| 1 | The Serum Exosome Derived MicroRNA-135a,-193b, and-384 Were Potential Alzheimer's Disease Biomarkers显示文摘Objective MicroR NAs(mi Rs) are attractive molecules to be considered as one of the blood-based biomarkers for neurodegenerative disorders such as Alzheimer's disease(AD). The goal of this study was to explore their potential value as biomarkers for the diagnosis of AD. Methods The expression levels of exosomal miR-135 a,-193 b, and-384 in the serum from mild cognitive impairment(MCI), dementia of Alzheimer-type(DAT), Parkinson's disease with dementia(PDD), and vascular dementia(VaD) patients were measured with a real-time quantitative reverse transcriptase PCR(qR T-PCR) method. Results Both serum exosome mi R-135 a and miR-384 were up-regulated while miR-193 b was down-regulated in serum of AD patients compared with that of normal controls. Exosome miR-384 was the best among the three miR s to discriminate AD, VaD, and PDD. Using the cut-off value could better interpret these laboratory test results than reference intervals in the AD diagnosis. ROC curve showed that the combination of mi R-135 a,-193 b, and-384 was proved to be better than a particular one for early AD diagnosis. | 4 | Association of Dietary Pattern during Pregnancy and Gestational Diabetes Mellitus: A Prospective Cohort Study in Northern China显示文摘Objective To examine the association of maternal dietary patterns during pregnancy with gestational diabetes mellitus(GDM) in northern China.Methods The dietary intakes of pregnant women were recorded twice by 24‐hour dietary recalls for three days prior to having been diagnosed with GDM, at 5‐15 and 24‐28 gestational weeks, respectively.GDM was diagnosed, and serum glycosylated hemoglobin(HbA1c) was measured at 24‐28 weeks.Dietary patterns were assessed by factor analysis. The association of the dietary pattern with GDM and HbA1c was examined by multiple logistic models.Results Of 753 participants, 64(8.5%) were diagnosed with GDM. Four dietary patterns were identified: Western pattern(dairy, baked/fried food and white meat), traditional pattern(light‐colored vegetables, fine grain, red meat and tubers), mixed pattern(edible fungi, shrimp/shellfish and red meat)and prudent pattern(dark‐colored vegetables and deep‐sea fish). Compared with the prudent pattern,both the Western pattern and the traditional pattern were associated with an increased risk of GDM(a OR = 4.40, 95% CI: 1.58‐12.22; a OR = 4.88, 95% CI: 1.79‐13.32) and a high level of HbA1c(a OR = 12.37,95% CI: 1.47‐103.91; a OR = 26.23, 95% CI: 2.54‐270.74). | 6 | Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-κB signaling pathway显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory disease that eventually leads to disability.Inflammatory cell infiltration,severe joint breaking and systemic bone loss are the main clinical symptoms.In this study,we established a collagen-induced arthritis(CIA)model and found a large number of M1 macrophages and pyroptosis,which are important sources of proinflammatory cytokines.Punicalagin(PUN)is an active substance extracted from pomegranate peel.We found that it inhibited joint inflammation,cartilage damage and systemic bone destruction in CIA mice.PUN effectively alleviated the high expression of inflammatory cytokines in synovial tissue in vivo.PUN treatment shifted macrophages from the M1 phenotype to the M2 phenotype after stimulation with lipopolysaccharide(LPS)and interferon(IFN)-γ.The expression of inducible nitric oxide synthase(i NOS)and other proinflammatory cytokines released by M1 macrophages was decreased in the PUN treatment group.However,simultaneously,the expression of markers of anti-inflammatory M2 macrophages,such as arginase(Arg)-1 and interleukin(IL)-10,was increased.In addition,PUN treatment attenuated pyroptosis by downregulating the expression of NLRP3 and caspase-1,thereby preventing inflammatory cell death resulting from the release of IL-1βand IL-18.Mechanistically,PUN inhibited the activation of receptor activators of the nuclear factor-κB(NF-κB)signaling pathway,which contributes to M1 polarization and pyroptosis of macrophages.We concluded that PUN ameliorated pathological inflammation by inhibiting M1 phenotype polarization and pyroptosis and has great potential as a therapeutic treatment for human RA. | Gaoran Ge Jiaxiang Bai Qing Wang Xiaolong Liang Huaqiang Tao Hao Chen Minggang Wei Junjie Niu Huilin Yang Yaozeng Xu Yuefeng Hao Yi Xue Dechun Geng | 2022 | Science China(Life Sciences)2022，65，3： | 14 |
| 7 | Interpretation Criteria for Standardized Western Blot for the Predominant Species of Borrelia Burgdorferi Sensu Lato in China显示文摘Objective Western blotting (WB;immunoblotting) is a widely used tool for the serodiagnosis of Lyme borreliosis (LB),but so far,no generally accepted criteria for its performance and interpretation have been established in China.The present study was designed to determine the criteria for standardized Western blot for the predominant species of Borrelia burgdorferi sensu lato in China,in which WB was produced with strain PD91 as the representative strain attributed to predominant genospecies Borrelia garinii of Borrelia burgdorferi sensu lato.Methods Approximately 13 bands between 14 and 100 kD were differentiated for strain PD91 by using Gel-Pro analysis software.In a study with 631 serum samples (taken from 127 patients with Lyme borreliosis and 504 controls),all observed bands were documented.To establish criteria for a positive WB result for strain PD91,receiver operating characteristic (ROC) curves were used.Results The following interpretation criteria were recommended:for IgG,at least one band of P83/100,P58,P39,P30,OspC,P17,P66,and OspA;for IgM,at least one band of P83/100,P58,OspA,P30,OspC,P17 or P41.In addition,syphilis,leptospirosis and other related diseases should be excluded when the positive band is P41 in IgM.For IgG criteria,the sensitivity is 73.2%,the specificity is 99.4% and Youden index is 0.726;for IgM criteria,the sensitivity is 50.6%,the specificity is 93.1% and Youden index is 0.437.Conclusion Standardization of WB assays is necessary for comparison of results from different laboratories.Moreover,the criteria of other genospecies of Borrelia burgdorferi sensu lato should be determined in the future to complete the criteria of WB for the diagnosis of the Lyme disease in China. | YI JIANG XUE-XIA HOU ZHEN GENG QIN HAO KANG-LIN WAN | 2010 | Biomedical and Environmental Sciences2010，23，5： | 13 |

| 15 | Area and volume ratios for prediction of visual outcome in idiopathic macular hole显示文摘AIM:To predict the visual outcome in patients undergoing macular hole surgery by two novel three-dimensional morphological parameters on optical coherence tomography(OCT): area ratio factor(ARF) and volume ratio factor(VRF).METHODS: A clinical case series was conducted,including 54 eyes of 54 patients with an idiopathic macular hole(IMH). Each patient had an OCT examination before and after surgery. Morphological parameters of the macular hole,such as minimum diameter,base diameter,and height were measured. Then,the macular hole index(MHI),tractional hole index(THI),and hole form factor(HFF) were calculated. Meanwhile,novel postoperative macular hole(MH) factors,ARF and VRF were calculated by three-dimensional morphology. Bivariate correlations were performed to acquire asymptotic significance values between the steady best corrected visual acuity(BCVA) after surgery and 2D/3D arguments of MH by the Pearson method with two-tailed test. All significant factors were analyzed by the receiver operating characteristic(ROC) curve analysis of SPSS software which were responsible for vision recovery. ROC curves analyses were performed to further discuss the different parameters on the prediction of visual outcome.RESULTS: The mean and standard deviation values of patients' age,symptoms duration,and follow-up time were 64.8±8.9y(range: 28-81),18.6±11.5d(range: 2-60),and 11.4±0.4mo(range: 6-24),respectively. Steady-postBCVA analyzed with bivariate correlations was found to be significantly correlated with base diameter(r=0.521,P<0.001),minimum diameter(r=0.514,P<0.001),MHI(r=-0.531,P<0.001),THI(r=-0.386,P=0.004),HFF(r=-0.508,P<0.001),and ARF(r=-0.532,P<0.001). Other characteristic parameters such as age,duration of surgery,height,diameter hole index,and VRF were not statistically significant with steady-post-BCVA. According to area under the curve(AUC) values,values of ARF,MHI,HFF,minimum diameter,THI,and base diameter are 0.806,0.772,0.750,0.705,0.690,and 0.686,respectively. However,Steady-post-BCVA analysis with bivariate correlations for VRF was no statistical significance. Results of ROC curve analysis indicated that the MHI value,HFF,and ARF was greater than 0.427,1.027 and 1.558 respectively which could correlate with better visual acuity.CONCLUSION: Compared with MHI and HFF,ARF could effectively express three-dimensional characteristics of macular hole and achieve better sensitivity and specificity. | 16 | A Study of the Technique of Western Blot for Diagnosis of Lyme Disease caused by Borrelia afzelii in China显示文摘Objective To study the technique of Western blot for the diagnosis of Lyme disease caused by Borrelia afzelii in China and to establish the standard criteria by operational procedure. Methods FP1, which is the representative strain of B. afzelii in China, was analyzed by SDS‐PAGE, electro transfer and immunoblotting assays. The molecular weights of the protein bands of FP1 were analyzed by Gel‐Pro analysis software. In a study using 451 serum samples (159 patients with Lyme disease and 292 controls), all observed bands were recorded. The accuracy of the WB as a diagnostic test was established by using the ROC curve and Youden index. Results Criteria for a positive diagnosis of Lyme disease were established as at least one band of P83/100, P58, P39, OspB, OspA, P30, P28, OspC, P17, and P14 in the IgG test and at least one band of P83/100, P58, P39, OspA, P30, P28, OspC, P17, and P41 in the IgM test. For IgG criteria, the sensitivity, specificity and Youden index were 69.8%, 98.3%, and 0.681, respectively; for IgM criteria, the sensitivity, specificity and Youden index were 47%, 94.2%, and 0.412, respectively. | 19 | Prediction of 10-year Atherosclerotic Cardiovascular Disease Risk among Adults Aged 40-79 Years in China:a Nationally Representative Survey显示文摘Objective To establish the distribution of 10-year atherosclerotic cardiovascular disease(ASCVD) risk among Chinese adults.Methods We estimated the 10-year ASCVD risk by applying the 2013 American College of Cardiology/American Heart Association pooled cohort equations(PCEs) to the data obtained from the 2010 China Chronic Disease and Risk Factor Surveillance that involved 61,541 participants(representing 520,158,652 Chinese adults) aged 40-79 years.We also compared the ASCVD risk with the 10-year ischemic cardiovascular disease(ICVD) risk,which was calculated using the simplified scoring tables recommended by the Chinese Guidelines for Prevention of Cardiovascular Diseases(Chinese model).Results Based on the PCEs,the average 10-year ASCVD risk among adults without self-reported stroke or myocardial infraction was 12.5%.Approximately 247 million(47.4%) and 107 million(20.6%) adults had ≥ 7.5% and > 20% 10-year ASCVD risks,respectively.The 10-year ASCVD risk > 20% was higher among men,less educated individuals,smokers,drinkers,and physically inactive individuals than among their counterparts.Overall,29.0% of adults categorized using the Chinese model were overclassified with the PCEs.Conclusion Our results define the distribution of 10-year ASCVD risk among Chinese adults.The 10-year ASCVD risk predicted by the PCEs was higher than the ICVD risk predicted by the Chinese model. | ZHANG Mei JIANG Yong WANG Li Min LI Yi Chong HUANG Zheng Jing LI Jian Hong ZHOU Mai Geng ZHAO Wen Hua 无 | 2017 | Biomedical and Environmental Sciences2017，30，4： | 7 |
