| 1 | Transgenic rhesus monkeys carrying the human MCPH1 gene copies show human-like neoteny of brain development显示文摘Brain size and cognitive skills are the most dramatically changed traits in humans during evolution and yet the genetic mechanisms underlying these human-specific changes remain elusive.Here,we successfully generated 11 transgenic rhesus monkeys(8 first-generation and 3 second-generation)carrying human copies of MCPH1,an important gene for brain development and brain evolution.Brain-image and tissue-section analyses indicated an altered pattern of neural-cell differentiation,resulting in a delayed neuronal maturation and neural-fiber myelination of the transgenic monkeys,similar to the known evolutionary change of developmental delay(neoteny)in humans.Further brain-transcriptome and tissue-section analyses of major developmental stages showed a marked human-like expression delay of neuron differentiation and synaptic-signaling genes,providing a molecular explanation for the observed brain-developmental delay of the transgenic monkeys.More importantly,the transgenic monkeys exhibited better short-term memory and shorter reaction time compared with the wild-type controls in the delayed-matching-to-sample task.The presented data represent the first attempt to experimentally interrogate the genetic basis of human brain origin using a transgenic monkey model and it values the use of non-human primates in understanding unique human traits. | Lei Shi Xin Luo Jin Jiang Yongchang chen Cirong Liu Ting Hu Min Li Qiang Lin Yanjiao Li Jun Huang Hong Wang Yuyu Niu Yundi Shi Martin Styner Jianhong Wang Yi Lu Xuejin Sun Hualin Yu Weizhi Ji Bing Su | 2019 | National Science Review2019,6,3: | 12 |
| 2 | Effects of Dietary Stachyose on Growth Performance, Digestive Enzyme Activities and Intestinal Morphology of Juvenile Turbot(Scophthalmus maximus L)显示文摘A 12-week feeding trial was conducted to evaluate the effects of dietary stachyose on the growth performance, digestive enzymes activities and intestinal structures of juvenile turbot(Scophthalmus maximus L). Five isonitrogenous(49.58% crude protein) and isolipidic(10.50% crude lipid) diets were formulated to contain 0(Control), 0.625%(S-0.625), 1.25%(S-1.25), 2.5%(S-2.5) and 5%(S-5) stachyose, respectively. With the increase of stachyose level, the growth performance and feed utilization of turbot, such as the specific growth rate, final mean body weight, weight gain rate and feed efficiency, increased significantly(P < 0.05) and then stabilized. The feed intake of fish fed S-5 was significantly higher(P < 0.05) than that of fish in other groups. The activities of trypsin, intestinal caseinolytic, stomach and intestinal amylase were significantly influenced by stachyose(P < 0.05). The highest values of trypsin and intestinal caseinolytic activities were observed in group S-1.25, while the highest activity of stomach amylase and the lowest activity of intestine amylase were observed in group S-5. No lesion or damage was found on the distal intestine structures of fish from all treatments, while the height of simple folds in the distal intestine was significantly increased(P< 0.05) when 1.25% or 2.5% stachyose was added in the diets. | 3 | Efficacy and safety of Reduqing granules in the treatment of common cold with wind-heat syndrome: a randomized, double-blind, double-dummy, positive-controlled trial显示文摘OBJECTIVE:To assess the efficacy and safety of Reduqing granules in patients with common cold with wind-heat syndrome(CCWHS).METHODS:A randomized, double-blind, double-dummy, parallel, positive-controlled trial included 72 CCWHS patients was performed.The participants were randomly assigned to two groups,Reduqing(RDQ) group and Lianhuaqingwen(LHQW) group, in a 1∶1 ratio.Patients in RDQ group received Reduqing granules and dummy Lianhuaqingwen capsules three times a day and patients in LHQW group received Lianhuaqingwen capsules and dummy Reduqing granules three times daily.The duration of treatment and follow-up were four days.RESULTS:There were no statistically significant differences in total markedly effective rate and total effective rate between RDQ group and LHQW group after treatment.Traditional Chinese Medicine(TCM) symptom score was significantly reduced after treatment in RDQ group, as well as in LHQW group.However, the difference of change in TCM symptom score between two groups was not statistically significant(P > 0.05).There were no significant differences between two groups in the median time to fever relief [RDQ group(4 ± 8) h vs LHQW group(4 ± 5) h] or the median time to fever clearance(RDQ group 47 h vs LHQW 36 h).No serious adverse events were reported during the study.CONCLUSION:Compared with Lianhuaqingwen capsules, Reduqing granules achieved similar therapeutic effect in the treatment of CCWHS and no drug-related adverse events were reported during the study.Therefore, Reduqing granules might be effective and safe in the treatment of CCWHS. | Ma Yanjiao Zhang Zhongyi Wei Lianbo He Shuai Deng Xun Ji Aimin Zhou Benjie Jiang Deqi Li Mingxing Wang Yong | 2017 | Journal of Traditional Chinese Medicine2017,37,2: | 5 |
| 4 | Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. | 6 | Facile fabrication of amphoteric semi-interpenetrating network membranes for vanadium flow battery applications显示文摘For improvement of vanadium redox flow battery(VRB) performance, novel amphoteric semiinterpenetrating membranes(ASIPN) were prepared using poly(ether ether ketone)(PEEK) and polysulfone(PSf), the former bearing sulfonic groups and the latter imidazolium. These two groups form ionic crosslinks between PEEK and PSf; meanwhile, covalent cross links were built between PSf chains with addition of N-(3-aminopropyl)-imidazole. The amphoteric nature of the membrane allows facile proton and anion transport; the IPN structure and the presence of imidazolium cation effectively suppress vanadium ion crossover through the membrane. Therefore, the ASIPN based VRBs show higher Coulombic efficiency and energy efficiency than that assembled with pristine SPEEK and Nafion 212 membranes. | 11 | Effect of Dietary Lipid on the Growth, Fatty Acid Composition and Δ5 Fads Expression of Abalone(Haliotis discus hannai Ino) Hepatopancreas显示文摘This study investigated the effect of dietary lipid on the growth, fatty acid composition and Δ5 fatty acyl desaturase genes(Fads) expression of juvenile abalone(Haliotis discus hannai Ino) hepatopancreas. Six purified diets were formulated to contain tripalmitin(TP), olive oil(OO, 72.87% 18:1n-9), grape seed oil(GO, 68.67% 18:2n-6), linseed oil(LO, 70.48% 18:3n-3), ARA oil(AO, 41.81% ARA) or EPA oil(EO, 44.09% EPA and 23.67% DAH). No significant difference in survival rate was observed among abalone fed with different diets. Weight gain rate(WGR) and daily growth rate of shell length(DGRSL) were significantly increased in abalone fed with diets containing OO, AO and EO, but decreased in abalone fed with LO diet(P < 0.05) in comparison with those fed with TP. High level of dietary 18:2n-6 resulted in higher content of n-6 polyunsaturated fatty acids(PUFAs) in abalone fed with GO than those fed with TP, OO, LO and EO(P < 0.05). n-3 PUFAs in abalone fed with LO was significantly higher than those in abalone fed with TP, OO, GO and AO(P < 0.05). The highest contents of 20:1n-9 and 22:1n-9 were observed in abalone fed with OO. The expression of Δ5 Fads in hepatopancreas of abalone was enhanced by high concentration of 18:3n-3 and suppressed by dietary LC-PUFAs; however it was not affected by dietary high concentration of 18:1n-9 or 18:2n-6.
