1SUMOylation-modified Pelota-Hbs1 RNA surveillance complex restricts the infection of potyvirids in plants显示文摘RNA quality control nonsense-mediated decay is involved in viral restriction in both plants and animals.However,it is not known whether two other RNA quality control pathways,nonstop decay and no-go decay,are capable of restricting viruses in plants.Here,we show that the evolutionarily conserved Pelota–Hbs1 complex negatively regulates infection of plant viruses in the family Potyviridae(termed potyvirids),the largest group of plant RNA viruses that accounts for more than half of the viral crop damage worldwide.Pelota enables the recognition of the functional G1-2A6-7 motif in the P3 cistron,which is conserved in almost all potyvirids.This allows Pelota to target the virus and act as a viral restriction factor.Furthermore,Pelota interacts with the SUMO E2-conjugating enzyme SCE1 and is SUMOylated in planta.Blocking Pelota SUMOylation disrupts the ability to recruit Hbs1 and inhibits viral RNA degradation.These findings reveal the functional importance of Pelota SUMOylation during the infection of potyvirids in plants.Linhao Ge Buwei Cao Rui Qiao Hongguang Cui Shaofang Li Hongying Shan Pan Gong Mingzhen Zhang Hao Li Aiming Wang Xueping Zhou Fangfang Li 2023Molecular Plant2023,16,3:2

4Intercellular transfer of activated STING triggered by RAB22A-mediated non-canonical autophagy promotes antitumor immunity显示文摘STING,an endoplasmic reticulum(ER)transmembrane protein,mediates innate immune activation upon cGAMP stimulation and is degraded through autophagy.Here,we report that activated STING could be transferred between cells to promote antitumor immunity,a process triggered by RAB22A-mediated non-canonical autophagy.Mechanistically,RAB22A engages PI4K2A to generate PI4P that recruits the Atg12–Atg5–Atg16L1 complex,inducing the formation of ER-derived RAB22A-mediated non-canonical autophagosome,in which STING activated by agonists or chemoradiotherapy is packaged.This RAB22A-induced autophagosome fuses with RAB22A-positive early endosome,generating a new organelle that we name Rafeesome(RAB22A-mediated non-canonical autophagosome fused with early endosome).Meanwhile,RAB22A inactivates RAB7 to suppress the fusion of Rafeesome with lysosome,thereby enabling the secretion of the inner vesicle of the autophagosome bearing activated STING as a new type of extracellular vesicle that we define as R-EV(RAB22A-induced extracellular vesicle).Activated STING-containing R-EVs induce IFNβrelease from recipient cells to the tumor microenvironment,promoting antitumor immunity.Consistently,RAB22A enhances the antitumor effect of the STING agonist diABZI in mice,and a high RAB22A level predicts good survival in nasopharyngeal cancer patients treated with chemoradiotherapy.Our findings reveal that Rafeesome regulates the intercellular transfer of activated STING to trigger and spread antitumor immunity,and that the inner vesicle of non-canonical autophagosome originated from ER is secreted as R-EV,providing a new perspective for understanding the intercellular communication of organelle membrane proteins.Ying Gao Xueping Zheng Boyang Chang Yujie Lin Xiaodan Huang Wen Wang Shirong Ding Weixiang Zhan Shang Wang Beibei Xiao Lanqing Huo Youhui Yu Yilin Chen Run Gong Yuanzhong Wu Ruhua Zhang Li Zhong Xin Wang Qiuyan Chen Song Gao Zhengfan Jiang Denghui Wei Tiebang Kang 2022Cell Research2022,32,12:1
5Plant and animal positive-sense single-stranded RNA viruses encode small proteins important for viral infection in their negative-sense strand显示文摘Positive-sense single-stranded RNA(+ssRNA)viruses,the most abundant viruses of eukaryotes in nature,require the synthesis of negative-sense RNA(-RNA)using their genomic(positive-sense)RNA(+RNA)as a template for replication.Based on current evidence,viral proteins are translated via viral+RNAs,whereas-RNA is considered to be a viral replication intermediate without coding capacity.Here,we report that plant and animal+ssRNA viruses contain small open reading frames(ORFs)in their-RNA(reverse ORFs[rORFs]).Using turnip mosaic virus(TuMV)as a model for plant+ssRNA viruses,we demonstrate that small proteins encoded by rORFs display specific subcellularlocalizations,and confirm the presence of rORF2 in infected cells through mass spectrometry analysis.The protein encoded by TuMV rORF2 forms punctuate granules that are localized in the perinuclear region and co-localized with viral replication complexes.The rORF2 protein can directly interact with the viral RNA-dependent RNA polymerase,and mutation of rORF2 completely abolishes virus infection,whereas ectopic expression of rORF2 rescues the mutant virus.Furthermore,we show that several rORFs in the-RNA of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)have the ability to suppress type l interferon production and facilitate the infection of ve-sicular stomatitis virus.In addition,we provide evidence that TuMV might utilize internal ribosome entry sites to translate these small rORFs.Taken together,these findings indicate that the-RNA of+ssRNA vi-ruses can also have the coding capacity and that small proteins encoded therein play critical roles in viral infection,revealing a viral proteome larger than previously thought.Pan Gong Qingtang Shen Mingzhen Zhang Rui Qiao Jing Jiang Lili Su Siwen Zhao Shuai Fu Yu Ma Linhao Ge Yaqin Wang Rosa Lozano-Durán Aiming Wang Fangfang Li Xueping Zhou 2023Molecular Plant2023,16,11:0

7Interplay of transcription factors and microRNAs during embryonic hematopoiesis显示文摘Hematopoietic stem cells (HSCs), which are localized in the bone marrow of adult mammals, come from hematopoietic endothelium during embryonic stages. Although the basic processes of HSC generation and differentiation have been described in the past, the epigenetic regulation of embryonic hematopoiesis remains to be fully described. Here, by utilizing an in vitro differentiation system of mouse embryonic stem cells (ESCs), we identified more than 20 microRNAs that were highly enriched in embryonic hematopoietic cells, including some (e.g. miR-10b, miR-15b, and miR-27a) with previously unknown functions in blood formation. Luciferase and gene expression assays further revealed combinational binding and regulation of these microRNAs by key transcription factors in blood cells. 9The novel C5 protein from tomato yellow leaf curl virus is a virulence factor and suppressor of gene silencing显示文摘Tomato yellow leaf curl virus(TYLCV)is known to encode 6 canonical viral proteins.Our recent study revealed that TYLCV also encodes some additional small proteins with potential virulence functions.The fifth ORF of TYLCV in the complementary sense,which we name C5,is evolutionarily conserved,but little is known about its expression and function during viral infection.Here,we confirmed the expression of the TYLCV C5 by analyzing the promoter activity of its upstream sequences and by detecting the C5 protein in infected cells by using a specific custom-made antibody.Ectopic expression of C5 using a potato virus X(PVX)vector resulted in severe mosaic symptoms and higher virus accumulation levels followed by a burst of reactive oxygen species(ROS)in Nicotiana benthamiana plants.C5 was able to effectively suppress local and systemic post-transcriptional gene silencing(PTGS)induced by single-stranded GFP but not double-stranded GFP,and reversed the transcriptional gene silencing(TGS)of GFP.Furthermore,the mutation of C5 in TYLCV inhibited viral replication and the development of disease symptoms in infected plants.Transgenic overexpression of C5 could complement the virulence of a TYLCV infectious clone encoding a dysfunctional C5.Collectively,this study reveals that TYLCV C5 is a pathogenicity determinant and RNA silencing suppressor,hence expanding our knowledge of the functional repertoire of the TYLCV proteome.Siwen Zhao Pan Gong Yanxiang Ren Hui Liu Hao Li Fangfang Li Xueping Zhou 2022Stress Biology2022,2,1:0
10Sonochemical synthesis and characterization of magnetic separable Fe_(3)O_(4)-TiO_(2)nanocomposites and their catalytic properties显示文摘A novel sonochemical method is described for the preparation of Fe_(3)O_(4)–TiO_(2)photocatalysts in which nanocrystalline titanium dioxide particles are directly coated onto a magnetic core.The Fe_(3)O_(4)nanoparticles were partially embedded in TiO_(2)agglomerates.TiO_(2)nanocrystallites were obtained by hydrolysis and condensation of titanium tetraisopropyl in the presence of ethanol and water under high-intensity ultrasound irradiation.This method is attractive since it eliminated the high-temperature heat treatment required in the conventional sol–gel method,which is important in transforming amorphous titanium dioxide into a photoactive crystalline phase.In comparison to other methods,the developed method is simple,mild,green and efficient.The magnetization hysteresis loop for Fe_(3)O_(4)–TiO_(2)nanocomposites indicates that the hybrid catalyst shows superparamagnetic characteristics at room temperature.Photocatalytic activity studies confirmed that the as-prepared nanocomposites have high photocatalytic ability toward the photodegradation of RhB solution.Furthermore,the photodecomposition rate decreases only slightly after six cycles of the photocatalysis experiment.Thus,these Fe_(3)O_(4)–TiO_(2)nanocomposites can be served as an effective and conveniently recyclable photocatalyst.Wanquan Jiang Xueping Zhang Xinglong Gong Fangfang Yan Zhong Zhang 2010International Journal of Smart and Nano Materials2010,1,4:0
