| 1 | Clinical study using mesenchymal stem cells for the treatment of patients with severe COVID-19显示文摘The coronavirus disease 2019(COVID-19)caused by SARS-CoV-2 was identified in December 2019.The symptoms include fever,cough,dyspnea,early symptom of sputum,and acute respiratory distress syndrome(ARDS).Mesenchymal stem cell(MSC)therapy is the immediate treatment used for patients with severe cases of COVID-19.Herein,we describe two confirmed cases of COVID-19 in Wuhan to explore the role of MSC in the treatment of COVID-19.MSC transplantation increases the immune indicators(including CD4 and lymphocytes)and decreases the inflammation indicators(interleukin-6 and C-reactive protein).High-flow nasal cannula can be used as an initial support strategy for patients with ARDS.With MSC transplantation,the fracrion of inspired O2(Fi02)of the two patients gradually decreased while the oxygen saturation(Sa02)and partial pressure of oxygen(P02)improved.Additionally,the patients9 chest computed tomography showed that bilateral lung exudate lesions were adsorbed after MSC infusion.Results indicated that MSC transplantation provides clinical data on the treatment of COVID-19 and may serve as an alternative method for treating COVID-19,particularly in patients with ARDS. | Lingling Tang Yingan Jiang Mengfei Zhu Lijun Chen Xiaoyang Zhou Chenliang Zhou Peng Ye Xiaobei Chen Baohong Wang Zhenyu Xu Qiang Zhang Xiaowei Xu Hainv Gao Xiaojun Wu Dong Li Wanli Jiang Jingjing Qu Charlie Xiang Lanjuan Li | 2020 | Frontiers of Medicine2020,14,5: | 8 |
| 2 | Influences of Sputtering Angles and Annealing Temperatures on the Magnetic and Magnetostrictive Performances of TbFe Films显示文摘To increase the low-field magnetostriction of TbFe films, the influences of sputtering angles and annealing temperatures on its magnetic and magnetostrictive performances were systematically investigated. With the change in detection of magnetic domains by MFM (magnetic force microscopy) indicates that the easy magnetization direction shifts gradually from perpendicular to parallel to the film plane with decreasing sputtering angles. Annealing can enhance the magnetization and magnetostriction of the TbFe films. | 3 | Influence of Annealing Temperature on the Microstructure and Electrical Properties of Indium Tin Oxide Thin Films显示文摘Indium tin oxide(ITO) thin films were prepared on alumina ceramic substrates by radio frequency magnetron sputtering.The samples were subsequently annealed in air at temperatures ranging from 500 to 1,100 °C for 1 h.The influences of the annealing temperature on the microstructure and electrical properties of the ITO thin films were investigated,and the results indicate that the as-deposited ITO thin films are amorphous in nature.All samples were crystallized by annealing at 500 °C.As the annealing temperature increases,the predominant orientation shifts from(222)to(400).The carrier concentration decreases initially and then increases when the annealing temperature rises beyond1,000 °C.The resistivity of the ITO thin films increases smoothly as the annealing temperature increases to just below900 °C.Beyond 900 °C,however,the resistivity of the films increases sharply.The annealing temperature has a significant effect on the stability of the ITO/Pt thin film thermocouples(TFTCs).TFTCs annealed at 1,000 °C show improved hightemperature stability and Seebeck coefficients of up to 77.73 μV/°C. | Yinzhi Chen Hongchuan Jiang Shuwen Jiang Xingzhao Liu Wanli Zhang Qinyong Zhang | 2014 | Acta Metallurgica Sinica(English Letters)2014,27,2: | 4 |
| 4 | Influences of Film Thickness on the Electrical Properties of TaN_x Thin Films Deposited by Reactive DC Magnetron Sputtering显示文摘TaNx thin films were deposited on commercial polished Al2O3 ceramic substrates by reactive dc magnetron sputtering. The influences of the film thickness on the electrical properties of the samples were examined in detail. It is found that the film thickness does not influence the phase structures of the TaNx thin films. The sheet resistances of the samples shift from 173 Ω/sq. to 7.5 Ω/sq. with the film thickness shifting from 30 nm to 280 nm. With the increase of the film thickness from 30 nm to 280 nm, the temperature coefficient of resistance (TCR) of the samples shifts from negative value to positive value. When the film thickness is about 100 nm, TaNx thin films exhibits a near-zero TCR value (approximately -15×10-6/℃). This fact implies that TaNx thin films with a null TCR can be obtained by adjusting the film thickness. | 5 | Adsorption isotherms and kinetic characteristics of methane on block anthracite over a wide pressure range显示文摘It is important to quantitatively understand the methane adsorption and transport mechanism in coal for an evaluation of the reserves and for its production forecast. In this work, a block coal sample was chosen to perform the CH4 adsorption experiments using the gravimetric method at temperatures of 293.60 K, 311.26 K, 332.98 K and 352.55 K and pressures up to 19 MPa. The excess adsorption capacity of CH4 in dry block anthracite increased, followed by a sequence decrease with the increasing pressure. High temperature restrained the growth of the excess adsorption due to that the adsorption is an intrinsically physical and exothermic process. The excess adsorption peak decreased slowly with the increase of temperature and intersected at a pressure of more than 18 MPa; meanwhile, the pressure at the excess adsorption peak increased.The existing correlations were examined in terms of density rather than pressure. The DR+k correlation, with an average relative deviation of ±0.51%, fitted our data better than the others, with an average relative deviation of up to 2.29%. The transportation characteristics of CH4 adsorption was also investigated in this study, including the adsorption rate and diffusion in block coal. The kinetic data could be described by a modified unipore model. The adsorption rates were found to exhibit dependence on pressure and temperature at low pressures, while the calculated diffusivities exhibited little temperature dependence. In addition, the kinetic characteristics were compared between CH4 and CO2 adsorption on the block coal. | 11 | A novel inhibitor of ARfl and ARv7 induces protein degradation to overcome enzalutamide resistance in advanced prostate cancer显示文摘Enzalutamide(ENZ) is a second-generation androgen receptor(AR) antagonist used for the treatment of castration-resistant prostate cancer(CRPC) and reportedly prolongs survival time within a year of starting therapy. However, CRPC patients can develop ENZ resistance(ENZR), mainly driven by abnormal reactivation of AR signaling, involving increased expression of the full-length AR(ARfl)or dominantly active androgen receptor splice variant 7(ARv7) and ARfl/ARv7 heterodimers. There is currently no efficient treatment for ENZR in CRPC. Herein, a small molecule LLU-206 was rationally designed based on the ENZ structure and exhibited potent inhibition of both ARfl and constitutively active ARv7 to inhibit PCa proliferation and suppress ENZR in CRPC. Mechanically, LLU-206 promoted ARfl/ARv7 protein degradation and decreased ARfl/ARv7 heterodimers through mouse double minute 2-mediated ubiquitination. Finally, LLU-206 exhibited favorable pharmacokinetic properties with poor permeability across the blood-brain barrier, leading to a lower prevalence of adverse effects, including seizure and neurotoxicity, than ENZ-based therapies. Single cell [Ca2+]i analysis and [Ca2+], measurement in cell populations revealed that EGF triggered a rapid [Ca2+]iincrease in the dose-dependent and time- dependent manner. Pretreatment of cells with an endoplasmic reticulum (ER) Ca2+-ATPase inhibitor, thapsigargin (TG) at 100 nM concentration for 20 min, completely abolished EGF-induced [Ca2+]i increase, and chelating extracellular calcium by excess EGTA partially inhibited the increase.Furthermore, the expression of antisense EGF receptor sequence in BEL-7404 cells suppressed the [Ca2+]i response to EGF. | 15 | Risk factors for disease severity in COVID-19 patients: A single-center retrospective study显示文摘Background:The outbreak of coronavirus disease 2019(COVID-19)has posed a huge threat to human health.However,little is known regarding the risk factors associated with COVID-19 severity.We aimed to explore early-stage disease risk factors associated with eventual disease severity.Methods:This study enrolled 486 hospitalized,non-intensive care unit(ICU)-admitted adult patients with COVID-19(age≥18 years)treated at Wuhan Jinyintan Hospital,who were divided into three groups according to disease severity.The demographic,clinical,and laboratory data at admission and clinical outcomes were compared among severity groups,and the risk factors for disease severity were identified by multiple regression analysis.Results:Of 486 patients with COVID-19,405(83.33%)were discharged,33(6.71%)died outside of the ICU,and 48(7.20%)were still being treated in the ICU by the time the study period ended.Significant differences in age,lymphocyte counts,and the levels of procalcitonin,aspartate aminotransferase,and D-dimer(P<0.001 for all)among the three groups.Further analysis showed that older age,decreased lymphocyte counts,and increased procalcitonin,aspartate aminotransferase,and D-dimer levels were significantly associated with disease progression.Conclusion:Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)may impair the immune system,the blood coagulation system,and hepatic and cardiac function.Some clinical characteristics and laboratory findings can help identify patients with a high risk of disease severity,which can be significant for appropriate resource allocation during the COVID-19 pandemic. | Jingqing Xu Yanping Cai Min Li Qing Li Yingrui Zhang Ying Jiang Wanli Yan Xuemei Chen Hongxuan Zhang Zhaonan Zeng Fayang Lian Xiuling Shang Rongguo Yu | 2021 | Journal of Intensive Medicine2021,1,2: | 0 |
| 16 | RNA interference affects tumorigenicity and expression of insulin-like growth factor-1,insulin-like growth factor-1 receptor,and basic fibroblast growth factor-2 in rat C6 glioma cells显示文摘BACKGROUND:Human gliomas are more likely to express basic fibroblast growth factor-2(FGF-2), insulin-like growth factor-1(IGF-1),and IGF-1 receptor(IGF-1R) than normal brain tissue.These factors activate signal transduction systems of Ras/MAPK and PI3K/Akt,which promote glioma growth. OBJECTIVE:To utilize RNA interference(RNAi) technique to down-regulate FGF-2,IGF-1,and IGF-1R gene expression,and to investigate the effects of these genes on rat C6 glioma cells,as well as the feasibility of RNAi for treating glioma. DESIGN,TIME AND SETTING:This neurooncological,randomized,controlled,in vivo and in vitro experiment,which used RNAi methodology,was performed at the Laboratory of Molecular Biology, Institute of Biochemistry,Chinese Academy of Sciences between August 2005 and February 2008. MATERIALS:Rat C6 cell lines were purchased from Shanghai Institute of Cellular Biology Affiliated to Chinese Academy of Sciences.Small interfering RNA(siRNA) was synthesized by Shanghai GenePharma.Anti-IGF-1,anti-IGF-1R,anti-FGF-2,anti-mouse and anti-rabbit IgG G1-HRP antibodies were provided by Santa Cruz Biotechnology,USA.Four to six week-old BALB/c nude mice were purchased from the Laboratory Animal Center,Chinese Academy of Sciences. METHODS:C6 glioma cells were transfected with siRNA,which was chemically synthesized in vitro to correspond to endogenous FGF-2,IGF-1,and IGF-1R genes.The inhibition ratio of targeting mRNA expression was detected by semiquantitative RT-PCR,and protein expression was determined by Western blot analysis.C6 glioma cell proliferation was observed using a growth curve. C6 glioma cell apoptosis rate and cell cycle were detected by flow cytometry.C6 glioma cell growth regression was observed by transwell migration assay.In addition,nude mouse subcutaneous tumor models were used in this study.For studying the anti-tumor effects of IGF-1 and IGF-1R siRNA,two blank control groups,with six mice each,were set up:A(2.5μg siRNA was injected one week after C6 cells were inoculated,i.e.,when tumor volume reached 8 mm×8 mm) and B(siRNA was injected at the same time with C6 cells were inoculated.To study the effects of FGF-2 siRNA, the groups consisted of a blank control group,negative control group,2.6μg siRNA group,4μg siRNA group,and 5.3μg siRNA group,with six mice each. MAIN OUTCOME MEASURES:mRNA and protein inhibition ratio of FGF-2,IGF-1,and IGF-1R;C6 glioma cell proliferation,apoptosis,and cycle growth arrest;C6 glioma cell growth regression and subcutaneous tumorigenicity rates. RESULTS:All siRNA constructs proved to be effective.After 48 hours,transfection of 200 nmol/L siRNA resulted in a FGF-2 or IGF-1R gene inhibition ratio>80%and an IGF-1 gene inhibition ratio of approximately 70%.Protein expression levels for FGF-2,IGF-1,and IGF-1R decreased in a dose-dependent manner following siRNA transfection,with an inhibition rate>85%,60%,and 50%, respectively.C6 glioma cell proliferation and apoptosis rates increased in proportion to siRNA.The apoptosis rate of C6 glioma cells induced by FGF-2,IGF-1,and IGF-1R siRNA was 39.96%,15.07%, and 22.47%,respectively(P<0.01).Transfection of 200 nmol/L IGF or IGF-1R siRNA for 48 hours suppressed C6 glioma cell migration.At 30 days after intratumoral injection of 2.6,4,and 5.3μg FGF-2 siRNA,tumor growth regression rate of FGF-2 siRNA was 56%,67%,and 86%,respectively. The tumor growth regression rate was 71.88%and 45.71%,respectively,when IGF-1 or IGF-1R siRNA was intratumorally injected 1 week after C6 glioma cell transplantation.When IGF-1 or IGF-1R siRNA was intratumorally injected during C6 glioma cell transplantation,the tumor growth regression rate was 78.13%and 74.29%,respectively. CONCLUSION:siRNA transfection downregulated gene expression of FGF-2,IGF-1,and IGF-1R.
