| 2 | Combination of ribavirin and reduning protects mice against severe pneumonia induced by H1N1 influenza a virus显示文摘OBJECTIVE:To investigate the effects of ribavirin administration combined with Reduning in a mouse model of influenza A(H1N1)-induced severe pneumonia.METHODS:Influenza A/Beijing/501/2009(H1N1)-infected C57BL/6 mice were randomly divided into four experimental groups treated with either a mock injection of phosphate-buffered saline(PBS),ribavirin(66.6 mg/kg daily)or Reduning(86.6 mg/kg daily),or a combination of both,for 7 days.Mice were monitored for clinical signs and survival,and body weight was measured daily for 14 days.Virus titer,lung wet-to-dry ratios,pathology and cytokines including interleukin(IL)-6,IL-10,and interferon(IFN)-γwere assayed on different days.RESULTS:In the untreated group injected with phosphate buffer saline,all the mice died of the infection.The survival rate of mice treated with Reduning was only 10%,whereas 100%of the ribavirin-and the combination-treated mice survived.Low lung viral loads indicated that ribavirin significantly inhibited virus replication,whereas Reduning did not.Lung wet-to-dry ratios demonstrated that both ribavirin and Reduning,administered together or separately,reduced acute lung edema compared with results in the untreated group.Pathology analyses also showed that treatment with a combination of both drugs relieved pathological lesions,whereas the single drug treatment did not.Levels of IL-6,IL-10 and IFN-γin mice treated with ribavirin or the combination of both ribavirin and Reduning were all significantly lower than in the untreated group,especially in the combination-treated group.In addition,Reduning administration significantly decreased both IL-6 and IL-10production but had no effect on IFN-γ.CONCLUSION:Due to the synergistic effect of antiviral and antiinflammation,the combination of ribavirin and Reduning could be an effective treatment for severe H1N1 which was considered to be significant to delayed antiviral and drug resistant. | Ma Yuexia Zhang Wei Zhao Zhongpeng Li Min Liu Jian Wang Yuguang | 2016 | Journal of Traditional Chinese Medicine2016,36,2: | 13 |

| 4 | Expression of human cytomegalovirus components in the brain tissues of patients with Rasmussen's encephalitis显示文摘Rasmussen's encephalitis(RE) is a rare and severe progressive epileptic syndrome with unknown etiology. Infection by viruses, including human cytomegalovirus(HCMV), has been speculated to be a potential trigger for RE. However, no viral antigens have been detected in the brains of patients with RE; thus, a possible clinical linkage between viral infections and RE has not been firmly established. In this study, we evaluated the expression of HCMV pp65 antigen in brain sections from 26 patients with RE and 20 non-RE patients by immunohistochemistry and in situ hybridization, and assessed the associations between HCMV infection and clinical parameters.Elevated expression of HCMV pp65 protein and DNA was observed in 88.5%(23/26) and 69.2%(18/26) of RE cases, respectively. In the non-RE group, HCMV pp65 antigen was detected only in two cases(10%), both of which were negative for DNA staining. Additionally, the intensity of HCMV pp65 staining was correlated with a shorter duration of the prodromal stage, younger age of seizure onset, and more severe unilateral cortical atrophy. Elevated expression of HCMV pp65 was observed in RE brain tissue and was correlated with the clinical features of RE disease. In summary, our results suggested that HCMV infection may be involved in the occurrence and progression of RE disease. | 6 | Efficacy and safety of Xinglouchengqi decoction for acute ischemic stroke with constipation:study protocol for a randomized con-trolled trial显示文摘OBJECTIVE: To investigate the efficacy and safety of Xinglouchengqi(XLCQ) decoction in treatment of acute ischemic stroke with constipation.METHODS: In this prospective, multicenter, assessor-blinded, randomized controlled trial, 360 eligible participants will be randomized to the XLCQ group or the control group. Participants in the XLCQ group will receive XLCQ decoction, while those in the control group will undergo clysis therapy using glycerin enemas or oral administration of lactulose solution. Both groups will undergo a treatment period of(5 ± 2) d and a 1-month follow-up.The primary outcome measure will be the Constipation Scale score. The secondary outcome measures will include scores on the National Institutes of Health Stroke Scale, the Traditional Chinese Medicine(TCM) Stroke Scale, the Diagnostic Scale for TCM Syndromes of Ischemic Stroke and TCM Scale for Syndrome of Phlegm-heat and Fu-organ Excess.Therapeutic mechanism outcomes and safety outcomes will also be assessed. Assessments will be conducted at baseline, at the end of the treatment period, and at the follow-up. | 8 | ~1H nuclear magnetic resonance-based metabolomic study on efficacy of Qingrehuatan decoction against abundant phlegm-heat syndrome in young adults with essential hypertension显示文摘OBJECTIVE: To observe the influence of Qingrehuatan decoction(QRHT) on serum metabolic profile in young essential hypertension(YEH) patients with abundant phlegm-heat syndrome and provide a basis for treatment with the decoction.METHODS: Twelve male YEH patients were randomly selected and serum samples were collected for examination before and after 4 weeks of thetreatment with QRHT. Twelve healthy males were randomly selected and their serum samples were collected as a control. All serum samples were detected using metabolomic technology with1 H nuclear magnetic resonance. Differences in metabolites were studied by principal component analysis and partial least squares-discriminate analysis,which produced scores and loadings plots.RESULTS: After 4 weeks of treatment, serum substances could be distinguished between the YEH patients with abundant phlegm-heat syndrome and the control patients. The specific serum endogenous metabolites tended to improve after the treatment. | 9 | Microbial community structure and diversity in deep-sea hydrothermal vent sediments along the Eastern Lau Spreading Centre显示文摘The aim of this study is to investigate microbial structures and diversities in five active hydrothermal fields' sediments along the Eastern Lau Spreading Centre (ELSC) in the Lau Basin (southwest Pacific). Microbial communities were surveyed by denatured gradient gel electrophoresis (DGGE) and clone library analysis of 16S rRNA genes. The differences in microbial community structures among sediment samples from the five deep-sea hydrothermal sites were revealed by DGGE profiles. Cluster analysis of DGGE profiles separated the five hydrothermal samples into two groups. Four different 16S rRNA gene clone libraries, representing two selected hydrothermal samples (19-4TVG8 and 19-4TVG11), were constructed. Twenty-three and 32 phylotypes were identified from 166 and 160 bacterial clones respectively, including Proteobacteria, Bacteroidetes, Firmicutes, Nitrospirae and Planctomycetes. The phylum Proteobacteria is dominant in both bacterial libraries with a predominance of Gamma-Proteobacteria. A total of 31 and 25 phylotypes were obtained from 160 and 130 archaeal clones respectively, including Miscellaneous Crenarchaeotic Group, Marine Group Ⅰ and Ⅲ, Marine Benthic Group E, Terrestrial Hot Spring Crenarchaeota and Deep-sea Hydrothermal Vent Euryarchaeota. Vertical dwelling burrows of opportunists dominate in active channel, whereas horizontal deposit-feeding, grazing, and vertical dwelling burrows occur in overbank deposits. Seven recurrent ichnofabrics were distingushed. The Palaeophycus tubularis ichnofabric preserves below scour interface between two fluvial cycles. The Skolithos linearis ichnofabric forms in middle to high-energy, soft substrates in the middle and upper parts of active channel deposits under semi-arid climate. The Skolithos linearis-Planolites montanus ichnofabric occurs in the sediments just a little above the Skolithos linearis ichnofabric and is formed in middle-energy, soft substrates in the upper part of active channel deposits. The Planolites montanus ichnofabric is mostly preserved in association with medium- to thick-bedded brown-red silty mudstones or siltstones in low-energy, perennial muddy bottom ponds in overbank settings. The Taenidium barretti ichnofabric is interpreted as periodically exposed ephemeral ponds or low-energy distal floodplain. The Skolithos isp. ichnofabric commonly occurs in middle to high-energy firm ground of proximal foodplain or crevasse-splay deposits. The root trace ichnofabric occurs in the crevasse splay deposits. | 14 | Imaging energetic electron spectrometer onboard a Chinese navigation satellite in the inclined GEO orbit显示文摘The energetic electron measurement is one of the most important issues to understand dynamics in space physics and the applications for space weather. In this study, the principle and functional components of the imaging energetic electron spectrometer(IES) onboard a Chinese navigation satellite in the inclined GEO orbit(IGSO) was introduced. The IES instrument is developed by the team in Peking University(BeiDa), thus it is named as BD-IES. Based on the pin-hole technique, the instrument can measure 50–600 keV electrons incident from 9 directions over a range of 180° in polar angle. With pulse height analysis(PHA), the spectrum can be determined for each direction. The energy and angular calibrations were performed, which show the good energy and angular characteristics of BD-IES. Monte Carlo simulations show that the anti-proton design of BDIES can effectively decrease the proton contamination on the electron measurements in the inclined GEO orbit. | 15 | Simplified mathematical model of inter-turn short circuit of field windings in hydro-generators and its application显示文摘Inter-turn short circuit of field windings is a common electrical fault of generators.Simulation is an important method of investigating the fault and providing data support for fault monitoring.However,huge numbers of pole pairs and damper loops in large hydro-generators would lead to lengthy calculation time,hindering scientific research and engineering application.To deal with this problem,we analyze a theoretical basis for a damper winding simplified model and then propose an equivalent treatment method.Through the analysis of steady-state current harmonic characteristics of generators with different stator winding configurations during the fault,the simplified models suitable for steady-state calculation are derived from two aspects,namely,additional rotor harmonic current frequency characteristics and the relationship of the amplitude as well as the phase of each branch current of the stator.The calculation and experimental results of the two simplified models are then compared to verify the models' correctness.A calculation example of the Three Gorges left bank VGS generator shows few deviations between the calculation results of the simplified model and the original model.Moreover,the calculation time using the simplified model is 1/1500 that using the original model,which provides a more effective tool for on-line fault monitoring.Finally,the sensitivity-verification application of the fault-monitoring scheme based on the stator steady-state unbalanced current RMS is depicted.The result shows that the scheme can monitor two-turn short circuits of field windings in the Three Gorges generator and provide high sensitivity. | HAO LiangLiang WU JunYong SUN YuGuang WANG XiangHeng | 2013 | Science China(Technological Sciences)2013,56,4: | 5 |

| 17 | The diversity of denitrifying bacteria in the alpine meadow soil of Sanjiangyuan natural reserve in Tibet Plateau显示文摘This is the first time to describe the diversity of denitrifying bacteria in Sanjiangyuan natural reserve in Tibet Plateau by investigating the molecular diversity and phylogenetic analysis of nirK and nosZ genes using PCR-RFLP and sequencing. Four soil samples were collected from alpine meadow communities from over an altitude of 4600 m which had different physicochemical properties by principal component analysis (PCA). For the genes fragment of nirK and nosZ, the diverse PCR products were characterized by cloning, restriction fragment length polymorphism (RFLP) analysis and sequenced. A total of 253 nirK clones and 283 nosZ clones were received in four samples, and 78 operational taxonomic units (OTUs) of nirK and 120 OTUs of nosZ by the restriction enzymes MspI and RsaI digested. The analysis of environmental factors showed that altitude and C/N ratio in soil may be the key factors to the denitrifying bacteria community. 36 nirK clones and 17 nosZ clones were sequenced, and their levels of nucleotide identity were from 69% to 98% and 57% to 97%, respectively. The phylogenetic tree was constructed using the Clustal W and Mega softwares, and all the sequenced clones could be subdivided into 4 groups. Some of clone sequences were related to the nirK and nosZ genes belonging to three phylogenetic subdivisions (α-, β- and γ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. | 19 | Detection of EBV and HHV6 in the Brain Tissue of Patients with Rasmussen's Encephalitis显示文摘Rasmussen's encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis,intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8?T cells. | 20 | Monte Carlo simulations of the sensor head of imaging energetic electron spectrometer onboard a Chinese IGSO navigation satellite显示文摘An imaging energetic electron spectrometer built by the Peking University team(BD-IES) onboard a Chinese navigation satellite in an inclined GEO orbit has been launched successfully in September 2015, which measures the spectra of the energetic electrons with the energy range of 50–600 keV in nine directions. In this study, Monte Carlo simulations of the BD-IES sensor head were performed using Geant4 and the corresponding characteristic responses to the isotropic energetic particles were derived. The effective geometric factors were estimated using the typical electron and proton spectra in the GEO orbit and the corresponding simulated sensor head responses. It was found that the average effective geometric factors of nine directions are close to the nominal geometric factors calculated with the traditional method, but the effective geometric factor decreases as the center energy of the energy channel decreases. The BD-IES sensor head also responses to the energetic protons, but the average contamination rate of all 72 channels is about 2%, which means that the proton contamination is acceptable.
