| 3 | Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images.Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases,comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance.Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. | 5 | Molecular characterization and expression of HSP70, HSF and HSBP genes in Octopus vulgaris during thermal stress显示文摘Temperature is an important environmental factor that affects the growth and survival of Octopus vulgaris, the common octopus. To understand the protective mechanism that O. vulgaris exhibits under heat stress, we used rapid amplification of c DNA ends(RACE) to obtain full-length sequences of three heat stress response related genes:(1) the heat shock protein 70(OvHSP70),(2) the heat shock transcription factor(OvHSF), and(3) the heat shock factor-binding protein(OvHSBP) of O. vulgaris. The OvHSP70, OvHSF, and OvHSBP proteins contained2 222 bp, 2 264 bp, 841 bp that encoded for 635, 458 and 90 amino acids, respectively. The results of multiple sequence alignment showed that the amino acid sequences of OvHSP70 were highly conserved with respect to other species. Similarly, the DNA binding domain, the trimerization domain of OvHSF, and the coiled coil region of OvHSBP also had highly conserved regions. The real-time polymerase chain reaction(PCR) results indicated that OvHSP70 was temperature-dependent and time-dependent, showing a positive response to heat stress. On exposure to 28°C and to 30°C, the m RNA expression levels of OvHSF and OvHSBP were higher than those in the control group at 24°C. The m RNA expression of OvHSBP significantly increased with heat treatment at 26°C, while the m RNA expression of OvHSF decreased. The experimental results indicated that the expression of OvHSP70,OvHSF and OvHSBP were all sensitive to heat stress, which suggests that these three genes may play an important role for O. vulgaris in responding to environmental stress. | 6 | Ablation of Mea6/cTAGE5 in oligodendrocytes significantly impairs white matter structure and lipid content显示文摘Lipid-rich myelin is a special structure formed by oligodendrocytes wrapping neuronal axons.Abnormal myelin sheath is associated with many neurological diseases.Meningioma-expressed antigen 6(Mea6)/cutaneous T cell lymphoma-associated antigen 5C(cTAGE5C)plays an important role in vesicle trafficking from the endoplasmic reticulum(ER)to Golgi,and conditional knockout(cKO)of Mea6 in the brain significantly affects neural development and brain function.However,whether the impaired brain function involves the development of oligodendrocytes and white matter beyond neurons remains unclear.In this study,by using different models of diffusion magnetic resonance imaging,we showed that cKO of Mea6 in oligodendrocytes leads to significant impairment of the gross and microstructure of the white matter,as well as a significant decrease of cholesterol and triglycerides in brains.Our lipidomic analysis of purified myelin sheath for the first time showed that Mea6 elimination in oligodendrocytes significantly altered the lipid composition in myelin lipidome,especially the proportion of very long chain fatty acids(VLCFAs).In particular,the levels of most VLCFA-containing phosphatidylcholines were substantially lower in the myelin sheath of the cKO mice.The reduction of VLCFAs is likely due to the downregulated expression of elongation of very long chain fatty acids(ELOVLs).Our study of an animal model with white matter malformation and the comprehensive lipid profiling would provide clues for future studies of the formation of myelin sheath,myelin lipids,and the pathogenesis of white matter diseases. | Tiantian Ma Wei Mao Shaohua Zhang Yaqing Wang Tao Wang Jinghua Liu Lei Shi Xiang Yu Rong Xue Guanghou Shui Zhiheng Xu | 2023 | Life Metabolism2023,2,2: | 0 |

Firstly, a sampled-data descriptor model with time-vary delay is proposed and transformed into a discrete-time non-singular one. Then, a robust sensor fault diagnosis observer is proposed based on the state estimation error and the measurement residual, this observer can guarantee the robustness of the residual against the augmented disturbance and the sensor fault, which means the H∞ performance index is satisfied. As the confining matrix of the designed observer parameters does not meet the Linear Matrix Inequality (LMI), a cone complementary linearization (CCL) algorithm is proposed to solve this problem. The decision logic of the residual is obtained by the residual evaluation function. | 10 | Chemotactic nanomotor for multimodal combined therapy of glioblastoma显示文摘Glioblastoma(GBM) is the most aggressive malignant brain tumor. Due to the infiltration and heterogeneity of GBM, the obstruction of the blood-brain barrier(BBB) and the unique immunosuppressive mechanism, it is hard to achieve significant effects of GBM treatment. Here, a kind of chemotactic nanomotor that loaded with glucose oxidase(GOx) and carboxylated cisplatin(Pt(IV)) prodrug on the L-arginine-derived polymer is proposed. The nanomotors are driven by catalysis of glucose decomposition and the positive chemotaxis towards the GBM microenvironment where inducible nitric oxide synthase and reactive oxygen species are highly expressed. This facilitates the BBB crossing and GBM targeting of the nanomotors. In addition, the released nitric oxide(NO) during propulsion as well as the loaded GOx and Pt(IV) can exert combined NO/starvation/chemotherapy.
