| 1 | China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy. | Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang | 2019 | Cancer Communications2019，39，1： | 35 |
| 2 | Autophagy activation aggravates neuronal injury in the hippocampus of vascular dementia rats显示文摘It remains unclear whether autophagy affects hippocampal neuronal injury in vascular dementia. In the present study, we investigated the effects of autophagy blockade on hippocampal neuronal injury in a rat model of vascular dementia. In model rats, hippocampal CA1 neurons were severely damaged, and expression of the autophagy-related proteins beclin-1, cathepsin B and microtubule-associated protein 1 light chain 3 was elevated compared with that in sham-operated animals. These responses were suppressed in animals that received a single intraperitoneal injection of wortmannin, an autophagy inhibitor, prior to model establishment. | 4 | Heat-stress-induced sprouting and differential gene expression in growing potato tubers: Comparative transcriptomics with that induced by postharvest sprouting显示文摘Crops face increased risk from heat stress due to climate change.Potato(Solanum tuberosum L.)tubers grown in hot summers often have defects including pre-harvest sprouting(“heat sprouts”).We have used 18 potato cultivars to investigate whether heat stress(HS)conditions alone could cause heat sprouting and dormancy changes in tubers.We also examined transcriptomic responses of potato to HS and whether these responses are like those induced by postharvest sprouting.We demonstrated that HS alone caused heat sprouts and shortened postharvest dormancy period,heat-sprouted tubers became dormant after harvest,and cultivars varied substantially for producing heat spouts but there was no clear association with cultivar maturity earliness.Cultivar Innovator did not show any heat sprouts and still had long dormancy.Dormancy-associated genes(DOG1 and SLP)were downregulated in HS tubers like in postharvest sprouting tubers.We have identified 1201 differentially expressed genes,14 enriched GO terms and 12 enriched KEGG pathways in response to HS in growing tubers of‘Russet Burbank’.Transcriptomic response of‘Russet Burbank’to HS showed significant similarities to that of postharvest non-HS sprouted tubers.Gibberellin biosynthesis pathway was enriched in heat-stressed tubers and was likely involved in heat sprouting and dormancy release.Heat sprouting and postharvest sprouting shared common candidate genes and had significant similarity in gene expression.Our study has significance for selecting potato cultivars for farming,planning storage and utilization of heat-stressed tubers,identifying sprouting-related genes,understanding heat-stress biology,and breeding heat-tolerant potato cultivars,especially for sustainable potato production under climate change. | Guodong Zhang Ruimin Tang Suyan Niu Huaijun Si Qing Yang Om P.Rajora Xiu-Qing Li | 2021 | Horticulture Research2021，8，1： | 1 |

| 9 | Single-bond-linked oligomeric donors for high performance organic solar cells显示文摘The effective design and synthesis of novel small-molecule donors(SMDs) is extremely essential for the in-depth study of the scientific problems of bulk heterojunction morphology and the improvement of photovoltaic performance in organic solar cells(OSCs). Importantly, developing a series of donors with different conjugated central donor(D) units is a remarkable strategy to obtain high-performance donors. Herein, two acceptor-donor-donor-acceptor(A-D-D-A) type oligomeric donors 2DTBDT and2DTBDT-2T with two dithieno[2,3-d:2,3-d’]benzo[1,2-b:4,5-b’]dithiophene(DTBDT) as D units, without and with bithiophene as the π bridge respectively are designed and synthesized successfully. The central linked-DTBDT unit can provide a larger conjugated plane and promote π electron delocalization, which can effectively improve π-π interactions between donors and regulate the crystallinity. And we found that the π bridge provided 2DTBDT-2T with 12.31% efficiency that is already a high efficiency in OSCs,whereas 2DTBDT with merely 3.63% efficiency, both with 2,2-((2Z,2 Z)-((12,13-bis(2-ethylhexyl)-3,9-diundecyl-12,13-dihydro-[1,2,5]thiadiazolo[3,4-e]thieno[2,'3 ’:4,5 ]thieno[2,3 :4,5]pyrrolo[3,2-g]thieno-[2,3 :4,5]thieno[3,2-b]indole,10-diyl)bis(methanylylidene))bis(5,6-difluoro-3-oxo-2,3-dihydro-1H-indene-2,1-diylidene)) dimalononitrile(Y6) as the acceptor. We conjecture that the main reason for the different device performance may be ascribed to the different molecular stacking orientation of the oligomeric donors and morphology features of the donors:Y6 blend films. Compared to the predominant face-on orientation of the 2DTBDT neat film, the 2DTBDT-2T neat film performed a preferential edge-on orientation, which obtained a smoother surface, stronger crystallinity and more uniform phase separation in the 2DTBDT-2T:Y6 blend films with nanofiber structure, which delivered higher and more balanced carrier mobilities, the more efficient exciton dissociation and reduced biomolecule recombination, therefore obtaining better power conversion efficiencies(PCEs). We speculate that the transformation of molecular stacking orientation of oligomeric donors is possibly due to that π bridge extended and twisted the molecular structure of 2DTDBT-2T, resulting in an edge-on orientation relative to the substrate.
