| 3 | The herbal compound geniposide rescues formaldehyde-induced apoptosis in N2a neuroblastoma cells显示文摘The herbal medicine Tong Luo Jiu Nao(TLJN)contains geniposide(GP)and ginsenoside Rg1 at a molar ratio of 10:1.Rg1 is the major component of another herbal medicine,panax notoginseng saponin(PNS).TLJN has been shown to strengthen brain function in humans,and in animals it improves learning and memory.We have previously shown that TLJN reduces amyloidogenic processing in Alzheimer’s disease(AD)mouse models.Together this suggests TLJN may be a potential treatment for patients with dementia.Because chronic damage of the central nervous system by formaldehyde(FA)has been presented as a risk factor for age-associated cognitive dysfunction,in the present study we investigated the protective effect of both TLJN and GP in neuron-like cells exposed to FA.FA-exposed murine N2a neuroblastoma cells were incubated with TLJN,its main ingredient GP,as well as PNS,to measure cell viability and morphology,the rate of apoptosis and expression of genes encoding Akt,FOXO3,Bcl2 and p53.The CCK-8 assay,cytoskeletal staining and flow cytometry were used to test cell viability,morphology and apoptosis,respectively.Fluorescent quantitative real-time PCR(qRT-PCR)was used to monitor changes in gene expression,and HPLC to determine the rate of FA clearance.Treatment of N2a cells with 0.09 mmol L?1 FA for 24 h significantly reduced cell viability,changed cell morphology and promoted apoptosis.Both TLJN and GP conferred neuroprotection to FA-treated N2a cells,whereas PNS,which had to be used at lower concentrations because of its toxicity,did not.Our data demonstrate that TLJN can rescue neuronal damage caused by FA and that its main ingredient,GP,has a major role in this efficacy.This presents purified GP as a drug or lead compound for the treatment of AD. | CHEN JinYan SUN MengRu WANG XingHua LU Jing WEI Yan TAN Yan LIU Ying GTZ Jürgen HE RongQiao HUA Qian | 2014 | Science China(Life Sciences)2014,57,4: | 15 |
| 4 | Near-surface sand-dust horizontal flux in Tazhong-the hinterland of the Taklimakan Desert显示文摘Tazhong is the hinterland and a sandstorm high-frequency area of the Taklimakan Desert. However, little is known about the detailed time-series of aeolian sand transport in this area. An experiment to study the sand-dust horizontal flux of near-surface was carried out in Tazhong from January to December 2009. By measuring the sand-dust horizontal flux throughout sixteen sand-dust weather processes with a 200-cm tall Big Spring Number Eight (BSNE) sampler tower, we quantitatively analyzed the vertical variation of the sand-dust horizontal flux. And the total sand-dust horizontal flux of different time-series that passed through a section of 100 cm in width and 200 cm in height was estimated combining the data of saltation movement continuously recorded by piezoelectric saltation sensors (Sensit). The results indicated that, in the surface layer ranging from 0-200 cm, the intensity of sand-dust horizontal flux decreased with the increase of the height, and the physical quantities obeyed power function well. The total sand-dust horizontal flux of the sixteen sand-dust weather processes that passed through a section of 100 cm in width and 200 cm in height was about 2,144.9 kg, the maximum of one sand-dust weather event was about 396.3 kg, and the annual total sand-dust horizontal flux was about 3,903.2 kg. The high levels of aeolian sand transport occurred during daytime, especially from 13:00 to 16:00 in the afternoon. | 7 | Influence of Al/Cu content on grain boundary diffusion in Nd-Fe-B magnet via in-situ observation显示文摘The influence of aluminum and copper content in the starting Nd-Fe-B magnet on grain boundary diffusion process(GBDP) was studied by observing the phase transformation behaviors of the magnets in-situ at high temperature. A higher coercivity increment is discovered in the sample with higher AI/Cu despite the fact that its Dy diffusion amount is the same as the other. DSC analysis shows an evident melting behavior in the higher Al/Cu sample. Laser scanning confocal microscopy(LSCM) in-situ characterization shows a large amount of melted intergranular phase spills out to the surface simultaneously at around 600 ℃ in the high Al/Cu sample, while the phase spills out gradually one after another in the range between 623 and680 ℃ in the other sample, which indicates that the intergranular phase can be more easily melted in the sample containing more AI/Cu. The area fraction of matrix phase remarkably shrinks while that of intergranular phase enlarges after LSCM heating, which demonstrates the outer region of the Nd_2 Fe_(14)B grains melt at the temperature of 900 ℃. Electron probe microanalyzer result(EPMA) shows that the Nd and Dy concentrate in edge regions and subsequently mix into the intergranular phase with the melting of the grain edge, while a large amount of AI and Cu in the intergranular phase spill out. Nevertheless, the sample with higher starting AI/Cu still remains higher residual contents after LSCM experiments, and that could probably be the main reason why the high AI/Cu magnet shows smaller coercivity decrement after LSCM experiment. | 10 | Screen-imaging guidance using a modified portable video macroscope for middle cerebral artery occlusion显示文摘The use of operating microscopes is limited by the focal length.Surgeons using these instruments cannot simultaneously view and access the surgical field and must choose one or the other.The longer focal length (more than 1 000 mm) of an operating telescope permits a position away from the operating field,above the surgeon and out of the field of view.This gives the telescope an advantage over an operating microscope.We developed a telescopic system using screen-imaging guidance and a modified portable video macroscope constructed from a Computar MLH-10 × macro lens,a DFK-21AU04 USB CCD Camera and a Dell laptop computer as monitor screen.This system was used to establish a middle cerebral artery occlusion model in rats.Results showed that magnification of the modified portable video macroscope was appropriate (5-20 ×) even though the Computar MLH-10 × macro lens was placed 800 mm away from the operating field rather than at the specified working distance of 152.4 mm with a zoom of 1-40 ×.The screen-imaging telescopic technique was clear,life-like,stereoscopic and matched the actual operation.Screen-imaging guidance led to an accurate,smooth,minimally invasive and comparatively easy surgical procedure.Success rate of the model establishment evaluated by neurological function using the modified neurological score system was 74.07%.There was no significant difference in model establishment time,sensorimotor deficit and infarct volume percentage.Our findings indicate that the telescopic lens is effective in the screen surgical operation mode referred to as 'long distance observation and short distance operation' and that screen-imaging guidance using an modified portable video macroscope can be utilized for the establishment of a middle cerebral artery occlusion model and micro-neurosurgery. | Xingbao Zhu Junli Luo Yun Liu Guolong Chen Song Liu Qiangjin Ruan Xunding Deng Dianchun Wang Quanshui Fan Xinghua Pan | 2012 | Neural Regeneration Research2012,7,12: | 6 |

| 20 | Radial Non-uniformity Index Research on High-density,High-flux CFB Riser with Stratified Injection显示文摘A high-density, high-flux circulating fluidized bed (CFB) riser (100 mm in ID and 10.614 m in height) was applied in a wide range of operating conditions (with solid fluxes up to 400 kg/m2s and superficial gas velocities up to 12 m/s) to examine its radial non-uniformity dynamics. The solids holdup was determined through the use of a fiber-optic probe at 11 axial levels. The results indicated that under all operating conditions, the high superficial gas velocity and low solid fluxes maintained a low radial non-uniformity index (RNI). The high-density/flux CFB riser had several unique characteristics, so that the peak of the radial solids holdup profile occurred at a position with r/R=0.8. The RNI and solids holdup at the cross-sectional position had a good logarithmic relationship at the low-density condition (with a mean solids holdup of <0.2), and the RNI decreased when the mean solids holdup exceeded 0.2. Investigation of the dynamics of stratified injection revealed that the feed ratio had an important effect on Gs and on solids holdup distribution. A novel '<' shaped axial solids holdup profile was found.
