| 1 | Peripheral CD4^(+) T cell signatures in predicting the responses to anti-PD-1/PD-L1 monotherapy for Chinese advanced non-small cell lung cancer显示文摘Limited benefit population of immune checkpoint inhibitors makes it urgent to screen predictive biomarkers for stratifying the patients.Herein,we have investigated peripheral CD4^(+) T cell signatures in advanced non-small cell lung cancer(NSCLC)patients receiving anti-PD-1/PD-L1 treatments.It was found that the percentages of IFN-γand IL-17A secreting naïve CD4^(+) T cells(Tn),and memory CD4^(+) T cells(Tm)expressing PD-1,PD-L1 and CTLA-4 were significantly higher in responder(R)than non-responder(NonR)NSCLC patients associated with a longer progression free survival(PFS).Logistic regression analysis revealed that the baseline IFN-γ-producing CD4^(+) Tn cells and PD-1^(+)CD4^(+) Tm cells were the most significant signatures with the area under curve(AUC)value reaching 0.849.This was further validated in another anti-PD-1 monotherapy cohort.Conversely,high percentage of CTLA-4^(+)CD4^(+) Tm cells was associated with a shorter PFS in patients receiving anti-PD-L1 monotherapy.Our study therefore elucidates the significance of functional CD4^(+) Tn and Tm subpopulations before the treatment in predicting the responses to anti-PD-1 treatment in Chinese NSCLC patients.The fact that there display distinct CD4^(+) T cell signatures in the prediction to anti-PD-1 and anti-PD-L1 monotherapy from our study provides preliminary evidence on the feasibility of anti-PD-1 and anti-PD-L1 combination therapy for advanced NSCLC patients. | Liliang Xia Hui Wang Mingjiao Sun Yi Yang Chengcheng Yao Sheng He Huangqi Duan Weimin Xia Ruiming Sun Yaxian Yao Zhiwei Chen Qiong Zhao Hong Li Shun Lu Ying Wang | 2021 | Science China(Life Sciences)2021,64,10: | 7 |

| 4 | Poisson's ratios and S-wave velocities of the Xishancun landslide,Sichuan,China,inferred from high-frequency receiver functions of local earthquakes显示文摘Landslides are recurrent geological phenomena on Earth that cause heavy casualties and property losses annually.In this study,we use the V_(p)-k stacking and nonlinear waveform inversion methods of high-frequency receiver functions extracted from local earthquakes,to sequentially invert Poisson’s ratios and S-wave velocities of the Quaternary Xishancun landslide,which is composed of three segments,i.e.,h1,h2,and h3 from bottom to top.Our results show that Poisson’s ratio values are generally higher than 0.33 and that the S-wave velocities vary from 0.1 to 0.9 km s^(-1).High Poisson’s ratios(>0.44)are mainly distributed in the juncture regions between different segments,as well as the western edge of h2.These zones show significant variation in landslide thickness and are potentially hazardous areas.Low velocities of 0.05–0.2 km s^(-1)with thicknesses of 10–30m are widely observed in the lower layer of the landslide.The high Poisson’s ratios and low-velocity layer may be related to water-rich materials in these areas.Our study suggests that the high-frequency receiver functions from local earthquakes can be used to delineate geotechnical structures,which is valuable for landslide stability analysis and hazard mitigation. | Zigen WEI Risheng CHU Zhiwei LI Minhan SHENG Qiu ZENG | 2021 | Science China Earth Sciences2021,64,7: | 3 |

| 7 | Science with the 2.5-meter Wide Field Survey Telescope(WFST)显示文摘The Wide Field Survey Telescope(WFST) is a dedicated photometric surveying facility being built jointly by University of Science and Technology of China(USTC) and the Purple Mountain Observatory(PMO). It is equipped with a 2.5-meter diameter primary mirror, an active optics system, and a mosaic CCD camera with 0.73 gigapixels on the primary focal plane for highquality image capture over a 6.5-square-degree field of view. The installation of WFST near the summit of Saishiteng mountain in the Lenghu region is scheduled in summer of 2023, and the operation is planned to start three months later. WFST will scan the northern sky in four optical bands(u, g, r and i) at cadences from hourly/daily in the deep high-cadence survey(DHS) program, to semi-weekly in the wide field survey(WFS) program. During a photometric night, a nominal 30 s exposure in the WFS program will reach a depth of 22.27, 23.32, 22.84, and 22.31(AB magnitudes) in these four bands, respectively, allowing for the detection of a tremendous amount of transients in the low-z universe and a systematic investigation of the variability of Galactic and extragalactic objects. In the DHS program, intranight 90 s exposures as deep as 23(u) and 24 mag(g), in combination with target of opportunity follow-ups, will provide a unique opportunity to explore energetic transients in demand for high sensitivities, including the electromagnetic counterparts of gravitational wave events, supernovae within a few hours of their explosions,tidal disruption events and fast, luminous optical transients even beyond redshift of unity. In addition, the final 6-year co-added images, anticipated to reach g■25.8 mag in WFS or 1.5 mags deeper in DHS, will be of fundamental importance to general Galactic and extragalactic science. The highly uniform legacy surveys of WFST will serve as an indispensable complement to those of the Vera C. | 8 | Small extracellular vesicles with nanomorphology memory promote osteogenesis显示文摘Nanotopographical cues endow biomaterials the ability to guide cell adhesion, proliferation, and differentiation. Cellular mechanical memory can maintain the cell status by retaining cellular information obtained from past mechanical microenvironments. Here, we propose a new concept “morphology memory of small extracellular vesicles (sEV)” for bone regeneration. We performed nanotopography on titanium plates through alkali and heat (Ti8) treatment to promote human mesenchymal stem cell (hMSC) differentiation. Next, we extracted the sEVs from the hMSC, which were cultured on the nanotopographical Ti plates for 21 days (Ti8-21-sEV). We demonstrated that Ti8-21-sEV had superior pro-osteogenesis ability in vitro and in vivo. RNA sequencing further confirmed that Ti8-21-sEV promote bone regeneration through osteogenic-related pathways, including the PI3K-AKT signaling pathway, MAPK signaling pathway, focal adhesion, and extracellular matrix-receptor interaction. Finally, we decorated the Ti8-21-sEV on a 3D printed porous polyetheretherketone scaffold. The femoral condyle defect model of rabbits was used to demonstrate that Ti8-21-sEV had the best bone ingrowth. In summary, our study demonstrated that the Ti8-21-sEV have memory function by copying the pro-osteogenesis information from the nanotopography. | 13 | Genetic Parameters of Body Length and Response to Selection for Growth Across Four Generations of Artemia sinica显示文摘To investigate the genetic components of growth in the brine shrimp Artemia sinica, we estimated the genetic parameters of body length and the response to selection using a fully pedigreed population of A. sinica. The base population was generated from four wild founder populations. We tested 4160 offspring in 360 families over four generations for growth and survival performance. Across four generations, we produced full-and half-sib families with nested mating, where two dams were mated to the same sire. Individual body length was measured for each nauplius at day 20 post-hatching. Heritability of body length was estimated across four generations with the restricted maximum likelihood method. The heritability of body length in A. sinica was low(0.14 ± 0.05), and the common environmental effect was 0.14 ± 0.02. We estimated the response to selection for body length by calculating the difference in the mean breeding values between different generations. The accumulated genetic gain in body length was 278.94 μm after three generations of selection. This low response to selection was probably caused by the low heritability of body length, small sample size, and the low selection intensity(50%). The results suggest that A. sinica selective breeding programs must be changed to generate any substantial, sustainable genetic increases in body length. In industrial settings, maldistribution is normally considered to be caused by vertical positional or coplanarity errors stemming from deflections associated with manufacture and installation, or even by excessive weight. The lack ofestimation protocols or standards impedes the description ofthis error, which influences the corresponding outflow rates. Given this situation, the paper proposes a lumped parameter, orifice position deviation(OPD), to facilitate the calculation of the relative discharge rate error(RDRE)based on a formula derivation, which allows the systematic analysis of the influence ofa single orifice or weir OPD.The paper introduces a sensitivity factor K as a concise and unified expression in theoretical RDREs for calibrating the influence of OPD on the RDREs ofgeometrically different orifices and weirs. With respect to the GTLD, a larger K indicates the need for more strict OPD requirements.
