2African Swine Fever Virus MGF360-12L Inhibits Type Ⅰ Interferon Production by Blocking the Interaction of Importin α and NF-κB Signaling Pathway显示文摘African swine fever(ASF)is an infectious transboundary disease of domestic pigs and wild boar and spreading throughout Eurasia.There is no vaccine and treatment available.Complex immune escape strategies of African swine fever virus(ASFV)are crucial factors affecting immune prevention and vaccine development.MGF360 genes have been implicated in the modulation of the IFN-Ⅰresponse.The molecular mechanisms contributing to innate immunity are poorly understood.In this study,we demonstrated that ASFV MGF360-12 L(MGF360 families 12 L protein)significantly inhibited the mRNA transcription and promoter activity of IFN-βand NF-κB,accompanied by decreases of IRF3,STING,TBK1,ISG54,ISG56 and AP-1 m RNA transcription.Also,MGF360-12 L might suppress the nuclear localization of p50 and p65 mediated by classical nuclear localization signal(NLS).Additionally,MGF360-12 L could interact with KPNA2,KPNA3,and KPNA4,which interrupted the interaction between p65 and KPNA2,KPNA3,KPNA4.We further found that MGF360-12 L could interfere with the NF-κB nuclear translocation by competitively inhibiting the interaction between NF-κB and nuclear transport proteins.These findings suggested that MGF360-12 L could inhibit the IFN-Ⅰproduction by blocking the interaction of importinαand NF-κB signaling pathway,which might reveal a novel strategy for ASFV to escape the host innate immune response.Yisha Zhuo Zeheng Guo Tongtong Ba Cheng Zhang Lihua He Cuiping Zeng Hanchuan Dai 2021Virologica Sinica2021,36,2:21
3The poisoning and regeneration effect of alkali metals deposed over commercial V_2O_5-WO_3/TiO_2 catalysts on SCR of NO by NH_3显示文摘In this study,commercial V2O5-WO3/TiO2catalysts were deactivated by loading with alkali metals(K and Na).These catalysts were then regenerated by washing with either deionized water or 0.5 mol/L H2SO4(through the ultrasonic-assisted method).The samples used in this research were characterized by NH3-temperature programmed desorption(TPD),and X-ray photoelectron spectroscopy(XPS).Results showed that Na2O and K2O doping can poison the V2O5-WO3/TiO2catalyst and that the poisoning effect of Na2O was stronger than that of K2O.However,the Na2O-loaded sample was easier to regenerate than the K2O-loaded sample.The surfaces of catalysts can be sulfated by washing with dilute sulfuric acid because strong acid sites adhere to the catalyst surface.SO42-could also promote catalyst activity.As indicated by the NH3-TPD findings,the deposition of Na2O and K2O could also reduce the amount of desorbed ammonia and destabilize the acid sites,especially strong chemisorption sites.XPS results revealed that catalysts were deactivated by the decrease in the concentration of chemisorbed oxygen[the Oa/(Oα+Oβ)ratio].In the Na2O-doped catalyst,much chemisorbed oxygen was lost(from 28.8%to10.6%).However,the decrease in the Oa/(Oα+Oβ)ratio was less significant in the K2O-doped catalyst(from28.8%to 23.5%).Nonetheless,the binding energies of O1s broadened with respect to both high and low energy.In particular,the binding energy of chemisorbed oxygen increased from 531.5 to 531.8 eV.Fengyu Gao Xiaolong Tang Honghong Yi Shunzheng Zhao Tongtong Zhang Dong Li Ding Ma 2014Chinese Science Bulletin2014,59,31:19

7Inhalation treatment of primary lung cancer using liposomal curcumin dry powder inhalers显示文摘Lung cancer is the leading cause of cancer-related deaths. Traditional chemotherapy causes serious toxicity due to the wide bodily distribution of these drugs. Curcumin is a potential anticancer agent but its low water solubility, poor bioavailability and rapid metabolism significantly limits clinical applications. Here we developed a liposomal curcumin dry powder inhaler(LCD) for inhalation treatment of primary lung cancer. LCDs were obtained from curcumin liposomes after freeze-drying. The LCDs had a mass mean aerodynamic diameter of 5.81 μm and a fine particle fraction of 46.71%, suitable for pulmonary delivery. The uptake of curcumin liposomes by human lung cancer A549 cells was markedly greater and faster than that of free curcumin. The high cytotoxicity on A549 cells and the low cytotoxicity of curcumin liposomes on normal human bronchial BEAS-2B epithelial cells yielded a high selection index partly due to increased cell apoptosis. Curcumin powders, LCDs and gemcitabine were directly sprayed into the lungs of rats with lung cancer through the trachea. LCDs showed higher anticancer effects than the other two medications with regard to pathology and the expression of many cancer-related markers including VEGF, malondialdehyde, TNF-α, caspase-3 and BCL-2. 8Mechanism of stall and surge in a centrifugal compressor with a variable vaned diffuser显示文摘To expand the stable operating range of compressors, understanding the mechanism of flow instability at low flow rates is necessary. In this paper, the mechanism of stall and surge in a centrifugal compressor with a variable vaned diffuser is experimentally investigated, where the diffuser blade setting angle can be adjusted. Many dynamic pressure transducers are mounted on the casing surface of the compressor. From the design condition to surge, dynamic pressure data is recorded throughout the gradual process. According to the signal developing status, the typical modes of compressor instability are defined in detail, such as stall, mild surge, and deep surge. A relatively high-frequency stall wave originates in the impeller and propagates to the diffuser, and finally stimulates a deep surge in the compressor. The compressor behavior during surge differs at different diffuser vane angles. When the diffuser vane angle is adjusted, both the unstable form and the core factor affecting the overall machine stability change. 10Applicability of five models to simulate water infiltration into soil with added biochar显示文摘As a soil amendment, biochar can reduce soil bulk density, increase soil porosity, and alter soil aggregates and thus affect the infiltration. Researchers have proposed and revised several theoretical models to describe the process of soil infiltration. Although these models have been successfully used to evaluate the soil infiltration in different scenarios in agricultural fields, little effort has been devoted to assess their performances in arid and semi-arid soils after the addition of biochar. A laboratory experiment was performed to study the infiltration characteristics of two typical Loess Plateau soils at three particle sizes(2–1, 1–0.25, and <0.25 mm) and five biochar application amounts(0, 10, 50, 100, and 150 g/kg). The performance of five models(i.e., the Philip model, Kostiakov model, Mezencev model, USDA-NRCS model, and Horton model) in simulating the infiltration process was then evaluated based on the adjusted coefficient of determination and a reduced Chi-Square test. Results indicated that the Horton model best simulated the water-infiltration process in an aeolian sandy soil with added biochar. However, the Mezencev model best simulated the infiltration process in a loamy clay soil(Eum-Orthic Anthrosol). The three-parameter model, i.e., Mezencev and Horton models can better describe the relationship between cumulative infiltration and infiltration time. 11Oxygen and nitrogen-doped metal-free carbon catalysts for hydrochlorination of acetylene显示文摘Activated carbon was tested as metal-free catalyst for hydrochlorination of acetylene in order to circumvent the problem of environment pollution caused by mercury and high cost by noble metals. Oxygen-doped and nitrogen-doped activated carbons were prepared and characterized by XPS, TPD and N2 physisorption methods.The influences of the surface functional groups on the catalytic performance were discussed base on these results.Among all the samples tested, a nitrogen-doped sample, AC-n-U500, exhibited the best performance, the acetylene conversion being 92% and vinyl chloride selectivity above 99% at 240 °C and C2H2 hourly space velocity30 h-1. Moreover, the AC-n-U500 catalyst exhibited a stable performance during a 200 h test with a conversion of acetylene higher than 76% at 210 °C at a C2H2 hourly space velocity 50 h-1. In contrary, oxygen-doped catalyst had lower catalytic activities. A linear relationship between the amount of pyrrolic-N and quaternary-N species and the catalytic activity was observed, indicating that these nitrogen-doped species might be the active sites and the key in tuning the catalytic performance. It is also found that the introduction of nitrogen species into the sample could significantly increase the adsorption amount of acetylene. 12Phytochrome B interacts with SWC6 and ARP6 to regulate H2A.Z deposition and photomorphogensis in Arabidopsis显示文摘Light serves as a crucial environmental cue which modulates plant growth and development, and which is controlled by multiple photoreceptors including the primary red light photoreceptor,phytochrome B(phyB). The signaling mechanism of phyB involves direct interactions with a group of basic helix-loop-helix(bHLH) transcription factors, PHYTOCHROME-INTERACTING FACTORS(PIFs), and the negative regulators of photomorphogenesis, COP1 and SPAs. H2 A.Z is an evolutionarily conserved H2 A variant which plays essential roles in transcriptional regulation. The replacement of H2 A with H2 A.Z is catalyzed by the SWR1 complex. Here, we show that the Pfr form of phyB physically interacts with the SWR1 complex subunits SWC6 and ARP6. phyB and ARP6 coregulate numerous genes in the same direction,some of which are associated with auxin biosynthesis and response including YUC9, which encodes a rate-limiting enzyme in the tryptophandependent auxin biosynthesis pathway. Moreover,phyB and HY5/HYH act to inhibit hypocotyl elongation partially through repression of auxin biosynthesis. 13A review of soil nematodes as biological indicators for the assessment of soil health显示文摘Healthy soils are essential for sustainableagricultural development and soil health requires carefulassessment with increasing societal concern over envir-onmentally friendly agricultural development. Soil healthis the capacity of soil to function within ecologicalboundaries to sustain productivity, maintain environmentalquality, and promote plant and animal health. Physical,chemical and biological indicators are used to evaluate soilhealth;the biological indicators include microbes, proto-zoa and metazoa. Nematodes are the most abundantmetazoa and they vary in their sensitivity to pollutants andenvironmental disturbance. Soil nematode communitiesare useful biological indicators of soil health, withcommunity characteristics such as abundance, diversity,community structure and metabolic footprint all closelycorrelated with the soil environment. The community size,complexity and structure reflect the condition of the soil.Both free-living and plant parasitic nematodes are effec-tive ecological indicators, contributing to nutrient cyclingand having important roles as primary, secondary andtertiary consumers in food webs. 14Facile Fabrication of Cellulosic Paper-based Composites with Temperature-controlled Hydrophobicity and Excellent Mechanical Strength显示文摘In this paper,we presented a novel strategy to employ a plantderived carbohydrate polymer,i.e.,cellulose,to prepare a hydrophobic composite.Cellulose was used as a scaffold,and ethylene-propylene side by side(ES)fiber was thermally melted and then coated on the cellulose surface to achieve hydrophobicity.Experimental results revealed that the thermocoating ES fibers greatly increased the water contact angle of the cellulose scaffold from 25°to 153°while simultaneously enhanced the wet tensile strength of the composite approximately 6.7-fold(drying temperature of 170℃)compared with the pure cellulose paper.In particular,compared with other related research,the prepared cellulose-based composite possessed excellent hydrophobicity and superior mechanical strength,which introduces a new chemical engineering approach to prepare hydrophobic cellulose-based functional materials.Tongtong Yun Yilin Wang Jie Lu Yi Cheng Yanna Lyu Haisong Wang 2020Paper And Biomaterials2020,5,2:6
15Predictors of post coronary artery bypass grafting atrial fibrillation显示文摘Objective: To investigate the incidence and relative risk factors of post coronary artery bypass grafting(post-CABG) atrial fibrillation(AF). Methods: 312 patients with CABG were reviewed and divided into an AF group and a non-AF group. Statistical analysis was used to compare the data between the two groups and screen for risk factors of post-CABG AF. Results: 103/312 (33.01%) patients developed post-CABG AF. Univariate analysis showed that patients in AF group compared with those in non-AF group were more likely to have advanced age (≥70 years), early postoperative withdrawal of β-blockers, hypertension, left atrial enlargement (≥40 mm), a history of AF, prolonged p-wave duration (≥120 ms) and increased number of grafts (≥3). Multivariate logistic regression analysis showed that advanced age (≥70 years), early postoperative withdrawal of β-blockers, hypertension, left atrial enlargement (≥40 mm) and a history of AF were highly related to post-CABG AF. Conclusion: The incidence of AF in patients following CABG was 33.01% in this study. 16Efficacy and safety of duloxetine in Chinese breast cancer patients with paclitaxel-induced peripheral neuropathy显示文摘Objective: Chemotherapy with paclitaxel is associated with significant neurotoxicity that may offset patients' quality of life and therapeutic benefits. This prospective, non-randomized control study evaluated the efficacy and safety of an antidepressant drug, duloxetine, at 30 or 60 mg/d, in the treatment of paclitaxel-induced peripheral neuropathy(PIPN) in Chinese breast cancer patients.Methods:A total of 102 patients with a median age of 50(range,25–60)years,treated in the Department of Medical Oncology,National Cancer Center/Cancer Hospital,Chinese Academy of Medical Sciences and Peking Union Medical College,between November 2014 and January 2017 were finally enrolled.Stratified by baseline characteristics,the patients were classified into two groups,receiving either duloxetine or alternative antineurotoxicity drugs.During the course of the paclitaxel regimen,the eligibility criteria included sensory neuropathy,as evaluated by the National Cancer Institute-Common Toxicity Criteria for Adverse Events.The treatment consisted of receiving 30 mg duloxetine(for the first 4 weeks)and 60 mg duloxetine for an additional 8 weeks,or any other anti-neurotoxicity drug daily during the same crossover period.The improvement associated with PIPN from the patient’s perspective were assessed by the Functional Assessment of Cancer Therapy-Taxane(FACT-Tax)Scales,which contained questions scored from 0 to 4(0,not at all;4,very much;total score range,0–44).Results:Duloxetine was more effective in decreasing PIPN(odds ratio=5.426;95%confidence interval,1.898–15.514;P=0.002).Between duloxetine group and control group,the median(25th–75th percentiles)decreasing difference in the FACT-Tax pain score was 4(2–6)vs.1(0–4)(P=0.005).Conclusions:Duloxetine is a promising and safe option with tolerable toxicity at a dose of 60 mg/d for Chinese breast cancer patients with PIPN.Non-neuropathy adverse events were mild and similar in both groups.The major toxicities of duloxetine included nausea,constipation,somnolence,dizziness and distention of the eyes.Further examination of the benefits of duloxetine in the prevention of PIPN is required.Jiani Wang Qing Li Binghe Xu Tongtong Zhang Shanshan Chen Yang Luo 2017Chinese Journal of Cancer Research2017,29,5:5
17Human genes influence the interaction between Streptococcus mutans and host caries susceptibility: a genome-wide association study in children with primary dentition显示文摘Streptococcus mutans is a well-known cause of dental caries,due to its acidogenicity,aciduricity,and ability to synthesize exopolysaccharides in dental plaques.Intriguingly,not all children who carry S.mutans manifest caries,even with similar characteristics in oral hygiene,diet,and other environmental factors.This phenomenon suggests that host susceptibility potentially plays a role in the development of dental caries;however,the association between host genetics,S.mutans,and dental caries remains unclear.Therefore,this study examined the influence of host gene-by-S.mutans interaction on dental caries.Genome-wide association analyses were conducted in 709 US children (<13 years old),using the dbGap database acquired from the center for oral health research in appalachia (COHRA) and the Iowa Head Start programmes (GEIRS).A generalized estimating equation was used to examine the gene-by-S.mutans interaction effects on the outcomes (decayed and missing/filled primary teeth due to caries).Sequentially,the COHRA and GEIRS data were used to identify potential interactions and replicate the findings.Three loci at the genes interleukin 32 (IL32),galactokinase 2 (GALK2),and CUGBP,Elav-like family member 4 (CELF4) were linked to S.mutans carriage,and there was a severity of caries at a suggestive significance level among COHRA children (P < 9×10?5),and at a nominal significance level among GEIRS children (P = 0.047–0.001).The genetic risk score that combined the three loci also significantly interacted with S.mutans (P < 0.000 1).Functional analyses indicated that the identified genes are involved in the host immune response,galactose carbohydrate metabolism,and food-rewarding system,which could potentially be used to identify children at high risk for caries and to develop personalized caries prevention strategies.Ying Meng Tongtong Wu Ronald Billings Dorota T. 18Bimetallic Nickel Cobalt Sulfide as E cient Electrocatalyst for Zn–Air Battery and Water Splitting显示文摘The development of e cient earth-abundant electrocatalysts for oxygen reduction, oxygen evolution, and hydrogen evolution reactions(ORR, OER, and HER) is important for future energy conversion and energy storage devices, for which both rechargeable Zn–air batteries and water splitting have raised great expectations. Herein, we report a single-phase bimetallic nickel cobalt sulfide((Ni,Co)S_2) as an e cient electrocatalyst for both OER and ORR. Owing to the synergistic combination of Ni and Co, the(Ni,Co)S_2 exhibits superior electrocatalytic performance for ORR, OER, and HER in an alkaline electrolyte, and the first principle calculation results indicate that the reaction of an adsorbed O atom with a H_2O molecule to form a *OOH is the potential limiting step in the OER. Importantly, it could be utilized as an advanced air electrode material in Zn–air batteries, which shows an enhanced charge–discharge performance(charging voltage of 1.71 V and discharge voltage of 1.26 V at 2 mA cm^(-2)), large specific capacity(842 mAh g_(Zn)^(-1) at 5 mA cm^(-2)), and excellent cycling stability(480 h). Interestingly, the(Ni,Co)S_2-based Zn–air battery can e ciently power an electrochemical water-splitting unit with(Ni,Co)S_2 serving as both the electrodes. 19Optimization of junction termination extension for ultrahigh voltage 4H-SiC planar power devices显示文摘Numerical simulations on the optimization of junction termination extension(JTE) have been performed.Various termination techniques have been applied and simulated in this paper,such as single-zone JTE(S-JTE),multi-zone JTE(M-JTE),and space-modulated JTE(SM-JTE).A completely novel and efficient method is demonstrated in this paper to determine total length of SM-JTE,and it is verified through simulation results.The simulation results show that the SM-JTE could provide a protection efficiency(defined in Section 2) of 95.2%,which is much higher than that of M-JTE(82.4%) and S-JTE(64.7%).Based on the fabricated MOSFETs,the interface charge density is extracted and the approximate range of charge density has been determined.The influences of different interface charge densities have been investigated for the three termination techniques respectively.According to the previous reports,the JTE is quite sensitive to the implanted dose,so the blocking capability of each termination structure with different implanted doses is also simulated.The results show that when interface charge is considered,the SM-JTE always shows an enormous advantage over the other two junction termination structures,however the interface charge densities varied.The space-modulated JTE is also applicable to the power planar devices such as MOSFETs and IGBTs,which would provide a very promising lower fabrication cost.Tongtong Yang Song Bai Runhua Huang 2017Journal of Semiconductors2017,38,4:4
20Sub-micrometer-scale chemical analysis by nanosecond-laser-induced tip-enhanced ablation and ionization time-of-flight mass spectrometry显示文摘The tip-enhanced near-field technique has drawn great attention recently becauseit is a promising technique for nanoscale chemical analysis when combined withother spectroscopic methods. Furthermore, this integration can improve the spatialresolution of mass spectrometry. In this stud~ a nanosecond-laser (nsqaser)-inducedtip-enhanced ablation and ionization source was coupled to an in-house-builtlaser ionization time-of-flight mass spectrometer. Sub-micrometer-sized craters(with diameters of 200-300 nm) were observed on titanium (Ti) film coated ontoa gold (Au) substrate. The corresponding mass spectra were acquired, whichverified that sub-micrometer-sized ablation (and subsequently ionization) couldbe achieved by the ns-laser-induced tip-enhanced electromagnetic field. Inaddition, formation of the crater was studied with an increased number of laserpulses.
