| 1 | Epalrestat protects against diabetic peripheral neuropathy by alleviating oxidative stress and inhibiting polyol pathway显示文摘Epalrestat is a noncompetitive and reversible aldose reductase inhibitor used for the treatment of diabetic neuropathy. This study assumed that epalrestat had a protective effect on diabetic peripheral nerve injury by suppressing the expression of aldose reductase in peripheral nerves of diabetes mellitus rats. The high-fat and high-carbohydrate model rats were established by intraperitoneal injection of streptozotocin. Peripheral neuropathy occurred in these rats after sustaining high blood glucose for 8 weeks. At 12 weeks after streptozotocin injection, rats were intragastrically administered epalrestat 100 mg/kg daily for 6 weeks. Transmission electron microscope revealed that the injuries to myelinated nerve fibers, non-myelinated nerve fibers and Schwann cells of rat sciatic nerves had reduced compared to rats without epalrestat administuation. Western blot assay and immunohistochemical results demonstrated that after intervention with epalrestat, the activities of antioxidant enzymes such as superoxide dismutase, catalase and glutathione peroxidase gradually increased, but aldose reductase protein expression gradually diminished. | 3 | Oral health status in Sichuan Province: findings from the oral health survey of Sichuan, 2015–2016显示文摘To investigate oral health status in the residents of Sichuan Province, southwest China, a cross-sectional study was performed using the latest Oral Health Survey Basic Methods recommended by the World Health Organization. A multistage stratified random cluster-sampling method was used to enroll participants from the following three groups: children aged 3–5 years,adolescents aged 12 years, and people aged 65–74 years. In these three groups, the mean numbers of teeth that were affected by caries were 3.28, 0.86 and 5.13, respectively, resulting in a prevalence of 63.47%, 37.20% and 83.20%, respectively.Relative to the high rate of decayed teeth, the prevalence of fillings was very low in all age groups(0.97%, 7.24% and 5.43%,respectively). In the 12-year-old adolescent group, only 3.61% had good pit and fissure sealing. In addition, the rate of dental fluorosis was 24.80%, and the Community Fluorosis Index value was 0.39. In the elder group, the community periodontal index was2.92. The prevalence in the elderly of having lost at least one tooth was 75.54%. Additionally, 4.44% of these participants were edentulous. The incidence of dental prosthesis was 51.75%, the proportion with a removable partial denture, a fixed denture, full dentures, dental implants and an informal fixed bridge was 21.59%, 11.45%, 4.64%, 0 and 16.67%, respectively. In this study,8.2% of the elderly participants were affected by different types of oral mucosal lesions. Among such lesions, recurrent aphthous ulcers were most prevalent(2.69%) and oral lichen planuses were second(1.6%). The conclusion presented in this survey is that oral diseases, especially dental caries and periodontal disease, are frequent and common in Sichuan province, China. | 4 | Value of contrast-enhanced ultrasound combined with elastography in evaluating cervical lymph node metastasis in papillary thyroid carcinoma显示文摘BACKGROUND Cervical lymph node metastasis in papillary thyroid carcinoma(PTC) affects the treatment and prognosis of patients. Ultrasound is a common imaging method for detecting cervical lymph nodes in PTC patients; however, it is not accurate in determining lymph node metastasis.AIM To evaluate the value of contrast-enhanced ultrasound combined with elastography in evaluating cervical lymph node metastasis in PTC.METHODS A total of 94 patients with PTC were recruited. According to pathological results,lymph nodes were divided into two groups: metastatic group(n = 50) and reactive group(n = 63). The routine ultrasound findings, contrast-enhanced ultrasound and elastography data were recorded and compared. Logistic regression was used to generate predictive probability distributions for the diagnosis of lymph node metastasis with different indicators. Receiver operating characteristic curve analysis was used to test the efficacy of contrast-enhanced ultrasound combined with elastography based on routine ultrasound in evaluating PTC cervical lymph node metastasis.RESULTS The ratio of long diameter/short diameter(L/S) ≤ 2, irregular marginal morphology, missing lymphatic portal, peripheral or mixed blood flow distribution, peak intensity(PI), non-uniform contrast distribution and elasticity score in the metastatic group were significantly higher than those in the reactive group(P < 0.05). L/S ratio, missing lymphatic portal, PI and elasticity score had a significant influence on the occurrence of PTC cervical lymph node metastasis(P< 0.05). Furthermore, the area under the curve(AUC) for lymph node metastasis diagnosed using the combination of PI ratio, elasticity score, missing lymphatic portal and LS was 0.936, which was significantly higher than the AUC for PI ratio alone. The difference was statistically significant(P < 0.05). | 13 | Tight sandstone gas accumulation mechanism and development models显示文摘Tight sandstone gas serves as an important unconventional hydrocarbon resource, and outstanding results have been obtained through its discovery both in China and abroad given its great resource potential.However, heated debates and gaps still remain regarding classification standards of tight sandstone gas, and critical controlling factors, accumulation mechanisms, and development modes of tight sandstone reservoirs are not determined. Tight sandstone gas reservoirs in China are generally characterized by tight strata, widespread distribution areas, coal strata supplying gas, complex gas–water relations, and abnormally low gas reservoir pressure. Water and gas reversal patterns have been detected via glass tube and quartz sand modeling, and the presence of critical geological conditions without buoyancy-driven mechanisms can thus be assumed. According to the timing of gas charging and reservoir tightening phases, the following three tight sandstone gas reservoir types have beenidentified:(a) ‘‘accumulation–densification''(AD), or the conventional tight type,(b) ‘‘densification–accumulation''(DA), or the deep tight type, and(c) the composite tight type. For the AD type, gas charging occurs prior to reservoir densification, accumulating in higher positions under buoyancy-controlled mechanisms with critical controlling factors such as source kitchens(S), regional overlaying cap rocks(C), gas reservoirs,(D) and low fluid potential areas(P). For the DA type, reservoir densification prior to the gas charging period(GCP) leads to accumulation in depressions and slopes largely due to hydrocarbon expansive forces without buoyancy, and critical controlling factors are effective source rocks(S), widely distributed reservoirs(D), stable tectonic settings(W) and universal densification of reservoirs(L). The composite type includes features of the AD type and DA type, and before and after reservoir densification period(RDP), gas charging and accumulation is controlled by early buoyancy and later molecular expansive force respectively. It is widely distributed in anticlinal zones, deep sag areas and slopes, and is controlled by source kitchens(S), reservoirs(D), cap rocks(C), stable tectonic settings(W), low fluid potential areas(P), and universal reservoir densification(L). Tight gas resources with great resource potential are widely distributed worldwide, and tight gas in China that presents advantageous reservoir-forming conditions is primarily found in the Ordos, Sichuan, Tarim, Junggar, and TurpanHami basins of central-western China. | 15 | Atorvastatin activates autophagy and promotes neurological function recovery after spinal cord injury显示文摘Atorvastatin, a lipid-lowering medication, provides neuroprotective effects, although the precise mechanisms of action remain unclear. Our previous studies confirmed activated autophagy following spinal cord injury, which was conducive to recovery of neurological functions. We hypothesized that atorvastatin could also activate autophagy after spinal cord injury, and subsequently improve recovery of neurological functions. A rat model of spinal cord injury was established based on the Allen method. Atorvastatin(5 mg/kg) was intraperitoneally injected at 1 and 2 days after spinal cord injury. At 7 days post-injury, western blot assay, reverse transcription-polymerase chain reaction, and terminal deoxynucleotidyl transferase-mediated dU TP nick-end labeling(TUNEL) staining results showed increased Beclin-1 and light chain 3B gene and protein expressions in the spinal cord injury + atorvastatin group. Additionally, caspase-9 and caspase-3 expression was decreased, and the number of TUNEL-positive cells was reduced. Compared with the spinal cord injury + saline group, Basso, Beattie, and Bresnahan locomotor rating scale scores significantly increased in the spinal cord injury + atorvastatin group at 14–42 days post-injury. The application of these technologies in basic biomedical research has yielded significant advances in identifying and studying key molecular targets relevant to human diseases and their treatment. The clinical translation of genome editing techniques offers unprecedented biomedical engineering capabilities in the diagnosis, prevention, and treatment of disease or disability. Here, we provide a general summary of emerging biomedical applications of genome editing, including open challenges.
