| 3 | Aspirin alleviates endothelial gap junction dysfunction through inhibition of NLRP3inflammasome activation in LPS-induced vascular injury显示文摘The loss of endothelial connective integrity and endothelial barrier dysfunction can lead to increased vascular injury, which is related to the activation of endothelial inflammasomes. There are evidences that low concentrations of aspirin can effectively prevent cardiovascular diseases. We hypothesized that low-dose aspirin could ameliorate endothelial injury by inhibiting the activation of NLRP3 inflammasomes and ultimately prevent cardiovascular diseases. Microvascular endothelial cells were stimulated by lipopolysaccharide(2 μg/mL) and administrated by 0.1–2 mmol/L aspirin. The wild type mice were stimulated with LPS(100 μg/kg/day), and 1 h later treated with aspirin(12.5, 62.5, or125 mg/kg/day) and dexamethasone(0.0182 mg/kg/day) for 7 days. Plasma and heart were harvested for measurement of ELISA and immunofluorescence analyses. We found that aspirin could inhibit NLRP3 inflammasome formation and activation in vitro in dose-dependent manner and has correlation between the NLRP3 inflammasome and the ROS/TXNIP pathway. We also found that low-concentration aspirin could inhibit the formation and activation of NLRP3 inflammasome and restore the expression of theendothelial tight junction protein zonula occludens-1/2(ZO1/2). | 7 | Impact of pre-oxidation on the formation of byproducts in algae-laden water disinfection:Insights from fluorescent and molecular weight显示文摘Pre-oxidation has been reported to be an effective way to remove algal cells in water, but the released algal organic matter (AOM) could be oxidized and lead to the increment in disinfection by-product (DBP) formation. The relationship between pre-oxidation and AOM-derived DBP formation needs to be approached more precisely. This study compared the impact of four pre-oxidants, ozone (O), chlorine dioxide (ClO), potassium permanganate(KMnO) and sodium hypochlorite (NaClO), on the formation of nitrogenous (N-) and carbonaceous (C-) DBPs in AOM chlorination. The characterization (fluorescent properties,molecular weight distribution and amino acids concentration) on AOM samples showed that the characterization properties variations after pre-oxidation were highly dependent on the oxidizing ability of oxidants. The disinfection experiments showed that Oincreased DBP formation most significantly, which was consistent with the result of characterization properties variations. Then canonical correspondent analysis (CCA) and Pearson’s correlation analysis were conducted based on the characterization data and DBP formation. CCA indicated that C-DBPs formation was highly dependent on fluorescent data. The formation of haloacetic acids (HAAs) had a positive correlation with aromatic protein-like component while trichloromethane (TCM) had a positive correlation with fulvic acid-like component.Pearson’s correlation analysis showed that low molecular weight fractions were favorable to form N-DBPs. Nitrate(NO-3) and natural organic matter(NOM) are widely present in water source.We investigated trichloronitromethane(TCNM) formation during chlorination after VUV photolysis, because TCNM is an unregulated highly toxic disinfection byproduct. In this study:(1) we found reactive nitrogen species that is generated under VUV photolysis of NO-3react with organic matter to form nitrogen-containing compounds and subsequently form TCNM during chlorination;(2) we found the mere presence of 0.1 mmol/L NO-3can result in the formation of up to 63.96 μg/L TCNM;(3) we found the changes in pH(6.0-8.0), chloride(1-4 mmol/L), and bicarbonate(1-4 mmol/L) cannot effectively diminish TCNM formation;and,(4) we established the quantitative structure-activity relationship(QSAR) model,which indicated a linear relationship between TCNM formation and the Hammett constant(σ) of model compounds;and,(5) we characterized TCNM precursors in water matrix after VUV photolysis and found 1161 much more nitrogen-containing compounds with higher aromaticity were generated. | 19 | Establishment and Evaluation of a Prediction Model of BLR for Severity in Coronavirus Disease 2019显示文摘Background:Coronavirus disease 2019(COVID-19)is an emerging infectious disease and has spread worldwide.Clinical risk factors associated with the severity in COVID-19 patients have not yet been well delineated.The aim of this study was to explore the risk factors related with the progression of severe COVID-19 and establish a prediction model for severity in COVID-19 patients.Methods:We retrospectively recruited patients with confirmed COVID-19 admitted in Enze Hospital,Taizhou Enze Medical Center(Group)and Nanjing Drum Tower Hospital between January 24 and March 12,2020.Take the Taizhou cohort as the training set and the Nanjing cohort as the validation set.Severe case was defined based on the World Health Organization Interim Guidance Report criteria for severe pneumonia.The patients were divided into severe and non-severe groups.Epidemiological,laboratory,clinical,and imaging data were recorded with data collection forms from the electronic medical record.The predictive model of severe COVID-19 was constructed,and the efficacy of the predictive model in predicting the risk of severe COVID-19 was analyzed by the receiver operating characteristic curve(ROC).Results:A total of 402 COVID-19 patients were included in the study,including 98 patients in the training set(Nanjing cohort)and 304 patients in the validation set(Nanjing cohort).There were 54 cases(13.43%)in severe group and 348 cases(86.57%)in nonsevere group.Logistic regression analysis showed that bodymassindex(BMI)and lymphocyte count wereindependent risk factors for severe COVID-19(all P<0.05).Logistic regression equation based on risk factors was established as follows:Logit(BL)=–5.552–5.473L+0.418BMI.The area under the ROC curve(AUC)of the training set and the validation set were 0.928 and 0.848,respectively(allP<0.001).The model was simplified to get a new model(BMI and lymphocyte count ratio,BLR)for predicting severe COVID-19 patients,and the AUC in the training set and validation set were 0.926 and 0.828,respectively(all P<0.001).Conclusions:Higher BMI and lower lymphocyte count are critical factors associated with severity of COVID-19 patients.The simplified BLR model has a good predictive value for the severe COVID-19 patients.Metabolic factors involved in the development of COVID-19 need to be further investigated. | Zebao He Fajuan Rui Hongli Yang Zhengming Ge Rui Huang Lingjun Ying Haihong Zhao Chao Wu Jie Li | 2022 | Infectious Diseases & Immunity2022,2,2: | 0 |
| 20 | Machine learning accelerated carbon neutrality research using big data-from predictive models to interatomic potentials显示文摘Carbon neutrality has been proposed as a solution for the current severe energy and climate crisis caused by the overuse of fossil fuels, and machine learning(ML) has exhibited excellent performance in accelerating related research owing to its powerful capacity for big data processing. This review presents a detailed overview of ML accelerated carbon neutrality research with a focus on energy management, screening of novel energy materials, and ML interatomic potentials(MLIPs), with illustrations of two selected MLIP algorithms: moment tensor potential(MTP) and neural equivariant interatomic potential(NequIP).
