1Development and Evaluation of a Loop-mediated Isothermal Amplification(LAMP) Assay for Rapid Detection of Chinese Giant Salamander Ranavirus显示文摘Loop-mediated isothermal amplification(LAMP) is a novel nucleic acid diagnostic method that can amplify rapidly a target template under isothermal conditions. In this study, a LAMP assay for rapid detection of Chinese giant salamander ranavirus(CGSRV) was developed from culture isolates and clinical samples. The LAMP assay was developed by designing one set of four specific primers, targeting the major capsid protein(MCP) gene of CGSRV. Reaction time and temperature were optimal for 40 min at 62°C. The developed LAMP assay is specific and highly sensitive for CGSRV detection, the detection limit could reach about 5 copies of cloned viral genomic fragments. The sensitivity of the LAMP assay was about 1000 and 10-fold higher than that of both conventional and nested PCR, respectively. The LAMP amplification produces a typical ladder-like pattern of products on an agarose gel that can be visually inspected after addition of ethidium bromide. 2Hypoxia response element-directed expression of bFGF in dental pulp stem cells improve the hypoxic environment by targeting pericytes in SCI rats显示文摘Cell-based transplantation strategies possess great potential for spinal cord injury(SCI)repair.Basic fibroblast growth factor(bFGF)has been reported to have multiple neuro-promoting effects on developing and adult nervous system of mammals and considered a promising therapy for nerve injury following SCI.Human dental pulp stem cells(DPSCs)are abundant stem cells with low immune rejection,which can be considered for cell replacement therapy.The purpose of this study was to investigate the roles of DPSCs which express bFGF under the regulation of five hypoxia-responsive elements(5HRE)using an adeno-associated virus(AAV-5HRE-bFGF-DPSCs)in SCI repairing model.In this study,DPSCs were revealed to differentiate into CD13^(+)pericytes and up-regulate N-cadherin expression to promote the re-attachment of CD13^(+)pericytes to vascular endothelial cells.The re-attachment of CD13^(+)pericytes to vascular endothelial cells subsequently increased the flow rate of blood in microvessels via the contraction of protuberance.As a result,increased numbers of red blood cells carried more oxygen to the damaged area and the local hypoxia microenvironment in SCI was improved.Thus,this study represents a step forward towards the potential use of AAV-5HRE-bFGF-DPSCs in SCI treatment in clinic.Sipin Zhu Yibo Ying Yan He Xingxing Zhong Jiahui Ye Zhiyang Huang Min Chen Qiuji Wu Yifan Zhang Ziyue Xiang Yurong Tu Weiyang Ying Jian Xiao Xiaokun Li Qingsong Ye Zhouguang Wang 2021Bioactive Materials2021,6,8:2
3Myocardial protection by heparin-based coacervate of FGF10显示文摘Heart disease is still the leading killer all around the world,and its incidence is expected to increase over the next decade.Previous reports have already shown the role of fibroblast growth factor10(FGF10)in alleviating heart diseases.However,FGF10 has not been used to treat heart diseases because the free protein has short half-life and low bioactivity.Here,an injectable coacervate was designed to protect growth factor from degradation during delivery and the effects of the FGF10 coacervate were studied using a mice acute myocardial infarction(MI)model.As shown in our echocardiographic results,a single injection of FGF10 coacervate effectively inhibited preserved cardiac contractibility and ventricular dilation when compared with free FGF10 and the saline treatment 6 weeks after MI.It is revealed in histological results that the MI induced myocardial inflammation and fibrosis was reduced after FGF10 coacervate treatment.Furthermore,FGF10 coacervate treatment could improve arterioles and capillaries stabilization through increasing the proliferation of endothelial and mural cells.However,with the same dosage,no statistically significant difference was shown between free FGF10,heparin+FGF10 and saline treatment,especially in long term.On another hand,FGF10 coacervate also increased the expression of cardiac-associated the mRNA(cTnT,Cx43 and α-SMA),angiogenic factors(Ang-1 and VEGFA)and decreased the level of inflammatory factor(tumor necrosis factor-α).The downstream signaling of the FGF10 was also investigated,with the western blot results showing that FGF10 coacervate activated the p-FGFR,PI3K/Akt and ERK1/2 pathways to a more proper level than free FGF10 or heparin+FGF10.In general,it is revealed in this research that one-time injection of FGF10 coacervate sufficiently attenuated MI induced injury when compared with an equal dose of free FGF10 or heparin+FGF10 injection.Zhouguang Wang Yan Huang Yan He Sinan Khor Xingxing Zhong Jian Xiao Qingsong Ye Xiaokun Li 2021Bioactive Materials2021,6,7:2
4Identification and characterization of novel fusion genes in prostate cancer by targeted RNA capture and next-generation sequencing显示文摘Gene fusions play critical roles in the development and progression of prostate cancer,and have been used as molecular biomarkers for diagnosis of the malignant disease.To further explore the novel fusions in prostate cancer,we performed targeted RNA capture and next-generation sequencing in a cohort of 52 prostate cancer patients,identified and validated 14 fusion events (7 types of fusion genes)in 12 cases,including three novel fusion genes.We characterized a chromosome rearrangement-induced trigenic KLK2-DGKB-ETV1 fusion,which may function as a noncoding RNA to upregulate the expression of the wild-type ETV1 protein in the tumor tissue. Additionally,we detected two novel fusion forms of HNRNPA2B1-ETV1 and SLC45A2-AMACR fusions,respectively.Interestingly,fusion events participated by kinase genes,which frequently occurred in other human cancers,were not present in these prostate cancer cases,suggesting discrepant gene fusion patterns in different cancers.These findings expand the genetic spectrum of prostate cancer and provide insight into diagnosis of this prevalent disease.Jie Yang Yun Chen Jingxiao Lu Xingxing Wang Lu Wang Jialong Liang Zhong Sheng Sun 2018Acta Biochimica et Biophysica Sinica2018,50,11:1
5Efficacy and safety of Sancai powder in patients with type 2 diabetes mellitus: a randomized controlled trial显示文摘OBJECTIVE: To assess the efficacy and safety of Sancai powder in patients with type 2 diabetes mellitus(T2DM) inadequately controlled with single oral metformin in a randomized controlled trial(RCT).METHODS: A total of 132 patients with T2 DM were enrolled in the study, who only took metformin(500-1000 mg/day) for at least three months and with inadequate glycemic control(7.0% ≤ hemoglobin A1 c ≤ 9.0%) in the past three months.The patients stopped taking metformin with lifestyle interventions for three weeks, and 105 patients qualified for the program. They were randomly divided into the Sancai powder group and the metformin group(1500 mg/day). The follow-up period was for 12 weeks. Comparisons of several variables were analyzed.RESULTS: No significant differences were found between the two groups in hemoglobin A1c(Hb A1c),fasting plasma glucose(FPG) and 2 h post-meal glucose(2h PG), although they had decreased significantly(P < 0.01). Homeostasis model assessment of beta cell function index was significantly improved in Sancai powder group(P < 0.01), and there were significant differences in the changes of homeostasis model assessment of insulin resistance and insulin sensitivity index in the two groups(P < 0.05). 6Multi-Omics-Guided Discovery of Omicsynins Produced by Streptomyces sp.1647:Pseudo-Tetrapeptides Active Against Influenza A Viruses and Coronavirus HCoV-229E显示文摘Many microorganisms have mechanisms that protect cells against attack from viruses.The fermentation components of Streptomyces sp.1647 exhibit potent anti-influenza A virus(IAV)activity.This strain was isolated from soil in southern China in the 1970s,but the chemical nature of its antiviral substance(s)has remained unknown until now.We used an integrated multi-omics strategy to identify the antiviral agents from this streptomycete.The antibiotics and Secondary Metabolite Analysis Shell(antiSMASH)analysis of its genome sequence revealed 38 biosynthetic gene clusters(BGCs)for secondary metabolites,and the target BGCs possibly responsible for the production of antiviral components were narrowed down to three BGCs by bioactivity-guided comparative transcriptomics analysis.Through bioinformatics analysis and genetic manipulation of the regulators and a biosynthetic gene,cluster 36 was identified as the BGC responsible for the biosynthesis of the antiviral compounds.Bioactivity-based molecular networking analysis of mass spectrometric data from different recombinant strains illustrated that the antiviral compounds were a class of structural analogues.Finally,18 pseudo-tetrapeptides with an internal ureido linkage,omicsynins A1–A6,B1–B6,and C1–C6,were identified and/or isolated from fermentation broth.Among them,11 compounds(omicsynins A1,A2,A6,B1–B3,B5,B6,C1,C2,and C6)are new compounds.Omicsynins B1–B4 exhibited potent antiviral activity against IAV with the 50%inhibitory concentration(IC_(50))of approximately 1μmol·L^(-1)and a selectivity index(SI)ranging from 100 to 300.Omicsynins B1–B4 also showed significant antiviral activity against human coronavirus HCoV-229E.By integrating multi-omics data,we discovered a number of novel antiviral pseudo-tetrapeptides produced by Streptomyces sp.1647,indicating that the secondary metabolites of microorganisms are a valuable source of novel antivirals.Hongmin Sun Xingxing Li Minghua Chen Ming Zhong Yihua Li Kun Wang Yu Du Xin Zhen Rongmei Gao Yexiang Wu Yuanyuan Shi Liyan Yu Yongsheng Che Yuhuan Li Jian-Dong Jiang Bin Hong Shuyi Si 2022Engineering2022,,9:0
7Defect-rich and highly porous carbon nanosheets derived from Ti_(3)AlC_(2) MAX with good lithium storage properties显示文摘Defect-rich,highly porous two-dimensional carbon nanosheets(CNS) have attracted tremendous research interests in catalysis and environmental purification and other fields,because of their unique micromorphology,chemical stability and high specific surface area.Herein,in this work,we report a new solution to synthesize an ultrathin two-dimensional CNS with rich defects and abundant pores via two-step etching the Ti_(3)AlC_(2)with the help of I2and NaOH.The CNS thickness,specific surface area and pore volume could be all tunable by adding the amount of I2.And the highest specific surface area and pore volume of the synthesized 2D CNS can be achieved 1134.4 m^(2)/g and 0.80 cm^(3)/g,with a thickness of only 0.64 nm and a yield of 35.9%.When employed as the anodes for lithium-ion batteries,the synthesized CNS anodes exhibit good cycling and rate capabilities.This work provides a novel and facile strategy for synthesizing highly porous and defective 2D carbon materials with good lithium storage properties.Zhihao Zhu Peng Liu Yucheng Liu Xingxing Gu Zhi Su Zhong Su Chao Lai 2023Chinese Chemical Letters2023,34,2:0
