| 1 | Exome sequencing confirms molecular diagnoses in 38 Chinese families with hereditary spherocytosis显示文摘Hereditary spherocytosis(HS), the most common cause of congenital hemolytic anemia, is caused by deficiency of the erythrocyte membrane proteins. Five causative genes(ANK1, SPTB, SPTA1, SLC4 A1, and EPB42) have been identified. To date,molecular genetic studies have been performed in different populations, including the American, European, Brazilian, Japanese and Korean populations, whereas only a few studies have been described in the Chinese population. Here, by reanalysis of the exome data, we revealed causative mutations and established a definitive diagnosis of HS in all 38 Chinese families. We found 34 novel mutations and four reported mutations in three known HS-causing genes—17 in ANK1, 17 in SPTB and four in SLC4 A1,suggesting that ANK1 and SPTB are the major genes in Chinese patients with HS. All of the ANK1 or SPTB mutations, scattered throughout the entire genes, are non-recurrent; and most of them are null mutations, which might cause HS via a haploinsufficiency mechanism. De novo mutations in ANK1 or SPTB often occur with an unexpected high frequency(87.5% and64.2%, respectively). | 3 | Mutation profile and its correlation with clinicopathology in Chinese hepatocellular carcinoma patients显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most common causes of cancer worldwide.Although many studies have focused on oncogene characteristics,the genomic landscape of Chinese HCC patients has not been fully clarified.Methods:A total of 165 HCC patients,including 146 males and 19 females,were enrolled.The median age was 55 years(range,27-78 years).Corresponding clinical and pathological information was collected for further analysis.A total of 168 tumor tissues from these patients were selected for next-generation sequencing(NGS)-based 450 panel gene sequencing.Genomic alterations including single nucleotide variations(SNV),short and long insertions and deletions(InDels),copy number variations,and gene rearrangements were analyzed.Tumor mutational burden(TMB)was measured by an algorithm developed in-house.The top quartile of HCC was classified as TMB high.Results:A total of 1,004 genomic alterations were detected from 258 genes in 168 HCC tissues.TMB values were identified in 160 HCC specimens,with a median TMB of 5.4 Muts/Mb(range,0-28.4 Muts/Mb)and a 75%TMB of 7.7 Muts/Mb.The most commonly mutated genes were TP53,TERT,CTNNB1,AXIN1,RB1,TSC2,CCND1,ARID1A,and FGF19.SNV was the most common mutation type and C:G>T:A and guanine transformation were the most common SNVs.Compared to wild-type patients,the proportion of Edmondson grade III-IV and microvascular invasion was significantly higher in TP53 mutated patients(P<0.05).The proportion of tumors invading the hepatic capsule was significantly higher in TERT mutated patients(P<0.05).The proportion of Edmondson grade I-II,alpha fetoprotein(AFP)<25μmg/L,and those without a history of hepatitis B was significantly higher in CTNNB1 mutated patients(P<0.05).CTNNB1 mutations were associated with TMB high in HCC patients(P<0.05).Based on correlation analysis,the mutation of TP53 was independently correlated with microvascular invasion(P=0.002,OR=3.096)and Edmondson grade III-IV(P=0.008,OR=2.613).The mutation of TERT was independently correlated with tumor invasion of the liver capsule(P=0.001,OR=3.030),and the mutation of CTNNB1 was independently correlated with AFP(<25μmg/L)(P=0.009,OR=3.414).Conclusions:The most frequently mutated genes of HCC patients in China were TP53,TERT,and CTNNB1,which mainly lead to the occurrence and development of HCC by regulating the P53 pathway,Wnt pathway,and telomere repair pathway.There were more patients with microvascular invasion and Edmondson III-IV grade in TP53 mutated patients and more patients with hepatic capsule invasion in TERT mutated patients,while in CTNNB1 mutated patients,there were more patients with Edmondson I-II grade,AFP<25μmg/L,and a non-hepatitis B background.Also,the TMB values were significantly higher in CTNNB1 mutated patients than in wild type patients. | Shuo Wang Huasheng Shi Tao Liu Manjiang Li Sanshun Zhou Xuan Qiu Zusen Wang Weiyu Hu Weidong Guo Xiaoqian Chen Honglin Guo Xiaoliang Shi Junping Shi Yunjin Zang Jingyu Cao Liqun Wu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,2: | 8 |
| 4 | Dynamics of ecosystem carbon stocks during vegetation restoration on the Loess Plateau of China显示文摘In the last few decades, the Loess Plateau had experienced an extensive vegetation restoration to reduce soil erosion and to improve the degraded ecosystems. However, the dynamics of ecosystem carbon stocks with vegetation restoration in this region are poorly understood. This study examined the changes of carbon stocks in mineral soil(0–100 cm), plant biomass and the ecosystem(plant and soil) following vegetation restoration with different models and ages. Our results indicated that cultivated land returned to native vegetation(natural restoration) or artificial forest increased ecosystem carbon sequestration. Tree plantation sequestered more carbon than natural vegetation succession over decades scale due to the rapid increase in biomass carbon pool. Restoration ages had different effects on the dynamics of biomass and soil carbon stocks. Biomass carbon stocks increased with vegetation restoration age, while the dynamics of soil carbon stocks were affected by sampling depth. Ecosystem carbon stocks consistently increased after tree plantation regardless of the soil depth; but an initial decrease and then increase trend was observed in natural restoration chronosequences with the soil sampling depth of 0–100 cm. | 11 | Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. | 13 | SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1显示文摘SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human malignancies.However,the biological role of SPDEF in head and neck squamous cell carcinoma(HNSCC)remains unclear.In this study,the expression level of SPDEF and its correlation with the clinical parameters of patients with HNSCC were determined using TCGA-HNSC,GSE65858,and our own clinical cohorts.CCK8,colony formation,cell cycle analysis,and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in HNSCC.ChIP-qPCR,dual luciferase reporter assay,and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in HNSCC.Compared with normal epithelial tissues,SPDEF was significantly downregulated in HNSCC tissues.Patients with HNSCC with low SPDEF mRNA levels exhibited poor clinical outcomes.Restoring SPDEF inhibited HNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest,while silencing SPDEF promoted cell proliferation in vitro.The xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates,smaller volumes,and lower weights.SPDEF could directly bind to the promoter region of NR4A1 and promoted its transcription,inducing the suppression of AKT,MAPK,and NF-κB signaling pathways.Moreover,silencing NR4A1 blocked the suppressive effect of SPDEF in HNSCC cells.Here,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in HNSCC.Our findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for HNSCC. | Yanting Wang Xianyue Ren Weiyu Li Ruoyan Cao Suyang Liu Laibo Jiang Bin Cheng Juan Xia | 2021 | International Journal of Oral Science2021,13,4: | 3 |

| 19 | Ultra-small hollow ternary alloy nanoparticles for efficient hydrogen evolution reaction显示文摘Hollow nanoparticles with large specific surface area and high atom utilization are promising catalysts for the hydrogen evolution reaction(HER). We describe herein the design and synthesis of a series of ultra-small hollow ternary alloy nanostructures using a simple one-pot strategy. The same technique was demonstrated for hollow PtNiCu nanoparticles, hollow PtCoCu nanoparticles and hollow CuNiCo nanoparticles. During synthesis, the displacement reaction and oxidative etching played important roles in the formation of hollow structures. Moreover, our hollow PtNiCu and PtCoCu nanoparticles were single crystalline, with an average diameter of 5 nm. Impressively, ultra-small hollow PtNiCu nanoparticles,containing only 10% Pt, exhibited greater electrocatalytic HER activity and stability than a commercial Pt/C catalyst. The overpotential of hollow Pt Ni Cu nanoparticles at 10 mA cm^(-2) was 28 mV versus reversible hydrogen electrode(RHE). The mass activity was 4.54 A mg Pt^(-1) at-70 mV versus RHE, which is 5.62-fold greater than that of a commercial Pt/C system(0.81 A mg Pt^(-1)). Through analyses of bonding and antibonding orbital filling, density functional theory calculations demonstrated that the bonding strength of different metals to the hydrogen intermediate(H*) was in the order of Pt > Co > Ni > Cu. The excellent HER performance of our hollow PtNiCu nanoparticles derives from moderately synergistic interactions between the three metals and H*. | 20 | The effects of two free-floating plants (Eichhornia crassipes and Pistia stratiotes) on the burrow morphology and water quality characteristics of pond loach (Misgurnus anguillicaudatus) habitat显示文摘Loach exhibit conspicuous drilling behaviors in the mud of shallow waters,yet their burrow morphology and the factors affecting this behavior have received little attention.We characterized the burrow morphology and water quality of the pond loach Misgurnus anguillicaudatus in three scenarios:in tanks without plants,tanks with the free-floating plant water hyacinth Eichhornia crassipes,and tanks with water lettuce Pistia stratiotes.Water hyacinth effectively removed water TN,COD,NO3-N and NH4-N,and water lettuce removed water TP and NH4-N.Water hyacinth and water lettuce markedly reduced water turbidity and DO,increased TOC and EC.Water hyacinth purified water more effectively than water lettuce,providing a suitable habitat for loach feeding,living and burrowing.The burrow structures were V-shaped,Y-shaped,inverted L-shaped,or complicated dendritic networks composed of multiple V shapes.The hyacinth treatment was characterized by the greatest burrow volume,length,depth,and structural complexity,but the opening size was reduced by dense root mat coverage.Burrows in the water lettuce treatment were characterized by intermediate volume,length,branches and sinuosity,but they had the largest opening and pit size.The control treatment had a flat bottom with the smallest,shortest burrows.This study indicates that free-floating plants improve habitat suitability and change burrow morphology and may be used to improve loach breeding methods。 | Jinqing Wang Guihua Fu Weiyue Li Ying Shi Jicai Pang Qiang Wang Weiguang Lü Change Liu Jiansheng Liu | 2018 | Aquaculture and Fisheries2018,3,1: | 2 |
