| 1 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 4 | Increased Expression of Intercellular Adhesion Molecule-1,Vascular Cellular Adhesion Molecule-1 and Leukocyte Common Antigen in Diabetic Rat Retina显示文摘Purpose: To understand the expression and distribution of intercellular adhesion molecule- 1(ICAM- 1),vascular cellular adhesion molecule- 1 (VCAM- 1)and CD45 (Leukocyte Common Antigen) in the control nondiabetic and various courses of diabetic rats retina. To explore the role of adhesion molecules (Ams) and the adhesion of leukocytes to vascular endothelial cells via Ams in diabetic retinopathy(DR).Methods: Sixty healthy adult male Wistar rats were randomly divided into diabetic groups(induced by Streptozotocin, STZ) and normal control groups. Rats in these two groups were further randomly divided into 3, 7, 14, 30, 90 and 180 days-group,including 5 rats respectively. The immunohistochemical studies of ICAM-1, VCAM-1 and CD45 were carried out in the retinal digest preparations or retinal paraffin sections, and the results were analyzed qualitatively, semi-quantitatively.Results: No positive reaction of VCAM-1 was found, and weak reactions of ICAM-1,CD45 were found in nondiabetic rats retina. The difference of 6 control groups had no statistical significance(P > 0.05). The increased ICAM-1 and CD45 staining pattern were detectable 3 days after diabetes induction, and a few VCAM-1 positive cells were observed in the retinal blood capillaries. The difference of diabetes and control is significant( P < 0.05).Following the course, the expressions of ICAM-1, VCAM-1 and CD45 were increasingly enhanced, reaching a peak at the 14th day.Conclusion: Increased expression of ICAM-1, VCAM-1 and leukocytes adhering and stacking in retinal capillaries are the very early events in DR. | 6 | Photodegradation of methylmercury in the water body of the Three Gorges Reservoir显示文摘Biogeochemical cycling of mercury in the young Three Gorges Reservoir(TGR), China, is strongly considered. Although methylmercury(MMHg) photodegradation(PD) is an important process involved in mercury cycling in this zone, little is known about this process. In situ incubation experiments were therefore performed to quantify the effect of different wavelength radiations and environmental factors on the PD process of MMHg in the water bodies of TGR. It was found that the effect of solar radiation on MMHg PD was highly dependent on wavelength and water depth. All PD-rate constants resulting from each wavelength range decreased rapidly with water depth. For surface water, UV-A radiation(320?400 nm) was the key driver, accounting for 49%?62% of MMHg PD. For the entire water column, both photosynthetically active radiation(PAR, 400?700 nm) and UV-A were responsible for MMHg PD. MMHg PD fluxes peaked in summer(7.5?18 ng m?2 d?1), followed by spring(3.3?8.0 ng m?2 d?1), autumn(1.0?2.7 ng m?2 d?1), and winter(0.060?0.15 ng m?2 d?1). The annual fluxes of MMHg PD were estimated to be 1.1?2.8 μg m?2 a?1. Filtering the reservoir water and amending it with chemicals(i.e., Cl?, NO3?, and dissolved organic matter(DOM)) showed significant effects on MMHg PD rate constants. Stepwise regression analysis showed that intensity of solar radiation, suspended particulate matter(SPM), DOM, Cl?, and NO3? were involved in the PD process. Path analysis clarified the relationship between MMHg PD rate constants and environmental variables, as well as the comparative strength of direct and indirect relationships among variables. | 7 | Photodegradation of methylmercury in Jialing River of Chongqing, China显示文摘Photodegradation(PD) of methylmercury(MMHg) is a key process of mercury(Hg) cycling i water systems, maintaining MMHg at a low level in water systems. However, we posses little knowledge of this important process in the Jialing River of Chongqing, China. In sit incubation experiments were thus performed to measure temporal patterns and influencin factors of MMHg PD in this river. The results showed that MMHg underwent a ne demethylation process under solar radiation in the water column, which predominantl occurred in surface waters. For surface water, the highest PD rate constants were observed i spring(12 × 10-3± 1.5 × 10^(-3)m^2/E), followed by summer(9.0 × 10^(-3)± 1.2 × 10^(-3)m^2/E), autum(1.4 × 10^(-3)± 0.12 × 10^(-3)m^2/E), and winter(0.78 × 10^(-3)± 0.11 × 10^(-3)m^2/E). UV-A radiatio(320–400 nm), UV-B radiation(280–320 nm), and photosynthetically active radiation(PAR400–700 nm) accounted for 43%–64%, 14%–31%, and 16%–45% of MMHg PD, respectively. PD rat constants varied substantially with the treatments that filtered the river water and amended with chemicals(i.e., Cl-, NO_3^-, dissolved organic matter(DOM), Fe(III)), which reveals tha suspended particulate matter and water components are important factors in affecting the PD process. For the entire water column, the PD rate constant determined for each wavelengt range decreased rapidly with water depth. UV-A, UV-B, and PAR contributed 27%–46%, 6.2%12%, and 42%–65% to the PD process, respectively. PD flux was estimated to be 4.7 μg/(m^2·yea in the study site. | 9 | Roles of chloride ion in photo-reduction/oxidation of mercury显示文摘Photo-reduction of divalent mercury(Hg(II)) in aquatic systems plays a key role in global biogeochemistry cycling of mercury(Hg) in the Earth's surface environment. The mechanisms of this process with various Clconcentrations([Cl-]) under different pH values and irradiation wavelength ranges are still unclear. In this work,photo-reduction and photo-oxidation experiments of Hg with different [Cl-] and pH values under various light conditions were conducted. The results show that photoreduction rate constants of Hg(II) decrease with the increasing of [Cl-] in neutral solution under full light spectrum. Photo-reduction rate constants of Hg(II) with Cl-is highly dependent on Hg(II) species, which is determined by [Cl-] and pH value. Irradiation wavelength ranges have significant effects on reaction processes of photo-reduction of Hg(II) and photo-oxidation of Hg(0) in the presence of Cl-. When cut off ultraviolet(UV) radiation(280–400 nm), the reduction rate constants decrease without Cl-, and increase with higher [Cl-], and the photooxidation rates of Hg(0) decrease with or without Cl-.Except Cl-complexation stabilize the reducible Hg ions in solutions, photo-oxidation is an important reason for Cllowering photo-reduction rate of Hg(II). | 10 | Management of Post-traumatic Aniridia with Retinal Detachment显示文摘Purpose: To reconstruct the anatomic and functional impairment in patients with post-traumatic aniridia, aphakia, and retinal detachment.
Methods: Four patients with unilateral aniridia and aphakia as well as retinal detachment as results of severe eye injuries underwent scleral buckling, vitrectomy, membrane peeling, endolaser photocoagulation, silicone oil or gas temponade, combined with iris diaphragm-IOL implantation.
Results: All four patients achieved successfully anatomic and functional reconstruction after surgery. During five to 22 months postoperative follow-up, all retinas remained attached. The final visual acuity increased from finger counting to 0.1 ~ 0.3.
Conclusions: The combination of vitreoretinal surgery and iris diaphrgm-IOL implantation is an effective method for post-traumatic aniridia, aphakia and traumatic retinal detachment. It could ameliorate photophobia and improve the biocular vision.Furthermore, artificial iris diaphragm implantation could prevent silicone oil-endothelia contact and salvage silicone keratopathy. | 12 | The Preliminary Report of Pathological Changes of Epiretinal Membranes and Internal Limiting Membrane Removed during Idiopathic Macular Hole Surgery显示文摘Purpose:To investigate the pathological changes of epiretinal membranes(ERM)and internal limiting membrane (ILM) removed during idiopathic macular hole surgery.Methods:Ten consecutive patients with a unilateral idiopathic macular hole underwent pars plana vitrectomy(PPV) with the surgical removal of the ERMs overlying the hole and ILM surrounding the hole. The pathological features of the excised tissues were examined under the microscope. Results:According to the morphological changes, four ERMs showed cellular elements which looked like glia cells, macrophages, plasma cells, lymphocytes and fibroblast cells. Two of the ILM appeared as transparent membranes without cellular elements. The other eight ILM showed cellular elements on the transparent membranes.Conclusion: Our study supports the hypothesis that the tangential traction of vitreous and proliferative cellular elements on the inner surface of ILM causes idiopathic macular holes. Methods: Perfluorodecalin (0.05 ml/each) was injected into the anterior chambers of eighteen rabbit eyes. Corneal morphology and endothelial cells were monitored clinically by slit-lamp biomicroscope and specular microscope for 26 weeks. Results: Perfluorodecalin droplets looking like “ fish eggs ” were found at about 1/4-1/2 of the corneal height in the inferior anterior chamber. Corneal opacification on the area contacted with perfluorodecalin was observed in five eyes five weeks after injection, and all in the 22nd week. Mutton fat KPs in one eye were seen in the 6th week firstly, and in all eyes in the 7th week. Corneal pannus formation in one eye was present in the 4th week, two eyes in the 5th week and three eyes in the 6th week. Retrocorneal fibrous membrane in one eye was detected at the 6th week and 3 eyes at the 7th week respectively. After injection of perfluorodecalin, endothelial cell density was sighificantly decreased (2789 ± 192 vs. 2341 ± 658, P < O. O1 ) and corneal thickness was increased. Conclusions: Perfluorodecalin injected into anterior chamber can lead to corneal damage and inflammatory reaction.
