| 1 | Development and molecular analysis of a novel acetohydroxyacid synthase rapeseed mutant with high resistance to sulfonylurea herbicides显示文摘With the increasing promotion of simplified rapeseed cultivation in recent years,the development of cultivars with high resistance to herbicides is urgently needed.We previously developed M342,which shows sulfonylurea herbicide resistance,by targeting acetohydroxyacid synthase(AHAS),a key enzyme in branched-chain amino acid synthesis.In the present study,we used a progeny line derived from M342 for an additional round of ethyl methane sulfonate mutagenesis,yielding the novel mutant DS3,which harbored two mutations in AHAS genes and showed high sulfonylurea resistance.One mutation was the substitution Trp574 Leu,as in M342,according to Arabidopsis protein sequencing.The other site was a newly recognized substitution,Pro197 Leu.A KASP marker targeting Pro197 Leu was developed and reliably predicted the response to sulfonylurea herbicides in the F2 population.The combination of Trp574 Leu and Pro197 Leu in DS3 produced a synergistic effect that greatly increased herbicide resistance.Analysis of the protein structures of AHAS1 and AHAS3 in wild-type and single-gene mutant plants revealed three-dimensional protein conformational changes that could account for differences in herbicide resistance characteristics including toxicity tolerance,AHAS enzyme activity,and AHAS gene expression. | Yue Guo Changle Liu Weihua Long Jianqin Gao Jiefu Zhang Song Chen Huiming Pu Maolong Hu | 2022 | The Crop Journal2022,10,1: | 3 |
| 2 | Derivation and characterization of human embryonic stem cell lines from poor quality embryos显示文摘Poor quality embryos discarded from in vitro fertilization (IVF) laboratories are good sources for deriving human embryonic stem cell (hESC) lines. In this study, 166 poor quality embryos donated from IVF centers on day 3 were cultured in a blastocyst medium for 2 days, and 32 early blastocysts were further cultured in a blastocyst optimum culture medium for additional 2 days so that the inner cell masses (ICMs) could be identified and isolated easily. The ICMs of 17 blastocysts were isolated by a mechanical method, while those of the other 15 blastocysts were isolated by immunosurgery. All isolated ICMs were inoculated onto a feeder layer for subcultivation. The rates of ICM at- tachment, primary ICM colony formation and the efficiency of hESC derivation were similar between the ICMs isolated by the two methods (P>0.05). As a result, four new hESC lines were established. Three cell lines had normal karyotypes and one had an unbalanced Robertsonian translocation. All cell lines showed normal hESC characteristics and had the differentiation ability. In conclusion, we established a stable and effective method for hESC isolation and culture, and it was confirmed that the mechanical isolation was an effective method to isolate ICMs from poor embryos. | 4 | Fabrication of nanofibrous sensors by electrospinning显示文摘This article reviews the techniques and applications of electrospinning for the fabrication of nanofibrous sensors. Considering that nanosensors require a large specific surface area and a continuous structure for the conduction of current signals, electrospun nanofibers have the dominant advantage. The device preparation is mainly divided into surface treatment and high-temperature sintering, which are, respectively, used for preparing composite conductive fibers and inorganic semiconductor fibers. Typical applications include pressure sensing, gas sensing, photoelectric sensing, and temperature sensing. In addition, nano-selfpowered systems have been mentioned to emphasize the good performance of smart nanosystems that do not require external power. | 16 | Investigations of the mechanical response of dummy HTPB propellant grain under ultrahigh acceleration overload conditions using onboard flight-test measurements显示文摘In this paper,to study the mechanical responses of a solid propellant subjected to ultrahigh acceleration overload during the gun-launch process,specifically designed projectile flight tests with an onboard measurement system were performed.Two projectiles containing dummy HTPB propellant grains were successfully recovered after the flight tests with an ultrahigh acceleration overload value of 8100 g.The onboard-measured time-resolved axial displacement,contact stress and overload values were successfully obtained and analysed.Uniaxial compression tests of the dummy HTPB propellant used in the gunlaunched tests were carried out at low and intermediate strain rates to characterize the propellant's dynamic properties.A linear viscoelastic constitutive model was employed and applied in finite-element simulations of the projectile-launching process.During the launch process,the dummy propellant grain exhibited large deformation due to the high acceleration overload,possibly leading to friction between the motor case and propellant grain.The calculated contact stress showed good agreement with the experimental results,though discrepancies in the overall displacement of the dummy propellant grain were observed.The dynamic mechanical response process of the dummy propellant grain was analysed in detail.The results can be used to estimate the structural integrity of the analysed dummy propellant grain during the gun-launch process. | Yiming Zhang Ningfei Wang Weihua Ma Ran Wang Long Bai Yi Wu | 2024 | Defence Technology(防务技术)2024,32,2: | 0 |
| 17 | A Pyrazine-Based 2D Conductive Metal-Organic Framework for Efficient Lithium Storage显示文摘The merits of intrinsic electrical conductivity, high specific surface area, tunable chemical composition and tailor-made properties enable two-dimensional conductive metal-organic frameworks (2D c-MOFs) as promising next-generation electrode materials in the field of energy storage and conversion. Herein, we have designed and synthesized a novel pyrazine-based 2D c-MOF (TPQG-Cu-MOF) bearing extended π-conjugated structure and abundant redox active sites. Thanks to the excellent redox reversibility of pyrazine units and CuO2 units, as well as the insolubility of the rigid framework skeleton, TPQG-Cu-MOF as the cathode material of lithium-ion battery exhibits a reversible specific capacity (150.2 mAh·g–1 at 20 mAh·g–1), good cycling stability (capacity retention of 82.6% after 500 cycles at 1 A·g–1) and excellent rate performance. Comprehensive ex-situ spectroscopic studies revealed the reversible redox activity of pyrazine units and CuO2 units of TPQG-Cu-MOF during the Li+ insertion/extraction process.
