1Changing Proportions of HIV-1 Subtypes and Transmitted Drug Resistance Among Newly Diagnosed HIV/AIDS Individuals—China,2015 and 2018显示文摘Introduction:With the expansion of human immunodeficiency virus(HIV)antiretroviral therapy(ART),HIV drug resistance is becoming more and more serious.This study describes the changing prevalence of HIV-1 subtypes and transmitted drug resistance(TDR)among newly diagnosed individuals in China,2015 and 2018.Methods:A total of 8,980 individuals in 2015 and 2018 from 31 provincial-level administrative divisions(PLADs)were enrolled in this study.Viral RNAs were amplified and sequenced using an in-house polymerase chain reaction(PCR)protocol.The Stanford HIV Drug Resistance Database(HIVdb)was used to predict susceptibility to 12 antiretroviral drugs.Results:The prevalence of TDR was not significantly increased over time.The prevalence of TDR was 3.8%and 4.4%in 2015 and 2018,respectively(P=0.13).The prevalence of CRF55_01B increased from 2.3%in 2015 to 3.9%in 2018(P<0.001).The drug resistance prevalence of nonnucleoside reverse transcriptase inhibitors(NNRTI)increased from 2.4%in 2015 to 3.3%in 2018(P<0.01).The prevalence of E138(P<0.001),H221(P=0.03),and V179(P<0.001)mutations increased from 0.30%,0.09%,and 0.70%in 2015 to 1.10%,0.30%,and 1.70%in 2018,respectively.Conclusions:HIV drug resistance affects the effect of antiretroviral treatment,so the monitoring of HIV TDR should be strengthened to control the transmission of HIV drug resistance.Jingjing Hao Shan Zheng Mengze Gan Aobo Dong Ruihua Kang Miaomiao Li Shuai Zhao Jing Hu Chang Song Lingjie Liao Yi Feng Yiming Shao Yuhua Ruan Hui Xing 2021China CDC weekly2021,3,53:13

3Interaction of Restin with transcription factors显示文摘Restin, a member of melanoma-associated antigen superfamily gene, was first cloned from differentiated leukemia cell induced by all trans-retinoic acid, and was able to inhibit cell proliferation, but the molecular mechanism was not clear. Since Restin was localized in cell nucleus, and its homolog member, Necdin (neuronal growth suppressor factor), could interact with transcription factors p53 and E2F1, we proposed that Restin might also function as Necdin through interacting with some transcription factors. In this study, transcription factors p53, AP1, ATFs and E2Fs were cloned and used in the mammalian two-hybrid system to identify their in- teraction with Restin. The results showed that only ATF3 had a strong interaction with Restin. It is interesting to know that ATF3 was an important transcription factor for G1 cell cycle initiation in physiological stress response. 5Aberrantly expressed GFRα-1/RET in patients with lacrimal adenoid cystic carcinoma is associated with high recurrence risk:a retrospective study of 51 LACC cases显示文摘Objective:Because of the poor prognosis of lacrimal adenoid cystic carcinoma(LACC),we aimed to investigate the effects of perineural invasion(PNI)and consequent aberrations in GDNF/GFRα-1/RET protein expression on LACC recurrence.Methods:Clinicopathological data for 51 histologically confirmed patients with LACC enrolled between 2001 and 2017 were retrospectively analyzed.Hematoxylin and eosin staining was applied to assess PNI.Tissue-based immunohistochemistry(IHC)detection of GDNF,GFRα-1,and RET proteins was performed on LACC formalin-fixed,paraffin-embedded specimens.We generated semi-quantitative data of the IHC results and compared them with the clinicopathological data for the 51 patients.Results:Of the 51 patients,19(37.3%)were PNI positive.Recurrence was more common for LACC with than without PNI(73.7%vs.37.5%,P=0.01).GDNF,GFRα-1,and RET proteins were expressed in 62.7%,62.7%,and 54.9%of the 51 patients with LACC,respectively.The expression of all 3 proteins was more common in patients with than without PNI.In agreement with previous findings,PNI-associated GFRα-1 and RET positivity,as detected by IHC,remained significantly associated with recurrence,whereas GDNF expression,as detected by IHC,was not correlated with LACC recurrence.Specifically,patients with concurrent GFRα-1 and RET expression may have a high risk of PNI(89.5%positivity rate)and recurrence(84.2%positivity rate).Conclusions:PNI may contribute to LACC recurrence.The concurrent expression of GFRα-1 and RET proteins,as detected by IHC,may potentially be associated with LACC PNI and recurrence.Lin Liu Liqiong Zhao Jie Zhang Guoxiang Song Carol LShields Ruihua Wei 2021Cancer Biology & Medicine2021,18,1:3

7Awareness intervention for Beijing neurologists regarding secondary prevention of cerebral infarction/transient ischemia Cross-sectional investigation显示文摘BACKGROUND: Stroke prevention guidelines should be made available to neurologists for clinical application to aid in uniformity, timing, preciseness, and acceptance of disease. OBJECTIVE: To investigate the awareness of neurologists in some Beijing hospitals of intervention in secondary prevention of cerebral infarction/transient ischemia. DESIGN: Cross-sectional study. SETTING: Beijing Tiantan Hospital, Beijing Anzhen Hospital, General Hospital of Beijing Military Area, Command of Chinese PLA, Beijing Chuiyangliu Hospital, Beijing 6th Hospital, Beijing Hepingli Hospital, and Beijing Daxing District Hospital. PARTICIPANTS: A total of 28 (associate) chief physicians, 58 attending physicians, and 54 resident physicians who engaged in clinical treatment of cerebrovascular diseases were selected from 8 hospitals in Beijing from March to April 2007. All physicians provided informed consent. METHODS: Self-made closed questionnaires were provided for data collection, consisting of 16 questions that were single choice or multiple choice. Specifically, questions 1-7 focused on awareness of blood pressure regulation in different patients and first choice of decompression drug; questions 8-12 focused on awareness of lipid regulation; and questions 13-16 focused on awareness of anti-blood platelet drugs applied in secondary prevention. The scores ranged from 0-100 points, and each question was worth 6.25 points. The scores positively correlated with the awareness rate. To test leveling real-time, the survey lasted for a maximum of 20 minutes. One questionnaire was independently finished by each subject in the survey. MAIN OUTCOME MEASURES: Awareness intervention among neurologists during secondary prevention of cerebral infarction/transient ischemia and questionnaire scores. RESULTS: 140 subjects were included in the final analysis. ① The awareness rate among neurologists for intervention during secondary prevention of cerebral infarction/transient ischemia ranged from 0.7-57.9%, the scores ranged between 0-56 points, and the mean score was 26 points. ② Scores of resident physicians were 0-56 points with a mean score of 26 points; attending physicians scored 6-50 points, with a mean score of 26 points; and chief physicians scored 6-50 points, with a mean score of 23 points. There were no significant differences among the various physicians (F = 0.771, t = 0.465, P > 0.05). CONCLUSION: Awareness among neurologists of intervention during secondary prevention of cerebral infarction/transient ischemia is not ideal. 8Distribution and spectral characteristics of chromophoric dissolved organic matter in a coastal bay in northern China显示文摘The absorption spectra of chromophoric dissolved organic matter(CDOM),along with general physical,chemical and biological variables,were determined in the Bohai Bay,China,in the springs of 2011 and 2012. The absorption coefficient of CDOM at 350 nm(a350) in surface water ranged from 1.00 to 1.83 m-1(mean: 1.35 m-1) in May 2011 and from 0.78 to 1.92 m-1(mean:1.19 m-1) in April 2012. Little surface-bottom difference was observed due to strong vertical mixing. The a350 was weakly anti-correlated to salinity but positively correlated to chlorophyll a(Chl-a) concentration. A shoulder over 260–290 nm,suggestive of biogenic molecules,superimposed the overall pattern of exponentially decreasing CDOM absorption with wavelength. The wavelength distribution of the absorption spectral slope manifested a pronounced peak at ca. 300 nm characteristic of algal-derived CDOM. All a250/a365 ratios exceeded 6,corresponding to CDOM molecular weights(Mw) of less than 1 kDa. Spectroscopically,CDOM in the Bohai Bay differed substantively from that in the Haihe River,the bay's dominant source of land runoff; photobleaching of the riverine CDOM enlarged the difference.Results point to marine biological production being the principal source of CDOM in the Bohai Bay during the sampling seasons. 9Investigation on Host Species and Damage Degree of Bemisia tabaci (Gennadius) in Shaanxi Area显示文摘The host plants of Bemisia tabaci( Gennadius) in Shaanxi were investigated,and its damage degree was evaluated by hazard index in 2013 and 2014.There were 73 species( variants) of host plants of B. tabaci belonging to 28 families,mainly concentrated in Cucurbitaceae,Cruciferae,Solanaceae,Laguminosae and Malvaceae. There were seven species of vegetables that suffered the damage of grade 4. B. tabaci was more serious in southern Shaanxi and lighter in northern Shaanxi. 13Erianin suppresses constitutive activation of MAPK signaling pathway by inhibition of CRAF and MEK1/2显示文摘Constitutive activation of RAS-RAF-MEK-ERK signaling pathway(MAPK pathway)frequently occurs in many cancers harboring RAS or RAF oncogenic mutations.Because of the paradoxical activation induced by a single use of BRAF or MEK inhibitors,dual-target RAF and MEK treatment is thought to be a promising strategy.In this work,we evaluated erianin is a novel inhibitor of CRAF and MEK1/2 kinases,thus suppressing constitutive activation of the MAPK signaling pathway induced by BRAF V600E or RAS mutations.KinaseProfiler enzyme profiling,surface plasmon resonance(SPR),isothermal titration calorimetry(ITC),cellular thermal shift assay,computational docking,and molecular dynamics simulations were utilized to screen and identify erianin binding to CRAF and MEK1/2.Kinase assay,luminescent ADP detection assay,and enzyme kinetics assay were investigated to identify the efficiency of erianin in CRAF and MEK1/2 kinase activity.Notably,erianin suppressed BRAF V600E or RAS mutant melanoma and colorectal cancer cell by inhibiting MEK1/2 and CRAF but not BRAF kinase activity.Moreover,erianin attenuated melanoma and colorectal cancer in vivo.Overall,we provide a promising leading compound for BRAF V600E or RAS mutant melanoma and colorectal cancer through dual targeting of CRAF and MEK1/2.Penglei Wang Xuechao Jia Bingbing Lu Han Huang Jialin Liu Xuejiao Liu Qiong Wu Yamei Hu Pan Li Huifang Wei Tingting Liu Dengyun Zhao Lingwei Zhang Xueli Tian Yanan Jiang Yan Qiao Wenna Nie Xinli Ma Ruihua Bai Cong Peng Zigang Dong Kangdong Liu 2023Signal Transduction and Targeted Therapy2023,8,4:1

16Hormone-like activities of Kuntai capsule in the uteri of ovariectomized rats and immature rabbits显示文摘OBJECTIVE: To investigate the hormone-like activities of Kuntai capsule(KTC) in the uteri of ovariectomized rats and immature rabbits.METHODS: Following bilateral ovariectomy, rats were randomly divided into six groups including sham group, control group, estradiol valerate group, KTC 0.24, 0.6, and 1.5 g/kg groups. The rats were treated with 0.5% CMC-Na, estradiol valerate and KTC(0.24, 0.6, and 1.5 g/kg), respectively for 28 consecutive days. Then the estrous cycle, uterine changes and pathological changes were examined.Serum levels of estradiol(E2), and progesterone(P4)were measured by enzyme-linked immunosorbent assay(ELISA). Protein levels of estradiol receptor(ER), progesterone receptor(PR), vascular endothelial growth factor(VEGF), proliferating cell nuclear antigen(PCNA) and nuclear-associated antigen-67(Ki-67) in uterine tissues were detected by western blot. Immature rabbits were estrogen-primed prior to intragastric administration with KTC for 6 d consecutively. Then, the uteri underwent hematoxylin-eosin staining to observe endometrial transformation.RESULTS: Compared with the control group, 0.6and 1.5 g/kg KTC markedly decreased the uterine organ coefficient and endometrial thickness(P <0.05). The serum level of P4 was increased in the KTC 0.6 g/kg group(P < 0.05). There were no significant variations in the serum level of E2 in the KTC groups compared with the control group. ERβ, but not ERα, was markedly upregulated after KTC administration(P < 0.05). Furthermore, 1.5 g/kg KTC significantly decreased the protein level of PRA(P <0.05) and 0.6 g/kg KTC increased the protein level of PRB in the uteri(P < 0.05). VEGF was highly expressed after treatment with 0.24 and 0.6 g/kg KTC,and Ki-67 was markedly reduced in ovariectomized rats treated with 1.5 g/kg KTC. No difference was found in the expression of PCNA. 20Simple self-assembly of 3D laminated CuO/SnO2 hybrid for the detection of triethylamine显示文摘Tin dioxide is important gas sensor material and has wide applications in the detection of toxic gases and volatile organic compounds.Here,we synthesized a 3D laminated structural CuO/SnO2 material possessing p-n heterostructures.The morphology and structure were characterized by XRD,SEM,TEM and XPS techniques and the sensing properties were investigated for the detection of triethylamine(TEA).The results indicate that 3D laminated CuO/SnO2 material,assembled by lamellae consisting of ordered nanoparticles,exhibit an enhanced sensing performance compared with SnO2,and notably,CuO/SnO2 with size less than 1μm has obvious high selectivity in the detection of 100 ppm TEA.Particularly,it has a high response and stability to 1 and 5 ppm TEA(S is 8 and 33),and that is higher than SnO2 material,suggesting 3D laminated CuO/SnO2 is an effective ca ndidate material served as sensor platform to detect low-concentration amines.Yangyang Shang Wenqiong Shi Ruihua Zhao Md.Maruf Ahmed Jinping Li Jianping Du 2020Chinese Chemical Letters2020,31,8:1
