| 1 | IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. | 2 | Effects of different litters on ammonia emissions from chicken manure显示文摘Experiments were conducted to investigate the influences of type of litter,initial moisture content(IMC)of litter,and dry weight ratio of manure to litter(DWRML)on ammonia emissions from chicken manure and the effects of pH values of tea leaves and the mixtures of tea leaves and other litter on the ammonia emissions from chicken manure.For the experiments,four kinds of litter,Northeast pine sawdust(sawdust),rice husk,tea leaves,and wheat straw,were selected.The IMCs of the litter were(20±2)%,(30±2)%,and(40±2)%;and the DWRML values were 1:4,1:6 and 1:8,respectively.The different litters adjusted at different moisture contents were mixed with chicken manure in different DWRML and then placed in different static test chambers,which were real-time monitored the ammonia concentrations.Pure chicken manure without any litter was used as a control group.The four kinds of litter had obvious inhibitory effects on the ammonia emissions from chicken manure under various conditions.There were significant differences among four kinds of litter(p<0.01).Under the same conditions,the best inhibitory effect was achieved by using tea leaves,followed by straw,rice husk,and sawdust.The IMC of litter had no significant effects on the ammonia inhibition(p>0.05).The DWRML had no significant effects on ammonia emission inhibition for tea leaves(p>0.05),but had a significant effects on the ammonia emission inhibition for the other three kinds of litter(p<0.05).The pH value of tea leaves had no significant effects on the inhibition of ammonia emissions(p>0.05).The mixed litter made of tea leaves and sawdust,rice husk,or straw were significantly better than the tea leaves and other single litter(p<0.01).It indicated that adding appropriate amount of tea leaves in the litter can effectively inhibit ammonia emissions from chicken manure. | Hequn Tan Meng Li Dengfei Jie Yafang Zhou Xin'an Li | 2019 | International Journal of Agricultural and Biological Engineering2019,12,4: | 2 |

| 8 | Anti-PD-L1 antibody enhances curative effect of cryoablation via antibody-dependent cell-mediated cytotoxicity mediating PD-L1^(high)CD11b^(+)cells elimination in hepatocellular carcinoma显示文摘Cryoablation(CRA)and microwave ablation(MWA)are two main local treatments for hepatocellular carcinoma(HCC).However,which one is more curative and suitable for combining with immunotherapy is still controversial.Herein,CRA induced higher tumoral PD-L1 expression and more T cells infiltration,but less PD-L1^(high)CD11b^(+)myeloid cells infiltration than MWA in HCC.Furthermore,CRA had better curative effect than MWA for anti-PD-L1 combination therapy in mouse models.Mechanistically,anti-PD-L1 antibody facilitated infiltration of CD8^(+)T cells by enhancing the secretion of CXCL9 from cDC1 cells after CRA therapy.On the other hand,anti-PD-L1 antibody promoted the infiltration of NK cells to eliminate PD-L1^(high)CD11b^(+)myeloid cells by antibody-dependent cell-mediated cytotoxicity(ADCC)effect after CRA therapy.Both aspects relieved the immunosuppressive microenvironment after CRA therapy.Notably,the wild-type PD-L1 Avelumab(Bavencio),compared to the mutant PD-L1 atezolizumab(Tecentriq),was better at inducing the ADCC effect to target PD-L1^(high)CD11b^(+)myeloid cells.Collectively,our study uncovered the novel insights that CRA showed superior curative effect than MWA in combining with anti-PD-L1 antibody by strengthening CTL/NK cell immune responses,which provided a strong rationale for combining CRA and PD-L1 blockade in the clinical treatment for HCC. | Jizhou Tan Ting Liu Wenzhe Fan Jialiang Wei Bowen Zhu Yafang Liu Lingwei Liu Xiaokai Zhang Songling Chen Haibiao Lin Yuanqing Zhang Jiaping Li | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 0 |

| 11 | Xiaoyu Jiangzhi capsule protects against heart failure via Ca^(2+)/CaMKII signaling pathways in mice显示文摘Objective: Heart failure(HF), a worldwide health condition, is the result of many cardiovascular diseases.The traditional Chinese medicine(TCM) Xiaoyu Jiangzhi capsule(XYC) has long been in use in China to treat hyperlipidemia and inhibit platelet aggregation. This study explores the effects of XYC on heart failure(HF) and its detailed mechanisms.Methods: Isoproterenol(ISO, 30 mg/kg) was injected intraperitoneally for 7 days to copy a HF model of 10-12 weeks old, 20-30 g male mice. We then compared the CON(control) group, ISO(HF model)group, MET(metoprolol) group, and XYC group. Cardiac systolic function and left wall thickness were evaluated by echocardiograph. Using western blot analysis, we detected the proteins of calmodulin dependent protein kinase Ⅱ(Ca MKII) and sarco/endoplasmic reticulum Ca^(2+)-ATPase(Serca). Furthermore, ts A201 cells were cultured and the human CaV1.2 calcium channel current(hCaV1.2) were detected by patch clamp experiments.Results: XYC reduced HF, inhibiting the protein expression of Ca MKII, but Serca did not change significantly. Moreover, XYC inhibited the peak amplitude of the hCaV1.2 current, depolarizing shifted the activation curve 27.6 mV, and shifted the inactivation curve toward a positive potential 17.6 mV.
