1Transgenic rhesus monkeys carrying the human MCPH1 gene copies show human-like neoteny of brain development显示文摘Brain size and cognitive skills are the most dramatically changed traits in humans during evolution and yet the genetic mechanisms underlying these human-specific changes remain elusive.Here,we successfully generated 11 transgenic rhesus monkeys(8 first-generation and 3 second-generation)carrying human copies of MCPH1,an important gene for brain development and brain evolution.Brain-image and tissue-section analyses indicated an altered pattern of neural-cell differentiation,resulting in a delayed neuronal maturation and neural-fiber myelination of the transgenic monkeys,similar to the known evolutionary change of developmental delay(neoteny)in humans.Further brain-transcriptome and tissue-section analyses of major developmental stages showed a marked human-like expression delay of neuron differentiation and synaptic-signaling genes,providing a molecular explanation for the observed brain-developmental delay of the transgenic monkeys.More importantly,the transgenic monkeys exhibited better short-term memory and shorter reaction time compared with the wild-type controls in the delayed-matching-to-sample task.The presented data represent the first attempt to experimentally interrogate the genetic basis of human brain origin using a transgenic monkey model and it values the use of non-human primates in understanding unique human traits.Lei Shi Xin Luo Jin Jiang Yongchang chen Cirong Liu Ting Hu Min Li Qiang Lin Yanjiao Li Jun Huang Hong Wang Yuyu Niu Yundi Shi Martin Styner Jianhong Wang Yi Lu Xuejin Sun Hualin Yu Weizhi Ji Bing Su 2019National Science Review2019,6,3:12
2Influences of working pressure on properties for TiO_2 films deposited by DC pulse magnetron sputtering显示文摘TiO2 films were deposited at room temperature by DC pulse magnetron sputtering system.The crystalline structures,morphological features and photocatalytic activity of TiO2 films were systematically investigated by X-ray diffraction(XRD),atomic force microscopy(AFM) and ultraviolet spectrophotometer,respectively.The results indicated that working pressure was the key deposition parameter in?uencing the TiO2 film phase composition at room temperature,which directly affected its photocatalytic activity.With increasing working pressure,the target self-bias decreases monotonously.Therefore,low temperature TiO2 phase(anatase) could be deposited with high working pressure.The anatase TiO2 films deposited with 1.4 Pa working pressure displayed the highest photocatalytic activity by the decomposition of Methyl Orange solution,which the degradation rate reached the maximum(35%) after irradiation by ultraviolet light for 1 h.ZHANG Can DING Wanyu WANG Hualin CHAI Weiping JU Dongying 2009Journal of Environmental Sciences2009,21,6:11

4Spatiotemporal differences and convergence of urban industrial land use efficiency for China's major economic zones显示文摘Based on an SBM model, this paper first analyzes the spatial differences of urban industrial land use efficiency(UILUE) in the six main economic zones of China. Then, we analyze the dynamic changes in the urban industrial land's total factor productivity(UILTFP) by using the Malmquist productivity index approach, and we examine the UILTFP's convergence. The results show that the Pearl River Delta and the Yangtze River Delta have a higher UILUEs but show downward trends in UILTFPs. The other four zones, Beijing-Tianjin-Hebei, Chengdu-Chongqing, Guanzhong-Tianshui and the Central Plains, show poorer UILUEs but have upward trends in UILTFPs. A significant excess supply of industrial land and labor exists in all six zones, and the zones in economically developed areas perform better in industrial output. The results of the convergence test do not support σ-convergence but support conditional β-convergence for all zones. These results indicate that the gaps in the UILTFP of industrial land in all six zones, in fact, do not narrow, and the UILTFPs of cities will converge to their own steady states. The regression results of the influencing factors show that the urbanization rate should be increased in the Pearl River Delta and reduced in Beijing-Tianjin-Hebei. The Yangtze River Delta and Chengdu-Chongqing should pay more attention to the problem of industrial labor surplus, and the Pearl River Delta should reduce the proportion of industrial output. 7A validated UPLC–MS/MS method for simultaneous determination of imatinib, dasatinib and nilotinib in human plasma显示文摘sensitive, rapid, simple and economical ultra-performance liquid chromatography–tandem mass spectrometric method(UPLC–MS/MS) was developed and validated for simultaneous determination of imatinib,dasatinib and nilotinib in human plasma using gliquidone as internal standard(IS). Liquid-liquid extraction method with ethyl acetate was used for sample pre-treatment. The separation was performed on an Xtimate Phenyl column using isocratic mobile phase consisting of A(aqueous phase: 0.15% formic acid and 0.05% ammonium acetate) and B(organic phase: acetonitrile)(A:B=40:60, v/v). The flow rate was 0.25 mL/min and the total run time was 6 min. The multiple reaction monitoring(MRM) transitions, m/z 494.5→394.5 for imatinib, 488.7→401.5 for dasatinib, 530.7→289.5 for nilotinib and 528.5→403.4 for IS, were chosen to achieve high selectivity in the simultaneous analyses. The method exhibited great improvement in sensitivity and good linearity over the concentration range of 2.6–5250.0 ng/mL for imatinib, 2.0–490.0 ng/mL for dasatinib, and 2.4–4700.0 ng/mL for nilotinib. The method showed acceptable results on sensitivity, specificity,recovery, precision, accuracy and stability tests. This UPLC–MS/MS assay was successfully used for human plasma samples analysis and no significant differences were found in imatinib steady-state trough concentrations among the SLC22A5-1889T > C or SLCO1B3 699G > A genotypes(P > 0.05). 8Exploration and practice of general practitioner responsibility system in an urban community of Shanghai显示文摘Objective:The Minhang district of Shanghai was selected as the subject and its public health management model was investigated.Methods:Minhang became a pilot location for the implementation of the general practitioner responsibility system(GPRS)in 2011.Since then,the management model of public health services in the district has gradually merged with the GPRS through a series of actions:the adjustment of organizations and the coordination mechanism in community public health services,the integration of basic public health services and clinic practice,the establishment of an informationized Internet service platform,comprehensive budgeting management of public health service programs,and performance assessment management of health workers.Results:By the end of 2014,the number of residents who had a contractual relationship with a general practitioner(GP)in Minhang had reached 1.4367 million and all of them had family health records.Compared with 2010,both the rate of maternal management and the rate of children management stayed at high level(≥96%),the rate of hypertension management increased(40.73%vs.28.70%),the rate of blood pressure control increased(82.13%vs.77.51%),the incidence of diabetes decreased(21.93%vs.37.85%),the rate of effective control of blood glucose for dia-betic patients increased(82.43%vs.62.77%),and the rate of child health management increased(99.79%vs.91.54%).Conclusion:The GPRS has helped to create a better community health environment.How-ever,the development of this system is limited by several factors,including the following:the shortage of GPs,most GPs do not have good public health service skills and a correct public health concept,unclear division of different services,and imperfect department coordination.Yan Wu Yanping Zhao Xiaoxia Huang Junyan Wang Huilin Xu Hualin Su 2015Family Medicine and Community Health2015,3,4:5
9Zika Virus Baculovirus-Expressed Virus-Like Particles Induce Neutralizing Antibodies in Mice显示文摘The newly emerged mosquito-borne Zika virus(ZIKV) strains pose a global challenge owing to its ability to cause microcephaly and neurological disorders. Several ZIKV vaccine candidates have been proposed, including inactivated and live attenuated virus vaccines, vector-based vaccines, DNA and RNA vaccines. These have been shown to be efficacious in preclinical studies in mice and nonhuman primates, but their use will potentially be a threat to immunocompromised individuals and pregnant women. Virus-like particles(VLPs) are empty particles composed merely of viral proteins, which can serve as a safe and valuable tool for clinical prevention and treatment strategies. In this study, we used a new strategy to produce ZIKV VLPs based on the baculovirus expression system and demonstrated the feasibility of their use as a vaccine candidate. The pre-membrane(prM) and envelope(E) proteins were co-expressed in insect cells and selfassembled into particles similar to ZIKV. We found that the ZIKV VLPs could be quickly and easily prepared in large quantities using this system. The VLPs were shown to have good immunogenicity in immunized mice, as they stimulated high levels of virus neutralizing antibody titers, ZIKV-specific IgG titers and potent memory T cell responses. 10Establishment of a Reverse Genetic System of Severe Fever with Thrombocytopenia Syndrome Virus Based on a C4 Strain显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV)is an emerging tick-borne bunyavirus that causes hemorrhagic fever-like disease(SFTS)in humans with a case fatality rate up to 30%.To date,the molecular biology involved in SFTSV infection remains obscure.There are seven major genotypes of SFTSV(C1-C4 and J1-J3)and previously a reverse genetic system was established on a C3 strain of SFTSV.Here,we reported successfully establishment of a reverse genetics system based on a SFTSV C4 strain.First,we obtained the 5’-and 3’-terminal untranslated region(UTR)sequences of the Large(L),Medium(M)and Small(S)segments of a laboratory-adapted SFTSV C4 strain through rapid amplification of cDNA ends analysis,and developed functional T7 polymerase-based L-,M-and S-segment minigenome assays.Then,fulllength cDNA clones were constructed and infectious SFTSV were recovered from co-transfected cells.Viral infectivity,growth kinetics,and viral protein expression profile of the rescued virus were compared with the laboratory-adapted virus.Focus formation assay showed that the size and morphology of the foci formed by the rescued SFTSV were indistinguishable with the laboratory-adapted virus.However,one-step growth curve and nucleoprotein expression analyses revealed the rescued virus replicated less efficiently than the laboratory-adapted virus.Sequence analysis indicated that the difference may be due to the mutations in the laboratory-adapted strain which are more prone to cell culture.The results help us to understand the molecular biology of SFTSV,and provide a useful tool for developing vaccines and antivirals against SFTS.Mingyue Xu Bo Wang Fei Deng Hualin Wang Manli Wang Zhihong Hu Jia Liu 2021Virologica Sinica2021,36,5:5

14Phylogenetic analysis revealed the central roles of two African countries in the evolution and worldwide spread of Zika virus显示文摘Recent outbreaks of Zika virus(ZIKV) infections in Oceania's islands and the Americas were characterized by high numbers of cases and the spread of the virus to new areas. To better understand the origin of ZIKV, its epidemic history was reviewed. Although the available records and information are limited, two major genetic lineages of ZIKV were identified in previous studies.However, in this study, three lineages were identified based on a phylogenetic analysis of all virus sequences from Gen Bank, including those of the envelope protein(E) and non-structural protein 5(NS5) coding regions. The spatial and temporal distributions of the three identified ZIKV lineages and the recombination events and mechanisms underlying their divergence and evolution were further elaborated. The potential migration pathway of ZIKV was also characterized. Our findings revealed the central roles of two African countries, Senegal and Cote d'Ivoire, in ZIKV evolution and genotypic divergence. Furthermore, our results suggested that the outbreaks in Asia and the Pacific islands originated from Africa. 16Electroacupuncture promotes the proliferation of endogenous neural stem cells and oligodendrocytes in the injured spinal cord of adult rats显示文摘A contusive model of spinal cord injury at spinal segment T8-9 was established in rats. Huantiao (GB30) and Huatuojiaji (Ex-B05) were punctured with needles, and endogenous neural stem cells were labeled with 5-bromo-2'-deoxyuridine (BrdU) and NG2. Double immunofluorescence staining showed that electroacupuncture markedly increased the numbers of BrdU+/NG2+ cells at spinal cord tissue 15 mm away from the injury center in the rostral and caudal directions. 19Critical impeller speeds for a gas-inducing stirring tank loaded with solid particles显示文摘The influence of solid particles size,density and loading on the critical gas-inducing impeller speed was investigated in a gas–liquid–solid stirring tank equipped with a hollow Rushton impeller.Three types of solid particles,hollow glass beads with diameters of 300 μm,200 μm,100 μm,and 60 μm,silica gel and desalting resin,were used.It was found that the adding solid particles would change the critical impeller speed.For hollow glass beads and silica gel,whose relative densities were less than or equal to 1.5,the critical impeller speeds increased with the solid loading before reaching the maximum values,and then decreased to a value even lower than that without added solids.The size of the solids also had apparent influence on the critical impeller speed,and larger solid particles correspond to a smaller critical impeller speed.The experimental data also showed that the gasinducing was beneficial to the suspension of the solid particles.Yanhong Zhang Zhengwei Zhang Chenwen Wei Hualin Wang 2018Chinese Journal of Chemical Engineering2018,26,6:3
20Improving Baculovirus Transduction of Mammalian Cells by Incorporation of Thogotovirus Glycoproteins显示文摘Baculovirus can transduce a wide range of mammalian cells and is considered a promising gene therapy vector. However,the low transduction efficiency of baculovirus into many mammalian cells limits its practical application. Co-expressing heterologous viral glycoproteins(GPs), such as vesicular stomatitis virus G protein(VSV G), with baculovirus native envelope protein GP64 is one of the feasible strategies for improving virus transduction. Tick-borne thogotoviruses infect mammals and their GPs share sequence/structure homology and common evolutionary origins with baculovirus GP64.Herein, we tested whether thogotovirus GPs could facilitate the entry of the prototype baculovirus Autographa californica multiple multiple nucleopolyhedrovirus(AcMNPV) into mammalian cells. The gp genes of two thogotoviruses, Thogoto virus and Dhori virus, were inserted into the AcMNPV genome. Both GPs were properly expressed and incorporated into the envelope of the recombinant AcMNPVs. The transduction rates of recombinant AcMNPVs expressing the two thogotovirus GPs increased for approximately 4–12 fold compared to the wild type AcMNPV in six of the 12 tested mammalian cell lines. It seemed that thogotovirus GPs provide the recombinant AcMNPVs with different cell tropisms and showed better performance in several mammalian cells compared to VSV G incorporated AcMNPV. Further studies showed that the improved transduction was a result of augmented virus-endosome fusion and endosome escaping, rather than increased cell binding or internalization. We found the AcMNPV envelope protein GP64-mediated fusion was enhanced by the thogotovirus GPs at relatively higher p H conditions.
