1A long-term Global LAnd Surface Satellite(GLASS)data-set for environmental studies显示文摘Recently,five Global LAnd Surface Satellite(GLASS)products have been released:leaf area index(LAI),shortwave broadband albedo,longwave broadband emissivity,incident short radiation,and photosynthetically active radiation(PAR).The first three products cover the years 19822012(LAI)and 19812010(albedo and emissivity)at 15 km and 8-day resolutions,and the last two radiation products span the period 20082010 at 5 km and 3-h resolutions.These products have been evaluated and validated,and the preliminary results indicate that they are of higher quality and accuracy than the existing products.In particular,the first three products have much longer time series,and are therefore highly suitable for various environmental studies.This paper outlines the algorithms,product characteristics,preliminary validation results,potential applications and some examples of initial analysis of these products.Shunlin Liang Xiang Zhao Suhong Liu Wenping Yuan Xiao Cheng Zhiqiang Xiao Xiaotong Zhang Qiang Liu Jie Cheng Hairong Tang Yonghua Qu Yancheng Bo Ying Qu Huazhong Ren Kai Yu John Townshend 2013International Journal of Digital Earth2013,6,S01:16
2Assessment of estrogen disrupting potency in animal foodstuffs of China by combined biological and chemical analyses显示文摘Food has been documented as one of major routes for human exposure to environmental estrogens(EEs), but information on the occurrence of EEs in animal foodstuffs is still scarce.This study analyzed estrogenic activity in 16 types of animal foodstuffs(n = 142) collected from four cities(Wuhan, Guangzhou, Wenzhou and Yantai) of China by combined yeast estrogen screen(YES) bioassay and chemical analysis. By bioassay, all samples' extracts were found to induce estrogenic activities and the bioassay-derived 17β-estradiol equivalent(EEQbio) ranged from 8.29 to 118.32 ng/g. In addition, the samples were analyzed by liquid chromatography coupled to tandem mass spectrometry for further chemical analysis. 17β-Estradiol was found in all samples in this survey at levels of 0.44 to 15.04 ng/g.All samples had 33.1% detection rate of 17α-ethinylestradiol(EE2), and the maximum concentration was 2.80 ng/g. Bisphenol A and 4-nonylphenols were detected in 83.8% and83.1% of samples, with concentrations up to 12.56 ng/g and 35.76 ng/g, respectively.However, the concentrations of estrone, diethylstilbestrol and 4-t-octylphenol were found to be below the limit of detection. A comparison of EEQbiomeasured from the YES assay and EEQchemcalculated from chemical analysis showed good correlation(R2= 0.84). 3Performance of BCC-CSM Models with Different Horizontal Resolutions in Simulating Extreme Climate Events in China显示文摘In this study,the performance of the Beijing Climate Center(BCC)Climate System Model version 1.1(BCCCSM1.1)(280-km resolution)and the BCC-CSM1.1 m(110-km resolution)in simulating extreme climate events over China in the last 40 years is compared.Both models capture the main spatial distribution features of heavy precipitation(R95T),the number of consecutive wet days(CWD),the annual count of days with precipitation≥1 mm(R1),the maximum consecutive 5-day precipitation(Rx5),and the numbers of frost days(FD)and summer days(SU).The BCC-CSM1.1 m has a better ability to simulate the detailed distribution of extreme climate events than the BCCCSM1.1,including R95T,CWD,R1,and the simple precipitation intensity index(SDII).However,the BCCCSM 1.1 m does not show an improvement in simulating the number of days with extreme precipitation(R90N),the number of consecutive dry days(CDD),the heat wave duration index(HWDI),the warm day frequency(TX90 P),and cold night frequency(TN10P).This indicates that the simulation of the R95T,CWD,R1,and SDII climate events is more sensitive to the resolution of the model.The improved BCC-CSM1.1 m is used to explore the projection of extreme climate change in China during the 21 st century under the RCP4.5(Representative Concentration Pathways)and RCP8.5 scenarios.The results show that extreme precipitation will increase dramatically over North and Southwest China in the late 21 st century.The CWD index will decrease on the Tibetan Plateau and in northeastern and central China and will increase in other parts of China;R1 will increase in northern China and decrease in southern China;Rx5 will increase dramatically in southern China;FD will decrease and SU will increase over China in the late 21 st century under both emission scenarios,with larger amplitudes in RCP8.5.Linxiao WEI Xiaoge XIN Chan XIAO Yonghua LI Yao WU Hongyu TANG 2019Journal of Meteorological Research2019,33,4:3
4Derivation and characterization of human embryonic stem cell lines from poor quality embryos显示文摘Poor quality embryos discarded from in vitro fertilization (IVF) laboratories are good sources for deriving human embryonic stem cell (hESC) lines. In this study, 166 poor quality embryos donated from IVF centers on day 3 were cultured in a blastocyst medium for 2 days, and 32 early blastocysts were further cultured in a blastocyst optimum culture medium for additional 2 days so that the inner cell masses (ICMs) could be identified and isolated easily. The ICMs of 17 blastocysts were isolated by a mechanical method, while those of the other 15 blastocysts were isolated by immunosurgery. All isolated ICMs were inoculated onto a feeder layer for subcultivation. The rates of ICM at- tachment, primary ICM colony formation and the efficiency of hESC derivation were similar between the ICMs isolated by the two methods (P>0.05). As a result, four new hESC lines were established. Three cell lines had normal karyotypes and one had an unbalanced Robertsonian translocation. All cell lines showed normal hESC characteristics and had the differentiation ability. In conclusion, we established a stable and effective method for hESC isolation and culture, and it was confirmed that the mechanical isolation was an effective method to isolate ICMs from poor embryos. 13SCGN deficiency is a risk factor for autism spectrum disorder显示文摘Autism spectrum disorder(ASD)affects 1-2%of all children and poses a great social and economic challenge for the globe.As a highly heterogeneous neurodevelopmental disorder,the development of its treatment is extremely challenging.Multiple pathways have been linked to the pathogenesis of ASD,including signaling involved in synaptic function,oxytocinergic activities,immune homeostasis,chromatin modifications,and mitochondrial functions.Here,we identify secretagogin(SCGN),a regulator of synaptic transmission,as a new risk gene for ASD.Two heterozygous loss-of-function mutations in SCGN are presented in ASD probands.Deletion of Scgn in zebrafish or mice leads to autism-like behaviors and impairs brain development.Mechanistically,Scgn deficiency disrupts the oxytocin signaling and abnormally activates inflammation in both animal models.Both ASD probands carrying Scgn mutations also show reduced oxytocin levels.Importantly,we demonstrate that the administration of oxytocin and anti-inflammatory drugs can attenuate ASD-associated defects caused by SCGN deficiency.Altogether,we identify a convergence between a potential autism genetic risk factor SCGN,and the pathological deregulation in oxytocinergic signaling and immune responses,providing potential treatment for ASD patients suffering from SCGN deficiency.Our study also indicates that it is critical to identify and stratify ASD patient populations based on their disease mechanisms,which could greatly enhance therapeutic success.Zhe Liu Shuai Tan Lianyu Zhou Li Chen Mingfeng Liu Wang Wang Yingying Tang Qin Yang Sensen Chi Peiyan Jiang Yue Zhang Yonghua Cui Junhong Qin Xiao Hu Shenglong Li Qi Liu Lu Chen Song Li Ezra Burstein Wei Li Xiaohu Zhang Xianming Mo Da Jia 2023Signal Transduction and Targeted Therapy2023,8,2:0
14Uncertainty analysis and visualization of geological subsurface and its application in metro station construction显示文摘To visualize and analyze the impact of uncertainty on the geological subsurface,on the term of the geological attribute probabilities(GAP),a vector parameters-based method is presented.Perturbing local data with error distribution,a GAP isosurface suite is first obtained by the Monte Carlo simulation.Several vector parameters including normal vector,curvatures and their entropy are used to measure uncertainties of the isosurface suite.The vector parameters except curvature and curvature entropy are visualized as line features by distributing them over their respective equivalent structure surfaces or concentrating on the initial surface.The curvature and curvature entropy presented with color map to reveal the geometrical variation on the perturbed zone.The multiple-dimensional scaling(MDS)method is used to map GAP isosurfaces to a set of points in lowdimensional space to obtain the total diversity among these equivalent probability surfaces.An example of a bedrock surface structure in a metro station shows that the presented method is applicable to quantitative description and visualization of uncertainties in geological subsurface.MDS plots shows differences of total diversity caused by different error distribution parameters or different distribution types.Weisheng HOU Qiaochu YANG Xiuwen CHEN Fan XIAO Yonghua CHEN 2021Frontiers of Earth Science2021,15,3:0
15Ionic liquid engineering enabled in-plane orientated 1D perovskite nanorods for efficient mixed-dimensional perovskite photovoltaics显示文摘Mixed-dimensional engineering of perovskite material has been demonstrated as a facile and promising strategy to improve both photovoltaic performance and long-term stability of perovskite solar cells(PSCs).In this study,we report an in-plane preferred orientation of 1D perovskite induced by an ionic liquid(IL)of 1-(3-cyanopropyl)-3-methylimidazolium chloride(CPMIMCl)for the first time via sequential deposition approach,leading to a mixed dimensional perovskite thin films.The generated one-dimensional(1D)CPMIMPbI3 with in-plane orientation resides at the grain boundaries of three-dimensional(3D)perovskite can be appreciably observed from the morphology level,leading to creation of high-quality films with large grain size with more efficient defect passivation.Moreover,the dispersion of IL in the bulk phase of perovskite material allows for the formation of 1D perovskite for multiple level passivation to inhibit non-radiative recombination and optimize carrier transport.This IL engineering strategy not only yields a mixed-dimensional perovskite heterostructure with in-plane orientation 1D perovskite nano-rods but also significantly improves the opto-electronic property with suppressed trap states.As a result,the CPMIMCl-treated PSCs show an enhanced photovoltaic performance with a champion power conversion efficiency(PCE)up to 24.13%.More importantly,benefiting from the hydrophobicity of formed 1D perovskite and defects suppression,the corresponding PSC demonstrates an excellent longterm stability and maintain 97.1%of its pristine PCE at 25C under 50%RH condition over 1000 h.This research provides an innovative perspective for employing the low dimensional engineering to optimize the performance and stability of photovoltaic devices.Fei Wang Dawei Duan Kang Zhou YZBXue Xiao Liang Xianfang Zhou Chuangye Ge Chao Zhou Jin Xiang Jiajie Zhu Quanyao Zhu Haoran Lin Yumeng Shi Yonghua Chen Gang Li Hanlin Hu 2023InfoMat2023,5,8:0
16Efficacy and mechanisms of Dingxian pill(定痫丸)combined with valproic acid on pentylenetetrazol-induced chronic epilepsy in rats显示文摘OBJECIVE:To investigate the efficacy and mechanisms of Dingxian pill(定痫丸)combined with valproic acid(VPA)on pentylenetetrazol-induced chronical epilepsy in rats.METHODS:A rat model of epilepsy was established by administering pentylenetetrazol(PTZ)water solution(35 mg/kg).Rats were divided into 4 groups,among which three groups were treated with different drugs once a day for 28 d including Dingxian pill(2.4 g/kg),VPA(0.2 g/kg),or a combination of Dingxian pill(2.4 g/kg)and VPA(0.2 g/kg)respectively,and the control group was given the same volume of saline.Rats in different groups were compared based on animal behavior,electroencephalograms,Morris water maze,immunohistochemistry,transcriptomics and real-time polymerase chain reaction.RSULTS:The combination therapy of Dingxian pill and VPA inhibited PTZ-induced seizure-like behavior and reduced seizure grades more significantly than VPA alone.Compared with the control group,the learning and memory ability of chronic PTZ-induced epileptic rats was improved in all the drug treatment groups,especially in the group that received both Dingxian pill and VPA.Similar to the results of MWM tests,expression of the neuroexcitability marker gene c-Fos was reduced after Dingxian pill and/or VPA treatment,and the effect was most pronounced in the combined treatment group.Transcriptomic analysis revealed that gene expression in the rodent hippocampus,which is involved in epilepsy,was upregulated by combined treatment with Dingxian pill and VPA,compared with VPA treatment alone.CONCLUSION:Our results not only highlight the antiepileptic effects of combined Dingxian pill and VPA treatment,but also shed light on the underlying molecular mechanisms and provide a way to apply Traditional Chinese Medicine in the treatment of epilepsy.QU Dongxiao GE Yiqin ZHUO Limin CHEN Liji XUE Yonghua CHENG Jiwei TAO Jie LI Guoyi ZHU Yudan XIAO Qian 2023Journal of Traditional Chinese Medicine2023,43,2:0
17NOGEA: A Network-oriented Gene Entropy Approach for Dissecting Disease Comorbidity and Drug Repositioning显示文摘Rapid development of high-throughput technologies has permitted the identification of an increasing number of disease-associated genes(DAGs),which are important for understanding disease initiation and developing precision therapeutics.However,DAGs often contain large amounts of redundant or false positive information,leading to difficulties in quantifying and prioritizing potential relationships between these DAGs and human diseases.In this study,a networkoriented gene entropy approach(NOGEA)is proposed for accurately inferring master genes that contribute to specific diseases by quantitatively calculating their perturbation abilities on directed disease-specific gene networks.In addition,we confirmed that the master genes identified by NOGEA have a high reliability for predicting disease-specific initiation events and progression risk.Master genes may also be used to extract the underlying information of different diseases,thus revealing mechanisms of disease comorbidity.More importantly,approved therapeutic targets are topologically localized in a small neighborhood of master genes in the interactome network,which provides a new way for predicting drug-disease associations.Through this method,11 old drugs were newly identified and predicted to be effective for treating pancreatic cancer and then validated by in vitro experiments.Collectively,the NOGEA was useful for identifying master genes that control disease initiation and co-occurrence,thus providing a valuable strategy for drug efficacy screening and repositioning.NOGEA codes are publicly available at https://github.com/guozihuaa/NOGEA.Zihu Guo Yingxue Fu Chao Huang Chunli Zheng Ziyin Wu Xuetong Chen Shuo Gao Yaohua Ma Mohamed Shahen Yan Li Pengfei Tu Jingbo Zhu Zhenzhong Wang Wei Xiao Yonghua Wang 2021Genomics, Proteomics & Bioinformatics2021,19,4:0

19Chromatin conformation of human oral epithelium can identify orofacial cleft missing functional variants显示文摘Genome-wide association studies(GWASs) are the most widely used method to identify genetic risk loci associated with orofacial clefts(OFC). However, despite the increasing size of cohort, GWASs are still insufficient to detect all the heritability,suggesting there are more associations under the current stringent statistical threshold. In this study, we obtained an integrated epigenomic dataset based on the chromatin conformation of a human oral epithelial cell line(HIOEC) using RNA-seq, ATAC-seq,H3K27ac Ch IP-seq, and DLO Hi-C. Presumably, this epigenomic dataset could reveal the missing functional variants located in the oral epithelial cell active enhancers/promoters along with their risk target genes, despite relatively less-stringent statistical association with OFC. Taken a non-syndromic cleft palate only(NSCPO) GWAS data of the Chinese Han population as an example, 3664 SNPs that cannot reach the strict significance threshold were subjected to this functional identification pipeline.In total, 254 potential risk SNPs residing in active cis-regulatory elements interacting with 1 718 promoters of oral epitheliumexpressed genes were screened. Gapped k-mer machine learning based on enhancers interacting with epithelium-expressed genes along with in vivo and in vitro reporter assays were employed as functional validation. Among all the potential SNPs, we chose and confirmed that the risk alleles of rs560789 and rs174570 reduced the epithelial-specific enhancer activity by preventing the binding of transcription factors related to epithelial development. 20Effects of Shenlian formula(参连方)on microbiota and inflammatory cytokines in adults with type 2 diabetes:a double-blind randomized clinical trial显示文摘OBJECTIVES:To observe the efficacy of Shenlian formula(SL formula,参连方),which consist of Huanglian(Rhizoma Coptidis)and Renshen(Radix Ginseng),in the treatment of type 2 diabetes mellitus(T2DM)and explore the effects on gut microbiota and serum inflammatory cytokines.METHODS:In a double-blind,randomized,placebocontrolled parallel-group clinical trial,31 adults with T2DM were randomly allocated to receive the SL formula or placebo for 12 weeks.Body mass index(BMI),blood lipid indices,glycemic biomarkers including glycated hemoglobin(HbA1C),fasting plasma glucose(FPG),postprandial blood glucose(PBG),fasting insulin levels(FIL),fasting C-peptide(C-P),homoeostasis model assessment for insulin resistance(HOMA-IR)and inflammatory cytokines were assessed at baseline and 12 weeks.The contents of gut microbiota were determined by pyrosequencing of the V3-V4 regions of 16S rRNA genes.RESULTS:Sixteen cases were allocated in the treatment group and 15 in the placebo group.Compared with the placebo,SL formula resulted in a higher significant reduction in PBG[(-1.318±0.772)vs(-0.008±1.404)mmol/L,P=0.003],BMI[(-0.611±0.524)vs(0.957±2.212)kg/m2,P=0.01],FIL[(-1.627±6.268)vs(3.976±6.85)μIU/mL,P=0.02],HOMA-IR[(-0.530±1.461)vs(1.511±2.288),P=0.006],and C-reactive protein(CRP)[(-1.307±0.684)vs(0.828±0.557)mg/L,P=0.04].In terms of gut microbiota,compared with the placebo,the SL formula resulted in a significant decrease in species richness and evenness.CONCLUSIONS:The SL formula showed the efficacy to improve postprandial blood glucose,insulin resistance,BMI and CRP levels.In addition,it could reduce the total number,richness and evenness of species,meanwhile increase the abundance of probiotics to modulate the structure of gut microbiota in patients with T2DM.However,further studies are required for exploring the deeper mechanism of TCM on gut microbiota.JIANG Li FU Qiang WANG Shidong ZHAO Jinxi CHEN Yu LI Jiayue XIAO Yonghua HUANG Weijun SUN Ruixi XIAO Yao SHEN Aijia WANG Junheng LIU Jiangteng FU Xiaozhe LI Yuanyuan ZHAO Yu XUE Taiqi 2023Journal of Traditional Chinese Medicine2023,43,4:0
