1A Plant Lectin Receptor-like Kinase Phosphorylates the Bacterial Effector AvrPtoB to Dampen Its Virulence in Arabidopsis显示文摘Plasma membrane-localized receptor-like kinases(RLKs)perceive conserved pathogen-associated molecular pattems(PAMPs)in plants,leading to PAMP-triggered immunity(PTI).TheArabidopsis thaliana lectin RLK LecRK-Ⅸ.2 has been shown to regulate the bacterial flagellin-derived peptide flg22-induced PTI.Here,we discover that Pseudomonas syringae effector AvrPtoB targets LecRK-Ⅸ.2 for degradation,which subsequently suppresses LecRK-Ⅸ.2-mediated PTI and disease resistance.However,LecRK-Ⅸ.2 can interact with and phosphorylate AvrPtoB at serine site 335(S335).AvrPtoB self-associates in vitro and in vivo,and the association appears to be essential for its E3 ligase activity in ubiquitinating substrate in plants.Phosphorylation of S335 disrupts the self-association and as a result,phosphomimetic AvrPtoBS335D cannot ubiquitinate LecRK-Ⅸ.2 efficiently,leading to the compromised virulence of AvrPtoB in suppressing PTI responses.fig22 enhances AvrPtoB S335 phosphorylation by inducing the expression and activating of LecRK-Ⅸ.2.Our study demonstrates that host RLKs can modify pathogen effectors to dampen their virulence and undermine their ability in suppressing PTI.Ning Xu Xuming Luo Wei Wu Yingying Xing Yingbo Liang Yanzhi Liu Huasong Zou Hai-Lei Wei Jun Liu 2020Molecular Plant2020,13,10:5
2Cloning and analysis of a cDNA encoding acetohydroxy acid isomeroreductase from G2 pea显示文摘Using cDNA representational difference analysis (cDNA RDA) method, we have successfully isolated a gene fragment whose expression was specifically induced by external GAa application. Screening a G2 pea cDNA library using this fragment as a probe, we obtained a 2036 bp full-length cDNA. It contains a 1746 bp open reading frame and encodes a protein of 581 amino acids with a theoretical molecular weight of 64 ku. It shares high-level sequenceidentity with AAIR genes from other plant species. 3SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX.Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He 2022Cellular & Molecular Immunology2022,19,1:4
4Microwave triggered multifunctional nanoplatform for targeted photothermal-chemotherapy in castration-resistant prostate cancer显示文摘Lacking a precise targeting strategy,castration-resistant prostate cancer(CRPC)is still hard to be treat effectively.Exploring treatment options that can accurately target CPRC is an important issue with urgent need.In this study,a novel nanotechnologybased strategy had been developed for the precise target treatment of CRPC.By combining microwaves and photothermal therapy(PTT),this nanoplatform,cmHSP70-PL-AuNC-DOX,targets tumor tissues with outstanding precision and achieves better anti-tumor activity by simultaneously eliciting photothermal and chemotherapeutic effects.From nanotechnology,cmHSP70-modified and thermo-sensitive liposome-coated AuNC-DOX were prepared and used for CRPC-targeted photothermal ablation and chemotherapy.Doxorubicin(DOX)was selected as the chemotherapeutic agent for cytotoxicity.In terms of the curative scheme,prostate tissues were firstly pre-treated with microwaves to induce the expression of heat shock protein 70(HSP70)and its migration to the cell membrane,which was then targeted by HSP70 antibody(cmHSP70)coated on the nanoparticles to achieve accurate drug delivery.The nanoplatform then achieved precise ablation and controlled release of DOX under external near-infrared(NIR)irradiation.Through the implementation,the targeting,cell killing,and safety of this therapeutical strategy had been verified in vivo and in vitro.This work establishes an accurate,controllable,efficient,non-invasive,and safe treatment platform for targeting CRPC,provides a rational design for CRPC’s PTT,and offers new prospects for nanomedicines with great precision.Pengyuan Liu Yajun Wu Xiaogang Xu Xudong Fan Chuan Sun Xiaojie Chen Jindan Xia Shiting Bai Li Qu Huasong Lu Jing Wu Jun Chen Ji-Gang Piao Zhibing Wu 2023Nano Research2023,16,7:2
