| 3 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 4 | Highly reversible lead-carbon battery anode with lead grafting on thecarbon surface显示文摘A novel C/Pb composite has been successfully prepared by electroless plating to reduce the hydrogen evolution and achieve the high reversibility of the anode of lead-carbon battery(LCB). The deposited lead on the surface of C/Pb composite was found to be uniform and adherent to carbon surface. Because lead has been stuck on the surface of C/Pb composite, the embedded structure suppresses the hydrogen evolution of lead-carbon anode and strengthens the connection between carbon additive and sponge lead.Compared with the blank anode, the lead-carbon anode with C/Pb composite displays excellent charge—discharge reversibility, which is attributed to the good connection between carbon additives and lead that has been stuck on the surface of C/Pb composite during the preparation process. | 5 | Pure drug nano-assemblies: A facile carrier-free nanoplatform for efficient cancer therapy显示文摘Nanoparticulate drug delivery systems(Nano-DDSs) have emerged as possible solution to the obstacles of anticancer drug delivery. However, the clinical outcomes and translation are restricted by several drawbacks, such as low drug loading, premature drug leakage and carrier-related toxicity.Recently, pure drug nano-assemblies(PDNAs), fabricated by the self-assembly or co-assembly of pure drug molecules, have attracted considerable attention. Their facile and reproducible preparation technique helps to remove the bottleneck of nanomedicines including quality control, scale-up production and clinical translation. Acting as both carriers and cargos, the carrier-free PDNAs have an ultra-high or even100% drug loading. In addition, combination therapies based on PDNAs could possibly address the most intractable problems in cancer treatment, such as tumor metastasis and drug resistance. In the present review, the latest development of PDNAs for cancer treatment is overviewed. First, PDNAs are classified according to the composition of drug molecules, and the assembly mechanisms are discussed. Furthermore, the co-delivery of PDNAs for combination therapies is summarized, with special focus on the improvement of therapeutic outcomes. | 6 | 2D/2D Bi_(2)MoO_(6)/g-C_(3)N_(4) S-scheme heterojunction photocatalyst with enhanced visible-light activity by Au loading显示文摘A novel 2D/2D Bi_(2)MoO_(6)/g-C_(3)N_(4) step-scheme(S-scheme)composite by loading Au as cocatalyst was successfully fabricated using a photoreduction and hydrothermal route.The obtained Bi_(2)MoO_(6)/g-C_(3)N_(4)/Au photocatalysts were characterized by X-ray diffraction(XRD),transmission electron microscope(TEM),X-ray photo-electron spectroscopy(XPS),UV–vis diffuse reflectance spectra(UV–vis),Fourier transform infrared spectroscopy(FTIR),photoluminescence(PL),photocurrent response(I-t),and electrochemical impedance spectroscopy(EIS).The HRTEM images revealed that an intimate interface in composites were formed.The optimum photocatalytic activity of Rhodamine B degradation over Bi_(2)MoO_(6)/g-C_(3)N_(4)/Au was about 9.7 times and 13.1 times as high as those of Bi_(2)MoO_(6) and g-C_(3)N_(4),respectively.The notably improved photocatalytic activity of Bi_(2)MoO_(6)/g-C_(3)N_(4)/Au could be mainly ascribed to the abundant active sites and the enhanced separation efficiency of photogenerated carriers in Bi_(2)MoO_(6)/g-C_(3)N_(4) S-scheme system.Notably,Au nanoparticles could act as a co-catalyst to further promote electron transfer and separation from the conduction band of g-C_(3)N_(4).Additionally,a possible step-scheme photocatalytic reaction mechanism of Rh B degradation over Bi_(2)MoO_(6)/g-C_(3)N_(4)/Au was tentatively proposed.PL and transient photocurrent analysis implied that Bi_(2)MoO_(6)/g-C_(3)N_(4)/Au photocatalysts possessed the lower recombination rate of photogenerated carriers compared with pure Bi_(2) MoO_(6) and g-C_(3)N_(4),respectively.The present work is expected to provide useful information in designing 2D/2D S-scheme heterojunction photocatalysts. | Qiaoqiao Li Wenli Zhao Zicheng Zhai Kaixu Ren Tingyu Wang Hao Guan Haifeng Shi | 2020 | Journal of Materials Science & Technology2020,56,21: | 4 |
| 7 | Design and application of 60mer oligonucleotide microarray in SARS coronavirus detection显示文摘The 60mer oligonucleotide microarray was designed and applied to detecting of SARS (severe acute res-piratory syndrome) coronavirus. Thirty 60mer specific oligos were designed to cover the whole genome of the first submit-ted coronavirus strain, according to the sequence of TOR2 (GENEBANK Accession: AY274119). These primers were synthesized and printed into a microarray with 12×12 spots. RNAs were extracted from the throat swab and gargling fluid of SARS patients and reverse-transcripted into the double strand cDNAs. The cDNAs were prepared as re-stricted cDNA fragments by the restriction display (RD) technique and labeled by PCR with the Cy5-universal primer. The labeled samples were then applied to the oligo microar-ray for hybridization. The diagnostic capability of the mi-croarray was evaluated after the washing and scanning steps. The scanning result showed that samples of SARS patients were hybridized with multiple SARS probes on the microar-ray, and there is no signal on the negative and blank controls. These results indicate that the genome of SARS coronavirus can be detected in parallel by the 60mer oligonucleotide mi-croarray, which can improve the positive ratio of the diagno-sis. | 13 | Asymmetric Distribution of Convection in Tropical Cyclones over the Western North Pacific Ocean显示文摘Forecasts of the intensity and quantitative precipitation of tropical cyclones(TCs) are generally inaccurate, because the strength and structure of a TC show a complicated spatiotemporal pattern and are affected by various factors. Among these, asymmetric convection plays an important role. This study investigates the asymmetric distribution of convection in TCs over the western North Pacific during the period 2005–2012, based on data obtained from the Feng Yun 2(FY2)geostationary satellite. The asymmetric distributions of the incidence, intensity and morphology of convections are analyzed.Results show that the PDFs of the convection occurrence curve to the azimuth are sinusoidal. The rear-left quadrant relative to TC motion shows the highest occurrence rate of convection, while the front-right quadrant has the lowest. In terms of intensity, weak convections are favored in the front-left of a TC at large distances, whereas strong convections are more likely to appear to the rear-right of a TC within a 300 km range. More than 70% of all MCSs examined here are elongated systems, and meso-β enlongated convective systems(MβECSs) are the most dominant type observed in the outer region of a TC. Smaller MCSs tend to be more concentrated near the center of a TC. | 16 | The Beaufort Gyre variation and its impacts on the Canada Basin in 2003–2012显示文摘The Beaufort Gyre(BG) was spun up in the last decade which is an important factor in regulating the variation of the upper ocean. The heat content and freshwater content of the upper ocean increased gradually in the Canada Basin, as did momentum input. Both the geostrophic wind curl and freshwater content could contribute to the spin-up of BG. However, even though there is no change of the wind field the increasing freshwater alone could result in the spin-up of BG. In this study we show that the Pacific Water is difficult to flow into the central basin as the BG spins up and the maximum temperature of the Pacific Summer Water(PSW) experienced a dramatic decrease inside the BG in 2005 and 2009 due to a change of flow pathway of PSW. The enhancement of Ekman Pumping(EP) contributed to the deepening of the Pacific Winter Water by piling up more freshwater. This change of water column dynamics has also contributed to the deepening ofthe Atlantic Water core after 2007. | 19 | Genome-wide identification of the CONSTANS-LIKE(COL)family and mechanism of fruit senescence regulation by PpCOL8 in sand pear(Pyrus pyrifolia)显示文摘Pyrus pyrifolia Nakai‘Whangkeumbae'is a sand pear fruit with excellent nutritional quality and taste.However,the industrial development of pear fruit is significantly limited by its short shelf life.Salicylic acid(SA),a well-known phytohormone,can delay fruit senescence and improve shelf life.However,the mechanism by which SA regulates CONSTANS-LIKE genes(COLs)during fruit senescence and the role of COL genes in mediating fruit senescence in sand pear are poorly understood.In this study,22 COL genes were identified in sand pear,including four COLs(Pp COL8,Pp COL9a,Pp COL9b,and Pp COL14)identified via transcriptome analysis and 18 COLs through genome-wide analysis.These COL genes were divided into three subgroups according to the structural domains of the COL protein.Pp COL8,with two B-box motifs and one CCT domain,belonged to the first subgroup.In contrast,the other three Pp COLs,Pp COL9a,Pp COL9b,and Pp COL14,with similar conserved protein domains and gene structures,were assigned to the third subgroup.The four COLs showed different expression patterns in pear tissues and were preferentially expressed at the early stage of fruit development.Moreover,the expression of Pp COL8 was inhibited by exogenous SA treatment,while SA up-regulated the expression of Pp COL9a and Pp COL9b.Interestingly,Pp COL8 interacts with Pp MADS,a MADS-box protein preferentially expressed in fruit,and SA up-regulated its expression.While the production of ethylene and the content of malondialdehyde(MDA)were increased in Pp COL8-overexpression sand pear fruit,the antioxidant enzyme(POD and SOD)activity and the expression of Pp POD1 and Pp SOD1 in the sand pear fruits were down-regulated,which showed that Pp COL8 promoted sand pear fruit senescence.In contrast,the corresponding changes were the opposite in Pp MADS-overexpression sand pear fruits,suggesting that Pp MADS delayed sand pear fruit senescence.The co-transformation of Pp COL8 and Pp MADS also delayed sand pear fruit senescence.The results of this study revealed that Pp COL8 can play a key role in pear fruit senescence by interacting with Pp MADS through the SA signaling pathway. | Yue Xu Shurui Song Huiying Wang Xilong Cao Xinran Zhao Wenli Wang Liyue Huo Yawei Li Misganaw Wassie Bin Lu Liang Chen Haiyan Shi | 2024 | Journal of Integrative Agriculture2024,23,4: | 0 |
| 20 | Cerium oxide nanozymes alleviate oxidative stress in tenocytes for Achilles tendinopathy healing显示文摘Background:Reactive oxygen species(ROS)is considered as ubiquitous and highly active chemicals that influence tendon integrity and orchestrate tendon repair.With significant recent advances in nanomaterials,cerium oxide nanoparticles(CeO_(2)NPs)exhibit superoxide dismutase-and catalase-like activities.Herein,we introduced a therapeutic approach of CeO_(2)NPs for Achilles tendinopathy(AT)healing.Methods:CeO_(2)NPs were synthesized to examine their effect as ROS scavengers on AT healing in vitro and in vivo.The mRNA levels of inflammatory factors were evaluated in AT after CeO_(2)NPs treatment in vitro.The mechanisms underlying CeO_(2)NPs-mediated stimulation of NRF2 translocation and ERK signaling were verified through immunofluorescence and Western blot analysis.The efficacy of CeO_(2)NPs was tested in an AT rat model in comparison with the control.Results:CeO_(2)NPs not only significantly scavenged multiple ROS and suppressed ROS-induced inflammatory reactions but also protected cell proliferation under oxidative stress induced by tert-butyl hydroperoxide(TBHP).Moreover,CeO_(2)NPs could promote NRF_(2)nuclear translocation for anti-oxidation and anti-inflammation through the ERK signaling pathway.In a rat model of collagenase-induced tendon injuries,CeO_(2)NPs showed significant therapeutic efficacy by ameliorating tendon damage.Conclusion:The present study provides valuable insights into the molecular mechanism of CeO_(2)NPs to ameliorate ROS in tenocytes via the ERK/NRF_(2)signaling pathway,which underscores the potential of CeO_(2)NPs for application in the treatment of enthesopathy healing. | Xingquan Xu Rongliang Wang Yixuan Li Rui Wu Wenjin Yan Sheng Zhao Quanyi Liu Yan Du Wenli Gong Weitong Li Hui Wei Dongquan Shi | 2023 | Nano Research2023,16,5: | 0 |
