1Major accident analysis and prevention of coal mines in China from the year of 1949 to 2009显示文摘From the year of 1949 to the present, the China national coal output has been increasing quickly and became first in the world in 2009. But at the same time, major coal mining accidents still exist nowadays. In order to review the overall situation and provide information on major accidents of coal mines in China, we investigated 26 major coal mining accidents in China between the years of 1949 and 2009 through statistical methods, each of which led to more than 100 fatalities. Statistical characteristics about accident-related factors such as time, death toll, accident reasons, characters and nature of enterprise were analyzed. And some special conclusions have been achieved. For example, although we have made great progress, the safety situation in China coal mining industry is still serious, and the reasons for the mining accidents are all human errors which are not inevitable. Such results may be helpful to prevent major accidents in coal mines. Moreso, based on both the knowledge of other countries which have good safety situation nowadays and the safety management situation of China, we made suggestion on safety management of China coal mining. 2Stem cell transplantation for treatment of cerebral ischemia in rats Effects of human umbilical cord blood stem cells and human neural stem cells显示文摘BACKGROUND: Exogenous neural stem cell transplantation promotes neural regeneration. However, various types of stem cells transplantation outcomes remain controversial. OBJECTIVE: To explore distribution, proliferation and differentiation of human neural stem cells (hNSCs) and human umbilical cord blood stem cells (hUCBSCs) following transplantation in ischemic brain tissue of rats, and to compare therapeutic outcomes between hNSCs and hUCBSCs. DESIGN, TIME AND SETTING: Randomized controlled animal studies were performed at the Experimental Animal Center of Nanjing Medical University and Central Laboratory of Second Affiliated Hospital of Nanjing Medical University of China from September 2008 to April 2009. MATERIALS: hNSCs were harvested from brain tissue of 10-13 week old fetuses following spontaneous abortion, and hUCBSCs were collected from umbilical cord blood of full-term newborns at the Second Affiliated Hospital of Nanjing Medical University of China. hNSCs and hUCBSCs were labeled by 5-bromodeoxyuridine (BrdU) prior to transplantation. METHODS: Rat models of cerebral ischemia were established by the suture method. A total of 60 healthy male Sprague Dawley rats aged 7-9 weeks were randomly assigned to hNSC transplantation, hUCBSC transplantation and control groups. The rat models in the hNSC transplantation, hUCBSC transplantation and control groups were infused with hNSC suspension, hUCBSC suspension and saline via the caudal vein, respectively. MAIN OUTCOME MEASURES: The distribution, proliferation and differentiation of hNSCs and hUCBSCs in ischemic brain tissue were observed using immunohistochemical methods. Neurological function in rats was assessed using the neurological severity score. RESULTS: The number of BrdU-positive cells was significantly greater in the hNSC transplantation group compared with hUCBSC transplantation group at 14 days following transplantation (P < 0.05). The number of BrdU-positive cells reached a peak at 28 days following transplantation. Nestin-positive, glial fibrillary acidic protein-positive, cyclic nucleotide 3' phosphohydrolase-positive and neuron specific enolase-positive cells were visible following transplantation. No significant difference was determined in the constituent ratio of various cells between hNSC and hUCBSC transplantation groups (P > 0.05). The neurological severity score was significantly decreased in rats at 21 days following transplantation (P < 0.05). No significant difference was detected in neurological severity score between hNSC and hUCBSC transplantation groups at various time points (P > 0.05). 4The effects of claudin 14 during early Wallerian degeneration after sciatic nerve injury显示文摘Claudin 14 has been shown to promote nerve repair and regeneration in the early stages of Wallerian degeneration(0–4 days) in rats with sciatic nerve injury, but the mechanism underlying this process remains poorly understood. This study reported the effects of claudin 14 on nerve degeneration and regeneration during early Wallerian degeneration. Claudin 14 expression was up-regulated in sciatic nerve 4 days after Wallerian degeneration. The altered expression of claudin 14 in Schwann cells resulted in expression changes of cytokines in vitro. Expression of claudin 14 affected c-Jun, but not Akt and ERK1/2 pathways. Further studies revealed that enhanced expression of claudin 14 could promote Schwann cell proliferation and migration. Silencing of claudin 14 expression resulted in Schwann cell apoptosis and reduction in Schwann cell proliferation. 5Peripheral CD4^(+) T cell signatures in predicting the responses to anti-PD-1/PD-L1 monotherapy for Chinese advanced non-small cell lung cancer显示文摘Limited benefit population of immune checkpoint inhibitors makes it urgent to screen predictive biomarkers for stratifying the patients.Herein,we have investigated peripheral CD4^(+) T cell signatures in advanced non-small cell lung cancer(NSCLC)patients receiving anti-PD-1/PD-L1 treatments.It was found that the percentages of IFN-γand IL-17A secreting naïve CD4^(+) T cells(Tn),and memory CD4^(+) T cells(Tm)expressing PD-1,PD-L1 and CTLA-4 were significantly higher in responder(R)than non-responder(NonR)NSCLC patients associated with a longer progression free survival(PFS).Logistic regression analysis revealed that the baseline IFN-γ-producing CD4^(+) Tn cells and PD-1^(+)CD4^(+) Tm cells were the most significant signatures with the area under curve(AUC)value reaching 0.849.This was further validated in another anti-PD-1 monotherapy cohort.Conversely,high percentage of CTLA-4^(+)CD4^(+) Tm cells was associated with a shorter PFS in patients receiving anti-PD-L1 monotherapy.Our study therefore elucidates the significance of functional CD4^(+) Tn and Tm subpopulations before the treatment in predicting the responses to anti-PD-1 treatment in Chinese NSCLC patients.The fact that there display distinct CD4^(+) T cell signatures in the prediction to anti-PD-1 and anti-PD-L1 monotherapy from our study provides preliminary evidence on the feasibility of anti-PD-1 and anti-PD-L1 combination therapy for advanced NSCLC patients.Liliang Xia Hui Wang Mingjiao Sun Yi Yang Chengcheng Yao Sheng He Huangqi Duan Weimin Xia Ruiming Sun Yaxian Yao Zhiwei Chen Qiong Zhao Hong Li Shun Lu Ying Wang 2021Science China(Life Sciences)2021,64,10:7

8Rupture process of the 2015 Pishan earthquake from joint inversion of InSAR, teleseismic data and GPS显示文摘An earthquake of Mw6.4 occurred in Pishan County in Xinjiang Province, northwestern Tibetan Plateau, on July 3,2015. The epicenter was located on an active blind thrust system located at the northern margin of the Western Kunlun Mountain Orogenic Belt southwest of the Tarim Basin. We constructed a shovel-shaped fault model based on the layered-crust model with reference to the seismic reflection profile, and obtained the rupture process of the earthquake from the joint inversion of Interferometric Synthetic Aperture Radar(InSAR) measurements, far-field waveform data, and Global Positioning System(GPS) data. The results show that the seismic fault dips southward with a strike of 109°, and the rupture direction was essentially northward. The fault plane rupture distribution is concentrated, with a maximum recorded slip of 73 cm. The main features of the fault are as follows: low inclination angle(25°–10°), thrust slip at a depth of 9–13 km, rupture propagation time of about 12 s, no significant slip in soft or hard sedimentary layers at 0–4 km depth and propagation from the initial rupture point to the surrounding area with no obvious directionality. The InSAR time-series analysis method is used to determine the deformation rate in the source region within 2 years after the earthquake, and the maximum value is ~17 mm yr-1 in the radar line-of-sight direction. Obvious post-earthquake deformation is evident in the hanging wall, with a similar trend to the coseismic displacement field. These results suggest that the Pishan earthquake has not completely released the accumulated energy of the region, given that the multilayer fold structure above the blind fault is still in a process of slow uplift since the earthquake. 9Cross-correlation function based multipath mitigation technique for cosine-BOC signals显示文摘We propose a new multipath mitigation technique based on cross-correlation function for the new cosine phased binary offset carrier(cosine-BOC) modulated signals, which will most likely be employed in both European Galileo system and Chinese Compass system. This technique is implemented to create an optimum cross-correlation function via designing the modulated symbols of the local signal. And the structure of the code tracking loop for cosine-BOC signals is quite simple including only two real correlators. 10Effects of Different Combinations of Red and Blue Light on Morphology and Photosynthetic Characteristic of Tomato Seedlings显示文摘Blue and red light are spectral wavelengths more effective for plants. The effects of different ratios of red and blue light( R/B = 2,R/B = 4,R/B = 8,R/B = 12) provided by LEDs on morphology and photosynthetic characteristics of tomato seedlings were studied. The results showed that plant height,stem diameter,fresh weight,dry weight,seedling index and G value increased with the increase of R/B ratio until 8. On the contrary,SPDA value decreased with the increase of R/B ratio. Photosynthetic characteristics were measured by CO_2 assimilation( Pn),stomatal conductance( gs) and intracellular CO_2 concentration( Ci). Pn and gs decreased with the increase of R/B ratio. 13Photothermal and pH dual-responsive self-healing coating for smart corrosion protection显示文摘A novel self-healing coating with photothermal and pH dual-responsive properties has been designed to protect carbon steel against corrosion by loading the stimuli-responsive microcapsules into a shape memory epoxy coating. The sandwich-like microcapsules were based on reduced graphene oxide/mesoporous silica(r GO@MS) assembled with a p H-responsive poly(N,N-dimethylaminoethyl methacrylate)(PDMAEMA) layer, and were loaded with benzotriazole(BTA) inhibitors(abbreviated as rGO@MS-P-BTA). Under near-infrared(NIR) light irradiation, the prominent photothermal effect of rGO could not only elevate the coating temperature to activate the shape memory effect and close the coating scratch, but also facilitate the release of corrosion inhibitors to suppress the corrosion activity. Moreover,the PDMAEMA as a p H-driven “gatekeeper” realized the controlled release of BTA from microcapsules at acid conditions. The surface morphology analysis, electrochemical impedance spectroscopy(EIS), and scanning electrochemical microscopy(SECM) were performed to evaluate the self-healing performance of the composite coatings. 19Junction plakoglobin regulates and destabilizes HIF2α to inhibit tumorigenesis of renal cell carcinoma显示文摘Background:Increased hypoxia-inducible factor 2α(HIF2α)activation is a common event in clear cell renal cell carcinoma(ccRCC)progression.However,the function and underlying mechanism of HIF2α in ccRCC remains uninvestigated.We conducted this study to access the potential link between junction plakoglobin(JUP)and HIF2αin ccRCC.Methods:Affinity purification and mass spectrometry(AP-MS)screening,glutathione-s-transferase(GST)pull-down and co-immunoprecipitation(Co-IP)assays were performed to detect the interacting proteins of HIF2α.Quantitative PCR(qPCR)and Western blotting were used to detect the expression of JUP in human ccRCC samples.Luciferase reporter assays,chromatin immunoprecipitation(ChIP),cycloheximide chase assays,and ubiquitination assays were conducted to explore the regulation of JUP on the activity of HIF2α.Cell Counting Kit-8(CCK-8)assays,colony formation assays,transwell assays,and xenograft tumor assays were performed to investigate the effect of JUP knockdown or overexpression on the tumorigenicity of renal cancer cells.Results:We identified JUP as a novel HIF2α-binding partner and revealed an important role of JUP in recruiting von Hippel-Lindau(VHL)and histone deacetylases 1/2(HDAC1/2)to HIF2α to regulate its stability and transactivation.JUP knockdown promoted and overexpression suppressed the tumorigenicity of renal cell carcinoma in vitro and in vivo.Importantly,the low expression of JUP was found in clinical ccRCC samples and correlated with enhanced hypoxia scores and poor treatment outcomes.Conclusion:Taken together,these data support a role of JUP in modulating HIF2α signaling during ccRCC progression and identify JUP as a potential therapeutic target.Ke Chen Jin Zeng Yi Sun Wei Ouyang Gan Yu Hui Zhou Yangjun Zhang Weimin Yao Wei Xiao Junhui Hu Jinchun Xing Kefeng Xiao Lily Wu Zhiqiang Chen Zhangqun Ye Hua Xu 2021Cancer Communications2021,41,4:1
20Regulation of the macrophage-related inflammatory microenvironment for atherosclerosis treatment and angiogenesis via anti-cytokine agents显示文摘Macrophages-mediated atherosclerosis(AS)is an inflammatory disease and the most common cause of ischemia.With the progress of basic and clinical research,anti-cytokine therapy has garnered considerable attention of the research community for the regulation of the inflammatory microenvironment for AS treatment.Despite of their promising potential,primary clinical trials have revealed that anti-cytokine drugs exhibit poor selectivity and thus affect other parts of the immune system,especially during long-term management.To circumvent these limitations,herein we exploited mesoporous silica nanoparticles(MSNs)with a pore size of 15.5 nm as carriers for the anti-interleukin-1β(anti-IL-1β)delivery to be the anti-cytokine agents.In vitro mechanistic studies indicated that the MSNs@anti-IL-1βcan regulate the macrophage-related inflammatory microenvironment,promote the viability of vascular endothelial cells(vECs),and reduce proliferation and phenotypic switching of vascular smooth muscle cells(vSMCs).In vivo evaluation further revealed that the MSNs@anti-IL-1βwere preferentially accumulated in macrophages,impeding the AS progress by maintaining the endothelium integrity and inhibiting the vSMCs proliferation.Besides,MSNs@antiIL-1βinduced neovascularization and improved hindlimb ischemia regeneration.Taken together,these MSNs affording the sustained release of anti-cytokine agents may have broad implications for the clinical management of the AS,including the reduction of the AS progression and alleviation of the ischemia.Hongji Pu Minghua Yao Zhaoyu Wu Zhijue Xu Chaoyi Cui Renhua Huang Muhammad Shafiq Weimin Li Xinwu Lu Bo Li 2022Nano Research2022,15,8:1
