| 1 | Phytochrome B interacts with SWC6 and ARP6 to regulate H2A.Z deposition and photomorphogensis in Arabidopsis显示文摘Light serves as a crucial environmental cue which modulates plant growth and development, and which is controlled by multiple photoreceptors including the primary red light photoreceptor,phytochrome B(phyB). The signaling mechanism of phyB involves direct interactions with a group of basic helix-loop-helix(bHLH) transcription factors, PHYTOCHROME-INTERACTING FACTORS(PIFs), and the negative regulators of photomorphogenesis, COP1 and SPAs. H2 A.Z is an evolutionarily conserved H2 A variant which plays essential roles in transcriptional regulation. The replacement of H2 A with H2 A.Z is catalyzed by the SWR1 complex. Here, we show that the Pfr form of phyB physically interacts with the SWR1 complex subunits SWC6 and ARP6. phyB and ARP6 coregulate numerous genes in the same direction,some of which are associated with auxin biosynthesis and response including YUC9, which encodes a rate-limiting enzyme in the tryptophandependent auxin biosynthesis pathway. Moreover,phyB and HY5/HYH act to inhibit hypocotyl elongation partially through repression of auxin biosynthesis. | 5 | Comparison of the differentiation of dental pulp stem cells and periodontal ligament stem cells into neuron-like cells and their effects on focal cerebral ischemia显示文摘Recent studies have reported an in creasing in cidence of ischemic stroke,particularly in younger age groups.Dental pulp stem cells(DPSCs)and periodontal ligament stem cells(PDLSCs)are the most comm on stem cells acquired from the teeth of adults,even elderly people.However,there are no detailed reports on whether DPSCs or PDLSCs are suitable for the treatment of ischemic stroke.In this study,the in vitro differentiation of DPSCs and PDLSCs into neuron-like cells was evaluated.Then,we established a rat model of cerebral ischemia.DPSCs or PDLSCs were administered to animals,and the therapeutic effects of these two types of cells were investigated.The results showed that PDLSCs had a higher differentiation rate than DPSCs.Immunofluorescence studies showed that the expression of the neuronal differentiation markerThy-1 was higher in PDLSCs than in DPSCs,and other gene markers of neuronal differentiation showed corresponding trends,which were confirmed by western blot analysis.In this process,the Notch and Wnt signaling pathways were inhibited and activated,respectively.Fin ally,rats with transient occlusion of the right middle cerebral artery were used as a model to assess the therapeutic effect of PDLSCs and DPSCs on ischemia.The results showed that rats in the PDLSC-treated group emitted significantly greater red fluorescence signal than the DPSC-treated group.PDLSC transplantation promoted the recovery of neurological function more effectively than DPSC transplantation.Hence,PDLSCs represent an autogenous source of adult mesenchymal stem cells with desirable biological properties and may be an ideal candidate for clinical applications. | Tingting Wu Wanting Xu Hanlin Chen Shasha Li Rengang Dou Hongtao Shen Xue Liu Xiaoyu Liu Yongfeng Hong Jiacai He | 2020 | Acta Biochimica et Biophysica Sinica2020,52,9: | 2 |
| 6 | Molecular Mechanism of Induction on Apoptosis of Human Esophageal Cancer HCE-4 Cells by Active Components from Astragalus membranaceus显示文摘[Objectives] To investigate the pharmacologic effects of active components from A. membranaceus on human esophageal cancer HCE-4 cells and its apoptosis mechanism. [Methods] The viabilities of HCE-4 cells were measured by MTT assay. The induction of active components from A. membranaceus on apoptosis of HCE-4 cells was detected by Annexin V-FITC/PI double staining. The apoptotic-related protein expression levels were determined by Western blotting. [Results] Formononetin and astragaloside IV suppressed the proliferation of HCE-4 cells in a dose-dependent manner. The Annexin V-FITC/PI double staining results showed that formononetin and astragaloside IV could induce HCE-4 cells apoptosis in a time-dependent manner. The Western blotting results showed that formononetin and astragaloside IV could significantly down-regulate p-AKT,pro-caspase-3,and increase cle-caspase-3 protein expression in HCE-4 cells. | 7 | Bearing Failure Optimization of Composite Double-Lap Bolted Joints Based on a Three-Step Strategy Marked By Feasible Region Reduction and Model Decoupling显示文摘To minimize the mass and increase the bearing failure load of composite double-lap bolted joints,a three-step optimization strategy including feasible region reduction,optimization model decoupling and optimization was presented.In feasible region reduction,the dimensions of the feasible design region were reduced by selecting dominant design variables from numerous multilevel parameters by sensitivity analyses,and the feasible regions of variables were reduced by influence mechanism analyses.In model decoupling,the optimization model with a large number of variables was divided into various sub-models with fewer variables by variance analysis.In the third step,the optimization sub-models were solved one by one using a genetic algorithm,and the modified characteristic curve method was adopted as the failure prediction method.Based on the proposed optimization method,optimization of a double-lap single-bolt joint was performed using the ANSYS®code.The results show that the bearing failure load increased by 13.5%and that the mass decreased by 8.7%compared with those of the initial design of the joint,which validated the effectiveness of the three-step optimization strategy. | Fengrui Liu Wanting Yao Xinhong Shi Libin Zhao Jianyu Zhang | 2020 | Computers, Materials & Continua2020,,2: | 1 |
| 8 | Drainage evolution in intermontane basins at the Qinling-Daba Mountains显示文摘River capture is of great significance to landform evolution and hominine migration.In the Qinling-Daba Mountains,there is a viewpoint that Jialing River captured Hanjiang River,but this is still controversial.In this paper,we discuss the drainage evolution processes in intermountain basins at the Qinling-Daba Mountains based on a combination of detrital zircon UPb geochronology and geomorphic indexes.We suggest that the Hanjiang River gradually captured the Jialing River from east to west,accompanied by the evolution of the ancient Yangtze River.In terms of geomorphic evidences,wide valleys did not match with discharge,and a series of wind gaps developed in the Shiquan-Ankang basin.In addition,the valley shapes and width-toheight ratios(Vf)indicate two possible rapid incisions.The hypsometric integrals(HI)reflect that the landform gradually changes from the old stage to the youth stage from west to east.Theχvalues show that the drainage divide is moving to the side of the Yuehe River,and the Yuehe River is gradually shrinking.According to the sedimentary records,the zircon U-Pb age distributions indicate the provenance change.The high-altitude terraces show three age peaks(200–250,400–505,and 700–900 Ma),with the dominant Indosinian age peak(200–250 Ma),while the modern fluvial sediments only show a single peak of Jinning(700–900 Ma).These data show that there are two major river captures:(1)The ancient Hanjiang River cut through the regional compression ridge,and then captured the Hanzhong Basin river system(a part of the ancient Jialing river system)from east to west,and(2)The southern tributary captured the trunk with the uplift of the divide in the Shiquan-Ankang Basin,forming the modern drainage pattern in the upper Hanjiang River.The activities of the regional strike-slip fault,and the associated compression uplift played a key role in the river captures,the drainage evolution,and related landforms in the Shiquan-Ankang basin.In addition,it is shown that the evolution of the upper tributary basins lagged behind the response of the trunk channel to the tectonic activities and river captures.The interconnected wide valleys caused by river capture may have provided convenient geomorphological conditions for human migration into the Qinling-Daba Mountains along those river valleys. | Wanting XIE Xianyan WANG Hanzhi ZHANG Quanyu LIU Shejiang WANG Huayu LU | 2021 | Science China Earth Sciences2021,64,11: | 1 |

| 10 | Novel heat shock protein Hsp70L1 activates dendritic cells and acts as a Th1 polarizing adjuvant显示文摘Heat shock proteins (HSPs) are reported to act as effective adjuvants to elicit anti-tumor and anti-infection immunity. Here, we report that Hsp70-like protein 1 (Hsp70L1), a novel HSP derived from human dendritic cells (DCs), has potent adjuvant effects that polarize responses toward Th1. With a calculated molecular weight of 54.8 kDa, Hsp70L1 is smaller in size than Hsp70 but resembles it both structurally and functionally. Hsp70L1 shares common receptors on DCs with Hsp70 and can interact with DCs, promoting DC maturation and stimulating secretion of the proinflammatory cytokines interleukin 12p70 (IL-12p70), IL-1beta, tumor necrosis factor-alpha (TNF-alpha), and the chemokines IP-10, macrophage inflammatory protein-1alpha (MIP-1alpha), MIP-1beta, and normal T cell expressed and secreted (RANTES). The induction of interferon-gamma-inducible protein 10 (IP-10) secretion by Hsp70L1 is not shared by Hsp70, and other functional differences include more potent stimulation of DC IL-12p70, CC-chemokine, and CCR7 and CXCR4 expression by Hsp70L1. Immunization of mice with the hybrid peptide Hsp70L1-ovalbumin(OVA)(257-264) induces an OVA(257-264)-specific Th1 response and cytotoxic T lymphocyte (CTL) that results in significant inhibition of E.G7-OVA tumor growth. | 15 | Lignin derived absorbent for efficient and sustainable CO_(2) capture显示文摘High and cost-efficient capture of CO_(2) is a prerequisite and an inevitable path of carbon emission reduction. To address the challenges(high cost, low efficiency, less sustainability, etc.) of existing petroleum-based CO_(2) absorbents, herein, a class of efficient and sustainable lignin-based absorbents were resoundingly prepared by grafting the active amine group on a lignin derived compound vanillin and alkali lignin. The results demonstrated that vanillin modified by acrylamide achieved the excellent absorption capacity among the three absorbents, whose ability was 0.114 g CO_(2) per gram of absorbent under 25 ℃ and 100 kPa. In addition, the absorbent retained stable absorbability of CO_(2) after 6 cycles.The absorbing capacity of the absorbent formed by the coupling of vanillin and acrylamide to CO_(2) was much greater than their own(i.e. 0 g CO_(2) ·g^(-1)vanillin, 0.01 g CO_(2) ·g^(-1) acrylamide, respectively).Detailed information revealed the multi-site synergistic absorption mechanism, in which CO_(2) has C and O double interactions with the amide group of the absorbent, and single interaction with the hydroxyl oxygen on the benzene ring of the absorbent. | 16 | Development and validation of prognostic implications of chromosome abnormalities algorithm for newly diagnosed multiple myeloma显示文摘Fluorescence in situ hybridization(FISH)evaluation is essential for initial risk stratification in multiple myeloma(MM).The presence of specific cytogenetic abnormalities(CA)confers a heterogeneity impact on prognosis.However,the cutoff values among different centers are not uniform.Therefore,we conduct this study to better predict the prognosis of newly diagnosed MM patients based on FISH results.The Kaps method was used to calculate the chromosomal abnormal cutoff values.A total of 533 participants were included in the study.The best cutoff value of overall survival were as follows:17p20.1%,13q85%,1q21+39%,t(11;14)55.5%,t(14;16)87%,and t(4;14)53.5%.The survival analysis showed that 17pand 1q21+were the independent factors affecting both OS and progress free survival(PFS)among CA.The analysis based on the cutoff value obtained by Kaps suggested that 13q,t(14;16),17p,and 1q21+were independent factors affecting OS among CA;t(14;16),17p,and 1q21+were independent factors affecting PFS among CA.The prognostic model was constructed by the Kaps method with the Harrell concordance index(c-index)at 0.719(95%CI,0.683–0.756;corrected 0.707),which was higher than that calculated by the European Myeloma Network criteria(0.714;95%CI,0.678–0.751;corrected 0.696).In conclusion,chromosomal abnormalities in different proportions and combinations can affect the prognosis ofMMpatients.Therefore,effective criteria should be formulated to evaluate the prognosis ofMMpatients better. | Tiancheng Luo Wanting Qiang Jing Lu Haiyan He Jin Liu Lu Li Hua Jiang Weijun Fu Juan Du | 2021 | Blood Science2021,3,3: | 0 |

| 20 | Higher efficacy of oral etoposide for mobilization of peripheral blood stem cells in patients with multiple myeloma显示文摘This study compares the efficacy,toxicity,hematopoietic recovery,and cost of stem-cell mobilization using intermediate-dose cyclophosphamide(IDCy)plus granulocyte colony-stimulating factor(G-CSF)compared with etoposide(VP-16)plus pegylated granulocyte colony-stimulating factor(PEG-rhG-CSF)in multiple myeloma(MM)patients.Two hundred forty-four consecutive patients undergoing mobilization with IDCy(3-3.5g/m^(2))plus G-CSF(n=155)were compared with patients receiving VP-16 plus PEG-rhG-CSF(n=89),including oral etoposide(n=65)and intravenous etoposide(n=24).Compared with IDCy,VP-16 use was associated with significantly higher median peak peripheral blood CD34+cell count(8.20[range:1.84-84]×10^(6)/kg vs 4.58[range:0.1-27.9]×10^(6)/kg,P=.000),and ideal CD34+cell yield of more than 6×10^(6)/kg(56.8%vs 35.1%,P=.001),notably with a higher efficacy in oral VP-16 use compared with IDCy use(CD 34+cell counts:median peak peripheral blood 5.87 vs 4.58?106/kg and≥6×10^(6)/kg[48.4%vs 35.1%]).The median number of apheresis courses was reduced from two in the IDCy group to one in the VP-16 group(P=.000).IDCy use was associated with significantly more frequent episodes of neutropenia(70.2%vs 35.2%;P=.000),intravenous antibiotic use(13.2%vs 11.4%;P=.672),and hospitalization(P=.000).The recoveries of neutrophils and platelets after autologous stem-cell transplantation were significantly faster in the VP-16 group compared with the IDCy group(P=.000).Our data indicate robust stem-cell mobilization in MM patients with VP-16 delivered either orally or intravenously.When compared with intravenous VP-16,oral VP-16 mobilization was associated with significantly more convenient,lower average total costs,and especially decreased the risk of hospital visits and exposure. | Wanting Qiang Hua Jiang Pei Guo Jing Lu Jin Liu Lu Li Haiyan He Xiaoxia Hu Weijun Fu Juan Du | 2022 | Blood Science2022,4,2: | 0 |
