2Correlation and prognostic significance of beta-galactoside alpha-2,6-sialyltransferase and serum monosialylated alpha-fetoprotein in hepatocellular carcinoma显示文摘AIM: To investigate the correlation between tissue ST6Gal I and serum msAFP in HCC patients, and to investigate their prognostic significance.METHODS: Preoperative sera,paired tumorous and non-tumorous tissues were collected from 19 consecutive patients who had undergone surgical resection of HCC.ST6Gal T activities in the tissues were measured by an in vitro microsomal enzyme activity assay. The percentages of tumor-specific msAFP in the sera were also estimated by an isoelectric focusing-immunoblotting assay.RESULTS: The tumor ST6Gal I activity was negatively correlated with serum msAFP percentage (r = -0.53,P = 0.019). Both decreased tumor ST6Gal I activity andincreased serum msAFP percentage were associatedwith poor tumor cell differentiation. Univariate analyses showed that both decreased tumor ST6Gal T activity (P = 0.028), increased serum msAFP percentage (P = 0.034) and poor tumor cell differentiation (P = 0.031)were associated with shorter overall survival. Multivariate analysis using the Cox regression model showed that the preoperative serum msAFP percentage (P = 0.022)and tumor cell differentiation status (P = 0.048) were independent prognostic indicators for patient overall survival.CONCLUSION: Our results indicate that the presence of msAFP in blood circulation is associated with a decreased activity of ST6Gal I activity in HCC. 3Acoustofluidic separation enables early diagnosis of traumatic brain injury based on circulating exosomes显示文摘Traumatic brain injury(TBI)is a global cause of morbidity and mortality.Initial management and risk stratification of patients with TBI is made difficult by the relative insensitivity of screening radiographic studies as well as by the absence of a widely available,noninvasive diagnostic biomarker.In particular,a blood-based biomarker assay could provide a quick and minimally invasive process to stratify risk and guide early management strategies in patients with mild TBI(mTBI).Analysis of circulating exosomes allows the potential for rapid and specific identification of tissue injury.By applying acoustofluidic exosome separation—which uses a combination of microfluidics and acoustics to separate bioparticles based on differences in size and acoustic properties—we successfully isolated exosomes from plasma samples obtained from mice after TBI.Acoustofluidic isolation eliminated interference from other blood components,making it possible to detect exosomal biomarkers for TBI via flow cytometry.Flow cytometry analysis indicated that exosomal biomarkers for TBI increase in the first 24 h following head trauma,indicating the potential of using circulating exosomes for the rapid diagnosis of TBI.Elevated levels of TBI biomarkers were only detected in the samples separated via acoustofluidics;no changes were observed in the analysis of the raw plasma sample.This finding demonstrated the necessity of sample purification prior to exosomal biomarker analysis.Since acoustofluidic exosome separation can easily be integrated with downstream analysis methods,it shows great potential for improving early diagnosis and treatment decisions associated with TBI.Zeyu Wang Haichen Wang Ryan Becker Joseph Rufo Shujie Yang Brian E.Mace Mengxi Wu Jun Zou Daniel T.Laskowitz Tony Jun Huang 2021Microsystems & Nanoengineering2021,7,2:3

5Dilemma of first line regimens in metastatic pancreatic adenocarcinoma显示文摘Pancreatic cancer is one of the deadliest cancers,ranking fourth among cancer-related deaths. Despite all the major molecular advances and treatment breakthroughs, mainly targeted therapies, the cornerstone treatment of metastatic pancreatic cancer(m PC) remains cytotoxic chemotherapy. In 2016, more than 40 years after the introduction of gemcitabine in the management of m PC, the best choice for first-line treatment has not yet been fully elucidated. Two main strategies have been adopted to enhance treatment efficacy. The first strategy is based on combining non-cross resistant drugs, while the second option includes the development of newer generations of chemotherapy. More recently, two new regimens, FOLFIRINOX and gemcitabine/nab-paclitaxel(GNP), have both been shown to improve overall survival in comparison with gemcitabine alone, at the cost of increased toxicity. Therefore, the best choice for first line therapy is a matter of debate. For some authors, FOLFIRINOX should be the first choice in patients with an Eastern Cooperative Oncology Group score(0-1) given its lower hazard ratio. However, others do not share this opinion. In this paper, we review the main comparison points between FOLFIRINOX and GNP. We analyze the two pivotal trials to determine the similarities and differences in study design. In addition, we compare the toxicity profile of the two regimens as well as the impact on quality of life. 6Acoustofluidic separation of cells andparticles显示文摘Acoustofluidics,the integration of acoustics and microfluidics,is a rapidly growing research field that is addressing challenges in biology,medicine,chemistry,engineering,and physics.In particular,acoustofluidic separation of biological targets from complex fluids has proven to be a powerful tool due to the label-free,biocompatible,and contact-free nature of the technology.By carefully designing and tuning the applied acoustic field,cells and other bioparticles can be isolated with high yield,purity,and biocompatibility.Recent advances in acoustofluidics,such as the development of automated,point-of-care devices for isolating sub-micron bioparticles,address many of the limitations of conventional separation tools.More importantly,advances in the research lab are quickly being adopted to solve clinical problems.In this review article,we discuss working principles of acoustofluidic separation,compare different approaches of acoustofluidic separation,and provide a synopsis of how it is being applied in both traditional applications,such as blood component separation,cell washing,and fluorescence activated cell sorting,as well as emerging applications,including circulating tumor cell and exosome isolation.Mengxi Wu Adem Ozcelik Joseph Rufo Zeyu Wang Rui Fang Tony Jun Huang 2019Microsystems & Nanoengineering2019,5,1:2

11Acoustofluidics for simultaneous nanopartide-based drug loading and exosome encapsulation显示文摘Nanocarrier and exosome encapsulation has been found to significantly increase the efficacy of targeted drug delivery while also minimizing unwanted side effects.However,the development of exosome-encapsulated drug nanocarriers is limited by low drug loading efficiencies and/or complex,time-consuming drug loading processes.Herein,we have developed an acoustofluidic device that simultaneously performs both drug loading and exosome encapsulation.By synergistically leveraging the acoustic radiation force,acoustic microstreaming,and shear stresses in a rotating droplet,the concentration,and fusion of exosomes,drugs,and porous silica nanoparticles is achieved.The final product consists of drug-loaded silica nanocarriers that are encased within an exosomal membrane.The drug loading efficiency is significantly improved,with nearly 30%of the free drug(e.g.,doxorubicin)molecules loaded into the nanocarriers.Furthermore,this acoustofluidic drug loading system circumvents the need for complex chemical modification,allowing drug loading and encapsulation to be completed within a matter of minutes.These exosome-encapsulated nanocarriers exhibit excellent efficiency in intracellular transport and are capable of significantly inhibiting tumor cell proliferation.By utilizing physical forces to rapidly generate hybrid nanocarriers,this acoustofluidic drug loading platform wields the potential to significantly impact innovation in both drug delivery research and applications.Zeyu Wang Joseph Rich Nanjing Hao Yuyang Gu Chuyi Chen Shujie Yang Peiran Zhang Tony Jun Huang 2022Microsystems & Nanoengineering2022,8,2:1

14Electrochemical micro-aptasensors for exosome detection based on hybridization chain reaction amplification显示文摘Exosomes are cell-derived nanovesicles that have recently gained popularity as potential biomarkers in liquid biopsies due to the large amounts of molecular cargo they carry,such as nucleic acids and proteins.However,most existing exosome-based analytical sensing methods struggle to achieve high sensitivity and high selectivity simultaneously.In this work,we present an electrochemical micro-aptasensor for the highly sensitive detection of exosomes by integrating a micropatterned electrochemical aptasensor and a hybridization chain reaction(HCR)signal amplification method.Specifically,exosomes are enriched on CD63 aptamer-functionalized electrodes and then recognized by HCR products with avidin-horseradish peroxidase(HRP)attached using EpCAM aptamers as bridges.Subsequently,the current signal that is generated through the enzyme reaction between the HRP enzyme and 3,3,,5/5,-tetramethylbenzidine(TMB)/H_(2)0_(2) directly correlates to the amount of bound HRP on the HCR products and thus to the number of target exosomes.By introducing anti-EpCAM aptamers,micro-aptasensors can detect cancerous exosomes with high specificity.Due to the micropatterned electrodes and HCR dual-amplification strategy,the micro-aptasensors achieve a linear detection response for a wide range of exosome concentrations from 2.5×10^(3) to 1×10^(7) exosomes/mL,with a detection limit of 5×10^(2) exosomes/mL.Moreover,our method successfully detects lung cancer exosomes in serum samples of early-stage and late-stage lung cancer patients,showcasing the great potential for early cancer diagnosis.Wenfen Zhang Zhenhua Tian Shujie Yang Joseph Rich Shuaiguo Zhao Mikael Klingeborn Po-Hsun Huang Zhishang Li Alexander Stout Quinn Murphy Edward Patz Shusheng Zhang Guozhen Liu Tony Jun Huang 2021Microsystems & Nanoengineering2021,7,4:1

17The evolving species concepts used for yeasts:from phenotypes and genomes to speciation networks显示文摘Here we review how evolving species concepts have been applied to understand yeast diversity.Initially,a phenotypic species concept was utilized taking into consideration morphological aspects of colonies and cells,and growth profiles.Later the biological species concept was added,which applied data from mating experiments.Biophysical measurements of DNA similarity between isolates were an early measure that became more broadly applied with the advent of sequencing technology,leading to a sequence-based species concept using comparisons of parts of the ribosomal DNA.At present phylogenetic species concepts that employ sequence data of rDNA and other genes are universally applied in fungal taxonomy,including yeasts,because various studies revealed a relatively good correlation between the biological species concept and sequence divergence.The application of genome information is becoming increasingly common,and we strongly recommend the use of complete,rather than draft genomes to improve our understanding of species and their genome and genetic dynamics.Complete genomes allow in-depth comparisons on the evolvability of genomes and,consequently,of the species to which they belong.Hybridization seems a relatively common phenomenon and has been observed in all major fungal lineages that contain yeasts.Note that hybrids may greatly differ in their post-hybridization development.Future in-depth studies,initially using some model species or complexes may shift the traditional species concept as isolated clusters of genetically compatible isolates to a cohesive speciation network in which such clusters are interconnected by genetic processes,such as hybridization.Teun Boekhout M.Catherine Aime Dominik Begerow Toni Gabaldón Joseph Heitman Martin Kemler Kantarawee Khayhan Marc-AndréLachance Edward J.Louis Sheng Sun Duong Vu Andrey Yurkov 2021Fungal Diversity2021,,4:0

19An acoustofluidic scanning nanoscope using enhanced image stacking and processing显示文摘Nanoscale optical resolution with a large field of view is a critical feature for many research and industry areas,such as semiconductor fabrication,biomedical imaging,and nanoscale material identification.Several scanning microscopes have been developed to resolve the inverse relationship between the resolution and field of view;however,those scanning microscopes still rely upon fluorescence labeling and complex optical systems.To overcome these limitations,we developed a dual-camera acoustofluidic nanoscope with a seamless image merging algorithm(alphablending process).This design allows us to precisely image both the sample and the microspheres simultaneously and accurately track the particle path and location.Therefore,the number of images required to capture the entire field of view(200×200μm)by using our acoustofluidic scanning nanoscope is reduced by 55-fold compared with previous designs.Moreover,the image quality is also greatly improved by applying an alpha-blending imaging technique,which is critical for accurately depicting and identifying nanoscale objects or processes.This dual-camera acoustofluidic nanoscope paves the way for enhanced nanoimaging with high resolution and a large field of view.Geonsoo Jin Joseph Rich Jianping Xia Albert JHe Chenglong Zhao Tony Jun Huang 2022Microsystems & Nanoengineering2022,8,4:0
20Non-typhoidal salmonella:an unusual cause of spontaneous bacterial peritonitis in decompensated cirrhosis显示文摘Salmonella typhimurium, a non-typhoidal salmonella, is an unusual cause of spontaneous bacterial peritonitis (SBP). It isusually reported in asymptomatic patients with normal or high ascitic fluid protein levels with underlying immunosuppression,as high opsonic activity in the ascitic fluid of these patients protects them from the usual organisms causingspontaneous bacterial peritonitis, unless they are exposed to a particularly virulent organism like salmonella.
