolecular complexes,namely[Nd(BTF)_(2)(H_(2)O)_(5)]_(n),[Sm(BTF)_(2)(H_(2)O)_(5)]_(n),and[Eu(BTF)_(2)(H_(2)O)_(5)]_(n),were synthesized.A single crystal was obtained by the solvent evaporation method,and the structure and coordination mode of metal complexes were determined by single crystal X-ray diffraction.Results show that the supramolecular complexes contain many hydrogen bonds and thus have good thermal stability(T_(dec)>540 K).The thermal decomposition of ammonium perchlorate(AP)catalyzed by the complexes was investigated by differential thermal analysis,which reveals a pre-eminent catalytic effect on AP.The high temperature decomposition peak of AP can be advanced by nearly 90 K at the amount of added complexes of 10 wt%,and the activation energy of AP descent range is from 70 to 150 kJ/mol.The other properties were fully characterized through elemental analysis and Fourier transform infrared spectroscopy.Wenjia Hao Tao Huang Bo Jin Jinhao Zhang Juan Shen Rufang Peng 2022Journal of Rare Earths2022,40,3:0
6Whole Exome Sequencing Identifies A Novel Pathogenic Bmpr2 Variant in Pulmonary Atresia显示文摘Objective:Pulmonary atresia(PA)is a rare type of complex cyanotic congenital heart defect characterized primarily by an undeveloped pulmonary valve or pulmonary artery.Therefore,defining a disease-causing gene mutation in a pulmonary atresia family is a possible method of genetic counseling,future prenatal diagnosis,and therapeutic approaches for pulmonary atresia.Methods:Blood samples were collected from six PA family members,and genomic DNA was extracted using the QIAamp DNA Blood Mini Kit.Gene detection was performed using a second-generation sequencing gene panel.Results:Genetic testing results indicated that a heterozygous mutation originating from maternal inheritance was detected in the BMPR2 gene of the proband’s genomic DNA.The pathogenic gene was c.2804C>T(p.A935V).The mutation was also detected in the genomic DNA of the proband’s elder brother(III-1),but not in other family members.Conclusion:To the best of our knowledge,this is the first study to report the BMPR2 variant responsible for pulmonary atresia.The frequency of the c.2804C>T(p.A935V)mutation detected in this family is extremely low in the normal population(1/246048).The mutation was highly conserved among different species.Sorting intolerant from tolerant(SIFT)predicts it to be a harmful mutation.Muyu Qi Xiaoping Lan Jia Li Junwen Ge Li Shen Rufang Zhang 2021Congenital Heart Disease2021,16,5:0

8Catalytic thermal decomposition of ammonium perchlorate by a series of lanthanide EMOFs显示文摘Given their unique and excellent properties,metal-organic frameworks(MOFs)materials have been used in many scientific fields.EMOFs use energetic materials as ligands,which can provide part of the energy for the system while catalyzing ammonium perchlorate.The energetic material 1.1'-dihydroxyazotetrazole(H_(2)AzTO),as a high-energy nitrogen-rich material,was selected as a ligand.Five kinds of La^(3+),Ce^(3+),Pr^(3+),Nd^(3+),and Sm^(3+)lanthanide EMOFs were synthesized and obtained.Single crystal X-ray diffraction tests were conducted to obtain the crystal structures of EMOFs 1-5,which indicate that they have similar crystal structures.The thermal stabilities of EMOFs 1-5,which are obtained by differential scanning calorimetry(DSC)tests,are improved compa red with that of the ligand.The results of thermicdecomposition of ammonium perchlorate(AP)and AP mixtures with 10 wt%EMOFs 1-5 show that except for AP mixed with 10 wt%co mpound 2,the high-temperature decomposition peak tempe rature of AP mixed with other compounds is significantly advanced(up to 59.3-88.3 K),and the decomposition of AP is continuous and violent.EMOFs 3-5 have good application prospects for the catalytic thermicdecomposition of AP.Jinhao Zhang Bo Jin Wenjia Hao Yulan Song Chenjie Hou Tao Huang Rufang Peng 2023Journal of Rare Earths2023,41,4:0
