3Potential chronic liver toxicity in rats orally administered an ethanol extract of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE: To investigate the potential chronic liver toxicity of oral administration of ethanol extract of Huangqin(Radix Scutellariae Baicalensis)(SBE) in Wistar rats.METHODS: SBE was administered to rats by gavage for 26 weeks, at doses of 300, 1250, or 2500 mg·kg-1·d-1 respectively. The rats were euthanized at the end of 13 and 26 weeks daily oral dosing and following 4 weeks of recovery time. The changes of hematology, urinary, blood biochemistry and histomorphology were examined at each time point and focus on liver function and histological changes.RESULTS: When SBE at a dose of up to 2500 mgkg-1 d-1 was fed to male and female rats for 2··6 weeks, the liver tissue showed some inflammatory change that predominated by leukocyte infiltrationbut returned to normal after withdrawal. In addition, high-dose SBE treatment of 26 weeks in rats,glucose, electrolyte and lipid levels also have some changes. In addition, there are no other functional or organic lesions related to SBE treatment.CONCLUSIONS: Long-term and high-dose SBE may cause liver damage, however, the structural damage of the liver can be restored after the ethanol extract stopping. 5Alleviative effects of a novel strain Bacillus coagulans XY2 on copper-induced toxicity in zebrafish larvae显示文摘Copper (Cu) is a kind of micronutrient element that is essential for human metabolism.However,it is also considered as an environmental pollutant which is toxic to organisms at a high concentration level.Probiotics,regarded as beneficial microorganisms for promoting human health,have functions of antioxidant capacity,immune-enhancing properties,intestinal barrier protection and regulation.Several studies have reported that probiotics show positive effects on alleviating and intervening heavy metals toxicity.However,evidence for relieving copper-induced toxicity by probiotics is still limited.In this study,we firstly conducted a zebrafish larvae model to screen out microorganisms which are helpful for CuSO_(4)toxicity resistance and one novel strain named as Bacillus coagulans XY2 was discovered with the best protective activity.B.coagulans XY2 significantly reduced the mortality of zebrafish larvae exposed to 10μmol/L CuSO_(4)for 96 hr,as well as alleviated the neutrophils infiltration in the larvae lateral line under a 2 hr exposure.B.coagulans XY2 exhibited a high in vitro antioxidant activity and against CuSO_(4)-induced oxidative stress in zebrafish larvae by up-regulating sod1,gstp1 and cat gene transcriptional levels and relevant enzymatic activities.CuSO_(4)stimulated the inflammation process resulting in obvious increases of gene il-1βand il-10 transcription,which were suppressed by B.coagulans XY2 intervention.Overall,our results underline the bio-function of B.coagulans XY2 on protecting zebrafish larvae from copper toxicity,suggesting the potential application values of probiotics in copper toxicity alleviation on human and the environment.Fang Ai Xuedi Huang Yalan Wu Chen Ji Yufang Gao Ting Yu Fujie Yan 2023Journal of Environmental Sciences2023,,3:1

8RING finger 138 deregulation distorts NF-κB signaling and facilities colitis switch to aggressive malignancy显示文摘Prolonged activation of nuclear factor(NF)-кB signaling significantly contributes to the development of colorectal cancer(CRC).New therapeutic opportunities are emerging from targeting this distorted cell signaling transduction.Here,we discovered the critical role of RING finger 138(RNF138)in CRC tumorigenesis through regulating the NF-кB signaling,which is independent of its Ubiquitin-E3 ligase activity involved in DNA damage response.RNF138^(−/−) mice were hyper-susceptible to the switch from colitis to aggressive malignancy,which coincided with sustained aberrant NF-кB signaling in the colonic cells.Furthermore,RNF138 suppresses the activation of NF-кB signaling pathway through preventing the translocation of NIK and IKK-Beta Binding Protein(NIBP)to the cytoplasm,which requires the ubiquitin interaction motif(UIM)domain.More importantly,we uncovered a significant correlation between poor prognosis and the downregulation of RNF138 associated with reinforced NF-кB signaling in clinical settings,raising the possibility of RNF138 dysregulation as an indicator for the therapeutic intervention targeting NF-кB signaling.Using the xenograft models built upon either RNF138-dificient CRC cells or the cells derived from the RNF138-dysregulated CRC patients,we demonstrated that the inhibition of NF-кB signaling effectively hampered tumor growth.Overall,our work defined the pathogenic role of aberrant NF-кB signaling due to RNF138 downregulation in the cascade events from the colitis switch to colonic neoplastic transformation and progression,and also highlights the possibility of targeting the NF-кB signaling in treating specific subtypes of CRC indicated by RNF138-ablation.Yalan Lu Rong Huang Jianming Ying Xingchen Li Tao Jiao Lei Guo Haitao Zhou Han Wang Amannisa Tuersuntuoheti Jianmei Liu Qichen Chen Yanhong Wang Luying Su Changyuan Guo Fu Xu Ziyi Wang Yan Lu Kai Li Junbo Liang Zhen Huang Xiao Chen Jinjie Yao Hanjie Hu Xiaowen Cheng Yufeng Wan Xinyan Chen Ning Zhang Shiying Miao Jianqiang Cai Linfang Wang Changzheng Liu Wei Song Hong Zhao 2022Signal Transduction and Targeted Therapy2022,7,7:0

10Toxicity study of ethanol extract from Oroxylumindicum(L.) Vent in rats显示文摘OBJECTIVE: To evaluate the safety of Oroxylumindicum(L.) Vent extract administered for 26 weeks in Wistar rats.METHODS: Oroxylumindicum(L.) Vent extract was administrated to male and female rats by gavage once daily at doses of 54, 225, and450 mg·kg-1·d-1.The rats were sacrificed after administration for13 weeks and 26 weeks. Part of the rats in each group were allowed to recover for 4 weeks after 26-week administration. Systematic examinations including haematology, urology, blood biochemistry and histomorphology were performed at the end of13, 26 weeks of administration and 4 weeks of recovery.RESULTS: No treatment related adverse effect shappened on rats' general status, body weight, food consumption, urinary index and histomorphologyexamination. Although during the administration,in some rats of extract's groups, the value of Red blood cell count(RBC), white blood cell counts(WBC), hemoglobin(HGB), haematocrit(HCT) and K iron were decreased, and biochemistry index,such as glucose(GLU), triglyceride(TG), alanine transaminase(ALT) and blood urea nitrogen(BUN)were increased, the above parameters were within the normal ranges and all returned to baseline after the drug stopping for 4 weeks.CONCLUSION: The administration of Oroxylumindicum(L.)
