1Temperature Variable Optimization for Precision Machine Tool Thermal Error Compensation on Optimal Threshold显示文摘Machine tool thermal error is an important reason for poor machining accuracy.Thermal error compensation is a primary technology in accuracy control.To build thermal error model,temperature variables are needed to be divided into several groups on an appropriate threshold.Currently,group threshold value is mainly determined by researchers experience.Few studies focus on group threshold in temperature variable grouping.Since the threshold is important in error compensation,this paper arms to find out an optimal threshold to realize temperature variable optimization in thermal error modeling.Firstly,correlation coefficient is used to express membership grade of temperature variables,and the theory of fuzzy transitive closure is applied to obtain relational matrix of temperature variables.Concepts as compact degree and separable degree are introduced.Then evaluation model of temperature variable clustering is built.The optimal threshold and the best temperature variable clustering can be obtained by setting the maximum value of evaluation model as the objective.Finally,correlation coefficients between temperature variables and thermal error are calculated in order to find out optimum temperature variables for thermal error modeling.An experiment is conducted on a precise horizontal machining center.In experiment,three displacement sensors are used to measure spindle thermal error and twenty-nine temperature sensors are utilized to detect the machining center temperature.Experimental result shows that the new method of temperature variable optimization on optimal threshold successfully worked out a best threshold value interval and chose seven temperature variables from twenty-nine temperature measuring points.The model residual of z direction is within 3 m.Obviously,the proposed new variable optimization method has simple computing process and good modeling accuracy,which is quite fit for thermal error compensation.ZHANG Ting YE Wenhua LIANG Ruijun LOU Peihuang YANG Xiaolan 2013Chinese Journal of Mechanical Engineering2013,26,1:11
2Preparation, characterization, pharmacokinetics and anticancer effects of PEGylated β-elemene liposomes显示文摘Objective:This study aimed to develop a new polyethylene glycol(PEG)ylatedβ-elemene liposome(PEG-Lipo-β-E)and evaluate its characterization,pharmacokinetics,antitumor effects and safety in vitro and in vivo.Methods:The liposomes were prepared by ethanol injection and high-pressure micro-jet homogenization.Characterization of the liposomes was conducted,and drug content,entrapment efficiency(EE),in vitro release and stability were studied by ultra-fast liquid chromatography(UFLC)and a liquid surface method.Blood was drawn from rats to establish the pharmacokinetic parameters.The anticancer effect was evaluated in a KU-19-19 bladder cancer xenograft model.Histological analyses were performed to evaluate safety.Results:The PEG-Lipo-β-E showed good stability and was characterized as 83.31±0.181 nm in size,0.279±0.004 in polydispersity index(PDI),-21.4±1.06 mV in zeta potential,6.65±0.02 in pH,5.024±0.107 mg/mL inβ-elemene(β-E)content,and 95.53±1.712%in average EE.The Fourier transform infrared spectroscopy(FTIR)and differential scanning calorimetry(DSC)indicated the formation of PEG-Lipo-β-E.Compared to elemene injection,PEG-Lipo-β-E demonstrated a 1.75-fold decrease in clearance,a 1.62-fold increase in half-life,and a 1.76-fold increase in area under the concentration-time curves(AUCs)from 0 hour to 1.5 hours(P<0.05).PEG-Lipo-β-E also showed an enhanced anticancer effect in vivo.Histological analyses showed that there was no evidence of toxicity to the heart,kidney,liver,lung or spleen.Conclusions:The present study demonstrates PEG-Lipo-β-E as a new formulation with ease of preparation,high EE,good stability,improved bioavailability and antitumor effects.Bingtao Zhai Qibiao Wu Wengang Wang Mingming Zhang Xuemeng Han Qiujie Li Peng Chen Xiaying Chen Xingxing Huang Guohua Li Qin Zhang Ruonan Zhang Yu Xiang Shuiping Liu Ting Duan Jianshu Lou Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,1:6

4Preparation of a novel PAN/cellulose acetate-Ag based activated carbon nanofiber and its adsorption performance for low-concentration SO_2显示文摘Polyacrylonitrile(PAN), PAN/cellulose acetate(CA), and PAN/CA-Ag based activated carbon nanofiber(ACNF) were prepared using electrostatic spinning and further heat treatment. Thermogravimetry–differential scanning calorimetry(TG–DSC) analysis indicated that the addition of CA or Ag did not have a significant impact on the thermal decomposition of PAN materials but the yields of fibers could be improved. Scanning electron microscopy(SEM) analysis showed that the micromorphologies of produced fibers were greatly influenced by the viscosity and conductivity of precursor solutions. Fourier transform infrared spectroscopy(FT-IR) analysis proved that a cyclized or trapezoidal structure could form and the carbon scaffold composed of C=C bonds appeared in the PAN-based ACNFs. The characteristic diffraction peaks in X-ray diffraction(XRD) spectra were the evidence of a turbostratic structure and silver existed in the PAN/CA-Ag based ACNF. Brunner–Emmett–Teller(BET) analysis showed that the doping of CA and Ag increased surface area and micropore volume of fibers; particularly, PAN/CA-Ag based ACNF exhibited the best porosity feature. 5PCDH17 increases the sensitivity of colorectal cancer to 5-fluorouracil treatment by inducing apoptosis and autophagic cell death显示文摘5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical success.Our previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in CRC.However,the relationship between PCDH17 and 5-FU resistance in CRC remains unclear.Here,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues,and high expression of PCDH17 was correlated with high BECN1 expression.Moreover,this expression profile contributed to superior prognosis and increased survival in CRC patients.Restoring PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell death.Furthermore,autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor Z-VAD-FMK.PCDH17 inhibition by siRNA decreased the autophagy response and 5-FU sensitivity.Mechanistically,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced autophagy.The compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC cells.Taken together,our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell death.PCDH17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients.Shuiping Liu Haoming Lin Da Wang Qiang Li Hong Luo Guoxiong Li Xiaohui Chen Yongqiang Li Peng Chen Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Mingming Zhang Xuemeng Han Qiujie Li Liuxi Chen Ying Liu Xiaying Chen Guohua Li Yu Xiang Ting Duan Jiao Feng Jianshu Lou Xingxing Huang Qin Zhang Ting Pan Lili Yan Ting Jin Wenzheng Zhang Lvjia Zhuo Yitian Sun Tian Xie Xinbing Sui 2019Signal Transduction and Targeted Therapy2019,4,1:4

7Joint association analysis method to dissect complex genetic architecture of multiple genetically related traits显示文摘Genome-wide association study(GWAS)has been a standard approach to discover the genetic determinants underlying complex traits.It is a major challenge in GWAS how to improve analysis power,uncover complex genetic correlation,and reveal gene-gene and gene-environment interactions through integrated analysis of multiple genetically related traits.To combat these challenges,we proposed a mixed linear model-based joint association analysis method for multiple traits,which include epistasis and geneenvironment interaction in the mapping model and utilize within-trait variance and between-trait covariance simultaneously;A F-statistics based on Wilks statistics is used to test the significance of each SNP and paired interacted SNPs,each genetic effects of QTS are estimated and tested by the MCMC method based on a QTS full model.Simulations showed that the multi-trait GWAS method could provide increased power in detecting pleiotropic loci affecting more than one trait,and can unbiasedly estimate effects of QTS.To demonstrate the performance of the proposed method,we analyzed four blood lipid traits in Multi-Ethnic Study of Atherosclerosis(MESA)Cohort and two yield-related traits in a rice immortalized F2 dataset.A software package was developed for the proposed method.Feng Lin Guoan Qi Ting Xu Xiangyang Lou Yongbo Hong Haiming Xu 2020The Crop Journal2020,8,5:2

10Molecular and structural characterization of a promiscuous chalcone synthase from the fern species Stenoloma chusanum显示文摘The key enzymes involved in the flavonoid biosynthesis pathway have been extensively studied in seed plants,but relatively less in ferns.In this study,two 4-Coumarate:coenzyme A ligases(Sc4CL1 and Sc4CL2)and one novel chalcone synthase(ScCHS1)were functionally characterized by mining the Stenoloma chusanum transcriptome database.Recombinant Sc4CLs were able to esterify various hydroxycinnamic acids to corresponding acyl-coenzyme A(CoA).ScCHS1could catalyze p-coumaroyl-CoA,cinnamoyl-CoA,caffeoyl-CoA,and feruloyl-CoA to form naringenin,pinocembrin,eriodictyol,and homoeriodictyol,respectively.Moreover,enzymatic kinetics studies revealed that the optimal substrates of ScCHS1were feruloyl-CoA and caffeoyl-CoA,rather than p-coumaroyl-CoA,which was substantially different from the common CHSs.Crystallographic and sitedirected mutagenesis experiments indicated that the amino acid residues,Leu87,Leu97,Met165,and Ile200,located in the substrate-binding pocket near the B-ring of products,could exert a significant impact on the unique catalytic activity of ScCHS1.Furthermore,overexpression of ScCHS1 in tt4 mutants could partially rescue the mutant phenotypes.Finally,ScCHS1 and Sc4CL1 were used to synthesize flavanones and flavones with multi-substituted hydroxyl and methoxyl B-ring in Escherichia coli,which can effectively eliminate the need for the cytochrome P450 hydroxylation/O-methyltransferase from simple phenylpropanoid acids.In summary,the identification of these important Stenoloma enzymes provides a springboard for the future production of various flavonoids in E.coli.Rong Ni Meng Niu Jie Fu Hui Tan Ting‐Ting Zhu Jing Zhang Hong‐Xiang Lou Peng Zhang Jian‐Xu Li Ai‐Xia Cheng 2022Journal of Integrative Plant Biology2022,64,10:1

16Plane Analysis for an Inclusion in ID Hexagonal Quasicrys tai Using the Hyp er singular Integral Equation Method显示文摘A model of a thin elastic inclusion embedded in an infinite ID hexagonal quasicrystal is discussed. The atomic arrangements of the matrix and the inclusion are both periodic along the xi-direction and quasiperiodic along the a;2-direction in the oxiX2-coordinate system. Using the hypersingular integral equation method, the inclusion problem is reduced to solving a set of hypersingular integral equations. Based on the exact analytical solution of the singular phonon and phason stresses near the inclusion front, a numerical method of the hypersingular integral equation is proposed using the finite-part integral method. 17Lysosomal Hydrolase Cathepsin D Non-proteolytically Modulates Dendritic Morphology in Drosophila显示文摘The main lysosomal protease cathepsin D(cathD)is essential for maintaining tissue homeostasis via its degradative function,and its loss leads to ceroid accumulation in the mammalian nervous system,which results in progressive neurodegeneration.Increasing evidence implies non-proteolytic roles of cathD in regulating various biological processes such as apoptosis,cell proliferation,and migration.Along these lines,we here showed that cathD is required for modulating dendritic architecture in the nervous system independent of its traditional degradative function.Upon cathD depletion,class I and class III arborization(da)neurons in Drosophila larvae exhibited aberrant dendritic morphology,including overbranching,aberrant turning,and elongation defects.Reintroduction of wild-type cathD or its proteolyticallyinactive mutant dramatically abolished these morphological defects.Moreover,cathD knockdown also led to dendritic defects in the adult mushroom bodies,suggesting that cathD-mediated processes are required in both the peripheral and central nervous systems.Taken together,our results demonstrate a critical role of cathD in shaping dendritic architecture independent of its proteolytic function.Ting Zhang Daxiao Cheng Cunjin Wu Xingyue Wang Qiang Ke Huifang Lou Liya Zhu Xiao-Dong Wang Shumin Duan Yi-Jun Liu 2020Neuroscience Bulletin2020,36,10:1
18Effective interventions on health effects of Chinese rural elderly under heat exposure显示文摘Due to climate change,the heatwave has become a more serious public health threat with aging as an aggravating factor in recent years.There is a pressing need to detect the most effective prevention and response measures.However,the specific health effects of interventions have not been characterized on an individual scale.In this study,an intervention experiment was designed to explore the health effects of heat exposure at the individual level and assess the effects of different interventions based on a comprehensive health sensitivity index(CHSI)in Xinyi,China.Forty-one subjects were recruited randomly,and divided into one control group and three intervention groups.Interventions included education(Educate by lecturing,offering relative materials,and communication),subsidy support(offer subsidy to offset the cost of running air conditioning),and cooling-spray(install a piece of cooling-spray equipment in the yard).Results showed that systolic blood pressure(SBP)and deep sleep duration(DSD)were significantly affected by short-term heat exposure,and the effects could be alleviated by three types of interventions.The estimated CHSI indicated that the effective days of the education group were longer than other groups,while the lower CHSI of the subsidy group showed lower sensitivity than the control group.These findings provide feasible implementation strategies to optimize Heat-health action plans and evaluate the intervention performance.Yujia Huang Ting Zhang Jianing Lou Peng Wang Lei Huang 2022Frontiers of Environmental Science & Engineering2022,16,5:1
